Effect of multidrug therapy on the levels of antibodies to Mycobacterium leprae glycolipid-1 in the leprosy spectrum.

Sera from one hundred and forty five untreated leprosy patients, ten proven cases of active pulmonary tuberculosis and twenty five healthy volunteers not exposed to M. leprae infection were assayed for PGL-1 antibodies. All available follow up samples after multidrug therapy were also assayed. A decline in the level of PGL-1 antibodies were seen in many of the post-treatment samples, giving an indirect assessment of the bacterial load and the prognosis of the disease.

Rhomboid fossa in the clavicles of North Indians.

The incidence and size of the rhomboid fossa were studied in paired clavicles of North Indians obtained from 20 neonates, 60 children, and 789 adult bodies of known age and sex on which postmortem examinations had been performed by the senior author (except in cases of neonates). The fossa was absent in the newborns but was present bilaterally in 44% of male and 54% of female children below 18 years of age and in 16.5% of male and 20.8% of female children unilaterally. Statistically the sexual differences were not significant (P greater than 0.05). In the adults, the incidence of bilateral fossa was 58.70% in males and 54.14% in females and a unilateral fossa was seen in 13.29% of males and 16.56% of females, but the sexual difference in both cases was insignificant (P greater than 0.05). However, in unilateral cases, the incidence of the fossa was greater on the right than on the left side in both male (P less than 0.001) and female (P less than 0.001) bones. Although racial differences cannot be ruled out, no sexual or side differences in the incidence of the fossa could be found. Factors responsible for creating a deep fossa unilaterally are not understood.

meso-Tetra(hydroxyphenyl)porphyrins, a new class of potent tumour photosensitisers with favourable selectivity.

We compared para-, meta- and ortho-isomers of meso-tetra(hydroxyphenyl)porphyrin (p-, m- and o-THPP) and the potassium salt of the para compound (K-p-THPP) with haematoporphyrin derivative (HpD) and Photofrin II in their ability to sensitise tumours, skin and brain to light. HpD and Photofrin II induced modest tumour photosensitisation at the cost of substantial skin and brain sensitisation. At doses low enough to keep sensitisation of these normal tissues within acceptable limits, tumour sensitisation was sufficient to give necrosis only approximately 2 mm deep after exposure to 10 J cm-2 light. In contrast, doses of p-THPP, K-p-THPP and m-THPP that produced skin and brain sensitivity within acceptable limits sensitised tumours enough to give 4-9 mm necrosis after 10 J cm-2 light. m-THPP was, on a molar basis, about 25-30 times as potent as HpD and Photofrin II in sensitising tumours. o-THPP was also a potent tumour photosensitiser, but induced a prohibitive degree of skin photosensitivity even at low doses. It is unlikely that these differences in relative selectivity are due to differences in such photophysical parameters as optimum activating wavelength (which would affect tissue penetration by light), or light absorption, and physicochemical factors that determine tissue localisation may be involved. The high tumour sensitising potency and favourable tissue selectivity of m-THPP, p-THPP and K-p-THPP make them promising candidates for clinical tumour phototherapy.

Inhibition of fusion of skeletal myoblasts by tunicamycin and its reversal by N-acetylglucosamine.

The differentiation of a permanent line of rat skeletal myoblasts is inhibited by a low concentration of tunicamycin in the growth medium. At a level of 0.9 micrograms/mL, mannose incorporation in trichloroacetic-acid-insoluble material is inhibited to the extent of about 50% by the antibiotic. Blotting of glycoproteins of the cytoplasmic membrane of myoblasts separated by gel electrophoresis by radioiodinated concanavalin A revealed that four major glycoproteins of 230,0000, 145,000, 119,000, and 46,000 daltons were present in lower relative amounts in the plasma membrane following tunicamycin treatment. The 119,000 dalton glycoprotein was a major radioiodinated protein in intact cells and was presumably localized on the periphery of the membrane. The effect of tunicamycin on both fusion and glycosylation of membrane proteins could be reversed by N-acetylglucosamine, but not by the protease inhibitors leupeptin and pepstatin.

Serum protein polymorphisms in breast cancer.

Serum haptoglobin, transferrin and albumin were studied electrophoretically in 50 patients with carcinoma of breast and in 50 normal healthy women. Results on haptoglobin show a preponderance of products of gene Hp1 (0.26) in patients as compared to controls (0.14). No variation was observed at the transferrin locus in either sample. The most notable finding was the detection of four cases of alloalbuminaemia (8%) in breast cancer Patients. The high incidence of alloalbuminaemia prompted us to extend our observations further. For this purpose sera from 41 patients was collected. One more case of alloalbuminaemia of the fast type was detected. However, in contrast to our first series of patients, quite high incidence (12.2%) of transferrin variants was recorded in this sample. Family studies were conducted on all the patients with protein anomalies.

Antigen Dd reactivity in selected diseases.

The distribution of detectable antibodies against antigen Dd has been studied in rheumatoid arthritis, goitre, nephrotic syndrome, cirrhosis, gastrointestinal tract diseases, neurological diseases, liver and gall bladder diseases, breast cancer, respiratory diseases and cardiovascular diseases. Except in rheumatoid arthritis, breast cancer and nephrotic syndrome, where the incidence of antigen Dd-reactivity did not differ much from that in the control group, in all other disease it was significantly lower.
