Chemotaxis of polymorphonuclear leukocytes to varicella-zoster virus antigens.

Chemotaxis of polymorphonuclear leukocytes (PMNs) to various varicella-zoster virus (VZV) antigens was studied using a membrane filter method. Chemotactic activity of PMNs was detected in the presence of sonicated VZV antigen and soluble VZV skin test antigen. This activity was reduced when sonicated VZV antigen was treated with human seropositive serum or murine monoclonal antibodies which reacted with glycoprotein (GP) I or GP II of VZV. However, chemotaxis of PMNs was not reduced when sonicated VZV antigen was treated with human seronegative serum or a murine monoclonal antibody which reacted with GP IV. These results suggest that GP I and GP II act as chemoattractants to PMNs, and this mechanism might contribute to the resolution of the skin lesions of varicella and herpes zoster.

Transforming activity of a synthetic c-Ha-ras gene containing O6-methylguanine in codon 12.

A mutagenic DNA-adduct, O6-methylguanine, was introduced into codon 12 of the synthetic c-Ha-ras gene by cassette mutagenesis. Transfection of this modified ras gene into normal NIH3T3 cells by the calcium phosphate procedure resulted in significant induction of focus formation. The ras gene inserted into the transformed cells was found to have a G to A transition at the position of the modified base. These results indicate that an O6-methylguanine residue in DNA may lead to a mutation and be one cause of activation of the ras gene.

Antibody response determined with antibody-dependent cell-mediated cytotoxicity (ADCC), neutralizing antibody, and varicella skin test in children with natural varicella and after varicella immunization.

Humoral and cellular immune responses were examined in 115 children with varicella and in 21 children who had been immunized with live varicella vaccine. Antibody determined with antibody-dependent cell-mediated cytotoxicity (ADCC) was detected earlier than neutralizing antibody (NT) in both groups. While humoral and cellular immunity were detected simultaneously in the natural varicella group, cellular immunity was detected earlier than humoral immunity in the immunized group. This suggests that ADCC, rather than NT, is involved in the early stages of recovery from varicella-zoster virus (VZV) infection.

Coronary angiography by means of the percutaneous transbrachial approach.

A simplified method of coronary angiography was performed by means of the percutaneous transbrachial approach with a 5-F catheter designed for selective cannulation of the right and left coronary arteries. On 70 serial coronary angiograms, visualization of the coronary arteries was sufficient in all cases with no major complication. The preformed catheter required only simple manipulation. This method was easy, safe, and rapid in the investigation of coronary artery disease.

Relationship between substance P-induced initial transient contractions and inositol phospholipid hydrolysis in guinea pig ileum.

The effects of neomycin on substance P (SP)-induced inositol phospholipid hydrolysis and muscle contraction were studied in longitudinal smooth muscle of guinea pig ileum. Various tachykinin peptides including SP stimulated inositol phospholipid hydrolysis in guinea pig ileum. Neomycin (2.5 mM) reduced both the contractions and the accumulation of [3H]inositol phosphates induced by SP. These results suggest that the initial transient contractions induced by SP are coupled to inositol phospholipid hydrolysis.

8-Hydroxyguanine, a DNA adduct formed by oxygen radicals: its implication on oxygen radical-involved mutagenesis/carcinogenesis.

Oxygen radicals have been suggested to be involved in mutation/carcinogenesis. The C-8 position of guanine residues in DNA is hydroxylated to produce 8-hydroxyguanine (8-OH-Gua) in DNA in vitro by various oxygen radical producing agents. The formation of 8-OH-Gua was also observed in cellular DNA in vivo by radiation or oxygen radical forming carcinogens. The 8-OH-Gua residue in DNA is often misread in the position of 8-OH-Gua residue itself but also at neighboring residues next to 8-OH-Gua. When second guanine in codon 12 was specifically replaced with 8-OH-Gua and transferred to NIH3T3, the recipient cells were transformed to malignant cell type. E. coli was found to contain an endonuclease which specifically recognizes 8-OH-Gua residue and cleave DNA strand before and after the modified base. The data obtained imply that 8-OH-Gua formed in DNA in vivo is recognized as an abnormal modified base which, if not repaired, play a role in the mediation of oxygen radical-involved mutation/carcinogenesis.

Contractile activity of Trimeresurus flavoviridis phospholipase A2 on guinea pig ileum and artery.

Trimeresurus flavoviridis phospholipase A2 (PLA2) induced strong contractions of the smooth muscles of guinea pig ileum and artery in a concentration-dependent manner (10(-10)-10(-6) M). When the same dose of PLA2 was administered in repetition to the ileal preparation, the contraction diminished progressively and was no longer recovered even by consecutive washings. The enzymatically inactive derivative of PLA2, in which His-47 was p-bromophenacylated, was unable to elicit contraction. Also, no activity was observed when the Ca(2+)-free medium was used. The contraction induced by PLA2 was inhibited completely by 1.0 x 10(-6) M indomethacin, but not by nordihydroguaiaretic acid. These results imply that the PLA2-induced contraction is due essentially to the hydrolytic action of the enzyme against phospholipid membranes to liberate arachidonic acid that is then converted to pharmacologically active prostaglandins. In guinea pig artery, PLA2 caused both contraction and relaxation.

Clinical use of Biken acellular DPT in two-month old infants.

We have examined the antibody titer and side effects in two-month-old infants (1.5-2.5) who received vaccinations of acellular DPT Combined Vaccine Adsorbed "Biken" Lots 22 and 23. We observed high-antibody titers far in excess of prevention levels. No particular increase in side effects was observed in the two-month-old infants, except a normal increase in once-only side effects. The results suggest that this vaccine can protect infants under one year of age, who are especially liable to be affected by this illness.

[Pharmacokinetic and clinical evaluations of flomoxef in neonates].

To evaluate pharmacokinetics and clinical efficacy of flomoxef (6315-S, FMOX) in neonates, FMOX was administered to 21 neonates. With 20 mg/kg and 40 mg/kg of intravenous drip-infusion of FMOX 60 minutes, half lives (T 1/2's) was 64.9 minutes and 130.3 minutes, respectively, and when 20 mg/kg of FMOX was infused intravenously to 2 cases, half lives were 70.8 minutes and 110.1 minutes, respectively. When 45-100 mg/kg of FMOX was administered to 17 neonates with infections (pneumonia 8, sepsis 1, sepsis suspected 2, intrauterine infection 2, urinary tract infection 2, omphalitis 2), the efficacy rate was 88.2% (15 of 17). No adverse reactions were observed clinically in the 21 neonates. Transient elevation of eosinophilia was observed in 1 case and transient elevation of S-GOT and S-GPT 1 in another. These results suggest that FMOX is an effective and safe antibiotic to use in neonates.

Application of alcohol- or acetone-fixed Schistosoma eggs as an alternative to lyophilized eggs for the circumoval precipitin test.

Antigenicity of Schistosoma mansoni and S. japonicum eggs preserved in ethanol or acetone were assessed in a circumoval precipitin (COP) assay. The egg antigens were found to retain sufficiently their COP reactivity for the diagnosis of both schistosomiasis mansoni and japonica, although their reactivity became lower than that of lyophilized eggs. These alternative preparations for COP tests have advantages, such as keeping eggs directly in fixatives soon after the egg-purification process. Furthermore, evaporation-process may cause eggshell cleavages which facilitate the reaction. The possible usefulness of those eggs in COP assays in local endemic areas is discussed.

[Mitral regurgitation due to redundant chordae].

This paper reports the etiology and the findings of phonocardiograms and echocardiograms in four cases with mitral regurgitation caused by the redundant chordae. Redundancy of the chordae were determined by the surgeon during operation. The results were as follows: 1. The etiology of redundant chordae was either congenital, rheumatic, or mucoid degeneration. 2. Phonocardiographic findings of this condition are a holosystolic murmur accompanied by a diastolic rumble at the apex. 3. Two-dimensional echocardiogram shows holosystolic bulging of the mitral leaflet to which redundant chordae are attached. 4. The findings of the M-mode echocardiogram are multiple linear echoes and bowing of the mitral valve in systole.

[Pharmacokinetics and clinical evaluation of cefpirome in children].

Pharmacokinetics and clinical effects of cefpirome (CPR, HR 810) in children were studied. When 20 mg/kg and 40 mg/kg doses of CPR were administered to 4 children through 30 minutes' drip infusion, half-lives were 1.23 +/- 0.23 (mean +/- S.D.) hours and 1.37 +/- 0.35 (mean +/- S.D.) hours, respectively for the 2 dose levels, and recovery rates in urine in the first 6 hours after administration were 74.8% and 56.1%, respectively. CPR was administered to 15 cases (3 tonsillitis, 3 bronchitis, 5 bronchopneumonia, 1 acute cystitis, 1 coxoiliatitis, 1 otitis media, 1 otitis externa). The efficacy rate was 86.7%. Seven strains of bacteria were isolated and identified 4 Haemophilus influenzae, 3 Staphylococcus aureus, 1 Pseudomonas sp. from these cases. These bacteria in children were followed after administration of CPR. Six strains were eradicated and one was reduced in number. No adverse effects of CPR were observed except in 2 cases, one of which showed transient eosinophilia and the other showed a transient increase of transaminase. These results suggest that CPR may be an effective and safe drug to use on children clinically.

[Clinical significance of manganese superoxide dismutase in ovarian carcinoma].

A monoclonal antibody to manganese superoxide dismutase (Mn-SOD) was measured in patients with epithelial ovarian carcinomas. An enzyme-linked immunosorbent assay has been developed to detect serum Mn-SOD. With this assay, only 0.9% of the 207 healthy females tested had more than 130 ng per milliliter of serum Mn-SOD. In contrast, 37 of 62 patients (59.7%) with epithelial ovarian carcinomas showed high levels of Mn-SOD. The serum Mn-SOD increased according to the clinical stage and declined to reflect the effects of therapy. Compared with CA-125, Mn-SOD showed a less frequent false positive rate (10%) in benign gynecological diseases. The determination of Mn-SOD levels proved to be a clinically useful marker for monitoring the response to treatment and to early detection of the recurrence of epithelial ovarian carcinomas.
