Persistence of retrieval enhancement by amphetamine following scopolamine-induced amnesia.

Little information is available on the permanence of pharmacologically-induced retrieval enhancement following amnesia. This was studied by comparing the rate of forgetting of a memory reactivated by d-amphetamine after amnesia with spontaneous forgetting of undisturbed fear conditioning. Mice were treated with either saline or scopolamine before conditioning and retention was tested three days later. Scopolamine-treated mice received either saline or amphetamine before testing while the saline controls received a second saline injection. The scopolamine-saline group exhibited robust amnesia, whereas both saline-saline and scopolamine-amphetamine groups showed good retention. To test the persistence of these effects mice in the three groups were subdivided and given a second retention test either 1 day, 1 week or 1 month after the first test. Amphetamine was not administered before the second test. The scopolamine-saline mice continued to exhibit amnesia for up to 1 month while the scopolamine-amphetamine and saline-saline groups continued to show strong memory with only a modest decrement in performance by 1 month after the first test. These results show that amphetamine results in a permanent recovery from scopolamine amnesia.

[Three-dimensional imaging in ultrasonic diagnosis. Initial results].

A new method of three-dimensional (3-D) reconstruction of 2-D ultrasound images of the kidney is described. It is based on a coordinated spatial reconstruction of sequential cross-sectional images. The ultrasound head is moved longitudinally between two rails (parallel sections) and rotated. With a suitable computer program and contouring of each cross-section (so that the organ limits are defined for the computer) these cross-sectional pictures can be reconstructed into 3-D organ images. The kidney can then be presented spatially either as a binary picture or with closed surface. Ultrasound investigators are still unaccustomed to colour reproduction of 3-D reconstructed organs. It remains to be seen whether the method is valuable in routine clinical use.

[Angiography in peripheral arterial occlusive disease. Comparison between large-format angiography, digital subtraction angiography and intermediate-format technic].

With peripheral venous injection of contrast medium, DSA serves to diagnose occlusive peripheral arterial disease in some 90% of cases. Indications for iv-DSA are occlusions and stenoses in the area of the iliac and femoral arteries, as well as in the proximal lower leg. Intraarterial DSA with fine needle is additionally indicated in some 10% of patients for examination of the distal lower leg arteries. In non-diagnostic DSA, conventional angiography using 100-mm technique may be performed with the same angiographic unit.

Correlation between polymerase beta activity and thermal radiosensitization in Chinese hamster ovary cells.

The enhancement of the radiosensitivity of CHO cells was determined for various single and multiple heat treatments followed by X-irradiation at 0 degrees C to prevent repair during exposure. Furthermore, the reduction in the activity of DNA polymerase beta was determined after the same heat treatments. The thermal enhancement ratios for cell killing, calculated on the 10% survival level for the various experimental conditions, showed a clear-cut correlation with the activity of DNA polymerase beta measured by the end of the various heat treatments. These results indicate that the thermal radiosensitization observed in many biological systems might be associated with the transient loss of polymerase beta activity.

Norethisterone-cholesterol eutectic mixture as an oral sustained-release hormonal preparation: bioequivalence study in humans.

A solid dispersion of norethisterone and cholesterol (NET:CHOL; eutectic 1:4 w/w) was prepared by melting and rapid cooling. The fused material was then mixed with lactose as vehicle. Soft gelatin capsules were filled with 55 mg of the final mixture to give 0.35 mg of NET. One control formulation prepared with fused NET and lactose (NET:LAC) was capsuled with the same NET dosage, and one commercial tablet (Dianor, Syntex) with 0.35 mg NET were used as reference formulations. In a cross-over study, five female volunteers received, one month apart, in fasting state, each one of the three formulations. Blood samples were drawn at O,O.5,1,1.5,2,4,8,12,24 and 36 hours after dosing. Immunoreactive plasma NET was measured by RIA to assess pharmacokinetic parameters. The NET:CHOL formulation showed a greater area under the serum concentration-time curve, lower peak concentrations and a smaller release rate constant as compared to the reference preparations. It is concluded that the NET:CHOL eutectic mixture is a modified release dosage form and a sound approach in regulating the drug access rate to the body's central compartment.

Amphetamine enhances retrieval following diverse sources of forgetting.

The generality of amphetamine-induced retrieval enhancement was investigated by determining whether pretest administration could alleviate different types of forgetting. Thirsty mice were punished for licking a water tube following a period of free drinking. Forgetting of the conditioned drink suppression was induced in different groups of animals by; protein synthesis inhibition, cholinergic receptor blockade, inhibition of norepinephrine synthesis, stimulation of serotonin receptors, electroconvulsive shock, a 2.5 month training to test interval and the use of senescent animals with an endogenous memory defect. Thirty min prior to testing mice were injected with either saline or with 2 mg/kg d-amphetamine sulphate. Results showed that amphetamine produced a highly significant improvement in remembering in all of the forgetting treatment groups. It is concluded that amphetamine can alleviate forgetting caused by widely diverse etiologies probably by activating a nonspecific general retrieval system.

[Visceral angiography with intra-arterial DSA and programmed 100-mm technic].

One hundred and seventy specially selected visceral angiograms were carried out on 96 patients using I-A DSA and 100 mm technique. 85.2% of the I-A DSA and 91.7% of the 100 mm images were of good quality. I-A DSA produced comparable or better quality than the 100 mm technique in 66% during the arterial phase, in 79% during the parenchymatous phase and in 70% during the venous phase. The 100 mm technique produced better quality in a third of the cases during the arterial phase and in a quarter of the cases during the parenchymal and venous phases. The indications for the 100 mm technique are failure of I-A DSA or the need for high spatial resolution.

[Rye and wheat in piglet rations supplemented by bisergon].

In a feeding experiment (54 piglets) and a slaughtering test for the registration of tract and digesta properties (15 piglets) the influence of rations with 69% rye (R) or wheat (W) with and without a supplement of 200 mg of the chinoxalin derivate Bisergon/kg DM (rations RB and WB) was studied. In the sequence R, RB, W and WB the feed intake in the test period of 3 weeks after weaning amounted to 581, 651, 583 and 764 g, weight gain to 314, 365, 319 and 424 g per animal and day. In the same sequence the concentration of organic acids in the stomach amounted to 41, 5, 25 and 18 mmol per 100 g dry matter and in the colon to 106, 83, 64 and 62. Bisergon caused a redistribution of the digesta from the stomach to the large intestine and a more rapid emptying of the stomach as well. Rye caused extreme fermentation processes in the front part of the tract and Bisergon clearly has ergotropic qualities for the stabilization of the intestinal flora.

Regulation of L-carnitine metabolism in Escherichia coli.

The metabolization of L-carnitine was studied using whole cells of Escherichia coli 044 K74. It showed features of an epigenetical control. L-carnitine and crotonobetaine were able to induce the carnitine-reducing system. Oxygen and nitrate as electron acceptors, gamma-butyrobetaine as final product of carnitine transformation in E. coli as well as glucose repress the carnitine metabolization. Other betaines and structurally related compounds did not show any effect neither as inductors nor as repressors.

Plasma levels of praziquantel decrease when dexamethasone is given simultaneously.

Treatment with praziquantel for neurocysticercosis frequently induces adverse reactions due to acute destruction of parasites; these reactions are suppressed by dexamethasone therapy. However, there is controversy about the most appropriate regimen with praziquantel and dexamethasone. We studied plasma levels of praziquantel in eight patients given the drug alone or with dexamethasone. Plasma levels of praziquantel were 50% lower in the same patient when dexamethasone was given simultaneously. Dexamethasone should not be added to praziquantel therapy as preventive treatment, but should be reserved for transient therapy of adverse reactions.
