[Colorimetric determination of cholesterol in biological fluids using 2,3-naphthalenedicarboxaldehyde (author's transl)].

The authors describe a new method of cholesterol determination, using the intense blue color given by interaction with 2,3-naphthalenedicarboxaldehyde in a sulphoacetic medium. The degree of precision of the method, as well as its reproducibility are discussed; a good correlation of the new method with two classical techniques, the colorimetric method of Liebermann and the enzymatic method, is observed. The high sensitivity and the specificity of the colour reaction allow the choice of the method for a direct determination of serum cholesterol using a 10 microliter sample.

Liposarcoma of the breast. Case report and review of the literature.

A case of primary liposarcoma of the breast in a 65-years-old woman is described by light- and electron microscopy. The patient had previously had a cystosarcoma phyllodes removed from the same area. A review of the literature showed 34 previously published cases of liposarcoma of the breast, which regarding age distribution, histological type and pattern of metastasis corresponded to liposarcomas of the lower limbs.

Presenile cataract. A follow-up investigation with special reference to occupational aspects.

A retrospective investigation was carried out of 100 patients below the age of 60 years operated for presenile cataract. The material has been divided into three occupational groups, depending on the visual requirements. The complaints prior to and after operation have been related to the patient ability to retain their employment. Of 78 patients who were still employed, only 45 were able to return to their former occupation after the first cataract extraction. The final result was that 62 patients could continue their former work, 12 changed their occupation due to aphakia problems, while four had to retire. The vision in the fellow eye, the type of work and the optical appliances have been found to be of primary importance. It is concluded that operation for unilateral cataract is inadvisable, if the vision of the fellow eye is good and contact lenses cannot be used, that the time between operations for bilateral cataract should be as short as possible, and that the use of contact lenses is essential.

Scintigraphic skeletal changes in non-dialyzed patients with advanced renal failure.

Technetium-99m-polyphosphate (Tc-PP) bone scintigraphy was performed in 51 patients with advanced renal failure in order to evaluate the applicability of this method in detection of metabolic bone changes in these patients. The creatinine clearance varied from 2 to 40 ml/min and none of the patients had previously been on dialysis treatment. The scintigrams were graded according to the focal and the generalized abnormal uptake of the tracer in the skeleton. 34 patients showed generalized scintigraphic changes and among these the changes in 18 patients were classified as severe. An inverse correlation was found between the kidney function and the generalized scintigraphic classification. Focal bone changes were found in 11 patients. In order to evaluate the influence of the lack of kidney function on the scintigraphic results, 3 patients with acute oliguric renal failure were examined. All had normal scintigrams. It is concluded that Tc-PP bone scintigraphy is a sensitive method in revealing renal osteodystrophy in non-dialyzed patients with advanced renal failure in agreement with previous reports on patients on chronic hemodialysis and after kidney transplantation.

Leptomeric fibrils and T-tubule desmosomes in the Z-band region of the mouse heart papillary muscle.

The papillary muscle of the heart of adult white mice is investigated. Intrafibrillary located leptomeric fibrils, frequently encountered in the Z-band region of the myofibrils. The leptomeric fibrils are always running in a transverse direction and often in close proximity to the transverse tubules (which are also located at this level). There seems to be a close connection between the dense striae of the leptofibrils and the Z-bands of ordinary myofibrils. The leptomeric fibrils are spindle-shaped and have a length varying between 0.6 and 1.2 microgrometer. The banding periodicity of the fibrils is approximately 0.16 micrometer. Occasionally desmosomes are observed in the T-tubule system.

Nerve-Purkinje fiber relationship in the moderator band of bovine and caprine heart.

The moderator band in the heart of the ox and goat contains bundles of Purkinje fibers and nerve fibers separated by connective tissue. The axons are mostly unmyelinated and embedded in the cytoplasm of Schwann cells. Small bundles of axons run close to the Purkinje fibers. The axons dilate into varicosities 0.5 to 1.6mu in diameter (mean 0.95mu), containing three types of vesicles: 1)agranular vesicles with a diameter of 400-500 A, 2) large dense-cored vesicles with a diameter of 800-1200 A, 3) small dense-cored vesicles with a diameter of 500 A. Most varicosities contain agranular vesicles together with a few large dense-cored vesicles. The gap between the varicosities and the nearest Purkinje fiber is unusually wide and normally varies between 0.3mu and 0.8mu. No intimate nerve-Purkinje fiber contacts, with a cleft of 200 A, were observed.

The diagnostic value of protein clearances in rejection of human renal allografts.

The relative clearances of transferrin, haptoglobin, IgG, and IgA were used as a diagnostic test for detection of acute rejection episodes after renal transplantation, using an automated immunoprecipitin reaction. Thirty-six out of 40 rejections were predicted by 0--5 days. The sensitivity and specificity were 90% and 98%, respectively. The predictive value of a positive diagnostic test was 86% and of a negative 99%. False positives caused by fever and urinary obstruction could probably be excluded by the finding of an increased urinary excretion of beta2-microglobulin prior to the increased relative clearance of transferrin, haptoglobin, IgG and IgA. This would improve the predictive value of a positive test to 95%. Daily or more frequent protein clearance determinations should be performed in renal transplant patients, since the method used was easy, rapid and suited for sequential analysis, and the procedure was without risk for the patient.

Urinary excretion of basement membrane antigen in normal persons and patients with febrile proteinuria--quantitated by means of rocket immunoelectrophoresis.

The urinary excretion of a glomerular basement membrane-like antigen was quantitated in 19 adult normal subjects and in 15 patients with febrile proteinuria by means of rocket immunoelectrophoresis. In the normal persons the excretion averaged 59 (S.D.8.9) U/24 hours' creatinine clearance. An increased excretion was demonstrated in 80% of the patients. There was a significant relation between the urinary excretions of albumin and protein in the patients with febrile proteinuria. The basement membrane antigen was also demonstrated in normal human serum, liver and placenta. No unusual basement membrane antigen could be demonstrated in the urine from patients with febrile proteinuria.

Structural changes in kidneys of patients with oliguric extracapillary glomerulonephritis during immunosuppressive therapy.

Structural changes in kidney biopsies were investigated from five patients with primary oliguric extracapillary glomerulonephritis in whom the renal function was adequately maintained during extended combined immunosuppressive treatment. The most important structural change was a pronounced decrease in the number of crescents. Reduction in numbers of crescents in the late biopsies was significantly greater than the increase in the number of hyalinized glomeruli. Tubular parenchyma showed only slight diffuse atrophy, and a moderate interstitial fibrosis was always present during the latter stages of treatment. Disappearance of crescents in the glomeruli was not accompanied by disappearance of immunoglobulins. Successfull immunosuppressive treatment of extracapillary glomerulonephritis causes the disappearance of structural characteristics of the kidney that are diagnostic for this disease.

Ultrastructure of the heart muscle cells of the cuttlefish Rossia macrosoma (Delle Chiaje) (Mollusca: Cephalopoda).

The muscle cells of the ventricle, the branchial heart and the branchial heart appendages of Rossia macrosoma (Delle Chiaje) are studied. The ventricle myocardium has three muscle layers, while the other two organs exhibited a loose arrangement of muscle cells. The muscle cells of the ventricle, the branchial heart and the branchial heart appendages are similar in structure. The nuclei are surrounded by myofibrils. In the myofibrils A-, I- and discontinuous Z-bands are seen. The diameters of the thick filaments are 300--400A, their length varies from 1.7 to 3.9 mu. Thin filaments have a diameter of approximately 85A. The ratio between thick and thin filaments is roughly 1 to 11. The SR runs mostly as a longitudinal network within the myofibrils. A few short T-tubules are observed in the Z-regions. Peripheral and internal couplings exist. The latter are few in number. Intercalated discs are small and rarely observed. They have been found in all three organs. A difference in the function of these organs is not reflected in the ultrastructure of the intercalated discs. These discs are often of the interdigitating type with interfibrillar junctions and unspecialized regions. Peripheral couplings are seen at the unspecialized regions. The intercalar surfaces of the muscle cells "shoulder off" into the lateral surface, and the transition between the two surfaces is not a sharp one. Attachment plaques are found scattered over the whole sarcolemma.

[Alpha-hydrazinophloretic acid, competitive inhibitor of fungal tyrosinase].

alpha-Hydrazinophloretic acid, the hydrazino analogue of tyrosine, was shown to behave as a competitive inhibitor of tyrosinase. Some closely related compounds, especially the hydrazino analogue of phenylalanine as well as other hydrazine derivatives, will also inhibit the enzyme, but in these cases, the inhibition observed belonged to the non-competitive type.

Ultrastructure of the myocardial cell and its membrane systems in the adult fly Calliphora erythrocephala (insecta: diptera).

Calliphora erythrocephala has cross-striated cardiac muscle cells with A, I and Z-bands. The diameters of the myosin and actin filaments are 200-250 A and 85 A respectively and the length of the myosin filaments (A-band) is approximately 1.5 micron. Usually 8-10 actin filaments surround each myosin filament. The myocardial cells show a well-developed membrane system and interior couplings. A perforated sheet of SR envelopes the myofibrils at the A-band, dilates into flattened cisternae at both A-I band levels before it merges into a three-dimensional net-work between the actin filaments of the I-bands and between the dense bodies of the discontinuous Z-discs. The T-system consists of broad flattened tubules running between the myofibrils at the A-I band levels forming dyads with the SR-cisternae. Longitudinal connections between the transverse (T-) tubules often occur. It is suggested that this well-developed SR may be an adaptation to facilitate a rapid contraction/relaxation frequency by an effective Ca2+ uptake.
