Promoting effects of bile acid to intestinal tumorigenesis in gnotobiotic ICR mice.

Gnotobiotes were produced by administrating Lactobacillus plantarum IAM 1041 in ICR strain male germfree mice which were fed by ordinary or high fat diet. Both groups were orally administered 0.3 mg/10 g of body weight (B.W.) of methylazoxymethanol (MAM) acetate. The oral administration of 0.3 mg/10 g/B.W. once a week for 11 consecutive weeks caused a total of 68 adenomatous polyps in the large intestine (an average of 11.4/mouse) of gnotobiotic high fat diet mice and a total of 32 adenomatous polyps (an average of 5.3/mouse) of the ordinary diet mice. There were no malignancies in either of the groups. Bile acids in the feces showed higher values in the high fat diet group than in the ordinary group. Bile acids are a factor which promotes the appearance of intestinal tumors. It was also assumed that the L. plantarum promoted the activation of beta-glucuronidase and alcohol dehydrogenase in the liver and intestine.

[A case of male breast cancer with special reference to the hormonal environment during chemoendocrine therapy].

Chemoendocrine therapy was performed on a man with advanced breast cancer, and partial response was observed for 11 months. Estrogen receptor in cancer tissue was detected before and after the therapy. Progesterone receptor, however, was not detected after the treatment, whether it had been there or not prior to it. The serum estrone level was continuously high during the treatment, but the serum testosterone level was obviously decreased after that.

[UFT, UFTM, and UFTM-Dpc treatment of gastric cancer].

It became increasingly evident that UFTM chemotherapy, a combination of UFT and mitomycin C is one of the most recommendable therapy for advanced gastric cancer, especially for the treatment of Borrmann type 4 gastric cancer. Objective responses were achieved in 18 out of 26 (69.2%) patients with Borrmann type 4 gastric cancer. Moreover, UFTM-Dpc treatment, a combination therapy of UFTM plus D-penicillamine (suppressor of collagen synthesis), is proposed for the treatment of Borrmann type 4 gastric cancer from the point of view of collagen synthesis inhibition. It is likely that D-penicillamine might inhibit the peritoneal metastatic involvements, which is lethal to the patients finally.

Preparation of eight ovalbumin subfractions by combined lectin affinity chromatography.

Ovalbumin was fractionated by successive lectin affinity chromatography using concanavalin A/Sepharose and wheat germ agglutinin/Sepharose. Eight glycoprotein fractions, all behaving as ovalbumin on polyacrylamide gel electrophoresis, were obtained. To characterize the carbohydrate chains, the asparaginyl-carbohydrates were prepared from the Pronase digests of the ovalbumin fractions and their dansyl derivatives were analyzed by high-performance liquid chromatography. The elution profile of the dansylated asparaginyl-carbohydrates from each subfraction was compared with that from the unfractionated ovalbumin. The results indicated that the above eight subfractions could be separated from each other according to their carbohydrate chains and that three of the subfractions were homogeneous with respect to their carbohydrate chains.

[The marginal utility of affinity chromatography using immobilized estradiol in the purification of human uterine estrogen receptor].

The purification of human uterine estrogen receptor (ER) was performed by way of the affinity chromatography technique using immobilized 17 beta-estradiol on Sepharose 4B, which was developed by Sica and Bresciani (Biochemistry, 18: 2369-2378, 1979). The anti-human ER antibody was induced and its immunological reactivity was also studied. About 300 micrograms of ER were obtained with the recovery of 18%. The purification magnitude of ER was 4000 times. The purified ER appeared as a complex with 17 beta-estradiol, and formed aggregated macromolecule which was eluate at void volume in Sephadex G-200 and Sephacryl S-300 gel filtration. In 7% approximately 18% linear gradient polyacrylamide gel electrophoresis after SDS treatment, two equally stained bands were observed at 90,000 and 70,000 MW areas. Although anti ER antiserum formed no detectable immunoreactive perception with any kinds of antigens, the gamma-globulin fraction of the antiserum produced a precipitate with human and cow uterine cytosols or their purified ERs. On the other hand, two other precipitin lines were observed in human uterine cytosol. One of the lines was also prevalently found with human muscle and liver cytosols, and human whole serum. The immunoreactive component in the human serum was detected in the globulin fraction, but not in the albumin fraction. These facts imply the immunological heterogeneity of the antibody induced by the purified human uterine ER using the affinity chromatography technique. The cytosol of human uterus obtained from hormonally treated women contains lesser amounts of ER than that of cow uterus in Dextran coated charcoal assay. The ER could not fully occupy the estradiol immobilized on Sepharose 4B in the purification of human uterine ER. Therefore, the non-specific estrogen binding proteins originated both from serum and cytoplasm such as steroid binding globulin, could bind to the remaining free estradiol-ligands of the affinity columns. Thus the marginal utility of the affinity chromatography using immobilized estradiol in the purification of human uterine ER was suggested.

Comparative study of avian ovalbumins by means of glycosidase treatment and following HPLC analysis of their dansyl glycopeptides.

Dansylated glycopeptides (DNS-GP) derived from ovalbumins (chicken, quail, goose and turkey) were treated with commercial endo-beta-N-acetyl-glucosaminidases (Endo-D and Endo-H). Susceptibility of DNS-GP components against those enzymes was examined by analysing the reaction product by HPLC. As reported previously (H. Iwase, Y. Kato and K. Hotta, 1984, Comp. Biochem. Physiol., 77B, 743-747), DNS-GP from chicken ovalbumin was separated into 13 peaks (C1-13) on HPLC. Beta-galactosidase treatment of the DNS-GP resulted in the disappearance of three peaks and the increase of their neighboring peaks. The results indicated that among 13 DNS-GP components from chicken ovalbumin, three (C6,8,11) were bearing galactose residue at the nonreducing terminal and another three (C7,9,12) would be bearing a galacto- sugar chain from each corresponded component.

Purification and characterization of two biosynthetic intermediates of ovalbumin.

Ovalbumin was extracted with a buffered Triton X-100 solution from oviduct slices incubated with [35S]methionine or [2-3H]mannose and was purified by CM-cellulose and Sephacryl S-200 column chromatography. The labeled ovalbumin was fractionated on a concanavalin A (Con A)/Sepharose column at room temperature. Six fractions were separated: Two unadsorbed fractions, OA and OB; and four adsorbed fractions, OC, OD, OE, and OF. Fractions OA, OB, OC, and OD corresponded to the four fractions prepared from unlabeled ovalbumin as previously reported (Iwase, H. et al. (1981) J. Biol. Chem. 256, 5638-5642). Fractions OE and OF were novel constituents of the labeled ovalbumin, although a small amount of OE was also present in unlabeled ovalbumin. Pulse-chase experiments indicated that both OE and OF behaved as biosynthetic intermediates of the other ovalbumin fractions, namely OA, OB, OC, and OD. Partial structural analyses involving endo-beta-N-acetylglucosaminidase treatment and subsequent Bio-Gel P-4 chromatography showed that both OE and OF were composed of components bearing high mannose-type sugar chains which consisted of Man9GlcNAc2, Man8GlcNAc2, or Man7GlcNAc2. We failed to distinguish OF from OE on the basis of their carbohydrate chains. However, on SDS slab gel electrophoresis, the OF band migrated slower than the OE band.

Leiomyosarcoma of pulmonary truncus.

Insidious and progressive right heart failure of obscure origin warrants differential diagnosis of primary sarcoma of the pulmonary artery in patients of both sexes and 20 years of age and older. We report a case of primary leiomyosarcoma of the pulmonary truncus in a 67 years old woman. She had been well in general until when she started having fluctuating and progressive right heart failure three years ago. Clinical data included systolic murmurs in the pulmonary valve area, right ventricular hypertrophy, diminished x-ray density of the pulmonary vasculature, moderate elevation of serum LDH, alkaline phosphatase, and uric acid, and prolonged or unobtainable circulation time from the arm to lungs. The immediate cause of death was attributable to right heart failure due to pulmonary arterial stenosis caused by a locally invasive leiomyosarcoma apparently arising in the pulmonary truncus. Contributing to this right heart failure were 280 ml of serosanguinous pericardial effusion and fibrinous pericarditis.

The use of mannan-Sepharose 4B affinity chromatography for the purification of endo-beta-N-acetylglucosaminidase from Bacillus alvei.

In order to facilitate the isolation of endo-beta-N-acetylglucosaminidase for the structural analysis of glycoconjugates, we have isolated a strain of Bacillus alvei which produces a high level of endo-beta-N-acetylglucosaminidase. We have also devised a simple procedure for the purification of endo-beta-N-acetylglucosaminidase from B. alvei using mannan-Sepharose affinity chromatography. By using this method, endo-beta-N-acetylglucosaminidase was purified 3300-fold with 85% yield from the crude enzyme obtained by ammonium sulfate precipitation of the culture medium. The molecular weight of this enzyme was estimated to be about 66 000 by gel filtration. When using (Man)6(GlcNAc)2-Asn-Dns as substrate, the optimal activity occurs at pH 6.5 with Km of 1.9 mM. The action of endo-beta-N-acetylglucosaminidase toward several glycopeptides was also studied.

Potentiation of large intestinal tumorigenicity of cycasin derivative by high-fat diet and Lactobacillus in germfree mice.

Large intestinal tumorigenesis was investigated in germ free (GF) mice following the administration of methylazoxymethanol (MAM) acetate, lard, and Lactobacillus arabinosus. Twelve weekly doses of 0.3 mg/10 g MAM acetate given to groups of male weanling, germfree ICR mice, fed either low-fat (LF) chow alone, or chow with added lard (HF). Additional groups of mice, on the low-fat diet, were monocontaminated with Lactobacillus arabinosus (GA) and they and their GF controls were given 12 weekly doses of 0.2 mg/10 g of MAM acetate. The HF and LF groups were sacrificed after 123 days, and the GA and GF groups after 214 days. Serial sections of the entire large intestine of the MAM acetate-treated mice revealed many more sessile polyps in the mice maintained on the high-fat diet. In the mice monocontaminated with Lactobacillus, numerous tumors were found, of which 25 percent were invasive. In addition, the polyps found in these animals were generally considerably larger than the polyps found in the GF mice. Atypia occurred in the large intestinal mucosa in all groups. Lard enhanced the large intestinal polypogenesis induced by MAM acetate in germfree mice. Gnotobiosis potentiated the formation of sessile polyps, and carcinogenesis.

Studies on the designing of chemotherapy for gastric cancer in man, based on the tumor tissue concentration of anticancer agents.

5-fluorouracil analogs investigated in this study include a combination of uracil and Ftorafur (UFT), Ethyl (+/-)-t-butoxy-5-fluoro-hexahydro-2, 4-dioxopyrimidine r-5-carboxylate (TAC-278), and 5'-deoxy-5-fluorouridine (5'-DFUR). In a total of 45 patients with gastric cancer, tumor tissue level of 5-fluorouracil (5-FU) was determined at 2, 4, 6, 8, and 12 hours following the oral administration of drug, using a resected stomach specimen as material. As a result, it was demonstrated that oral administration of 200 mg/m2 of UFT maintained above 0.05 microgram/g (minimum inhibitory concentration: MIC) of 5-FU in tumor tissues over 12 hours in 11 of 13 patients. On the contrary, 133 mg/m2 of TAC-278, and 200 mg/m2 or 300 mg/m2 of 5'-DFUR (which is activated by thymidine phosphorylase in man) did not produce an effective 5-FU concentration in tumor tissues. Serum 5-FU level was high in order of TAC-278, UFT, and 5'-DFUR. Clinical response rates obtained with UFT (200 mg/m2 twice a day), or TAC-278 (133-200 mg/m2, 3 times daily) were 27.5% (49 of 178 cases), and 8.3% (3 of 36 cases, by Koyama et al.), respectively. Fisher's direct probability test revealed that there was a significant difference (p less than 0.05) between them in the response rate. It was considered that the measurement of concentration of anticancer drugs in tumor tissues might provide us useful information for the designing of chemotherapy of gastric cancer.

Ovalbumin subfractionation and individual difference in ovalbumin microheterogeneity.

Purified ovalbumin from hen egg white was fractionated by concanavalin A (Con A)/Sepharose chromatography. Four major fractions were separated, one unadsorbed fraction OA, followed by a minor fraction OA', and three adsorbed fractions OB, OC, and OD. The recovery was over 90%. Fractional ratios (OA:OB:OC:OD) were 16:6:37:41. The amino acid composition of the four fractions were quite similar. Galactose was only detected in OA, and the mannose content in OD was high as compared to that of the other fractions. The glycopeptide resulting from pepsin digestion of ovalbumin was also fractionated by Con A/Sepharose. Four fractions were obtained, and they corresponded to fractions OA, OB, OC, and OD. The amino acid compositions of the peptides were identical, indicating that they were derived from the same region in ovalbumin. In order to study individual differences in ovalbumin microheterogeneity, ovalbumin samples prepared from individual eggs were subfractionated on Con A/Sepharose. Four ovalbumin samples prepared from the eggs of a single hen revealed the same elution profile and the same fractional ratios. Fractional ratios of 17 individual preparations varied from 7:2:19:72 to 27:5:40:28 (OA:OB:OC:OD). A constant relation was found among the fractional ratios: the OB, OC and OD contents were dependent on the OA content, and the proportion of OD was in inverse relation to those of OA and OC. The OB content was low and did not change in all ranges of OA content. The present observations indicate that a single hen produces ovalbumin with qualitatively and quantitatively similar carbohydrate chains by unknown mechanisms; moreover, individual differences exist in ovalbumin microheterogeneity which follow constant patterns.
