Detection of UDP-N-acetylgalactosamine-oligosaccharide N-acetylgalactosaminyltransferase activity in rat stomach and human serum by high-performance liquid chromatography.

The enzymatic transfer of the sugar portion from UDP-N-acetylgalactosamine to pyridylamino (PA) lacto-N-fucopentaose I (Fuc alpha 1-2Gal beta 1-3GlcNAc beta 1-4Glc-PA) was detected by high-performance liquid chromatography. Separation of the fluorescent product from the fluorescence-labeled acceptor was achieved within 10 min by reversed-phase high-performance liquid chromatography. Rat stomach enzyme activity was detected in the microsomal fraction from antrum but not corpus. Ohara et al. (1986, Comp. Biochem. Physiol. 83B, 273-275) reported that the N-acetylgalactosamine content in antrum mucin was greater than that in corpus mucin and antrum mucin had strong blood group A activity. The prominent asymmetrical distribution of the enzyme detected here well supports these findings. The elution position of the fluorescent product was the same as that of the product formed by the action of type A human serum toward the acceptor. Its hydrolysis by alpha-N-acetylgalactosaminidase yielded the acceptor. It is thus evident that the detected enzyme is the same as that producing the blood group A structure.

Comparative study of chicken ovalbumin subfractions having different carbohydrate chain from each other by high performance anion exchange chromatography.

1. Ovalbumin was fractionated to six fractions according to their phosphate content by high performance anion exchange chromatography. 2. This method was applied to analyze the phosphate content of ovalbumin subfractions having different carbohydrate chain from each other which were prepared by concanavalin A/Sepharose chromatography from oviduct slices incubated with [2-3H]mannose. 3. Most biosynthetic intermediates bearing a carbohydrate chain of Man8 or 9 GlcNAc2 was not phosphorylated while other fractions bearing differently processed carbohydrate chains such as Man5 or 6 GlcNAc2 or hybrid type carbohydrate chain was phosphorylated at their peptide portion.

[A case of stromal sarcoma of the breast occurring after augmentation mammaplasty].

The first case of stromal sarcoma of breast occurring after augmentation mammaplasty is reported. The patient is 55-year-old Japanese woman who had received injection augmentation mammaplasty 19 years earlier. Both breasts indurated soon after mammaplasty and the patient felt a severe pain in her right breast for about 6 months before seeking treatment. Stromal sarcoma of the breast was proven by a subcutaneous mastectomy, and the determined histological subclassification was a malignant fibrous histiocytoma (MFH). The patient died with tumor recurrence 14 months after her first admission. The tumor cells were found to have invaded the surrounding tissue, such as the nerves, the muscle, the lymphatic vessels, and a severe reactive inflammation, caused by the injected material, was noticed.

[A case of retroperitoneal chylous cyst developed after cholecystectomy and choledochotomy].

A 43-year-old woman complaining of upper abdominal pain was referred to our clinic. Well movable mass was detected in the right hypochondria region by palpation. Three years before, she underwent cholecystectomy and choledochotomy for cholelithiasis. After that she had been aware of painless mass in the right hypochondria region, but because of no symptom she regarded it as operative scar and had no treatment. Abdominal CT, ultrasonography, upper GI series and barium enema revealed a retroperitoneal cyst compressing the 3rd portion of the duodenum upward and the vena cava backward. Laparotomy showed an unilocular chylous cyst, 5 cm in diameter in the retroperitoneal cavity. Histologically, the cyst was cystic lymphangioma. Etiologically, the previous operation of cholecystocholedocholithiasis was supposed to be a trigger of the development of the chylous cyst.

Subfractionation of the dansylated derivatives of glucosyl galactosyl hydroxylysine by liquid chromatography and its application to a specific alpha-1,2-glucosidase assay.

The dansyl derivative of glucosyl galactosyl hydroxylysine (GGH) was separated into two components, as GP-I (monodansyl GGH) and GP-II (didansyl GGH) by paper chromatography. GP-I was further fractionated into four peaks (a, b, c and d) by reversed-phase liquid chromatography. These peaks corresponded to the dansyl derivatives at the alpha-amino (a and b) and epsilon-amino (c and d) groups of their hydroxylysine residues. There is the possibility that the fractions for b and d are diastereoisomers of a and c, respectively, since the monodansyl derivative from human urine consists of a and c. GP-II was fractionated into two peaks, e and f, which may possibly be diastereoisomers of each other. Treatment of the a, b, c and d fractions with crude chicken liver enzyme resulted in the preferential cleavage of a and b and the production of monodansyl galactosyl hydroxylysine. Components c and d were also cleaved slowly, resulting in the production of monodansyl hydroxylysine by the successive action of beta-galactosidase on dansyl galactosyl hydroxylysine. The detected alpha-glucosidase activity was strongly inhibited by free mannosamine. The method developed using the monodansyl GGH fraction a (or b) and high-performance liquid chromatography facilitated the detection of alpha-1,2-glucosidase, which acts specifically toward GGH even in a crude enzyme preparation.

Comparative study of specific alpha-1,2-glucosidase activity toward glucosyl galactosyl hydroxylysine in various animal species.

1. Assay of the activity of alpha-1,2-glucosidase was completed within 10 min using reversed-phase high performance liquid chromatography and purified dansylated glucosyl galactosyl hydroxylysine as the substrate. 2. A comparative study was made on the enzyme activity of liver homogenate from eight animal species, mouse, frog, chicken, rabbit, pig, rat, human, bovine and that of a spinach leaf homogenate. alpha-1,2-glucosidase activity in human and bovine liver was very low, and that of alpha-1,2-glucosidase could not be detected in the spinach homogenate as expected. 3. 1-deoxynojirimycin, a well known potent inhibitor of alpha-1,2-glucosidases which act on the N-glycosidic type carbohydrate chain, also inhibited alpha-1,2-glucosidase acting specificity on glucosyl galactosyl hydroxylysine derived from the collagen molecule.

[Complete response of unresectable gastric cancer (Borrmann's type III) treated with combination chemotherapy of UFT and mitomycin--a case report].

A 69-year-old male visited our hospital in December, 1983, complaining of dysphasia. X-ray and endoscopic examination of the stomach revealed Borrmann's type 3 of the cardia. A biopsy specimen revealed typical features of adenocarcinoma. Because the tumor could not be resected, chemotherapy was started in February, 1984 with a combination of UFT (600 mg/day) and mitomycin (6 mg/week). After five courses of the combination chemotherapy, UFT therapy was continued. Follow-up endoscopy in August, 1985, showed a discolored area of the cardia. A biopsy specimen revealed no evidence of carcinoma. UFT-M therapy is a useful chemotherapy for gastric cancer.

[Clinical studies on cefuzonam in obstetric and gynecological infections].

Seventeen patients with gynecological infections (5 cases each of Bartholin's abscess and adnexitis, 4 cases of endometritis and 1 case each of bartholinitis, pyosalpinx and vulvar abscess) were treated with cefuzonam (CZON, L-105) administered by intravenous drip infusion, 1-2 g twice daily, for 4 to 12 days (6 g to 23 g in total). The clinical effectiveness reached 94.1%, including 1 excellent case, 15 cases good, and a poor case. Bacteria were detected in 9 cases, and with CZON treatment, bacterial eradications were obtained in 6 case, but, in 3 cases, bacterial replacements were observed. No abnormal laboratory findings were noted. From the above results, CZON seemed to be a highly effective and useful agent for gynecological infections.

Characterization of a highly glycosylated biosynthetic intermediate of ovalbumin.

Ovalbumin extracted from oviduct slices incubated with [35S]methionine or [2-3]mannose contained two biosynthetic intermediates, OE and OF (Y. Kato, H. Iwase, and K. Hotta, 1984, J. Biochem. (Tokyo) 95, 455-463). In the present study, these intermediates are further characterized. The 3H dpm/35S dpm ratio of OF labeled with both [35S]methionine and [3H]mannose was twice that or greater than that of OE. The tritium-labeled OF migrated more slowly than OE on sodium dodecyl sulfate-gel electrophoresis and had a molecular weight exceeding that of OE by about 1500. These findings, along with the fact that [3H]OF had sugar chains similar to those of [3H]OE, suggest that OF may possibly have two sugar chains in one molecule. For confirmation of this, the glycosylation sites of OF were examined. Peptic and tryptic glycopeptides were prepared from [2-3H]mannose-labeled OF and the other ovalbumin subfractions--OA, OC, OD, and OE--and then analyzed by high-performance liquid chromatography. The peptic glycopeptides prepared from [3H]OF contained glycopeptides in addition to those derived from the other subfractions although tryptic glycopeptides obtained from [3H]OF were similar to those from the other subfractions. This shows that the above hypothesis is valid.

[Fundamental and clinical studies on imipenem/cilastatin sodium in the field of obstetrics and gynecology].

Fundamental and clinical studies on imipenem/cilastatin sodium (MK-0787/MK-0791) in the field of obstetrics and gynecology were carried out. The concentration of MK-0787 in uterine tissue was 5.9-12.2 micrograms/g at 30 minutes after an administration of 500 mg/500 mg of MK-0787/MK-0791 by a 30-minute intravenous drip infusion and 0.9-1.1 micrograms/g at 185 minutes. The clinical application of the drug to 9 patients with gynecological infections produced "good" results with all the patients clearly showing clinical or bacteriological improvement. Neither side effects nor abnormal laboratory findings were observed. Based on these findings, MK-0787/MK-0791 appeared to be a useful antibiotic for the treatment obstetrical and gynecological infections.

[Histologic analysis of microinvasive carcinoma of the cervix uteri with graphic reconstruction].

To seize the aspect of microinvasive carcinoma in relation to C.I.S., graphic reconstruction of 37 cases, comprising 31 cases of Stage Ia and 6 cases with confluent invasion to a depth of 5mm or less, were represented in detailed schematic drawings. From a new approach to analysis, the following conclusions could be drawn. Only 1/37 cases (2.7%) revealed the transition from C.I.S. to Stage Ia. Configurations of the remaining 36 cases were quite different from those of C.I.S. Their distinctive features were represented in transversal linear spread, parallel to the S.C.J. line. Their extent and multiple foci regulated ramified lesions. According to the degree of ramification, 36 cases were divided into zonal type (33 cases) and multiple type (3). Inceptive foci of invasion from glandular involvement were noticed in 28/33 cases (84.8%). In 17/33 cases (51.5%), lesions were limited to the columnar epithelia region. They provided evidence of the origin of a sub-opening cleft area. The ratio of numbers of early stromal invasion to those of glandular involvement indicated the speed of progression. Early stromal invasion from surface epithelial lesion was discernible in 9/33 cases (27.3%). In the 5 cases without any early stromal invasion from glandular involvement, they might originate from surface epithelia.

[A case of intracranial amelanotic melanoma].

Malignant melanoma is usually black in color because of the existence of melanin pigments in it. Differing from such a usual melanoma, amelanotic melanoma, which has no melanin pigments and is not black-colored, is rarely described in literature. In this report, a case of intracranial amelanotic melanoma of unknown origin is presented and discussed. This represents the seventh case report of an intracranial amelanotic melanoma. On May 28, 1984, a 55-year-old man was admitted to our department because of repeated left hemiconvulsion. Neurological and physical examination disclosed left hemiparesis, poor concentration and right inguinal tumors. CT scan revealed a right posterior frontal mass with ring like enhancement. On June 7, the patient underwent craniotomy. The brain tumor, partly emerged from right frontal lobe, was reddish and easy to bleed. Macroscopically there were no abnormal findings over leptomeniges. The right inguial tumors, that were lymph nodes, were also removed simultaneously. Microscopic interpretation at that time was that of a malignant tumor of ectodermal origin. Although no melanin pigments were demonstrated in any specimens except for a part of lymph nodes, dopa reaction was positive in both specimens from the brain tumor and the inguial lymph nodes, strongly suggesting amelanotic melanoma. Postoperative course was uneventful, but CT scan of Aug. 31 again showed multiple intracerebral enhanced lesion and ventricular hemorrhage. Aphasia, right hemiparesis and consciousness disturbance developed gradually, and he died on Sept. 21, 1984. Autopsy demonstrated multiple intracerebral hematomas, containing much tumor cells in lung, thyroid and subcutaneous tissue. Primary lesion remained unknown in spite of an extensive examination in autopsy.(ABSTRACT TRUNCATED AT 250 WORDS)

[The localization of estrogen receptor (ER) in target tissues as visualized by immunoperoxidase method using anti-ER antibody].

It is very important to detect the intracellular distribution and dynamics of estrogen receptor (ER) in order to make the mechanisms of estrogen action clear. In a recent report, monoclonal antibodies localized ER only in the nucleus of target cells under both the presence and absence of estrogen. We purified cow and human uterine ER and induced specific polyclonal anti-ER antibodies. In this report, the specificity of immunological reactivity of anti-ER antibodies was examined, and the distribution of ER in the target tissues was detected by the indirect immunoperoxidase method. The specific staining was observed in the nucleus and cytoplasm of the endo- and myometrium of cow, rat and human uterus. In the nontarget organs, such as kidney and muscle, the specific staining was not detected. The cytoplasm of MCF-7 cells and immature rat uterine tissues were mainly stained without estrogen; however, the nuclear staining increased obviously after the estrogen priming. The cytoplasmic and/or nuclear staining was positive in 63% of 21 human breast cancer cases. This immunohistochemical assay (IHC) method of ER measurement was compared with the DCC assay method, a biochemical assay based on hormone binding ability. The concomitant positivity or negativity in DCC and IHC were observed as 61.9% (13 cases) or 19.0% (4 cases), respectively. Therefore, these two methods showed the same assay results in 81.0% of human breast cancer tissues.
