Conglutinin microtiter plate ELISA system for detecting circulating immune complexes.

A new technique for a conglutinin ELISA using a microtiter plate has been developed for measuring circulating immune complexes. The sensitivity of our system ranges between 0.2 and 3.2 micrograms equivalents of aggregated human IgG. We have applied this system to 21 normal healthy controls and 80 neurological patients and have found high percentages of positivity in multiple sclerosis and amyotrophic lateral sclerosis.

Long term effects of thorium dioxide (thorotrast) administration on human liver. Ultrastructural localization of thorium dioxide in human liver by analytical electron microscopy.

The ultrastructual localization of thorium dioxide was examined in liver biopsy specimens from two patients injected more than 30 years before with thorotrast. An energy dispersive X-ray microanalysis spectrometer (Kevex 5100) was used to identify thorium in the liver tissue. By electron microscopy, most of the thorium particles were found in the cells of the reticuloendothelial system, such as macrophages of the portal triad and Kupffer cells in the hepatic sinusoid. Thorium dioxide particles were mainly located within the phagosomes, but larger aggregates existed in the cytoplasma with no visible limiting membrane. In addition, the deposition of thorium granules in a few hepatocytes was also confirmed by electron microscopy. This fact indicates that two main pathways of elimination are still functioning in the late period, but the hepatocytic pathway appears to be less effective than the reticuloendothelial system.

Sakyomicins A, B, C and D: new quinone-type antibiotics produced by a strain of Nocardia. Taxonomy, production, isolation and biological properties.

Actinomycete strain M-53, a new soil isolate, was found to produce four quinone-type antibiotics. Antibiotic sakyomicin components, A, B, C and D were isolated from the fermentation broth of strain M-53 by XAD-2 column chromatography, silica gel column chromatography and Sephadex LH-20 column chromatography. The components are active against Gram-positive bacteria. Strain M-53 was identified as a strain of genus Nocardia.

C1q microtiter plate ELISA system for detecting circulating immune complexes and its application of testing the sera in neurological diseases.

We have developed a C1q microtiter plate ELISA system for detecting circulating immune complexes. The sensitivity of our system ranges between 0.5 and 32 micrograms equivalents of aggregated human IgG per ml. We have applied this system to 21 normal healthy controls and 80 neurological patients. Average level of circulating immune complexes in normal healthy controls is 4.67 +/- 2.67 micrograms per ml. We choose the upper limit of normality as 10.01 micrograms per ml (M + 2SD) and define the cases above this limit as positive cases. We have found high percentages of positivity in multiple sclerosis and amyotrophic lateral sclerosis.

Therapeutic activity of pretazettine on Rauscher leukemia: comparison with the related Amaryllidaceae alkaloids.

Comparative studies on the cytotoxicity and antileukemic activity of limited numbers of Amaryllidaceae alkaloids with pretazettine, a narcissus alkaloid, have been performed on the systems of Rauscher virus-carrier cells and the leukemic mice. Only precriwelline, a stereochemical epimer of pretazettine, has been found to be therapeutically active as that of pretazettine. The natural precursors such as haemanthamine, crinamine and 6-hydroxycrinamide were also moderately active, but the artificial final product, tazettine, was confirmed to be inert. The structure-activity relationship of pretazettine or precriwelline has been partially analyzed. Also, the predominancy of antiviral activity relative to cytotoxicity of the alkaloids has been demonstrated when compared with some standard antileukemic drugs.
