Limb bud chondrogenesis in cell culture, with particular reference to serum concentration in the culture medium.

When limb bud mesodermal cells of stages 23-24 chick embryos were plated at low cell density (2 X 10(5) cells/cm2) and cultured in medium containing 10% fetal calf serum (FCS) (serum-rich medium), all cells became fibroblastic and no chondrocyte differentiation occurred in the culture. However, when cells of the same origin were cultured in a medium containing only 0.1% FCS (serum-poor medium), almost all the cells formed aggregates which developed further to form cartilage nodules. The loss of chondrogenic activity in serum-rich medium culture was irreversible: cultivation of the limb bud cells in serum-rich medium for 12 h abolished chondrogenic activity completely and these cells could not resume activity on re-cultivation in serum-poor medium. Calf, horse and chick serum at a concentration of 10% also induced the loss of chondrogenic activity in low cell density culture. Failure of chondrogenesis in serum-rich medium culture seemed to be due to the commitment of bipotential limb bud mesodermal cells to fibroblastic cells rather than to selective detachment of pre-committed chondroblasts.

[Mechanisms of high hemoglobin A1 in alcohol drinkers].

Hemoglobin A1 (HbA1) levels were significantly higher in healthy alcohol drinkers (HbA1 = 7.50%, n = 11) than in normal non-drinkers (HbA1 = 6.62%, n = 13). Ethanol was not able to change HbA1 level when ethanol was added to human whole blood in vitro. Acetaldehyde (AcCHO), although, markedly increased it. Glucose utilization in erythrocytes was stimulated by AcCHO. While it was completely blocked by sodium fluoride in the presence of AcCHO in the incubation medium, but sodium fluoride did not affect the formation of HbA1. AcCHO formed HbA1 with human purified hemoglobin in vitro. The level of HbA1 formed by AcCHO was significantly low when purified human hemoglobin used as a substrate in comparison with the use of whole blood. AcCHO and dihydroxyacetone phosphate reacted in the presence of aldolase. The reacted product, 5-deoxy-D-xylulose-1-phosphate, increased HhA1 level of human purified hemoglobin. It is suggested, the high level of HbA1 in healthy drinkers was caused by AcCHO, the first metabolite of ethanol. AcCHO formed addicts with human hemoglobin directly, and there might be other mechanisms of HbA1 formation due to AcCHO, such as 5-deoxy-D-xylulose-1-phosphate, which is the reacted product of AcCHO.

Proliferation in vitro of melanophores from Xenopus laevis.

Melanophores of wild-type and periodic albino mutants of Xenopus laevis were successfully cultured in vitro. They proliferated in the presence of alpha-melanocyte-stimulating hormone (alpha-MSH or cyclic adenosine monophosphate (cAMP) at a doubling time of 8-10 days. These proliferating melanophores retained their phenotypes, ability to synthesize melanin, and melanin-dispersing response to MSH stimulation. Neither depigmentation nor selective cell death of periodic albino melanophores was observed for at least 4 months during the cultivation.

Long-term cultivation of amphibian melanophores. In vitro ageing and spontaneous transformation to a continuous cell line.

Pure melanophore populations isolated from the tail skin of the tadpole, Rana catesbeiana, were mass cultured for a period of 2-3 years. All cell lines of amphibian melanophores studied exhibited growth crisis (in vitro ageing) followed by spontaneous transformation to a continuous cell line, as shown by changes in growth characteristics in mass culture and in clone culture, by the appearance of the cells, and by measurements of cell volumes. Even after becoming a continuous cell line, amphibian melanophores continued to have a diploid chromosome number (2n = 26) in three of four cell lines examined. The chromosome mode in one cell line, however, changed to thirty. Measurement of melanin dispersion after the addition of alpha-melanocyte-stimulating hormone suggested that the mechanism for melanin dispersion in melanophores changed during in vitro ageing.

Radiation-induced reduction of nitroimidazole derivatives in aqueous solution.

The radiation-induced reduction of N1-alkyl substituted 2- and 5-nitroimidazoles in aqueous solution containing sodium formate or 2-propanol was studied at pH 7.0 +/- 0.1 under deaerated conditions. Irrespective of 2- or 5-nitroimidazole, N1-unsubstituted nitroimidazoles (2-nitro-(1a), 4(5)-nitro-(2a), 2-methyl-4(5)-nitro (3a), and 4(5)-methyl-5(4)-nitro- (4a) imidazoles) were reduced stepwise to consume 6 electrons per molecule, whereas N1-alkyl substituted nitroimidazoles (1-methyl-2-nitro (5a) and 1-methyl-5-nitro (6a) imidazoles, misonidazole (7a), and metronidazole (8a] reacted with consumption of 4 electrons. In accord with the stoichiometry for the reduction of N1-unsubstituted nitroimidazoles, the formation of the amino derivatives of (1a)-(4a) was shown by HPLC or colour identification tests. 4-Electron-reduction products of N1-alkyl substituted 2-nitroimidazoles (5a) and (7a) were characterized by 13C and 1H n.m.r. and FDMS measurements, indicating that the hydroxyamino derivative of (5a) as a 4-electron-reduction product isomerizes to an oxime form. The formation of an analogous oxime-type product was also suggested for (7a) together with a product bearing the partially cleaved imidizole ring. The HPLC analysis showed that 4-electron-reduction products of N1-alkyl substituted 5-nitroimidazoles (6a) and (8a) are unstable relative to those of the corresponding 2-nitroimidazoles (5a) and (7a).

Radiation-induced hydroxylation of thymine promoted by electron-affinic compounds.

The effect of 20 electron-affinic compounds including nitroimidazoles, nitrofurans, nitrobenzenes, and quinones on the radiation-induced reaction of thymine in aqueous solution was studied under deaerated and N2O-saturated conditions. The radiolysis of thymine in aerated aqueous solution was also performed for comparison. Thymine decomposition was depressed to some extent by the addition of electron-affinic compounds in both deaerated and N2O-saturated solutions, while promoted in aerated solution. The radiolyses with varying concentration of misonidazole indicated that the depression of thymine decomposition can be attributed to a competition between thymine and electron-affinic compounds for the reactions with .OH. Among the radiolysis products, the formation of thymine glycol was remarkably promoted by the addition of electron-affinic compounds. Irrespective of structures of the electron-affinic compounds, the G-value of thymine glycol increased in sigmoidal form with increasing one-electron reduction potential of the electron-affinic compounds and attained the ultimate values of ca. 1.1 and 1.8 in deaerated and N2O-saturated solutions, respectively. The results are in accord with one-electron oxidation of the hydroxythymyl radical, produced by the reaction of thymine with .OH, to the corresponding cation by electron-affinic compounds. The so-formed hydroxythymine cation undergoes solvolytic substitution to give thymine glycol. Based on the ultimate G-values of thymine glycol, the difference in reactivity between hydroxythymine-5-yl and 6-yl radicals toward electron-affinic compounds is discussed.

Plasma cyclic AMP responses to adrenaline administration in patients with spinocerebellar degeneration, bronchial asthma, and diabetes mellitus--a preliminary report of a clinical test for detecting beta-adrenergic dysfunctions.

Plasma cyclic AMP responses to adrenaline administration in normal volunteers, patients with spinocerebellar degeneration, bronchial asthma, pulmonary emphysema, and diabetes mellitus were studied. Intramuscular administration of low doses (0.1--0.4 mg/person) of adrenaline caused a dose-dependent increase in plasma cyclic AMP. The increase in cyclic AMP was completely prevented by propranolol, while it was not affected by phentolamine or atropine. In patients with spinocerebellar degeneration, the concentrations of plasma cyclic AMP both before and after adrenaline administration were lower than in normal subjects. In asthmatic patients, the plasma cyclic AMP increase after adrenaline administration was smaller than that of the healthy controls. The plasma concentration of cyclic AMP in patients with insulin-dependent diabetes reached the peak level more slowly than in diabetic patients with dietary control alone. Examining changes in the plasma cyclic AMP level after adrenaline administration appears to be a useful means for assessing the degree of beta-adrenergic dysfunction.

Melanophore differentiation in Xenopus laevis, with special reference to dorsoventral pigment pattern formation.

During the development of Xenopus laevis, after stage 26, a large number of dopa-positive cells were observed in the ventral region. Electron microscopic observations revealed that these cells became localized in the epidermal layer and contained premelanosomes. In cultured ventral skin, fully matured melanophores appeared. These results strongly suggest that a large number of melanoblasts are present in the ventral epidermis and remain there without final differentiation into melanized melanophores. Thus the positional difference of melanoblasts differentiation mainly contributes to dorsoventral pigment pattern formation of Xenopus laevis.
