Comparative effects of dehydroepiandrosterone and related steroids on peroxisome proliferation in rat liver.

Dehydroepiandrosterone (DHEA) is known to induce peroxisome proliferation and peroxisomal enoyl-CoA hydratase/3-hydroxyacyl-CoA dehydrogenase (PBE) mRNA in the rat liver. We have compared the effects of 6 intermediate metabolites of DHEA on the induction of peroxisome proliferation and PBE mRNA. Administration of epiandrosterone, etiocholanolone, androstenedione, estrone or estradiol for 2 weeks in the diet at 0.45% concentration to adult male F-344 rats failed to induce significant increases in peroxisome proliferation and PBE mRNA when compared to the parent compound DHEA. Dietary administration of 5-androstene-3 beta,17 beta-diol (ADIOL) for 2 weeks at 0.45% concentration caused an increase in PBE mRNA and peroxisome proliferation but to a lesser extent than DHEA. Following a single intragastric dose of DHEA an increase in PBE mRNA level was observed in the liver at 1 hr and continued to 16 hrs., but not with its metabolites. These results strongly suggest that DHEA or possibly another yet to be identified metabolite might be responsible for peroxisome proliferation.

Correlation between somatosensory-evoked potentials and magnetic resonance imaging of focal cerebral ischemia in cats.

Serial measurements of somatosensory-evoked potentials (SEPs) and magnetic resonance imaging (MRI) were performed in 17 cats for 24 h after unilateral middle cerebral artery occlusion. Intravoxel incoherent motion (IVIM) imaging demonstrated ischemic cerebral injury 2 h post-occlusion in all cats, while T2-weighted imaging failed to show clear evidence of injury until 2-6 h. In the severely affected group of eight cats as determined by SEP criteria, ischemic injury evaluated by MR imaging was detected extensively in not only the basal ganglia but also the frontoparietal cortex including part of the somatosensory center. In contrast, in the mildly affected group of nine other cats, ischemic injury was limited to the basal ganglia with or without some involvement of the lower temporal cortex. The quantitative signal intensity ratios in the frontoparietal cortex were significantly greater in the severely affected group than in the mildly affected group, but showed no difference between them in the basal ganglia.

Thymine ring saturation and fragmentation products: lesion bypass, misinsertion and implications for mutagenesis.

We have used thymine glycol and dihydrothymine as representative ring saturation products resulting from free-radical interaction with DNA pyrimidines, and urea glycosides and beta-ureidoisobutyric acid (UBA) as models for pyrimidine-ring fragmentation products. We have shown that thymine glycol and the ring-fragmentation products urea and beta-ureidoisobutyric acid, as well as abasic sites, are strong blocks to DNA polymerases in vitro. In contrast, dihydrothymine is not a block to any of the polymerases tested. For thymine glycol, termination sites were observed opposite the putative lesions, whereas for the ring-fragmentation products, the termination sites were primarily one base prior to the lesion. These and other data have suggested that thymine glycol codes for an A, and that a base is stably inserted opposite the damage, whereas when a base is inserted opposite the non-coding lesions, it is removed by the 3-->5 exonuclease activity of DNA polymerase I. Despite their efficiency as blocking lesions, thymine glycol, urea and UBA can be bypassed at low frequency in certain specific sequence contexts. When the model lesions were introduced individually into single-stranded biologically active DNA, we found that thymine glycol, urea, beta-ureidoisobutyric acid, and abasic sites were all lethal lesions having an activation efficiency of 1, whereas dihydrothymine was not. Thus the in vitro studies predicted the in vivo results. When the survival of biologically active single-stranded DNA was examined in UV-induced Escherichia coli cells where the block to replication was released, no increase in survival was observed for DNA containing urea or abasic sites, suggesting inefficient bypass of these lesions. In contrast, beta-ureidoisobutyric acid survival was slightly enhanced, and transfecting DNA containing thymine glycols was significantly reactivated. When mutation induction by unique lesions was measured using f1-K12 hybrid DNA containing an E. coli target gene, thymine glycols and dihydrothymine were found to be inefficient as premutagenic lesions, suggesting that in vivo, as in vitro, they primarily code for A. In contrast, urea and beta-ureidoisobutyric acid were efficient premutagenic lesions, with beta-ureidoisobutyric acid being about 4-5-fold more effective than urea glycosides, which have approximately the same rate of mutation induction as abasic sites from purines. Sequence analysis of the mutations resulting from these ring-fragmentation products shows that the mutations produced are both lesion and sequence context dependent. The possible roles that bypass efficiency and lesion-directed misinsertion might play in mutagenesis are discussed.

Acute stroke in cats: comparison of dynamic susceptibility-contrast MR imaging with T2- and diffusion-weighted MR imaging.

PURPOSE: To evaluate the hemodynamics of acute stroke in cats by means of dynamic susceptibility-contrast magnetic resonance (MR) imaging compared with standard T2- and diffusion-weighted MR imaging. MATERIALS AND METHODS: Unilateral permanent occlusion of the middle cerebral artery was performed in 18 cats. MR imaging was performed with each method 2 hours (n = 2), 6 hours (n = 10), and 24 hours (n = 11) after occlusion. RESULTS: At every time point, dynamic MR images clearly showed ischemic regions of steady-state signal hyperintensity and areas of less dynamic decrease in signal intensity that corresponded closely to those seen on diffusion-weighted MR images. T2-weighted MR images did not reveal regions of high signal intensity 2 hours after occlusion. The measurements of relative blood volume obtained with the dynamic method were significantly lower in steady-state hyperintense ischemic areas of the affected side than those in the contralateral normal areas at 6 and 24 hours (P < .001). CONCLUSION: This dynamic method can advance the time of detection of early ischemia in comparison with T2-weighted MR imaging and provides functional information about hemodynamics not provided by other methods of MR imaging.

An occurrence of equine transport pneumonia caused by mixed infection with Pasteurella caballi, Streptococcus suis and Streptococcus zooepidemicus.

An acute death occurred in a racehorse with pneumonia after long-distance transportation in December, 1990. Pasteurella caballi, Streptococcus suis and Streptococcus zooepidemicus were isolated from the lung at high rate. Specific antigens of these bacteria were also demonstrated immunohistologically in the pneumonic lesion. These findings indicated that the disease is equine transport pneumonia caused by a mixed infection of the three bacterial species. This is the first report on the isolation of P. caballi and S. suis from a racehorse in Japan.

Detection of specific base sequences in solution using DNA probes labeled with d- and/or 15N-substituted spin-labels.

In order to enhance the sensitivity and the accuracy of the detection by the DNA probe method in which spin-labeled oligonucleotides are used as probes, 4-amino-2,2,6,6-tetramethylpiperidine-15N-oxyl-d16 (4-amino-TEMPO-15N-d16) and 4-amino-2,2,6,6-tetramethylpiperidine-14N-oxyl-d16 (4-amino-TEMPO-14N-d16) were incorporated into the phosphate linkage of oligonucleotides by the hydrogen phosphonate method and these DNA probes were used for the detection of target DNAs in solution. The intensities of the electron paramagnetic resonance (EPR) signals of the oligonucleotides labeled with 4-amino-TEMPO-14N-d16 and 4-amino-TEMPO-15N-d16 were 3-fold and 4-fold larger than that of the oligonucleotide labeled with 4-amino-TEMPO-14N, respectively. Also, the EPR lines of these labeled oligonucleotides do not overlap each other, allowing the detection of two different regions in the same target DNA by the single EPR measurement.

Oligonucleotides site-specifically spin-labeled at 5'-terminal or internucleotide linkage and their use in gene analyses.

Spin-labeled oligonucleotides (S-probes) were synthesized and examined as DNA probes to monitor hybrid formation. TEMPO was introduced either at the internucleotide linkage of 5'-terminus (Type 1) or at the 5'-terminal hydroxyl group (Type 2) and both types of S-probes were used in this study. The presence of target DNA was detected in solution by EPR spectroscopy for both types of S-probes. Hybridization of the S-probes resulted in notable broadening of EPR line width, accompanied by a decrease in the EPR signal height ratio for I(-1)/I(0).I(-1)/I(0) of S-probes having no spacer between oligonucleotide and TEMPO decreased more markedly than that of S-probes with a spacer, indicating that TEMPO should be introduced to an oligonucleotide directly to monitor hybrid formation. When M13mp8 single-stranded DNA with or without an EcoRI recognition site was selected as a target DNA, hybrid formation was detected only for DNA containing EcoRI site in solution using spin-labeled oligonucleotides.

Histological studies of intracranial vessels in primates following transluminal angioplasty for vasospasm.

Percutaneous transluminal angioplasty for treatment of cerebral vasospasm was performed in primates. Chronic cerebral vasospasm was induced by placement of an autologous blood clot over the right internal carotid artery (ICA), middle cerebral artery (MCA), and anterior cerebral artery (ACA). Cerebral angiography on Day 7 showed that the diameters of the ICA, MCA, and ACA were reduced to 55.7% +/- 1.3%, 55.3% +/- 2.6%, and 59.6% +/- 1.3%, respectively, of baseline. The angioplasty was carried out with a silicone microballoon attached to a microcatheter under somatosensory evoked potential (SEP) monitoring on Day 7. The angioplasty for ICA was performed satisfactorily; however, the balloon could not be advanced to the spastic M1 or A1 portions of the cerebral artery. Following angioplasty, the diameters of the ICA, the M1 segment, and the A1 segment were 79.6% +/- 2.9% (p < 0.001), 67.6% +/- 4.3% (p < 0.05), and 61.7% +/- 2.2% (not significant), respectively, of baseline. Histological studies demonstrated that the vessels were well dilated and patent without endothelial cell damage.

Prevalence and clinical significance of anomalous muscular band in the left atrium.

The significance of left ventricular false tendon and right atrial anomalous muscular band has been previously reported. As well as in the right atrium, anomalous muscular band is sometimes observed in the left atrium. To evaluate the prevalence and clinical significance of the left atrial anomalous band, a clinico-pathologic study was performed among 1,100 autopsies over 60 years. The following results were obtained: (1) Pathologically, the incidence of the left atrial anomalous band was 2% (22 cases among 1,100 autopsies). The anomalous band connected the left atrial side of fossa ovalis with the other areas of left atrial endocardium in most of the cases (19 cases). The cases with the bands had Chiari's network and patent foramen ovale at the same time more frequently than those without the bands (Chiari's network: 27% vs 8%, p < 0.01, patent foramen ovale: 23% vs 8%, p < 0.05). Microscopically, the anomalous band was composed of fibrous and muscular tissues. (2) Clinically, premature atrial complex was more frequently observed among the cases with left atrial anomalous bands than those without the bands (41% vs 11%, p < 0.01). In conclusion, left atrial anomalous band seemed to be one of congenital abnormalities which was associated with Chiari's network and patent foramen ovale in origin, and might be one of the causes of supraventricular arrhythmias.

Cooperative activation of Chox-4 homeobox genes by factors from the polarizing region and the apical ridge in chick limb morphogenesis.

When a mouse zone of polarizing activity (ZPA) at the posterior margin of the limb bud was grafted at the anterior margin of the chick limb bud, we found that expression of the chick homeobox genes, Chox-4.7 and -4.8, was induced prior to the formation of chick extra digits. The induction was observed in the restricted domain close to both grafted mouse ZPA and the chick apical ectodermal ridge (AER). When the posterior half of the AER was removed, the normal expression was diminished in the posterodistal region. Thus, it is likely that at least two distinct factors, one from the ZPA and the other from the AER, provide positional information to induce cooperatively the sequential expression of the Chox-4 genes.

Synthesis and characterization of oligonucleotides containing the alpha-anomer of deoxyadenosine to study its influence on DNA replication.

The alpha-anomer of 2'-deoxyadenosine 1 is a major adenine lesion produced by hydroxyl radicals in DNA. To assess its biochemical effects on DNA replication, 1 was site-specifically incorporated into oligonucleotide templates using phosphoramidite chemistry. The templates were purified and extensively characterized to ensure their homogeneity and nucleotide sequences.

[Post-sternotomy mediastinitis treated by omental transposition].

Mediastinitis after cardiac surgery is a serious complication with high morbidity and mortality rates. Between December 1989 and April 1992, of 264 patients who underwent cardiac or aortic surgery with median sternotomy, seven (2.6%) developed deep sternal infections with mediastinitis. Six of these patients were treated with debridement, irrigation and delayed wound closure with mediastinal transposition of the greater omentum. Most of the infections were associated with several predisposing factors: cardiac cachexia in the preoperative state, prolonged perfusion time, and respiratory insufficiency in the postoperative period. One of the six patients died of adult respiratory distress syndrome in the postoperative course, but the remaining five patients were discharged with their wounds well healed. Acute and deep mediastinal infection occurred in one patient on the fifth day after cardiac surgery. The infection was successfully treated with radical debridement, two days of intermittent mediastinal irrigation and staged wound closure with omental transposition into the mediastinum. The omental transfer was achieved as follows: after the greater omentum was divided into two pedicles one pedicle was used to fill in the pericardial space and the other was placed between the sternum and the undermined skin. We conclude that early debridement and omental flap transposition are effective in the management of mediastinitis after cardiac surgery.

[Preoperative evaluation of a distal aortic arch aneurysm by spiral CT with three-dimensional reconstruction].

Sixty-seven-year-old male with a distal aortic arch aneurysm was evaluated by spiral CT with three-dimensional reconstruction. Spiral CT allows rapid data acquisition through an entire anatomic region during a single breath hold. The helically acquired data undergo interpolation. Transverse images are reconstructed with slice intervals 1-4 mm apart. Three-dimensional images were generated by these data with the transverse images. The entire anatomical correlation with the aneurysm and the aortic arch branches was clearly recognized by these three-dimensional images. Preoperative evaluation of the distal aortic arch aneurysm by spiral CT with three-dimensional reconstruction was useful for the surgical treatment.

Abnormal activation of lipoprotein lipase by non-equilibrating apoC-II: further evidence for the presence of non-equilibrating pools of apolipoproteins C-II and C-III in plasma lipoproteins.

Using artificial triglyceride emulsions, we have demonstrated the presence of non-equilibrating pools of apolipoproteins C-II and C-III in human plasma lipoproteins. As the concentrations of acceptor triglycerides were increased, a greater fraction of both apoC-II and apoC-III shifted away from the native plasma lipoproteins to the artificial lipid emulsions. All of the apoC-II and apoC-III in very low density and high density lipoproteins (VLDL and HDL), however, could not be removed from native plasma lipoproteins. The percent of total plasma apoC-II and apoC-III that could be recovered in the VLDL and HDL density fractions varied when plasma from different individuals was used. When plasma samples from normotriglyceridemic subjects were used, HDL was the primary donor of apoCs. The percent of total plasma apoCs associated with HDL decreased from 60% to 25% for apoC-II and from 65% to 15% for apoC-III. When plasma samples from hypertriglyceridemic subjects were incubated with artificial lipid emulsions, VLDL was the primary donor of apoCs. HDL from hypertriglyceridemic subjects only accounted for 5-10% of total fasting plasma apoCs and did not contribute significantly to the final apoC contents of the artificial triglyceride emulsions. To evaluate the significance of the depletion of exchangeable apoCs from plasma HDL, we also examined the ability of control and apoC-depleted HDL to serve as activator for bovine milk lipoprotein lipase (LPL) in vitro. When HDL depleted of exchangeable apoCs were used as the source of plasma apolipoproteins for the activation of LPL in vitro, only 5-10% of the maximal activity obtained with native HDL was demonstrated. In fact, in the presence of comparable concentrations of HDL apoC-II, activation of LPL was the least with HDL which lacked exchangeable apoCs. Our data thus indicated that the presence of exchangeable apoC-II on HDL is necessary for the activation of LPL in vitro. This finding is consistent with our data that suggest that HDL from hypertriglyceridemic subjects do not stimulate LPL as well as HDL from normolipidemic subjects.

Misincorporation of ribonucleotides by DNA polymerase during in vitro DNA replication.

The fidelity of DNA replication with respect to misincorporation of ribonucleotides into DNA has been studied. Primed M13 DNA was replicated by E. coli DNA polymerase I in the presence of four dNTPs, a single rNTP and Mg2+ as a cofactor. All four rNTPs were incorporated into DNA in place of the corresponding dNTPs when rNTP/dNTP ratio was 1000:1. The frequency of rNTP misincorporation was C > G > A > U.
