Sound scattering by bubble clouds near the sea surface

The classical exact formulation required to evaluate the form function (or the scattering cross-section, SCS) of a single, ideal, air bubble in a boundless liquid is briefly recalled. It is then immediately generalized to the case of a round cloud of many possibly interacting such bubbles of known volume concentration, contained within the same boundless medium. This is further generalized to the case when the bubble cloud is near a free surface. The presence of the nearby pressure release surface, assumed flat, substantially alters the cloud's scattering cross-section relative to its value in the absence of boundaries. We then use an earlier technique of ours [i.e., see I.E.E.E. J. Ocean. Eng. 20, 285-293 (1995)] based on the method of images that uses the addition theorem for the spherical wave functions, to relate all the scattered sound fields to a common origin and thus obtain the (modified) SCS of the cloud now near the boundary. This formulation accounts for all orders of multiple scattering and yields an infinite set of coupled algebraic equations for the coupling coefficients. This set is then solved for the coupling coefficients in terms of infinite sums of products of pairs of Wigner 3-j symbols, which are then used to construct and evaluate the form function. We display numerical results in four cases that correspond to geographical sites in which the bubble concentrations within the cloud have been measured along a couple of oblique upward directions, or have been assumed to have increasing (and in a few instances, purposely unrealistically high) values. In all cases considered here the bubble clouds are only a few meters beneath the sea surface and consist of ideal bubbles. The results are also compared to those found in the absence of a boundary in all the cases considered.

Identification of a boundary domain adjacent to the potent human cytomegalovirus enhancer that represses transcription of the divergent UL127 promoter.

Transcriptional repression within a complex modular promoter may play a key role in determining the action of enhancer elements. In human cytomegalovirus, the major immediate-early promoter (MIEP) locus contains a highly potent and complex modular enhancer. Evidence is presented suggesting that sequences of the MIEP between nucleotide positions -556 and -673 function to prevent transcription activation by enhancer elements from the UL127 open reading frame divergent promoter. Transient transfection assays of reporter plasmids revealed repressor sequences located between nucleotides -556 and -638. The ability of these sequences to confer repression in the context of an infection was shown using recombinant viruses generated from a bacterial artificial chromosome containing an infectious human cytomegalovirus genome. In addition to repressor sequences between -556 and -638, infection experiments using recombinant virus mutants indicated that sequences between -638 and -673 also contribute to repression of the UL127 promoter. On the basis of in vitro transcription and transient transfection assays, we further show that interposed viral repressor sequences completely inhibit enhancer-mediated activation of not only the homologous but also heterologous promoters. These and other experiments suggest that repression involves an interaction of host-encoded regulatory factors with defined promoter sequences that have the property of proximally interfering with upstream enhancer elements in a chromatin-independent manner. Altogether, our findings establish the presence of a boundary domain that efficiently blocks enhancer-promoter interactions, thus explaining how the enhancer can work to selectively activate the MIEP.

A protein kinase activity associated with Epstein-Barr virus BGLF4 phosphorylates the viral early antigen EA-D in vitro.

The Epstein-Barr virus (EBV) open reading frame BGLF4 was identified as a potential Ser/Thr protein kinase gene through the recognition of amino acid sequence motifs characteristic of conserved regions within the catalytic domains of protein kinases. In order to investigate this potential kinase activity, BGLF4 was expressed in Escherichia coli and the purified protein was used to generate a specific antiserum. Recombinant vaccinia virus vTF7-3, which expresses the T7 RNA polymerase, was used to infect 293 and 293T cells after transient transfection with a plasmid containing BGLF4 under the control of the T7 promoter. Autophosphorylation of the BGLF4 protein was demonstrated using the specific antiserum in an immune complex kinase assay. In addition, EBNA-1-tagged BGLF4 and EBNA-1 monoclonal antibody 5C11 were used to demonstrate the specificity of the kinase activity and to locate BGLF4 in the cytoplasm of transfected cells. Manganese ions were found to be essential for autophosphorylation of BGLF4, and magnesium can stimulate the activity. BGLF4 can utilize GTP, in addition to ATP, as a phosphate donor in this assay. BGLF4 can phosphorylate histone and casein in vitro. Among the potential viral protein substrates we examined, the EBV early antigen (EA-D, BMRF1), a DNA polymerase accessory factor and an important transactivator during lytic infection, was found to be phosphorylated by BGLF4 in vitro. Amino acids 1 to 26 of BGLF4, but not the predicted conserved catalytic domain, were found to be essential for autophosphorylation of BGLF4.

Partitioning of pyruvate between oxidation and anaplerosis in swine hearts.

The goal of this study was to measure flux through pyruvate carboxylation and decarboxylation in the heart in vivo. These rates were measured in the anterior wall of normal anesthetized swine hearts by infusing [U-(13)C(3)]lactate and/or [U-(13)C(3)] pyruvate into the left anterior descending (LAD) coronary artery. After 1 h, the tissue was freeze-clamped and analyzed by gas chromatography-mass spectrometry for the mass isotopomer distribution of citrate and its oxaloacetate moiety. LAD blood pyruvate and lactate enrichments and concentrations were constant after 15 min of infusion. Under near-normal physiological concentrations of lactate and pyruvate, pyruvate carboxylation and decarboxylation accounted for 4.7 +/- 0.3 and 41.5 +/- 2.0% of citrate formation, respectively. Similar relative fluxes were found when arterial pyruvate was raised from 0.2 to 1.1 mM. Addition of 1 mM octanoate to 1 mM pyruvate inhibited pyruvate decarboxylation by 93% without affecting carboxylation. The absence of M1 and M2 pyruvate demonstrated net irreversible pyruvate carboxylation. Under our experimental conditions we found that pyruvate carboxylation in the in vivo heart accounts for at least 3-6% of the citric acid cycle flux despite considerable variation in the flux through pyruvate decarboxylation.

Stereotactic biopsy for intracranial space-occupying lesions: clinical analysis of 550 cases.

OBJECT: To investigate the role of stereotactic biopsy in planning the optimal management of intracranial space-occupying lesions. PATIENTS AND METHODS: Between December 1989 and December 1999, stereotactic biopsy was performed in 550 patients with intracranial mass lesions that were deep-seated or located in the functional area. There were 340 males and 210 females, and their ages ranged from 4 to 75 years. All the procedures were done under local anesthesia with a Leksell stereotactic system. A CT scan was used to determine the coordinates in the first 420 cases and the Aero Tech Stereotactic Surgical Plan System in the subsequent 130 patients. RESULTS: Brain tumors were diagnosed pathologically in 475 patients (86.4%), inflammatory process in 44 (8.0%), other lesions in 12 (2.2%) and no conclusive diagnosis was found in 19 (3.4%). The overall positive rate of biopsy was 96.6%, and the positive rate for brain tumor was 86.4%. Intracranial hematomas after biopsy were found in 13 cases (2.4%). Seizures occurred during the operation in 7 cases (1.2%), and slight and transient neurological deficits were found in 23 cases (4.2%). There were no deaths or other serious complications. CONCLUSIONS: The results suggest that the stereotactic biopsy is a reliable method to obtain histopathological diagnosis of intracranial mass lesions, and it is also of great help in selecting the appropriate management.

Osteoprotegerin and osteoclast differentiation factor in tooth eruption.

A critical cellular event in tooth eruption is the formation of osteoclasts that are needed for bone resorption to form an eruption pathway. To analyze molecular regulation of osteoclast formation and activation, we examined the expression of osteoprotegerin (OPG), an inhibitor of osteoclast formation. In vivo, the gene expression of OPG is reduced in the dental follicle of the first mandibular molar of the rat at day 3 post-natally and in the mouse at day 5. This correlates with the days of maximal mononuclear cell influx and osteoclast numbers in the rat and mouse. Thus, inhibition of OPG gene expression on these days might allow osteoclasts to be formed and/or activated. In vitro studies demonstrated that both colony-stimulating factor-1 and parathyroid hormone-related protein reduced OPG gene expression in follicle cells, suggesting that these are candidate molecules for the in vivo inhibition of OPG expression. Osteoclast differentiation factor (ODF) immunolocalizes to the alveolar bone stromal cells adjacent to the follicle, whereby it might act to stimulate fusion of the mononuclear cells in the follicle.

Differential effects of dexamethasone and clenbuterol on rat growth and on beta2-adrenoceptors in lung and skeletal muscle.

Beta-adrenergic agonists increase growth rate, but their efficacy is reduced over time as the number of beta2-adrenoceptors in muscle decreases. Dexamethasone increases beta2-adrenoceptor density in many tissues, but this effect has not been reported in skeletal muscle. In this study, male rats were treated daily for 10 d with either clenbuterol (4 mg/kg of feed), dexamethasone (.2 mg/kg BW, s.c.), or clenbuterol plus dexamethasone. Untreated rats served as controls. Dexamethasone caused a marked suppression of growth rate, which resulted in decreased (P < .001) body weight (-29%), carcass weight (-30%), hind-limb muscles (-22%), omental fat (-22%), and heart weight (-10%). Feed intake was reduced (-26%), but feed conversion efficiency was also impaired (P < .001). Clenbuterol caused a small increase in growth rate (+6%; P < .05), with an increase in leg muscle (+7%; P < .01) and heart mass (+8%; P < .05). Feed efficiency was improved (P < .001) by clenbuterol. Rats given the combined treatment still showed a reduction in growth rate (-81%). Clenbuterol caused only a mild attenuation of the effects of dexamethasone on feed intake, BW, and carcass weight, but reduced the catabolic effect of dexamethasone on hind-limb muscle to only -8%. Clenbuterol caused a slight increase in the affinity beta2-adrenoceptors in lung for binding to the radioligand (-)[125I]iodocyanopindolol. Relative to control values, the density of beta2-adrenoceptors in lung was +31% with dexamethasone treatment, -45% with clenbuterol, and -23% with the combined treatment. Clenbuterol also decreased beta2-adrenoceptors in skeletal muscle (-35%), but so did dexamethasone (-13%), so the effects of the beta-adrenergic agonist were not attenuated through use of the combined treatment (-40%). The results show that the inductive effect of glucocorticoids on beta2-adrenoceptors is tissue-specific and that glucocorticoid treatment is not a useful adjunct to beta-adrenergic agonist treatment in animal production.

Mechanotransduction and arterial smooth muscle cells: new insight into hypertension and atherosclerosis.

Vascular cells depend on multiple stimuli to maintain a biomechanically and biologically stable environment. Mechanical stresses contribute significantly to multiple cellular processes that regulate vascular structure and function. For example, fluid shear stresses control endothelial cell molecular responses. Less attention has focused on responses of the smooth muscle cell, the 'other' major vascular cell, to mechanical stimuli, in part because of the experimental difficulties in applying precisely controlled deformation. With the advent of new bioengineered devices, combined with modern technologies for studying molecular expression, we are beginning to understand how the smooth muscle cell responds to and controls the biomechanical environment. These studies will help us to understand vascular diseases where vascular mechanics plays a prominent role, such as hypertension, aneurysm formation and atherosclerotic plaque rupture.

Consistent and selective expression of the discoidin domain receptor-1 tyrosine kinase in human brain tumors.

OBJECTIVE: Few molecular targets are both consistently and selectively expressed in a majority of central nervous system (CNS) neoplasms. Receptor tyrosine kinases have been implicated in brain tumor oncogenesis. We previously isolated one such receptor, discoidin domain receptor-1 (DDR1), from high-grade pediatric brain tumors. Here, we analyze the cellular origin and distribution of DDR1 expression in human brain tumors and its expression in tumor cells relative to surrounding brain. METHODS: By use of a digoxigenin-labeled DDR1 riboprobe, we investigated the expression of DDR1 messenger ribonucleic acid in a prospective series of 30 resected human primary and metastatic brain neoplasms, nonneoplastic human brain, and mouse embryonic brain, as well as a mouse glioblastoma model, by in situ hybridization. RESULTS: All the high-grade primary brain and metastatic brain tumors showed unequivocal, intense DDR1 expression within the majority of tumor cells, whereas expression was not observed in hyperplastic tumor blood vessels, normal brain blood vessels, inflammatory cells, or in the normal brain tissue that surrounded the tumor. Receptor expression was limited to tumor cells located within solid tumor tissue. Overall, 27 of 29 resected CNS tumors exhibited tumor cell-specific DDR1 expression, whereas one specimen composed of isolated glioblastoma cells within invaded brain parenchyma showed no detectable staining for this receptor. DDR1 was also expressed preferentially in the ventricular zone (a region of highly proliferating precursor cells) of mice at embryonic Day 15.5. CONCLUSION: We found that DDR1 is consistently expressed in all high-grade brain neoplasms studied and is selective for tumor cells in the specimens analyzed. The expression of DDR1 by tumor cells of CNS neoplasms and by primitive cells of the embryonic ventricular zone suggests that DDR1 is a potentially useful marker of tumor cells within the CNS.

SPIKET and COBRA compounds as novel tubulin modulators with potent anticancer activity.

Agents that either promote or inhibit tubulin polymerization exhibit anticancer activity by disrupting normal mitotic spindle assembly and cell division as well as inducing apoptosis. Recently identified novel agents that target tubulin include synthetic spiroketal pyrans (SPIKET), targeting the spongistatin binding site of beta-tubulin, and COBRA compounds, targeting a unique binding cavity on alpha-tubulin. At nanomolar concentrations, the SPIKET compound SPIKET-P caused tubulin depolymerization in cell-free turbidity assays and exhibited potent cytotoxic activity against cancer cells as evidenced by destruction of microtubule organization, and prevention of mitotic spindle formation in human breast cancer cells. Molecular modeling studies predicted a high-affinity interaction of the first COBRA compounds, COBRA-0 and COBRA-1, with a unique hydrophobic binding site on alpha-tubulin located between the GTP/GDP binding site and the M-loop. Further studies showed that COBRA-1 inhibited GTP-induced tubulin polymerization in cell-free tubulin turbidity assays. Treatment of human breast cancer and brain tumor (glioblastoma) cells with COBRA-1 caused destruction of microtubule organization and apoptosis. COBRA-1 activated the pro-apoptotic c-Jun N-terminal kinase (JNK) signal transduction pathway. COBRA and SPIKET compounds represent two new classes of tubulin targeting agents that show promise as anticancer drugs.

Abnormalities of the human Rb tumor suppressor gene in oral squamous cell carcinoma.

OBJECTIVE: To investigate the status of Retinoblastoma (Rb) gene in 30 patients with primary oral squamous cell carcinoma (OSCC) and 3 healthy control samples. MATERIAL AND METHODS: We determined the status of Rb tumor suppressor gene in oral squamous cell carcinoma using a DNA southern blotting assay with Digoxin (Dig) labeled Rb cDNA probe. RESULTS: The structural abnormalities of Rb tumor suppressor gene were detected in 8 of the 30 invasive OSCC samples analyzed (8/30). 7 cases contained partial gene deletion, and there was 1 case of gene rearrangement. The structural abnormality rate of Rb is 26.7%. No correlation was found between the alteration of Rb gene with the sex, age, site of primary tumor, level of histopathological differentiation, and TNM staging. CONCLUSIONS: Our study shows that abnormalities of the Rb tumor suppressor gene occur in oral squamous cell carcinoma, which include Rb deletion and Rb rearrangement, but its abnormality rate is low.

[The characteristics diagnosis and treatment of hepatic metastasis of simple immature ovarian teratoma].

OBJECTIVE: To analyze the characteristics of hepatic metastasis of pure immature ovarian teratoma and explore proper diagnosis and treatment. METHODS: A total of 18 cases with hepatic metastasis of pure immature ovarian teratoma were involved in this study. The clinical stage, operation, chemotherapy and histopathology of primary and secondary tumor as well as the data of long term follow-up were analyzed retrospectively. RESULTS: All of the hepatic metastatic tumor located on the surface of liver, 66.1% (11/18) of them was clinically stage 3 and 44.4% (8/18) was grade 1 at first operation. The hepatic metastatic rate was 16.7% (3/18) in standard adjuvant chemotherapy group but increased obviously to 31.2% (15/48) in irregular chemotherapy group. Auxiliary diagnostic methods could not indicate correct results. The surgical resection rate of hepatic metastasis of pure immature ovarian teratoma was 94.4% (17/18). There was less complication occurred in the group with the diameter of tumor less than 15 cm. The follow-up time ranged from 3-205 months with the mean of 20.9 months. The 3 year survival rate was 77.8% (14/18), lost of follow-up rate was 0.0% (0/18); The 5 year and 10 year survival rate was 55.6% (10/18) and 38.9% (7/18) respectively, lost of follow-up rate was 22.2% (4/18) and 38.9% (7/18) respectively, one of them had survived for more than 17 years. CONCLUSIONS: The hepatic metastatic rate of pure immature ovarian teratoma could be decreased by using standard adjuvant chemotherapy. Suitable surgical treatment could reduce complication and improve the prognosis of this kind of patients.

Embolic events in 93 elderly Chinese patients with atrial fibrillation.

OBJECTIVES: To evaluate the prevalence of embolic events and relevant factors in elderly Chinese patients with atrial fibrillation(AF), and to provide evidence on ways to prevent embolic events. METHODS: Autopsy data from ninety-three continous elderly Chinese patients with AF were analysed. The incidence of embolic events and its relationship to underlying disease, pathologic changes in the heart, and other clinical characteristics were examined. RESULTS: Embolism were observed in 27 of 93 cases, with an incidence of 29.03%. The incidence of embolic events was higher in elderly patients with rheumatic heart disease than those with coronary artery disease, hypertensive myocardiopathy and heart diseases. Patients with chronic AF, with a course of AF > or = 3 years, and those with heart failure or diabetes had a higher incidence of embolic events than those without these complications. There was significant difference in incidence between paroxysmal and chronic AF. Patients with left atrial or ventricular enlargement, mural thrombosis in cardiac chambers, valvular calcification and valvular vegetation also had a higher incidence of embolic events. Oral dipyridamole (75-150 mg/d) or aspirin (50-150 mg/d) showed no definite effects in preventing embolism in some patients. CONCLUSIONS: There was a high incidence of embolic events in elderly Chinese patients with AF. Anticoagulation therapy should be provided to the elderly patients with AF, especially to the patients with risk factors for embolism.

[Study on the amplification uniformity of whole genome in a single cell by degenerate oligonucleotide primed polymerase chain reaction].

OBJECTIVE: To investigate the uniformity of whole genome amplification from a single cell by degenerate oligonucleotide primed polymer ase chain reaction (DOP-PCR). METHODS: Amplification of the whole genomic DNA from a single cell with XX, XY, XO, XXY, +13 or +21 was performed by DOP-PCR. Comparative genomic hybridization (CGH) of a single cell DOP-PCR product against the genomic DNA or a single cell DOP-PCR product from normal male as carried out respectively. RESULTS: The average profile of fluorescence intensive ratio in CGH with the genomic DNA as the reference was greatly fluctuated and the standard deviation in about 30% haploid was beyond the normal limits. False positive hyperrepresentation was existed in X chromosome while trisomy 13 and 21 were failed to be detected. However, the distributions of the mean and the standard deviation of the ratio in the CGH with DOP-PCR product as the reference were acceptable. The copy number changes of chromosome X, Y, 13 and 21 were demonstrated. CONCLUSION: Although the single cell DOP-PCR product is not completely uniform, it would be reliable to study the whole genome of a single cell by controlling the conditions of PCR and selecting the suitable control reference because the unequal amplification is unrandom.

[High-dose chemotherapy with peripheral-blood stem cells transplantation in patients with advanced gynecological malignant tumors].

OBJECTIVE: To evaluate the clinical effectiveness of high dose chemotherapy(HDC) with peripheral-blood stem cells transplantation(PBSCT) in the patients with advanced malignant tumors. METHODS: Four patients with advanced or refractory ovarian carcinoma and 1 patient with recurrent fallopian tube carcinoma received the HDC with PBSCT after optimal tumor debulking. Of whom 4 patients were with the recurrent or progressive tumors, 1 patient received HDC as primary chemotherapy. The response and toxicity were assessed after treatment. RESULTS: Among 5 patients, four patients are survival. One case died from the recurrence of juvenile granulosa cell tumor. The average survival periods were 21 months. The tumors were all recurrent among the 4 survival patients. The average periods of recurrence after HDC treatment was 9 months. The main toxicity of HDC was severe marrow suppression. The lowest value of the neutrophil count and platelet count was 0.05-0.2 x 10(9)/L and 7 x 10(9)/L-8 x 10(9)/L respectively. The median time to recover a neutrophil count greater than 0.5 x 10(9)/L was 14 days and a platelet count greater than 50 x 10(9)/L was 17 days. None of the patients was complicated with severe complications. CONCLUSIONS: HDC and PBSCT used for ovarian carcinoma was flexibility and security if the patient was selected strictly though its toxicity was severe. Further study among the chemorefactory recurrent patients need to try in the future.

[Effect of low dose of inhaled corticosteroid combined with small dose of oral theophylline on treatment of bronchial asthma].

OBJECTIVE: To investigate the effect of small dose of oral theophyllin combined with low dose of inhaled beclomethasone dipropionate (BDP) on clinical symptoms, bronchial responsiveness and hypothalamic-pituitary-adrenal axis (HPAA). METHODS: 43 patients with mild-moderate bronchial asthma were randomly divided into A and B groups. 21 subjects in group A were treated with oral substained release theophyllin (Protheo) (200 mg/night) and inhaled BDP (300 micrograms/day); 22 cases in group B received inhaled BDP (600 micrograms/day) and oral placebo. RESULT: Before and 13 weeks after the treatment, symptom scores, peak flow(PEF), and peak flow rate (PEFR) for group A were (1.8 +/- 0.9) and (0.10 +/- 0.30), (295 +/- 98) L/min and (444 +/- 150) L/min, (22 +/- 8)% and (9 +/- 3)% respectively, and those for group B were (1.90 +/- 0.70) and (0.10 +/- 0.30), (328 +/- 129) L/min and (441 +/- 146) L/min, (24 +/- 7)% and (9 +/- 3)% respectively, which were remarkably improved in both groups (P < 0.01), and bronchial provocation responsiveness to histamine tests (BHR) in both groups were also improved significantly (P < 0.01). There were no differences between two groups (P < 0.05). Frequency of using inhaled beta 2 agonist to relieve nocturnal asthmatic attacks in group B (3.9 +/- 1.7) was significantly greater than that in group A (1.3 +/- 1.0, P < 0.05). Plasma ACTH concentrations, basic cortisol levels and cortisol responses to ACTH before and 13 weeks after the treatment in group B were (31 +/- 13) ng/L and (20 +/- 8) ng/L, (95 +/- 50) micrograms/L and (86 +/- 48) micrograms/L, (156 +/- 98)% and (74 +/- 46)% respectively, which were decreased significantly after the treatment (P < 0.05). There were no such changes in group A. CONCLUSIONS: It was suggested that small dose of oral theophylline combined with low dose of inhaled BDP might have the same effect on relief of clinical symptoms and bronchial responsiveness, without suppression on HPAA function, comparing with relatively higher dose of inhaled BDP.
