Lateral diffusion of membrane lipids changes with aging in C57BL mouse dorsal root ganglion neurons from a fetal stage to an aged stage.

The membranes of dorsal root ganglion (DRG) neurons dissected from different age groups of C57BL mice, from 18-day fetal to 30-month-old mice, were labeled with the fluorescent analog of fatty acids, F18, after a day in culture. The fluorescent probe specifically labeled the cell surface. The lateral diffusion of F18 (5-(octadecylthiocarbamoylamino) fluorescein) was measured with fluorescence photobleaching recovery (FPR) method. During development, the lateral diffusion coefficients of F18 rapidly decreased from (0.34 +/- 0.07) X 10(-8) cm2/s (18-day-old fetus) to (0.22 +/- 0.07) X 10(-8) cm2/s (3-day-old newborn). Then the values slowly decreased and reached (0.13 +/- 0.05) X 10(-8) cm2/s in a 6-month-old stage. In stages older than 6-months the lateral diffusion coefficients scarcely changed with aging. As this decrease in the membrane fluidity with increasing age is parallelled to that in capacities of extending neurites, it is thought that the membrane fluidity might change with aging in concert with changes in important cell functions.

Inhibition of pentagastrin-stimulated gastric acid secretion by intraileal administration of bile and elevation of plasma concentrations of gut glucagon-like immunoreactivity in anesthetized dogs.

The effects of intraileal administration of bile on gastric acid secretion stimulated by a submaximal dose of intravenous pentagastrin infusion and on plasma concentrations of gut glucagon-like immunoreactivity (gut GLI) were studied in anesthetized dogs. Gastric acid secretion was measured for a 2-h period at 15-min intervals before and after intraluminal instillation of test solutions. 100 ml of canine bladder bile diluted to 10% in saline evoked a significant inhibition (20%) of gastric acid secretion. The inhibition of gastric acid secretion was accompanied by an elevation of plasma concentration of gut GLI, whereas saline instillation (in controls) caused no responses. Although the inhibition of gastric acid secretion and the elevation of plasma gut GLI are parallel phenomena, gut GLI can be reasonably postulated as one of the candidate mediators of bile-induced inhibition of gastric acid secretion, since its structurally related peptides, pancreatic glucagon, glicentin and oxyntomodulin have been reported as inhibitors of gastric acid secretion.

Carcinoid in a gastrointestinal duplication.

This is a report of a 12-year-old boy having gastric duplication with foci of carcinoid. He complained of right abdominal pain. Physical examination showed a cystic mass in the right abdomen. It was excised operatively. The mass was a duplication cyst composed of gastric mucosa with a muscle layer. Foci of carcinoid was detected in the muscle layer. The tumor cells involved nerve fibers but did not extend to the subserosa. Postoperative blood serotonin and histamine levels were normal. The patient is healthy without signs of recurrence 17 months after operation. This is the second case of gastrointestinal duplication with foci of carcinoid. The findings indicate that careful histologic examination is necessary to determine the exact nature of the cyst.

Mapping and sequencing of RNAs without recourse to molecular cloning: application to RNAs of the Sabin 1 strain of poliovirus and its defective interfering particles.

Complementary DNA to the genome of the Sabin 1 strain of poliovirus was prepared by reverse transcription with oligo(dT)10 as a primer and separated into six classes of DNA by their size. Each class of the DNA, after digestion with restriction endonuclease HaeIII, was analyzed by two-dimensional polyacrylamide gel electrophoresis. Comparison of the patterns of the restriction fragments led us to compose a possible arrangement of the restriction fragments on the viral genome. Sequence analysis of these fragments indicated that the arrangement was consistent with the known total nucleotide sequence of the genome. In the determined sequences, two bases were observed to differ from those of a cloned complementary DNA of the Sabin 1 genome. This suggested that the sequence of the cloned DNA reflected that of a mutated virus genome that was a minor component in the virus inoculation stock. The genomes of defective interfering particles generated from the Sabin 1 strain were also analyzed by this technique. The results suggested that the RNAs lacked an internal region of the Sabin 1 RNA encoding viral capsid proteins. The location of the deletion was further confirmed by determination of the nucleotide sequence of a cloned complementary DNA copy of the defective interfering particle RNA. Thus, the method described here is useful for mapping and sequencing of RNAs and for knowing whether cloned cDNAs represent the major population of RNA molecules or not.

Hepatic microcirculation of liver cirrhosis studied by corrosion cast/scanning electron microscope examination.

Changes of hepatic microcirculations in 22 autopsy cases of liver cirrhosis were analyzed by corrosion cast/scanning electron microscope (SEM) examination. By this method, the site of arterioportal (A-P) communication in liver cirrhosis was clearly demonstrated between proliferated portal venules and arterial capillaries. The communications were observed at the same site as in the normal liver and were not at larger arterial and portal vein branches. The findings indicate that the increase of A-P communication in liver cirrhosis may be called "capillary shunting". On the basis of the findings, it was postulated that the A-P shunt could not assist in the development of portal hypertension by the transmission of high arterial pressure to the portal vein but could only compensate for decreased portal flow and/or elevate the oxygen concentration in the sinusoids to improve the hypoxic state of the liver parenchyma. It was also demonstrated that the arterial capillarization of the interstitial septa in micronodular wide septal cirrhosis was more prominent than that in macronodular thin septal cirrhosis. A grade of portal vein reduction and compensatory arterialization in a fibrous septum have been regarded as an index to estimate the advancement of liver cirrhosis. Therefore, if alcoholic micronodular cirrhosis could change into macronodular, the process should have occurred at least before the establishment of micronodular wide septal cirrhosis.

Endolymphatic stromal myosis of the uterus with metastasis to ovary and recurrence in vagina.

A 24-year-old woman underwent a supracervical hysterectomy with findings of an endolymphatic stromal myosis (ESM) with vascular invasion as a submucosal tumor of the uterine fundus. Four years later, metastasis of ESM in the right ovary was found after salpingo-oophoréctomy. Parametrial masses and a tumor infiltrated cervical stump revealed the same structure of ESM after operation. Though subtotal excision of the pelvic and vaginal recurrences nine years after the first operation for ESM and in spite of oral administration of danazol and duphaston (hydrogesterone), a recurrent vaginal tumor developed after the fourth operation and local application of chemotherapy with 5-fluorouracil was given. At present, the patient's condition is stable without distant metastasis.

Malignant clear cell cystadenoma of the ovary.

A 50-year-old female had suffered from abdominal distension and a lower abdominal mass for 3 months. Ultrasonographic examination demonstrated a myoma uteri pattern with a large cystic ovarian tumor. Laparotomy was done on August 30, 1983 and simple total hysterectomy with bilateral salpingo-oophorectomy was carried out. FIGO stage IIC ovarian cancer was stated clinically. Histologically, clear cell cystadenoma of right ovary having benign, proliferative and malignant epithelial elements without dense ovarian stroma was found and diagnosed as malignant clear cell cystadenoma. In spite of Cisplatin therapy, the patient expired on April 6, 1984.

Benign hemangioendothelioma of the pancreas with obstructive jaundice.

A 3-year-old boy with benign hemangioendothelioma of the pancreas is reported. He showed the presenting sign of obstructive jaundice. The boy was treated with temporary choledochojejunal bypass without additional treatment. In a 16-month follow-up the child has been well and asymptomatic. Laboratory data showed no abnormality. Biopsied specimen of pancreatic head showed hemangiomatous pattern histologically. Immunohistochemical studies of the biopsy revealed positive reaction to factor VIII-related antigen (FVIII-RAG), and endothelial cell marker, supporting an endothelial origin. Hemangioendothelioma has a favorable prognosis and spontaneous regression can be expected. So the first treatment of the tumor should be conservative.

Proliferative Brenner tumor of the ovary.

A 72-year-old woman was admitted with abdominal distention and a hard mass in the pelvis. Abdominal laparotomy was performed, and a large solid left ovarian tumor was found. Left salpingo-oophorectomy was carried out. Histology showed cords and nests of stratified high cellular epithelium with dense eosinophilic material in epithelial nests and dense fibrous stroma identified as a Brenner tumor of the ovary. Because mild to moderate cytologic atypia with mitotic activity was present, a diagnosis of proliferative Brenner tumor was made. After the operation, the patient was in good condition with no recurrence or metastasis so far.

Niemann-Pick disease associated with liver disorders.

We report a case of Niemann-Pick disease (NPD) with accumulation of sphingomyelin in reticuloendothelial system (RES), hepatocellular giant cell transformation (GCT), cirrhosis, and multiple hepatocellular adenomata in a 19-month-old girl. GCT, but no NP-cells, were seen at age 3 months by biopsy. Cirrhosis and hepatocellular adenomata were demonstrated in the liver at 19 months of age. Cytoplasmic, probably locally synthesized, globules of alpha-1-antitrypsin (A-1-AT) were accumulated in the hepatocellular adenomata. A-1-AT and alpha-fetoprotein (AFP) were present in the serum.

Organelle motility in rat pituitary clonal cells. I. Dynamic movements of intracellular organelles.

Intracellular organelle motion within clonal pituitary tumor cells (GH3) was observed directly with a contrast enhancement, computer-video microscope system. All particles except nuclei moved in a complex fashion. Two types of particles predominated; one large and round, the other small and elongated. We classified the movements of these particles as saltation, oscillation and slow translocation. Saltation was directional movement with velocity of the order of 1 micron/sec. Oscillation was local motion occurring within 1 micron that showed no specific direction. Its velocity was similar to that of saltation. Large particles, in particular, showed the 3rd type of movement, slow translocation. The velocity appeared to be one order slower than that of saltation. We also examined the cells with fluorescent, dark-field and electron microscopies. We concluded that the large round particles were lysosomes and the small elongated ones mitochondria. The microtubule depolymerizer, vinblastine and the microfilament depolymerizer, cytochalasin D, completely inhibited all the types of organelle movement. The mechanism and significance of these organelle movements are discussed.

Lactic acidosis and hypoglycemia associated with acute leukemia.

A 42-year-old man was admitted because of episodic attack of general malaise. He was lethargic and had a severe lactic acidosis and hypoglycemia. Blood chemistry and endocrinological data were normal. Glucose administration led to an improvement in the hypoglycemia but not the lactic acidosis. At autopsy, there was a massive infiltration of leukemic cells in both kidneys and in liver. Phosphoenolpyruvate carboxykinase, pyruvate carboxylase and glucose-6-phosphatase activities in patient's liver were much the same as in the control liver, but fructose-1, 6-diphosphatase activity was slightly reduced. Since circulatory failure was absent, type B lactic acidosis has to be considered. Since hypoglycemia was associated with acidosis, the severe lactic acidosis in our patient may have been due to an overproduction of lactic acid as well as to an impaired hepatic gluconeogenesis in the presence of leukemic cells.

Improved survival and differentiation of newborn and adult mouse neurons in F12 defined medium by fibronectin.

Few newborn DRG neurons attached on polylysine coated coverslips in Ham's F12 medium, although most adult neurons attached on them. Fibronectin improved the attachment of newborn neurons and improved differentiation capacity of both neurons. Under this condition, newborn DRG neurons developed their processes more rapidly, but died more prematurely than adult neurons. Neural cell adhesion molecules and differentiation capacity of DRG neurons might change with aging.

Osteogenesis bioassay and immunohistochemical and radioisotopic studies of parotin, parotid gland extract, and subunit.

Bioassay data support the hypothesis that the salivary gland participates in endocrine regulation of the development and maintenance of connective tissues. Epiphyseal cartilages become dystrophic in young growing rats after the resection of all major salivary glands. A subunit obtained from parotin, an extract of bovine parotid glands, contains the active agent for the presumed endocrine function of the salivary gland. Daily injections of 3 mg/rat of parotin or the subunit allow normal epiphyseal endochondral osteogenesis in salivary gland-deprived rats. The active agent appears to be secreted by the salivary acinar cells and resorbed through the striated ducts. Pancreatic islets and striated ducts of salivary glands share immunohistochemical activities for insulin, glucagon, and the subunit of parotin. Hyperglycemia and hypocholesterolemia occur in intact rats given 5 mg/day of parotin for 30 days. Together these data suggest endocrine functions of the salivary gland and possible interactions between the pancreatic islet and salivary gland.
