Structural homology of microtubule-associated proteins 1 and 2 demonstrated by peptide mapping and immunoreactivity.

The major high molecular weight microtubule-associated polypeptides from hog brain (MAP-1 and MAP-2) were compared by one- and two-dimensional peptide mapping under varied conditions and by immunological techniques. Partial digestion of MAP-1 and MAP-2 with Staphylococcus aureus V8 protease and analysis in one dimension gave rise to very similar peptide maps independent of whether 125I-, 3H-, or 32P-labeled proteins were used. One-dimensional cleavage patterns of significant similarity were also obtained by partial digestion of MAP-1 and MAP-2 using trypsin or chymotrypsin. Furthermore, a pronounced similarity, although clear nonidentity, of MAP-1 and MAP-2 was also revealed after exhaustive digestion of 125I-labeled proteins with S. aureus V8 protease or trypsin followed by analysis of peptides in two dimensions. For immunological comparison, antisera were used that had been raised in rabbits using electrophoretically purified MAP-1 and MAP-2 components as immunogens. As determined by immunoprecipitation, the antiserum raised to MAP-1 was equally reactive with MAP-1 and MAP-2 components, whereas the antiserum to MAP-2 reacted primarily with MAP-2. Indicating the presence of common as well as unique antigenic determinants on MAP-1 and MAP-2, these results, therefore, were in agreement with the peptide mapping data. Implications of these results for biosynthetic mechanisms as well as differential distribution and functions of MAPs in cells are discussed.

Ethnic distribution of phenylketonuria in the north German population.

Results of neonatal screening for phenylketonuria (PKU) suggest a west-east gradient of PKU gene frequency in central Europe. In order to test the hypothesis that the unexpectedly high prevalence of PKU in northwestern Germany (northern region of the FRG) is due to the migration of Germans from eastern regions of prewar Germany in the decade after World War II, grandparental origin was determined in a group of 87 pediatric PKU patients and in a control group of 210 children. Grandparents of east German origin were significantly more frequent among the PKU patients. The observed frequency distribution of grandparental subgroups was described by a theoretical distribution in order to obtain a likely set of values for the ratio between the frequency of the PKU gene in the autochthonous populations of prewar northeastern and northwestern Germany. The most likely value for the PKU gene frequency ratio was 1.37, which indicates that the prevalence for PKU in prewar northeastern Germany was almost twice as high as in the autochthonous population of the northwest.

High frequency FP2 donor of Pseudomonas aeruginosa PAO.

After NG mutagenesis an FP2 donor was isolated which exhibited an enhanced conjugational capacity for chromosomal genes. The recombination frequency was increased by two orders of magnitude as compared to the parental strain. In plate matings recombinants arose at a frequency up to 5 X 10(-1) per donor cell. Late markers also recombined efficiently. An Hfr state of the donor strain was supported by (i) the high recombination frequency, (ii) the incompatibility reaction with plasmid pRO271 (= FP2::Tn401) and (iii) the clearcut transfer kinetics in interrupted matings, even for a late marker.

Mapping of cysteine genes on the chromosome of Pseudomonas aeruginosa PAO.

Three loci coding for different steps in the pathway of cysteine biosynthesis have been mapped by R68.45-mediated coconjugation analysis. The cysteine auxotrophic mutants could be subdivided into sulfite and sulfide-requiring mutants. Sulfide-requiring mutants (cysIV group) were localized at a single position between pyrF and pur-67, while sulfite-requiring mutants (cysI and cysII) mapped at two different regions. The cysI group was also localized between pyrF and pur-67, although more distal to pyrF than the cysIV group. This group included the cys-54 marker, which has been mapped previously. The second group of sulfite-requiring mutants, designated as cysII, was cotransducible with hisI and localized at the end of the PAO chromosomal map. This location was also confirmed for the marker cys-59. The marker cys-59 (which was cotransducible with hisI) was cotransferred by R68.45-mediated conjugations with both the late marker pur-67 and the early marker ilv-226. As the late marker hisI was positioned at about 60-65 min (Herrmann and Günther, in press) the length of the PAO chromosome was estimated to be about 70 min.

Adjustment of the oral temperature rhythm to a fixed watchkeeping system on board ship.

Oral temperature data were collected from 12 members of the crew of an oil tanker at sea. Most of the personnel examined were engaged on watchkeeping duties on a '4 on, 8 off' fixed-hours system; the remainder included 3 "day-workers". The study commenced after the subjects had been following their particular work schedules continuously for several weeks, thus providing good opportunity for adaptation to them. Observations were made at 4-hourly intervals during waking hours, over a period ranging from 8 to 13 days in individual cases. The form of the mean curves produced by averaging the readings over all days indicated that a reasonable degree of adjustment of the temperature rhythm to the different sleep/wake routines imposed by the work system had occurred. Estimates of rhythm phase and amplitude obtained by "single cosinor" time series analyses of the sequential data supported this impression. However, further investigations are needed to substantiate these findings, and also to determine how long it takes for the rhythm adjustment process to reach completion in inexperienced workers.

[Psychosocial situation of married couples prior to artificial insemination with donor sperm].

57 couples who attended our outpatient department for artificial insemination with donor sperm (AID) had a detailed semistandardized interview prior to treatment. The Giessen Test was used as personality test, as well as a questionnaire concerning the sexual and partner relationship. We found that it was the woman who had to bear the major part of the intrapsychic burden and who primarily suffered because of the problem which was actually caused by male sterility. However, 54 of 57 couples had no psychological difficulties to overcome the sterility problem. These couples developed individual and partnership strategies to deal with the burden of unwanted childlessness. They preferred AID rather than adoption, because for the woman this alternative offers the chance of real parenthood. AID seems to provide a medico-technical solution for couples with sterility problems: the reproductive process at the biological level was regarded as distinctly separated from their future role as parents which they considered to be a social task.

Specific in situ phosphorylation of plectin in detergent-resistant cytoskeletons from cultured Chinese hamster ovary cells.

Plectin (Mr = 300,000) was found to be an abundant polypeptide component of Chinese hamster ovary cells accounting for up to 1% of cellular protein. Seventy-five per cent of the plectin were present in cytoskeletons prepared by extraction of attached cells with 0.15% Triton X-100. As shown by immunofluorescence microscopy, plectin's spatial arrangement within these cytoskeletal preparations appeared well preserved, though slightly more filamentous compared to nonextracted cells. Upon in situ incubation of cytoskeletons with [gamma-32P] ATP followed by solubilization and immunoprecipitation, plectin was identified as one of the major phosphoacceptors. A basic phosphorylation of the protein was accomplished by a type I cAMP-independent protein kinase, while a cAMP-dependent protein kinase enhanced its phosphorylation up to 2-fold. Peptide mapping revealed that the two kinases phosphorylated different molecular sites. Peptide maps generated from cytoskeletal plectin phosphorylated in vitro using [gamma-32P]ATP and plectin phosphorylated in vivo using 32Pi were virtually identical demonstrating that the in situ phosphorylation of plectin in preparations of cytoskeletons was specific. Moreover, the specific radioactivity of cytoskeletal plectin was three times higher than that of detergent-extracted plectin, suggesting that phosphorylation is important for the protein's association with the cytoskeleton.

[The strategy of tuberculosis control in East Germany after 1982].

100 years after ROBERT KOCH'S detection of the tubercle bacillus the tuberculosis situation in GDR has so improved due to the progress in tuberculosis control, systematic activities in prevention, casefinding, and care and due to the comprehensive social measures that tuberculosis cannot be any more regarded as a "Volkskrankheit". The reduction of the annual infection rate to less than 0.1%, the decrease of infectious tuberculosis to less than 5/100,000 inhabitants, the shortening of the periods of chemotherapy and the further regression of relapses has made it possible to rationalize tuberculosis control by giving up indiscriminate mass x-ray examinations of the whole population in favour of the supervision of endangered person groups, by reducing the extent of risk groups according to stricter criteria, by making shorter the follow-up of the several forms of tuberculosis. Primary health care services have to be involved still more into tuberculosis control, especially in casefinding and outpatient treatment.

[Importance of communal environmental factors in the development of chronic lung diseases].

After a review on the problem the results of a longitudinal study are reported performed from 1971 to 1980 in men of two towns with an extremely contrasting emission situation. The investigations showed a higher prevalence of chronic bronchitis, an aggravation of clinical symptoms of bronchitis, more social effects of chronic bronchitis, a higher deterioration of pulmonary function (FEV1), and an increase of the average annual decline of FEV1 when the emission load was high. But also in areas with a very strong emission load the false individual behaviour (smoking) is of highest importance for the development of chronic bronchitis.

Incorporation of [1-14C]acetate into fatty acids and aliphatic moieties of glycerolipids in Leishmania donov ani promastigotes.

1. Short-time incubations (20-300 sec) of Leishmania donovani with [1-14C] acetate showed lauric, myristic and palmitic acid to be labeled first, whereas from 150 sec on the radioactivity was mainly found in stearic and oleic acid. 2. After an incubation period of 7 days the distribution of radioactivity in fatty acids was similar to the mass distribution of fatty acids. 3. Up to 300 sec 14C-activity was present mainly in acyl moieties of lipids (94%). 4. Ether moieties of radylglycerols, diradylglycerophosphoethanolamine and -inositol slowly incorporated radioactivity in pulse-chase experiments from 1 to 24 hr. 5. Pulse-chase experiments revealed significant differences in plasmanylethanolamine, acylglycerol and sterol metabolism of cells in the logarithmic or stationary phase of growth.

[The development of tuberculosis epidemiology in the German Democratic Republic].

The development of tuberculosis in GDR is analysed upon the different points of view. After assessing the validity of the parameters for evaluating the tuberculosis problem (mortality, incidence and prevalence of disease and infection) some aspects remarkable for tuberculosis control are discussed. Tuberculosis has been vastly reduced in all parameters since 1949. However, the regression of tuberculosis did not occur with the same rate in the various age and finding groups. By this the efficacy of the control measures has changed making necessary a reconsideration of the methods. The Volkskrankheit (common disease in the population) tuberculosis has become a vastly reduced endemic disease in GDR. For future tuberculosis control conclusions are drawn from the analysis.

[Relevance of the maximal expiratory flow-volume curve in an epidemiological study].

In an epidemiological study a random sample (1,554 males aged 35 to 54 years) of the population of a hydrogen chloride air-polluted area was investigated by means of the modified MRC questionnaire and maximal expiratory flow-volume curves. The MEF-values were dependent on age and height. We stated no significant differences between potash miners and the remaining population. In contrast we found significant differences between smokers and non-smokers and also between persons with and without respiratory symptoms. Persons with bronchitis and chronic bronchitis most differed from reference groups in MEF 75 (flow at high lung volume). Comparing smokers with non-smokers MEF 25 attained the highest significance level. In our investigation the maximal expiratory flow-volume curve was only a little more sensitive than conventional spirometry.
