Myoclonus in a case of suspected progressive rubella panencephalitis.

Predominantly unilateral myoclonus induced by movement of the affected parts in a 21-year-old Japanese man with suspected progressive rubella panencephalitis appeared in the course of treatment for epilepsy with anticonvulsant drugs after five years. Antirubella antibody levels in the serum and CSF were elevated and IgM antibody against rubella was found in the serum. An EEG recorded while the patient was awake showed diffuse 3- to 4-Hz theta activity with high voltage. The neurological symptoms are progressing slowly.

Selective loss of acetylcholine sensitivity in a nerve cell line cultured in hormone-supplemented serum-free medium.

When clonal rat pheochromocytoma PC12h cells were cultured in a hormone-supplemented serum-free medium, the carbamylcholine-elicited catecholamine release from cells cultured in serum-free medium was completely abolished. On the other hand, the high potassium-induced catecholamine release was not changed, even in PC12h cells cultured in serum-free medium. The lack of carbamylcholine sensitivity was confirmed directly by measuring carbamylcholine-induced 22Na influx, which was completely abolished in PC12h cells cultured in serum-free medium. The loss of carbamylcholine-induced 22Na influx seemed to obey nearly first-order kinetics and was fully restored upon a re-exposure to serum. The half-time for the loss was about 1 day, and the cultivation for 5 days in serum-free medium caused a 95% decrease of the nicotinic sensitivity in PC12h cells. The carbamylcholine-induced 45Ca influx into cells also was lost due to the serum-free cultivation. By contrast, the binding of alpha-bungarotoxin, which is an antagonist of nicotinic acetylcholine receptor in muscular cells, remained and did not change in PC12h cells cultured, even in the serum-free medium. In addition, veratridine-dependent 22Na influx and high potassium-induced 45Ca influx into cells, and high potassium-induced 86Rb efflux from cells cultured in the serum-free medium were also perfectly preserved. These results suggest that PC12h cells cultured in the serum-free medium seemed to be a useful model for comparing the differential mechanisms between acetylcholine sensitivity and other membranous functions on this cell.

Nerve growth factor induces specific enkephalin binding sites in a nerve cell line.

Specific enkephalin binding was found in a rat pheochromocytoma cell line, PC12 (subclone h), when cultured in the presence of nerve growth factor (NGF; 50 ng/ml). Specific binding of [3H][D-Ala2]Met-enkephalinamide was saturable with a KD=2.4 nM and Bmax = 866 fmol/mg of protein. The binding was displaced completely by morphine and naloxone at a concentration of 10(-4) and 10(-6) M, respectively. beta-Endorphin and dynorphin [1-13] also displaced the binding; KI = 9.6 and 81.6 nM, respectively. Enkephalin binding in PC12h cells cultured in the absence of NGF was very low. Binding was dramatically increased when cultured in the presence of NGF for 7 to 10 days. The enkephalin binding was induced by NGF in the cells cultured in a hormone-supplemented serum-free medium. In contrast, epidermal growth factor was of two orders lower potency, and insulin had no effect on the expression of enkephalin binding sites. Since the differentiation effects of NGF on PC12h cells are though to offer a model system to investigate the effect of NGH on neurons, the present observations may suggest a physiological role for NGF in the induction and maintenance of specific opiate receptors during development of the autonomic nervous system.

Nerve growth factor-mediated stimulation of tyrosine hydroxylase activity in a clonal rat pheochromocytoma cell line.

In order to confirm the multiple neurotransmitter biosynthetic ability, the possibility to separation of the activities of tyrosine hydroxylase (TH), choline acetyltransferase and glutamic acid decarboxylase was tested by subcloning of a clonal rat pheochromocytoma PC12 cell line. All of 9 subclones obtained showed significant activities of above 3 enzymes, indicating that the PC12 cell has multi-functional properties of neurotransmitter syntheses. One of the subclones, designated PC12h, was demonstrated to have nerve growth factor- (NGF) responsive TH activity. The ED50 value of NGF to increase the TH activity was 1.7 ng/ml (6.5 X 10-11 M). A simultaneous addition of saturating amounts of NGF (50 ng/ml) and dexamethasone (10-6 M) resulted in the increase of TH activity that is equal to the sum of those achieved when either effector was added separately, indicating that the NGF- mediated increase of TH activity in PC12h cells was independent upon the effect of dexamethasone. And also, the TH activity increased by NGF was somewhat potentiated in PC12h cells cultured in a hormone- supplemented serum-free medium.

A mouse neuroblastoma x rat glioma hybrid cell produces immunoreactive substance P-like material.

A new hybrid cell line, designated NG115-401, produced a significant amount of immunoreactive (IR) substance P-like material. The production was measured by radioimmunoassay using specific antisera raised to substance P. The production rate was dependent upon cell growth, and maximum production occurred at the same stage (late logarithmic stage) as the time of appearance of choline acetyltransferase activity, one of the characteristic neuronal properties of the cells. On the other hand, no IR-substance P was found in hybrid cells other than NG115-401, i.e. NG115-301 and NG115-303 cells, which were derived from the same parent mouse neuroblastoma N115TG-2 cells. Furthermore, in an attempt to survey IR-substance P production in several clonal cells, consisting of some mouse neuroblastoma cells and a rat glioma cell, and several of their hybrid cells, we could find no cell line producing a significant amount of IR-substance P except for NG115-401 hybrid cells. The cellular production of IR-substance P in NG115-401 hybrid cells was also confirmed by detecting the material in all 14 subpopulations of this hybrid cells. Partial characterization of IR-substance P-like material produced by NG115-401 hybrid cells was carried out by its application to gel-filtration column chromatography. Only high molecular weight material (approximately 26,000) appeared in the column eluates.

[A case of arteriovenous malformation, grade IV, with two giant aneurysms in dominant side speech and motor-senosry area--evaluation of grades, operability with microsurgical technique and pre- and post-operative performance status in 37 cases of cerebral vascular malformation (author's transl)].

A case of grade IV arteriovenous malformation(AVM) with 2 giant aneurysms, deeply seated in the dominant hemisphere speech and motor-sensory area, improved remarkably by a total excision, after an incapacitating rebleeding which occurred 4 months after a subtotal excision of the AVM. The author has learned importance of audacity on the part of the neurosurgeon. Personal experience of 37 CVM's (including 30 AV angiomas, 4 cryptic AVM's, and 3 other types of CVM's was analyzed. Of 37, 34 were operated on. Three were considered inoperable and were treated with radiotherapy (including proton beam). Of the 34 operated (30 AVM), 31 (24 AVM) were totally removed. None of them died or exhibitied deterioration. Of the 3 partially operated cases, one improved, two remained unchanged, but one of the two died of rebleeding 3 months later. Two of the radiated cases were later severely incapacitated due to rebleeding. One that underwent proton therapy is still under observation without deterioration after 18 months.

[A case of multiple fusiform aneurysms presenting homonymous hemianopsia (author's transl)].

Fusiform Aneurysms are encountered in elderly persons with advanced arteriosclerosis. When they are enlarged to compress the neighboring structures, they may resemble tumors symptomatically. Especially when situated in close relation to the optic nerve or chiasmatic region, pituitary tumor is likely to be suspected. This is a case of 62 year old female with a giant fusiform aneurysm of the right internal carotid artery extending to the middle cerebral artery which caused left homonymous hemianopsia and a large aneurysm in the cavernous portion of the left internal carotid artery with fusiform extension of the distal portion of the carotid artery which presented left blepharoptosis, dilated pupil and total ophthalmoplegia. The basilar artery and the right meningohypophyseal artery showed fusiform dilatation as well. Systemic atherosclerotic change, abdominal aortic aneurysm, nephrosclerosis were also accompanied. The computed tomography demonstrated the aneurysms distinctly. Left oculomotor palsy disappeared after six months with residue of left homonymous hemianopsia. Surgical intervention was not attempted, because of the widely distributed constitutional aneurysms in the whole body.
