Inhibition of mitochondrial respiration by asteltoxin, a respiratory toxin from Emericella variecolor.

Asteltoxin, a respiratory toxin from Emericella variecolor, was examined for an inhibitory effect on mitochondrial function. Asteltoxin strongly inhibited state 3 respiration, which was released by an uncoupling agent, 2,4-dinitrophenol (DNP), indicating that the action site of asteltoxin is localized in the energy transfer system of mitochondria. Asteltoxin strongly depressed Mg2+-ATPase activity in mitochondria and only slightly affected Na+, K+-activated ATPase in microsomes at the concentration range for inhibition of mitochondrial respiration.

Molecular cloning of a cDNA for human alpha-L-fucosidase.

A lambda gt11 human hepatoma cDNA expression library was screened with antibodies to human alpha-L-fucosidase, a lysosomal enzyme whose activity is deficient in the human autosomal recessive disease fucosidosis. Three positive clones were identified after screening 9 X 10(6) plaques. One of these was sequenced and found to be spurious, probably representing an out-of-frame cDNA that gave rise to amino acid sequences of unknown length that crossreacted with alpha-L-fucosidase. A second clone, lambda AF3, was isolated which, after establishment in Escherichia coli BNN103, gave rise to a fusion protein of Mr 154,000 containing a human fragment of Mr 40,000 that represented 80% of the mature processed enzyme (Mr 50,000). Southern blot analysis of mouse and human chromosomal DNA confirmed the human origin of insert AF3. The nucleotide sequence of AF3 was determined and colinearity was established between 270 nucleotides and 90 amino acids in alpha-L-fucosidase. AF3 was found to contain 1058 base pairs and to code for 347 amino acids of alpha-L-fucosidase. Four potential glycosylation sites were identified. The frequency of lambda AF3 in the hepatoma library was 0.0018%.

Direct isolation of Yersinia enterocolitica and Yersinia pseudotuberculosis from meat.

Studies were done to determine the usefulness of dilute alkali (KOH) treatment of meat samples for direct isolation of Yersinia enterocolitica and Yersinia pseudotuberculosis, without enrichment. Virulent Y. enterocolitica and Y. pseudotuberculosis in pork contaminated with 10(2), 10(3), and 10(4) cells per g survived the direct KOH treatment and were never recovered by using KOH postenrichment treatment. From 6 (4.8%) of 125 samples of retail ground pork, four biotype 4 serotype O3 and one biotype 3B serotype O3 strains of Y. enterocolitica and one Y. pseudotuberculosis serotype 4b strain were recovered by using direct KOH treatment without enrichment. As these isolations were attained without using enrichment cultural procedures, they represent an important time-saving alternative to simplify and speed isolation of Yersinia spp. from meat.

Effect of simultaneous infusion of a somatostatin analogue, insulin and glucose on serum triglycerides and blood glucose levels in man.

Nine non-diabetic, non-obese, normocholesterolemic normal male subjects with varied triglycerides levels were subjected to a simultaneous infusion test with a synthetic somatostatin analogue [des(Ala1, Gly2)-D-Trp8, D-Asn3, 14-somatostatin], insulin and glucose under ambulatory conditions. The levels of C-peptide reactivity, immunoreactive glucagon and growth hormone were reduced, and the level of immunoreactive insulin remained constant during the infusion. The blood glucose reached a constant value at 110-120 minutes (steady state blood glucose, SSBG) after the commencement of the infusion. The total cholesterol (TC) levels decreased slightly in the 30 minutes after the experiments were begun, and the triglycerides (TG) levels decreased gradually throughout the infusion period, due mainly to the reduction of very low density lipoprotein (VLDL). The most striking finding was the highly significant positive correlation (p less than 0.005, r = 0.868) between SSBG and the serum TG level prior to the infusion. These results indicate an important relationship between insulin sensitivity and serum TG level. High TG level may be regarded as one of the indices of insulin resistance.

Epidemiological study of Yersinia enterocolitica and Yersinia pseudotuberculosis infections in Shimane Prefecture, Japan.

In Shimane Prefecture, Japan, 175 Yersinia enterocolitica isolates from 6,479 stools and 347 appendixes of patients were separated into biotype 3B (VP-, sorbose-, inositol-) serotype O3 phage type II (22 isolates); biotype 4 (84 isolates), biotype 4 (ornithine-) (16 isolates) and biotype 4 (maltose-) (9 isolates) of serotype O3 phage type VIII, biotype 2 serotype O9 (1 isolate) and biotype 1 (43 isolates). This may be the first documentation of isolation of Y. enterocolitica biotype 3B serotype O3 phage type II from patients. Twenty-three Yersinia pseudotuberculosis isolates from stools of 23 patients were grouped into serotypes IB (13 isolates), III (2 isolates) and IVB (8 isolates). Clinical manifestations were more numerous in Y. pseudotuberculosis infections than Y. enterocolitica O3 infections. Y. enterocolitica O3 was more frequently isolated between summer and autumn but Y. pseudotuberculosis and Y. enterocolitica biotype 1 were isolated between winter and spring. Secondary infection with Yersinia among close family members was frequent. Y. enterocolitica O3 and Y. pseudotuberculosis infected persons excreted 10(4) to 10(9) viable cells per g stool during 27 and 19 days, but Y. enterocolitica biotype 1 strains were detected only when using enrichment techniques. Serological responses were not observed in under 2 year old persons infected with Y. enterocolitica O3.

The inhibition of mitochondrial respiration by indomethacin, a non-steroid anti-inflammatory agent possessing inhibitory effect on prostaglandin biosynthesis.

It was found that indomethacin, a potent inhibitor to cyclo-oxygenase in prostaglandin biosynthesis system, disturbed mitochondrial respiration and its farnesyl ester, E-0710 which no longer exhibited the inhibitory effect on cyclo-oxygenase, did not affect the mitochondrial respiration, indicating a good correlation between the inhibitory effect on ATP biosynthesis in mitochondria and the anti-inflammatory activity of indomethacin.

A rare suicidal case of a ten-year-old child stabbing himself in the throat.

A rare case is reported of a ten-year-old boy who committed suicide by stabbing himself in the throat with a pointed knife. Possibility of an accidental injury was excluded by the autopsy findings; suspicion of a homicide by his father was cleared through the deposition of his sister. The reason of suicide was attributed directly to his father's severe scolding and indirectly to his mother's death two years before.

Presence of zoonotic pathogens (Yersinia spp., Campylobacter jejuni, Salmonella spp., and Leptospira spp.) simultaneously in dogs and cats.

The purpose of this study was to determine the presence of zoonotic pathogens simultaneously in animals. The isolation of human pathogenic Yersinia enterocolitica (Ye), Yersinia pseudotuberculosis (Yp), Campylobacter jejuni (Cj), Salmonella spp. (Sal) and Leptospira spp. (Lep) in 318 cats and 252 dogs were performed in Shimane Prefecture, Japan. A total of 13 isolates of Yp (4 strains) and Sal (9 strains) were recovered from intestine and/or mesenteric lymph nodes (MLN) of 13 cats (4.1%) but not Ye, and Cj was not examined. A total of 88 isolates of Ye (15 dogs, 15 strains), Yp (16 dogs, 16 strains), Cj (10 dogs, 13 strains) and Sal (39 dogs, 44 strains) were recovered from intestine and/or MLN of 76 dogs (30.2%). Two species of Ye O3, Cj and Sal were recovered from either intestine or MLN of 4 dogs but not from cats. Lep was not detected in dogs and cats kidney. The 101 isolates from dogs and cats belonged to Ye 3B/O3/II (biotype/serotype/phage type) (2 strains), 4/O3/VIII (10 strains) and 2/O5,27 (3 strains), Yp serotypes 1b, 2b (each 3 strains), 2c (2 strains), 4a (2 strains), 4b (4 strains), 5a (5 strains) and 7 (1 strain), Cj serotypes TCK 9, 13, 26 (each 1 strain), 21 (2 strains) and untypable (8 strains) and 24 serotypes of Sal. Ye O3 and Yp were detected frequently in cold months. There was no definite seasonal variation of Ye O5,27 Cj and Sal in internal origin of dogs and cats. Ye O3, Yp and Cj were counted at 10(2) to 10(7) cells per g of the jejunal-to-rectal contents, but Sal at less than 10(2) cells per g of the intestinal contents. Ye O3, Yp and Sal were recovered from mesenteric lymph nodes, but not Ye O5,27 and Cj.

[A case of iodide myxedema observed for 3 years under a low iodide diet--especially on the restoration of the mechanism of escape from the Wolff-Chaikoff effect].

A 47-year-old housewife was admitted to our hospital because of general fatigue and constipation suggesting hypothyroidism. For 3 years before admission, general fatigue, arrhythmia, dry skin, drowsiness, cold intolerance and hypermenorrhea occurred insidiously. She had habitually taken considerable amounts of seaweed every day, e.g. more than 50 g of " Kombu " for more than 5 years and at least 1 g of " Wakame " for 6 months. On admission, serum thyroxine (T4) was 1.3 micrograms/dl, serum triiodothyronine (T3) was 47 ng/dl, TSH was 132 microunits/ml, and 123I thyroidal uptake was 60% at 3 hr. and 75% at 24 hr. Anti-thyroglobulin hemagglutination antibodies and anti-thyroid microsomal hemagglutination antibodies were both negative. When seaweed was omitted from her diet, T4 rose to 6.3 micrograms/dl and T3 rose to 113 ng/dl, whereas TSH lowered to 11 microunits/ml in 2 weeks. The seaweed-free diet was continued and 4 months later, when she had become euthyroid, an open biopsy of the thyroid gland was carried out. Histological examination of the specimen revealed a marked colloid deposition without characteristic features of Hashimoto's disease. Five months after admission, with the daily administration of 100 mg potassium iodide (KI), the effects of inorganic iodide on thyroid function had begun to be seen. On the 16th day of the KI regimen, palpitation and tachycardia (pulse rate 160/min.) with multifocal ventricular premature beat appeared, and T4 on the 11th day was 5.9 micrograms/dl, which was clearly lower than the pretreatment level of 8.4 micrograms/dl. KI was discontinued on the 16th day, and one week after the withdrawal, T4, T3 and TSH all returned to the pretreatment level. For more than 3 years on a seaweed-free diet, she remained euthyroid without any thyroid regimen. To see the effects of inorganic iodide on thyroid function after this long period on a seaweed-free diet, KI was again administered. One hundred mg/day KI for 14 days followed by 200 mg/day for 21 days had virtually no effect on T4, T3 and free T4 and she remained well. None of the perchlorate discharge tests performed on 3 occasions during the 6 month period after the initiation of the seaweed-free diet showed a discharge.(ABSTRACT TRUNCATED AT 400 WORDS)

A dissecting aneurysm of the vertebral artery.

A rare case of dissecting aneurysm of the vertebral artery evidenced by ischemic attacks is reported. The aneurysm was treated by proximal ligation of the artery. There have been no repeated ischemic attacks and bleeding for 20 months. Analysis of 51 cases of vertebral artery aneurysm failed to disclose any comparable case. A review of the literature also revealed that clinically diagnosed dissecting aneurysms of the vertebrobasilar system have been reported rarely. Most of these cases were found at autopsy.

Bone resorption in osteogenic sarcoma--II. Resorption of the bone collagenous matrix by tumor cells, normal fibroblasts and macrophages.

Three osteogenic sarcoma cell lines of human and canine origin were compared to normal fibroblastic cells and peptone-induced murine peritoneal macrophages in terms of bone collagenous matrix (BCM) resorption capacity. The dissolution of the BCM was measured in an in vitro system consisting of the tested cells and live or killed [3H]-proline-labeled fetal mouse long bones. Experiments with osteogenic sarcoma cells revealed a paradoxical phenomenon indicating an inverse relationship between the number of tumor cells and the rate of collagen resorption from live bones. On the other hand, collagen matrix of devitalized bones, particularly those denatured by exposure to heat, is strongly resorbed by osteogenic sarcoma cells even in the presence of serum. Contrary to osteogenic sarcoma cells, normal fibroblasts do not resorb collagen from either live or killed bones, regardless of the devitalization method and culture conditions utilized. Macrophages resorb collagenous matrix from live bones, but their collagen resorption activity from devitalized bones depends greatly on choice of the incubation condition. Results have shown that osteogenic sarcoma tumor cells, when acting alone, may not have the capacity to destroy healthy bone. We suggest, therefore, that bone destruction seen in osteogenic sarcoma patients depends on the metabolic condition of the affected bone, and the interaction between the tumor and normal host cells and tissues.

Studies on Tetrahymena microsomal electron transport systems: solubilization of microsomal electron transport enzymes involved in fatty acid desaturation.

Tetrahymena microsomes were solubilized with five different detergents and the effect on electron transport enzymes involved in fatty acid desaturation was studied. Cytochrome b560ms and NADPH-cytochrome c reductase were solubilized with a low concentration detergent (0.25%), in the order of sodium deoxycholate greater than Renex 690 greater than Triton X-100 greater than octylglucoside greater than sodium cholate, whereas all of these detergents at the high concentration (1%) could solubilize preferentially both enzymes (70-100%). Increasing the concentration of various detergents from 0.5 to 1.0% did not produce an incremental change in NADH-ferricyanide reductase solubilization. NADH-cytochrome c reductase system, which would be catalyzed by the cooperation action of NADH-ferricyanide and cytochrome b560ms, was relatively inactivated by all detergents. Compared to the other four detergents, octylglucoside has a much higher recovery of stearoyl-CoA desaturase activities in the supernatant. Our study suggests that octylglucoside may be more useful for the isolation in active form of cyanide-sensitive factor (CSF) from Tetrahymena microsomes.

Ecological studies of Yersinia enterocolitica. II. Experimental infection with Y. enterocolitica in pigs.

Fattening pigs were inoculated with the human pathogenic strains of Yersinia enterocolitica, biovar 4 serovar 3 phagovar 8 and biovar 2 serovar 5.27. Each pig received 2.6 X 10(9) organisms by catheter into the stomach and thereafter 10% sodium bicarbonate solution, by the same route. The serovar 3 strain became established in the intestines of four out of six 11-week-old pigs and in all three 24-week-old pigs. Serovar 5.27 established in the intestines of all four 10-week-old pigs. In these pigs, both serovars were excreted in the feces at 10(2)-10(6) cells g-1 for a few weeks. However, the serovar 5.27 was excreted in the feces at less than 10(3.3) cells g-1 for 2 weeks by 24-week-old pigs. Horizontal transmission with the serovar 5.27 strain was observed on the 5th-11th day, and the bacteria were present in the intestines during the 2nd week. However, the serum titers to O-agglutinin were 1/10 and less, but in one pig, the titer was 1/40 against serovar 5.27 strain.

Ecological studies of Yersinia enterocolitica. III. Cross-protection against fecal excretion between Y. enterocolitica serovars 3 and 5.27 in pigs.

Yersinia enterocolitica biovar 4 serovar 3 and biovar 2 serovar 5.27 failed to establish in the intestines of pigs when challenged with the homologous or heterologous strains. After inoculation, the serum O-agglutinin titers were 1/10 or less and were not boosted by challenge with either serovar. Y. enterocolitica were not recovered in any specimens at the time of slaughter.

Significance of milk as a possible source of infection for human yersiniosis. I. Incidence of Yersinia organisms in raw milk in Shimane Prefecture, Japan.

Twelve strains of Yersinia enterocolitica and 4 strains of a Y. enterocolitica-like organism (Y. intermedia) were isolated from 374 pooled raw milk samples. Growth occurred in a cold enrichment system using phosphate buffer solution (7 strains), peptone solution (5 strains) and yeast extract-casein-cystine broth (5 strains). Serovar 13,7 was most frequently isolated (8 strains) followed by serovars 5,27 (2 strains), 6,31, 7,8, 14 and 22 (each one strain). Two strains were untypable, serologically. Seasonal variation was of little consequence, but the regional incidence was an important feature of the bacteriological findings. Autoagglutination tests gave negative results with all isolates. Although so-called "clinical strains" were not identified, the relationship of the strains of Yersinia to milk-borne infections in humans cannot be excluded.
