Molecular aspects of erythroenzymopathies associated with hereditary hemolytic anemia.

Since the discovery of glucose 6-phosphate dehydrogenase (G6PD) and of pyruvate kinase deficiencies, erythroenzymopathies associated with hereditary hemolytic anemia have been extensively investigated. Kinetic and electrophoretic studies have shown that most, if not all, erythroenzymopathies are caused by the production of a mutant enzyme. Except for a few enzymes that are abundant in blood and tissues, it is difficult to obtain enough sample to study the functional and structural abnormalities of mutant enzymes associated with genetic disorders in man. The primary structures of only two normal red cell enzymes which can cause hereditary hemolytic anemia, phosphoglycerate kinase (PGK) and adenylate kinase, have been determined. Single amino acid substitutions of PGK variants have been found, and the identification of the exact molecular abnormalities of such variants has helped us to understand the accompanying functional abnormality. Gene cloning makes possible the identification of the DNA sequence that codes for enzyme proteins. Recently, human complementary DNA (cDNA) for aldolase, PGK, G6PD, and adenosine deaminase (ADA) have been isolated, and the nucleotide sequences for PGK and ADA determined. In the near future, human cDNA sequencing should permit identification of the gene alteration that gives rise to the mutant enzymes.

One- and two-dimensional EPR imaging studies on phantoms and plant specimens.

The advantages of electron paramagnetic resonance (EPR) imaging at L-band frequencies are discussed. The construction and calibration of a low-field L-band EPR imaging spectrometer is described with capillary phantoms containing aqueous nitroxides as paramagnetic imaging agents. The peak separation induced by the magnetic field gradient is related to the object separation. The linewidth of each (first-derivative) line is used to calculate the dimensions of each paramagnetic object, which is the sum of an intrinsic and a gradient-induced component. By extrapolating the linewidth back to zero field gradient, one obtains the intrinsic linewidth correction factor in computing image size. The nonuniformity caused by deterioration of biological substructure was examined with plant stems where one capillary vessel had become leaky. Spin-label destruction, particularly by biological reducing agents, was compared for three species of plant stems.

The bursa of Fabricius as a trapping mechanism for environmental antigens.

The bursa of Fabricius was isolated from gut-derived antigens by ligating the bursal duct on the 18th day of incubation. Ligation of the bursal duct (BDL) suppressed the bursal development, spontaneous germinal centre formation in the spleen and the development of serum "natural" agglutinins for bacteria or heteroerythrocytes. Moreover, administration of sterilized cecal contents into the bursal lumen at BDL led to normal level of serum "natural" agglutinins. These findings strongly suggest that the bursa of Fabricius possesses an antigen-dependent process which modulates the development of immune system. Antigen-trapping mechanism of the bursa may supply information about environmental antigens for this process.

A maximum entropy criterion of filtering and coding for stationary autoregressive signals: its physical interpretations and suggestions for its application to neural information transmission.

The operations of encoding and decoding in communication agree with filtering operations of convolution and deconvolution for Gaussian signal processing. In an analogy with power transmission in thermodynamics, an autoregressive model of information transmission is proposed for representing a continuous communication system which requires a pair of an internal noise source and a signal source to encode or decode a message. In this model transinformation (informational entropy) equals the increase in stationary nonequilibrium organization formed through the amplification of white noise by a positive feedback system. The channel capacity is finite due to the existence of inherent noise in the system. The maximum entropy criterion in information dynamics corresponds to the 2nd law of thermodynamics. If the process is stationary, the communication system is invertible, and has the maximum efficiency of transformation. The total variation in informational entropy is zero in the cycle of the invertible system, while in the noninvertible system the entropy of decoding is less than that of encoding. A noisy autoregressive coding which maximizes transinformation is optimum, but is also ideal.

The fundamental factor determining the clinical prognosis of prolonged jaundice--relation to the arterial ketone body ratio.

Factors related to the prognosis of patients with hyperbilirubinemia were investigated in 16 highly jaundiced patients. Patients who died within 3 weeks showed a deterioration of the hepatic energy status, measured by the arterial ketone body ratio, but patients who did not die within 3 weeks after the measurement of the ketone body ratio had a ratio within the normal range, despite high total bilirubin levels (18 mg/0.1 L). C3, C4 and CH50 in the former were also significantly lower than those in the latter. However, endotoxin and high fever occurred to the same extent, in the both groups. Thus, hepatic energy balance is the most pertinent factor related to prognosis, and is concerned with both the hepatocyte and reticuloendothelial systems. On the other hand, infection or endotoxin, when the energy balance is disturbed, becomes an aggravating but not a fundamental factor.

Microsomal monooxygenase system in Morris hepatoma: purification and characterization of cytochromes P-450 from Morris hepatoma 5123D of 3-methylcholanthrene-treated rats.

Two forms of cytochrome P-450 (hepatoma P-450MCI and P-450MCII) were purified from hepatoma 5123D microsomes of tumor-bearing rats treated with 3-methylcholanthrene. Hepatoma P-450MCI had a specific content of 18.4 nmol/mg protein and showed a main protein band with a minimum molecular weight of 56,000 on sodium dodecyl sulfate-polyacrylamide gel. Hepatoma P-450MCII had a specific content of 7.38 nmol/mg protein and a minimum molecular weight of 50,000. The carbon monoxide-reduced difference spectral peak of hepatoma P-450MCI was at 446.5 nm, whereas the peak of hepatoma P-450MCII was at 451 nm. In the reconstituted system, hepatoma P-450MCI catalyzed 3-hydroxylation of benzo[a]pyrene and O-deethylation of 7-ethoxycoumarin, but showed low activities for N-demethylation of benzphetamine and aminopyrine, O-demethylation of p-nitroanisole, and p-hydroxylation of aniline. On the other hand, hepatoma P-450MCII did not catalyze hydroxylation of any of the substrates tested. By Ouchterlony double-diffusion analysis, hepatoma P-450MCI was immunologically indistinguishable from rat liver cytochrome P-450c, but hepatoma P-450MCII was distinct from hepatoma P-450MCI and rat liver cytochrome P-450c. Peptide maps of hepatoma P-450MCI and rat liver cytochrome P-450c after proteolysis with Staphylococcus aureus V8 protease demonstrated the similarity of the two cytochromes P-450.

Phagocytic myeloma cells. Report of a case and review of the literature.

The authors have reviewed 17 cases of the phagocytic multiple myeloma, including seven cases reported in Japan but not cited in the English literature and an additional new case investigated in the authors' laboratory. They would like to add these Japanese cases to the English literature. The salient features of these 17 cases were summarized to have a better understanding of the clinical aspects as well as of the pathogenesis of the phagocytic myeloma cells. The following two clinical points should be noted: The erythrophagocytosis by myeloma cells may be responsible for the hemolytic anemia in multiple myeloma. Myeloma cells were found in the peripheral blood in more cases of this series than expected. The pathogenesis of this phenomenon, related to the cellular origin of plasma cells, may be interpreted differently, as discussed in this report.

Structural study on teichoic acids of Listeria monocytogenes types 4a and 4d.

The chemical compositions of the cell walls obtained from 10 strains (serotypes 1a, 3a, 4a, 4b, 4c, 4d, 4e, 4e, 4f, 6, and 7) of Listeria monocytogenes were analyzed. These cell walls were shown to be mainly composed of peptidoglycan and ribitol teichoic acids. Considerable variations in the composition of neutral sugars were observed among these cell walls. Chemical and NMR analyses indicated that the teichoic acids from L. monocytogenes serotypes 4a and 4d are composed of the following repeating units: Formula: See Text.

Structure of acidic polysaccharide from cell wall of Propionibacterium acnes strain C7.

The structure of polysaccharide prepared by lysozyme digestion from the cell wall of Propionibacterium acnes strain C7 was examined. The polysaccharide fraction was composed of glucose, galactose, mannose, galactosamine, and diaminomannuronic acid in a molar ratio of 1:1:0.3:1:2. By Smith degradation of the polysaccharide, diaminouronic acid-containing fractions were obtained, and the configuration of diaminouronic acid was identified as 2,3-diacetamido-2,3-dideoxymannuronic acid [Man(NAc)2A] by means of 1H-NMR and 13C-NMR spectroscopic analyses. The results of analyses involving methylation and partial acid hydrolysis led to the conclusion that the polysaccharide has the repeating unit----6)Gal(alpha 1----4)Man(NAc)2A(beta 1----6)Glc(alpha 1----4)Man(NAc)2A (beta 1----3)GalNAc(beta 1--. In addition, a portion of the galactose residues were substituted at C-4 by alpha 1----2 linked mannotriose.

Enzyme immunoassay of teichoic acids from Listeria monocytogenes.

Amino groups were introduced into Listeria monocytogenes teichoic acids by reductive amination, and the product was coupled to biotin. Teichoic acids were assayed by their binding to specific antibody adsorbed to a solid phase, followed by detection of the antigen-antibody complex by horseradish peroxidase-avidin. Less than 20 ng of teichoic acid was detectable.

Stimulating effect of natural estrogens on proliferation of hepatocytes in adult mice.

A single oral administration of natural principal estrogens, estrone (E1), 17 beta-estradiol (E2) and estriol (E3), caused active proliferation of hepatocytes of adult mouse liver. Steroid hormones tested (testosterone, progesterone and cortisone) other than estrogens were not stimulants of proliferation of hepatocytes. Among these three natural estrogens, E3 was found to be the most potent stimulant of hepatocyte proliferation and E1 was the weakest. The possible mechanism of the hepatocyte-proliferating potency of estrogens was discussed in close relationship to their stimulating effect on the reticuloendothelial system.

Orbital pseudotumor: histopathological classification and treatment.

43 cases of orbital pseudotumor have been studied clinicopathologically during the past 20 years. Orbital pseudotumor might be histopathologically divided into three principal types: lymphoid, granulomatous and sclerosing types. Lymphoid and granulomatous types might be transformed into sclerosing type in the end stage, respectively. Lymphoid type showed good response to radiotherapy and poor or temporary response to steroid therapy. Granulomatous type indicated frequently good response to steroid and poor response to radiotherapy. Sclerosing type demonstrated poor response to steroid and radiation therapy. Therefore, orbital pseudotumor must be treated at an early stage.
