Application of flow cytometry for rapid detection of Lactococcus garvieae.

Flow cytometry (FCM) technique was applied to rapid determination of cell number of Lactococcus garvieae. An antiserum against L. garvieae was prepared and its immunological property was examined. The present antibody would recognize some epitopes of L. garvieae with a high specificity. The optimum conditions for the FCM assay were as follows: discriminate value, 60; dilution ratio of the antiserum, 1.0 x 10(4). Calibration curve for L. garvieae cells was linear, in the range of 2.4 x 10(4)-1.5 x 10(7) cells/mL. The detection of L. garvieae in cell suspensions contaminated with Escherichia coli was carried out. A good correlation was observed in the range of 20-90% for the mixing ratio of L. garvieae. One FCM assay could be completed within 2 min, and the total assay time, including the preparation of bacterial sample, was within 3 h.

Hepatic artery embolization for postoperative hemorrhage in upper abdominal surgery.

BACKGROUND: The purpose of this study was to evaluate angiographic findings and radiologic and clinical results of transcatheter arterial embolization (TAE) of the hepatic artery for hemorrhage after upper abdominal surgery. METHODS: Ten patients (nine male, one female, mean age = 63.5 years) with postoperative hemorrhage underwent emergency hepatic artery embolization. We retrospectively analyzed the angiographic findings and the effectiveness of TAE. RESULTS: Angiography demonstrated pseudoaneurysms (n = 6) and extravasation (n = 5). Portal veins were not compromised in any patient. The bleeding points were at the gastroduodenal artery (n = 3), proper hepatic artery (n = 3), right hepatic artery (n = 2), left hepatic artery (n = 1), and intrahepatic artery (n = 2). Embolization was performed at the extrahepatic site (n = 6) and intrahepatic site (n = 4). TAE was successful in eight patients. Hemostasis was achieved in eight patients. Two patients in whom TAE was not successful died of bleeding after TAE. In six patients whose collateral arteries were visualized at TAE, hypoxic hepatic failure was avoided and the patients had a favorable clinical course. CONCLUSION: TAE is a useful treatment for postoperative hemorrhage, but the presence of collateral arteries at the time of TAE can affect the patients' prognoses.

A molecular hybridization study for the presence of Herpes simplex, cytomegalovirus and Epstein-Barr virus in brain and blood of Alzheimer's disease patients.

Among several hypothesis for the development of Alzheimer's disease is a viral hypothesis. The present study was designed to detect nucleic acid sequences for conventional viruses in peripheral blood cells and brain of Alzheimer's disease patients. DNA was isolated from peripheral blood cells and brain tissue from control individuals and Alzheimer's disease patients. Southern blot analysis was performed using radiolabeled probes for various conventional viruses. The results fail to detect the presence of Herpes simplex 1 (HSV-I), Herpes simplex II (HSV-II), Epstein-Barr virus (EBV) and cytomegalo virus (CMV) at a sensitivity level of detecting 1 genome/100 cells. We exclude conventional viruses as a cause of Alzheimer's disease at this level of detection.

Determination of phosphate ions with an enzyme sensor system.

An enzyme sensor system for the determination of phosphate ions was constructed using immobilized enzymes and an oxygen electrode. The principle of this method is based upon the nucleoside phosphorylase catalyzed reaction for which the presence of inorganic phosphorus is indispensable. One assay could be completed within 3 min. This enzyme sensor was able to withstand at least 70 assays. This system was applicable to simple, rapid and continuous determination of phosphate ions in food.

Anatomical and histological characterization of the female reproductive organs of mouse deer (Tragulidae).

The Tragulidae may be a type that represents the earliest ruminant families to evolve. The female reproductive organs including ovary, oviduct, uterus and placenta were anatomically and histologically investigated. The structures of the ovary and oviduct were very similar to other ruminants. However, the gross structure of the placenta was diffuse and thus noticeably different from other ruminants which are polycotyledonary. Histologically, the placenta of Tragulidae appears to be epitheliochorial and therefore similar to other ruminants. Numerous trophoblastic binucleated cells which are characteristic of all other ruminants were observed. These results suggest that the placenta of Tragulidae is a transitional type between diffuse epitheliochorial and polycotyledonary synepitheliochorial categories.

IGFBP1 and Follistatin-like 3 genes are significantly up-regulated in expression profiles of the IUGR placenta.

To date, the clinicopathological features of intrauterine growth restriction (IUGR) are not clearly understood, and no effective therapy has been established for IUGR. This is the first study that uses microarray analysis to identify differentially expressed genes in the IUGR placenta. The expression profiles of a total of 9121 genes were examined by cDNA microarray analysis, using mRNA from an appropriate gestational age (AGA) placenta and an IUGR placenta from discordant dichorionic twins. Up-regulation of the IGFBP1 and Follistatin-like 3 genes was detected in the IUGR placenta, with a balanced differential degree of 20.7+/-1.3 and 13.1+/-2.1, respectively, while the balanced differential degrees of other genes were 2.6 or less. The expressions of the IGFBP1 and Follistatin-like 3 genes in four single IUGR and four AGA placentas were also examined by RT-PCR. Consistent with our data in discordant chorionic twin placentas, three of four IUGR placentas showed up-regulation of the IGFBP1 and all four IUGR placentas showed upregulation of Follistatin-like 3 genes when compared to the AGA placentas. Our results suggest that IGFBP1 and Follistatin-like 3 are highly up-regulated in IUGR in the placenta. IGFBP1 and Follistatin-like 3 are known critical regulators of fetal growth and differentiation. Pathways associated with these genes might be important for the pathogenesis of IUGR.

The successful treatment of diffuse neonatal hemangiomatosis with flashlamp pulsed dye laser.

Diffuse neonatal hemangiomatosis is a rare disease with the distinctive features of multiple hemangiomas of the skin and visceral organs. These lesions have been treated with systemic corticosteroids, interferon-alpha, and their combination. We report a patient with diffuse neonatal hemangiomatosis who had multiple cutaneous and hepatic hemangiomas. Single therapy with the flashlamp pulsed dye laser was effective for cutaneous hemangiomas, whereas the hemangiomas of the liver remained stable and no liver dysfunction or hemorrhage has occurred so far, even with no treatment.

Effects of intragastric ammonia on collagen metabolism of gastric ulcer base in rats.

We studied the chronic effect of ammonia, which is one of the pathogenic factors of Helicobacter pylori in gastroduodenal diseases, on the healing of acetic acid-induced gastric ulcers in rats with special reference to the collagen metabolism. Rats were given either tap water or 0.1 or 0.02% ammonia solution before and after ulcer induction. We measured the ulcer index, hydroxyproline concentrations and type III collagen/total hydroxyproline ratios in the ulcer base. Epithelialization was achieved 4 weeks later under the influence of ammonia. Nevertheless, ammonia treatment significantly increased the collagen deposition in the ulcer base compared with the nonammonia treatment group, 4 weeks after ulcer induction. In the nonammonia treatment group, the type III collagen/total hydroxyproline ratio decreased 3 weeks after ulcer induction. On the contrary, in the groups treated with ammonia throughout the experiment, type III collagen/total hydroxyproline ratio remained high until 4 weeks after ulcer induction. In conclusion, intragastric ammonia delayed the ulcer healing process in its early phase, but did not affect the achievement of re-epithelialization. However, it disturbed the collagen metabolism in the ulcer base. Excessive deposition of collagen and an abnormally high proportion of its immature type may be unfavorable for ulcer healing. These findings may explain in part why H. pylori-induced ulcer is so difficult to treat.

Antitumor effect of biological response modifier, PSK, on C57BL/6 mice with syngeneic melanoma B16 and its mode of action.

The effects of PSK, a protein-bound polysaccharide, on the survival period and effector cell activity were examined using C57BL/6 mice with melanoma B16. PSK prolonged the survival period of the mice with tumors in a schedule- and dose-dependent manner. However, no life-prolonging effect was observed when carrageenan-treated mice or congenitally athymic mice were used as hosts. PSK enhanced the cytostatic activity and interleukin-1-producing capacity of peritoneal exudate plastic-adherent cells in C57BL/6 mice with tumors. These findings suggested that PSK prolongs the survival period of mice with B16 tumors through T-cell- and macrophage-dependent mechanisms.
