[The significance of p16, cyclin D1 and pRb expression in soft tissue leiomyosarcoma].

OBJECTIVE: To evaluate the significance of p16, cyclin D1 and pRb expression in human soft tissue leiomyosarcoma (LMS) and the relationship among these three proteins. METHODS: 42 cases of LMS were studied for p16, cyclin D1 and pRb expression with immunohistochemical method. RESULTS: The total abnormality percentage of p16, cyclin D1 and pRb expression in all 42 cases of LMS was 97.6%. The negative rate of p16 and pRb protein expression was 35.7% and 47.6% respectively, and the overexpression rate of cyclin D1 was 57.1%. Overexpression of cyclin D1 was also noticed in 55% leiomyomas. In the 20 cases of LMS which were pRb negative, 19 cases were moderately or strongly positive and one faintly positive for p16. Contrastly, in the 15 cases of LMS negative to p16, 12 cases were moderately or strongly positive and 3 cases faintly positive for pRb. Higher rates of loss of pRb protein were observed in the low-differentiated group than in the high-differentiated one. CONCLUSIONS: (1) The abnormality of p16-cyclin-pRb pathway is highly related to the pathogenesis of LMS. (2) Loss of p16 or pRb and/or overexpression of cyclin D1 may be involved in the pathogenesis of LMS. (3) There is a negative reciprocity between p16 and pRb protein expression in LMS. (4) Overexpression of cyclin D1 is a common molecular alteration that may occur in the early stage. (5) Expression of pRb is inversely related to the differentiation degree of LMS. (6) The expression of the 3 proteins studied has no relation to tumor location, recurrence, invasion or metastasis.

[Expression and distribution of human blood type antigens in Chinese Neijiang pig tissues].

OBJECTIVE: The expression and distribution of human blood type A, B, O antigens in Chinese Neijiang porcine were investigated. METHODS: The anti-human blood type A, B, H monoclonal antibodies were used as the primary antibody, and the immunohistochemistry EnVision technique was used to detect the expression of A, B, H antigens in tissues from 10 pigs. RESULTS: The B and H antigens were not found in all 10 pigs' tissues investigated. While the A antigen was found in all above tissues except vascular endothelial cells. CONCLUSION: The A antigen did not exist on vascular endothelial cells, so it did not play a role as a hyperactive antigens in pig to human xenotransplantation. However, in other tissues it might play roles in acute cellular rejection or chronic rejection. The future study on its role in pig to human xenotransplantation was needed. The expression of human blood type antigens in Neijiang pigs was different from other pig strains reported, it was suggested that it was possible to find a pig strain that contained less xenoantigen.

[Expression of vascular endothelial growth factor in brain astrocytoma and its clinical evaluation].

Angiogenesis is often involved in tumor growth. Neovascularization plays an important role in the prognosis of patients with brain astrocytoma. Vascular endothelial growth factor (VEGF) has been known as the potent angiogenetic factor and mitogen. To gain a deep insight into the possible relation between expression of VEGF and pathological characterization/prognosis in brain astrocytoma, the authors investigated 107 cases of brain astrocytoma and 25 cases of normal brain, tissue via a combination of immunohistochemical staining of paraffin-embedded tissue and follow-up survey. The results showed that the positive staining rate and PU (positive unit) were obviously higher in astrocytoma than that in normal brain (86.92% vs 16.00%, P < 0.05), that the value of the positive staining rate and PU in astrocytoma corresponded closely with its pathological grade and stage (P < 0.01), and that the survival rates (6, 12, 24 months) for the patients suffering from astrocytoma gradually decreased when the VEGF positive rate increased. These suggest that the expression of VEGF may have close relationship with brain astrocytoma and the measurement of the VEGF expression may provide another potential standard for judgement about pathological grade/stage and clinical prognosis in brain astrocytoma.

[The RYR1 genotype of the pigs from Neijiang of China].

The aim of this study was to test the RYR1 genotype of 25 Neijiang pigs ao as to provide data for the selective breeding of medical experimental pig strains. PCR-RFLP technique was used. The results showed that the RYR1 genotype of all the 25 porcine cases was HALNN and all of the 25 porcine cases were Halothane Negative. These suggested that Neijiang pig could be one of the porcine strains selectively bred for medical experiments and clinically for pig-to-human xenotransplantation. PCR-RFLP technique is a reliable and simple method for RYR1 gene testing.

[Changes of F-actin in neutrophils under fluid shear stress].

Neutrophils are the main defender in the body, and their physiological characters are associated with functions. Because the environments of neutrophils maturation and storage are different from their function environment, they have to adapt thess changes. Filamentous actin (F-actin), as one of the important cytoskeleton components of neutrophils, has different quantity and distribution under different physiological conditions. Previous researches are all focused on static activation of neutrophils using various stimulants such as formyl-methionyl-leucyl-phenylalanine (f-MLP) and tumor necrosis factor(TNF). In our study, Low-Shear 30 and NXE-1 rheometers were used to provide steady or sinusoidaloscillatory fluid shear stress on separated neutrophils. Fluorescent agent Tritc-Phalloidin was added to label F-actin and the mean fluorescent intensity tested by flow cytometry was used as the indication of F-actin quantity and confocal laser scan microscope was used to detect the distribution of F-actin in neutrophils. We found that F-actin polymerization was significantly decreased under both steady and sinusoidal-oscillatory shear stress when the shear stress was below certain level. The density of the cortex F-actin near cell membrane in cells sheared became thinner than in that without shear stress. f-MLP and TNF both could increase actin polymerization in neutophils due to activation. When neutorphils were activated with f-MLP or TNF under shear stress, the quantity of F-actin in the cells was also decreased, but it was still higher than that in the cells absent of f-MLP or TNF. We concluded that neutrophils had different mechanical properties in different fluid environments. F-actin content and distribution would change according to the shear stress values to play their determinant role in neutrophils mechanical adaptation. The mechanism of actin polymerization in neutrophils for mechanical action is probably different from that for chemoattractants and cytokine activation.

[Reactivity of tissues of chinese NJZ pig to human serum].

Insufficient supply of organ for allotransplantation made the study on finding new organ resources from animal progress. Pig is regarded as one of the optimal donor animals for human. The major obstacle in this field is hyperacute reaction (HAR), which is triggered after the xenogenic natural antibodies preexisting in recipient blood combine to the antigens on the surface of the endothelium and activate the complement system. alpha-Galactose residues (alpha-Gal) on the endothelial cell have been identified as the major xenoantigens. NJZ Pig has been closely breed since 1938, whose family history is clear. Tissue samples from heart, liver, kidney, pancreas, lung, small intestine, skin, spleen, thymus and lymph node were obtained and embedded in paraffin. The sections were performed the immunohistochemical staining with the sera from health volunteers (including all the blood types) as the primary antibodies as well as the biotin labeled bandeirae simplicifolia I isolectin B4 (BS I-B4), which has specific affinity to alpha-galactose. All the staining sections were compared with the tissues digested with alpha-galactosidase. There was no difference between the antigens recognized by sera of different blood types. alpha-Gal was still the major xenoantigen on the endothelial cells. There might exist non-alpha-Gal antigens on the distal convoluted tubules and collecting tubules of the kidney. There was no alpha-Gal distributing on the secreting part of pancreas, either the islet cells or the matrix cells, but surely on pancreatic duct and vessels. All the antigenity was destroyed after the enzyme digestion except that the small intestine gland still positive with the BS I-B4. alpha-Gal is the major xenogenic antigen in NJZ Pigs. There exist some unknown antigens on the distal convoluted tubules and collecting ducts of the kidney. The blood type of recipient is not the first affair to be considered in pig-to-human xenotransplantation. The specificity of BS I-B4 for the alpha-galactose needs more detail research.

[Surface phenotype on dendritic cells and their immunological significance].

Dendritic cells (DC) are very important antigen presenting cells(APC) in immune response. DC gradually gets mature while it migrates in vivo. Compared with mature DC, immature DC plays a different role in immune response. In this review, the recent advances in researches on specific cell surface marker and costimulatory molecules on DC as well as their immunological significance and methodology of propagation in vitro are discussed. It may be beneficial for further study of DC.

[Determination of trace Al in serum of cancer patients by graphite furnace atomic absorption spectrometry using K2Cr2O7 and surfactant as matrix modifier with molybdenate coated tubes].

Different kinds of graphite tubes and of matrix modifiers have been tested for optimization of operation condition for determination of Al in serum by GFAAS. The effects of different interference ions have been eliminated by adding K2Cr2O7 and Triton X-100 as matrix modifier. The method is suitable for determination of serum of cancer patients. The characteristic mass is 21.15pg/0.0044 A x S. The recovery rate is 104.92. The relative standard deviation is less than 8.17%. The result obtained is satisfactory.

[Laboratory diagnosis of tuberculous peritonitis].

OBJECTIVE: To evaluate the diagnostic value of polymerase chain reaction (PCR) assay in combination with Southern hybridization technique and enzyme-linked immunosorbent assay (ELISA) in tuberculous peritonitis. METHODS: Fourty-two tuberculous ascites were detected for M. tuberculosis by PCR technique in combination with Southern hybridization, in which nuclear acid probe was digoxigenin-labeled, and the result was compared with conventional bacteriological methods and ELISA. IS6110, a specific insert sequence of M. tuberculosis, was used as primer. The specificity of PCR products was confirmed by hybridization and digestion with Sal I restrictive endonuclerase, and the sensitivity of hybridization was compared with gel electrophoresis. RESULTS: The sensitivities were 69% by PCR, 71% by ELISA, 9% by culture and 0 by acid fast staining respectively. Hybridization was found more sensitive than gel electrophoresis. CONCLUSIONS: Both PCR technique and ELISA are valuable in diagnosis of tuberculous peritonitis, and the former is more specific than the latter. PCR technique in combination with Southern hybridization can improve both the sensitivity and the specificity of dectection.

[Effects of repeated nicotine uptake on brain inositol levels].

In the acute experiments, 30 minutes after nicotine was intraperitoneally injected at the doses of 0.5, 1.0, 1.0, 2.0, 2.0 mg/kg with 5 min interval, the inositol levels were increased in rats cerebral cortex and hippocampus rather than striatum. While the inositol level was decreased in rat cerebral cortex 30 minutes after lithium chloride was given at the dose of 10 mmol/kg. In the chronic experiments, after nicotine at the doses of 2.0-5.0 mg/kg was subcutaneouly injected twice a day for 14 days the inositol level was increased in rat cerebral cortex. In other experiments, nicotine was orally given at the dosese of 2.69, 6.26, 11.53 mg/kg.d for 64 days, the inositol level of rat cerebral cortex was also increased. These indicated that the effects of nicotine on inositol level of rat brain are different from those of lithium chloride, the inositol level of rat cerebral cortex can be increased by repeated administration of nicotine.

[Mycobacterium paratuberculosis in the intestine of patients with Crohn's disease].

Crohn's disease has long been suspected of having a mycobacteria, specifically M. paratuberculosis cause. In this study polymerase chain reaction (PCR) assay was used to detect M. paratuberculosis DNA in 74 paraffin wax embedded gut tissues from endoscopic biopsy or surgical resection specimens, including 36 Crohn's disease tissues, 18 ulcerative colitis tissues and 20 non-inflammatory bowel disease tissues. Oligonucleotides derived from IS900 sequence, which is a repeated in M. paratuberculosis chromosome and highly specific for the M. paratuberculosis, were used as primer. The specificity of products was confirmed by southern blot hybridization by using a biotin-labeled probe. The results show that M. paratuberculosis DNA was detected in 17 Crohn's disease (47.2%), 2 ulcerative colitis (11.1%) and 3 non-inflammatory bowel disease control subjects (15.0%). PCR positive or negative results in 36 Crohn's disease show that there is no relation with small or large gut involvement, and also the presence or absence of granulomata. The results suggest that M. paratuberculosis is surely present in tissues from some patients with Crohn's disease, which may have a specific association with Crohn's disease.

[The recent advances in the study of dendritic cells and induction of donor antigen-specific T cell tolerance].

The induction of donor antigen specific T cell tolerance will be one of the effective treatment for overcoming rejection and improving the alive quality of long-term recipients. It is increasingly noted that dendritic cell may play a two way regulation role in the balance between transplantation rejection and tolerance as the advances of knowledge of dentritic cell maturation stages. This review will focus on the recent advances in the relation between dendritic cell and induction of donor antigen specific tolerance; its mechanism and potential therapeutic application in transplantation.

[Human papillomavirus infection and esophageal squamous cell carcinoma].

Human papillomavirus (HPV) infection, especially high risk types HPV 16 and 18, have been studied widely in cervical cancer. However, HPV infection in esophageal cancer has not been well defined. In the present study, immunohistochemistry, PCR and Southern blot hybridization methods were used to detect HPV infection in 127 cases of esophageal squamous cell carcinoma. Immunohistochemistry results indicated that the virus was detected frequently in well differentiated carcinoma. The positive rates for BPV and HPV E6 protein were 60.6% (77/127) and 43% (54/127) respectively. Meanwhile, PCR and Southern hybridization showed that 35.9% (37/103) of esophageal squamous cell carcinomas have HPV DNA, which included 20.4% (21/103) HPV 16 and 7.8% (8/103) HPV 18. Of the 103 cases, only 1 had both HPV 16 and HPV 18 DNA. Our results suggest that HPV infection is present in esophageal squamous cell carcinoma and may play a role in its pathogenesis.

[The value of polymerase chain reaction assay in the diagnosis of intestinal tuberculosis and differentiation from Crohn's disease].

It is difficult to make a precise diagnosis for intestinal tuberculosis and differentiate it from Crohn's disease. For evaluating the efficacy of PCR assay in these two aspects, 36 specimens of intestinal tuberculosis and 26 of Crohn's disease from surgical resections and endoscopic biopsies were subjected to PCR assay. Oligonucleotides derived from IS 6110 sequence, which is a repeated in M. tuberculosis chromosome and highly specific for the M. tuberculosis complex, were used as a primer. The specificity of PCR products was confirmed by digestion with Sal I restrictive endonuclease and southern blot hybridization by using digoxigenin-labeled probe. The results showed that M. tuberculosis DNA was identified in 27 of the 36 specimens of intestinal tuberculosis, but none in those of 26 Crohn's disease. In conclusion, PCR is a rapid, sensitive and specific method for the diagnosis of intestinal tuberculosis, and it is also valuable in differentiating intestinal tuberculosis from Crohn's disease.

[Tuberculous lymphadenitis: a sarcoidosis-like variation].

By means of polymerase chain reaction technique to amplify DNA sequence specific for mycobacterium tuberculosis (M. TB-PCR), BCG immunohistochemistry (BCG-IHC) as well as acid fast stain (AF), mycobacterium other than tuberculosis (MOTT) and mycobacterium tuberculosis (M. TB) were detected in paraffin blocks from 12 cases diagnosed by consultant pathologists as lymph node "sarcoidosis". Of the 5 cases which consulting pathologists unanimously agreed to the lymph node "sarcoidosis" diagnosis, 1 case showed BCG-IHC (+) and M. TB-PCR (+); Of the 7 cases which most of the consulting pathologists agreed to be "sarcoidosis", 1 case showed AF (+), BCG-IHC (+) and M. TB-PCR (+). The results suggest: (1) some tuberculous lymphadenitis cases may present as "sarcoidosis"-like lesions; (2) lymph node sarcoidosis may be related to MOTT and M. TB infection.

Value of polymerase chain reaction assay in diagnosis of intestinal tuberculosis and differentiation from Crohn's disease.

It is difficult to make a precise diagnosis of intestinal tuberculosis and to differentiate it from Crohn's disease. For evaluating Polymerase Chain Reaction (PCR) assay in these two aspects, 36 specimens of intestinal tuberculosis from surgical resections and endoscopic biopsies and 26 Crohn's disease samples were subjected to PCR assay. 21 specimens of normal colon tissue surrounding cancer were used as the control. Oligonucleotides derived from the IS 6110 sequence, which is repeated in M. tuberculosis chromosome and highly specific for the M. tuberculosis complex, were used as a primer. The amplified PCR products were detected by examination of ethidium-bromide-stained polyacrylamide gels. The specificity of PCR products was confirmed by digestion with Sal I restrictive endonuclease and southern blot hybridization using digoxigenin-labeled probe. The results showed that the M. tuberculosis DNA was identified in 27/36 intestinal tuberculosis, but none of 26 Crohn's disease. Acid fast bacilli were only found in 16/36 intestinal tuberculosis. In conclusion, as a rapid, sensitive, and specific pathogenic method in diagnosis of intestinal tuberculosis, PCR assay has been developed in this study, and is considered valuable in the differentiation between intestinal tuberculosis and Crohn's disease.

[The establishment of ADBS data bank system for autopsy records].

The ADBS data bank system consists of PC 286 microcomputer, (or 386 and other more advanced machine) VGA color monitor, printer and ADBS V. 2.0 software. According to the international classification of diseases (ICD-9 or ICD-10, SNOMED), pathological diagnoses are coded and put into computer. This system has five functions: search, statistics, edition, print and system restore. For facilitating the operation of this system, all orders are listed as window menu. The statistic results can be shown as tables or figures and put out as disk files or prints. The system is currently an advanced computerized system of histopathological and autopsied records in Chinese.
