Some aspects of division and cell density, expression and motility of certain surface structures were studied on lymphocytes of non-Hodgkin's lymphoma of high and low degrees of malignancy. The neoplastic lymphocytes of the two lymphoma types differed in these aspects.
Hydrolysis of glycerol trioleate by human leucocytes was characterized and the enzymes responsible for this activity were obtained in a purified form by means of gel chromatography on Sephadex G-100 as well as by zonal ultracentrifugation followed by gel chromatography. The activity is localized in the granule fraction of leucocytes (15 000 X g, 20 min) and shows a sharp pH optimum at pH 5.25. As judged from the elution profile obtained by gel chromatography, two proteins are likely to contribute to the hydrolysis of glycerol trioleate. The approximate molecular weights of the two enzymes are 74 100 and 60 300, respectively. The activity is reduced in the presence of NaCl, KCl, CaCl2 as well as of p-hydroxymercuribenzoate. The enzymes are stable at -25 degrees C but loose about 50% of their activity within 48 h at 4 degrees C.
In 47 patients with acute lymphoblastic leukemia surface markers were evaluated on mononuclear cells of the peripheral blood as well as in some cases on bone marrow lymphocytes. The lymphocytes were characterized by their binding capacity for sheep red blood cells, the demonstration of Fc-receptors, complement receptors as well as surface immunoglobulins. In 6 of 23 untreated patients the blasts bound sheep red blood cells spontaneously (T-ALL), in two of these six cases the lymphoblasts had simultaneously receptors for complement. In a further patients the lymphoblasts had complement- and Fc-receptors. The blasts of 16 of 23 patients were negative in respect to the markers tested (O-ALL). By comparing two groups of patients--one with positive cells, one unreactive--the clinical features differed: the marker positive group showed a predominance of male patients, 5 of 7 patients had a massive mediastinal mass and the remission rate was lower than in the group with positive blasts. 24 patients in remission under maintance treatment had a decreased percentage of rosette forming lymphocytes as well as lymphocytes with surface immunoglobulins and Fc-receptors. There existed some correlation between the percentage of rosette forming lymphocytes and the clinical course: patients with complications had lower percentages of rosette forming lymphocytes than patients with a favourable course.
An assay of lipolytic activity in human adipose tissue is described, in which native homogenates of the adipose tissue yield the enzyme, as well as the triglyceride substrate, and the emulsifying phospholipids. The lipolytic activity in the presence of serum is characterized mainly as lipoprotein lipase activity by a pH-optimum of 8.0, by the fact that serum is necessary for full activity, and that it is inhibited by 1 M NaCl and by protamine. At serum concentrations of higher than 50% a marked inhibition of the lipolysis is observed. Noradrenaline, insulin, and heparin have no effect on the serum-stimulated lipolytic activity.
The percentage of T- and B-lymphocytes was determined by means of surface markers in 190 patients suffering from various lymphoproliferative disease. Characteristic T-cell lymphomas were diagnosed in 8 patients: 2 of these patients suffered from acute leukaemia, 2 from lymphoblastic lymphoma, 3 from mycosis fungoides and one from the Sézary syndrome. The clinical course of the disease in these patients, the clinical picture and the results obtained with various surface markers and with unspecific mitogens are described and discussed.

Linear density gradients were used to separate either unsensitized human peripheral blood lymphocytes or cells sensitized in mixed leukocyte culture (MLC), at different time points in the immune response. Testing of cells from the various density fractions for their ability to respond in MLC and cell-mediated lympholysis (CML) revealed that (a) activity between individual fractions differed, as well as activity between individual fractions and a suspension of unfractionated cells, (b) although precursor cells of MLC and CML were maximally enriched in closely associated light density regions, their respective distribution profiles were strikingly different, and (c) the density of cells responding in MLC and CML differed depending upon the point of the immune response at which the cells were separated. Whereas effector cells were of light density relative to the precursor cells, putative memory cells for CML exhibited a high density.

Tibric acid [CP-18.524; 2-chloro-5-(3,5-dimethyl-piperidino-sulfonyl-benzoic acid], a new hypolipidemic drug was given to 20 patients with primary endogenous hyperlipoproteinemia of type II, III, IV and V Fredrickson in a daily dose of 1.5 g for 1 year. As compared to a 4-week placebo period, the treatment with tibric acid led to a significant decrease in plasma cholesterol in 6 type-II patients (p less than 0.02) and to a significant decrease in plasma cholesterol and triglycerides in 6 type-III patients (p less than 0.001 and p less than 0.001, resp.) and in 4 type-IV patients (p less than 0.001 and p less than 0.001, resp.). No significant effect of tibric acid on plasma lipids was noted in 4 type-V patients. Apart from a moderate nausea in two patients, no side effects occurred.
Human plasma proteins were isolated by zonal ultracentrifugation and the fractions obtained examined under the electron microscope. HDL2 had a high degree of uniformity in normal persons, characterised by numerous two-dimensional, hexagonal arrangements. Optical diffraction analysis of HDL2 gave a centre distance of 11.3 nm, individual HDL2 particles being spherical with a diameter of 9.8 nm. HDL3 represented a somewhat less uniform lipoprotein population with a particle diameter between 6.5 and 9.5 nm. LDL had a diameter of 20-25 nm. In patients with hyperlipoproteinaemia type III a third peak was isolated, in addition to the two normal lipoprotein peaks VLDL and LDL. It lies between them and has been called Lp III. VLDL from type III plasma consisted of spherical particles, diameter 31-81 nm, morphologically indistinguishable from normal VLDL. Lp III consisted of a relatively homogeneous spherical lipoprotein population with a diameter of 25-30 nm. LDL consisted of spherical particles whose size was highly homogeneous (18-23 nm diameter), indistinguishable from normal LDL.

In 1971 a population-based Ischaemic Heart Disease (IHD) Register was established in the Innsbruck area as part of the WHO international collaborative study. Demographic and geographic data covering the area, as well as the locally-applied methods are described. One hundred and seventy four cases (133 males and 41 females) of acute myocardial infarction (AMI) were registered in the age group 20-64 years. This corresponds to an annual incidence rate of 1.9(0/00) in men and 0.6(0/00) in women. These results confirm indications from the national mortality statistics that Innsbruck belongs to the group of areas in Europe with a relatively low incidence of AMI. The epidemiology of AMI in Europe is discussed. The frequency distribution of AMI according to month of the year, day of the week and hour of the day is reported for this area.
The lipoproteins of very low density, low density and intermediate density were isolated by zonal ultracentrifugation from the plasma of subjects with familial type III hyperlipoproteinaemia and studied by electron microscopy. Each of these lipoproteins exhibited a spherical shape when free-standing. The very low density, low density and intermediate density lipoproteins ranged in diameter from 31 - 80, 17 - 23, .and 21 - 35 nm, respectively. These results are consistent with our previous findings that the intermediate density lipoprotein particles characteristic of plasma from type III subjects exhibit properties which are intermediate between those of very low density and low density lipoproteins.
