A new simplified technique recording readaptation time after photo stress, RAT, is described. The psychometric properties in terms of internal consistency and retest reliability were tested, and the effect on RAT after intake of melperon at two different dose levels was investigated and correlated to blood plasma levels. The results show that there was a satisfactory consistency of RAT at each occasion but stability over a 1 month period could not be demonstrated. Significantly dose-dependent changes were recorded after intake of melperon but the prolongation of RAT was not significantly correlated to blood plasma levels.
In a group of 55 consecutively admitted patients who exclusively abused sedatives or hypnotics, neuropsychological performance was significantly lower and signs of intellectual impairment significantly more often diagnosed than in a matched control group taken from the general population. These results suggest a relationship between abuse of sedatives or hypnotics and cerebral disorder.
A group of 47 patients exclusively abusing sedatives-hypnotics (not alcohol or hard drugs) has been studied with regard to field dependence as assessed by a rod-and-frame test. In comparison to a pairwise matched control group from the general population these sedative-hypnotic abusers were not more field dependent, even though they in a previous study were shown to have a higher incidence of intellectual impairment.
From 90Sr-induced primary tumours, three transfer series were established by serial in vivo transplantation. Chromosome counts were obtained from 2 of the primary tumours and 284 transplanted tumours. The recording of abnormalities was limited to numerical chromosome deviations and the occurrence of metacentric configurations. By means of the serial tumour transplantation the numerical chromosome progression was also analysed. Though appearing at different stages of the tumour evolution, similarities in chromosome pattern were observed.
Highly inbred CBA mice were used. The registration of chromosome abnormalities was limited to numerical deviations and the occurrence of metacentric chromosomes. By separate serial transplantation from a 90Sr-induced osteosarcoma two parallel transfer series (B and b) were established. From these series transplantation was also performed to hyperimmunized hosts B (Hi) and b (Hi). Besides differences in mean outgrowth period between B-and b-generations, a variation in chromosome pattern was observed. However, this variation should not be evaluated as a typical chromosomal progression of fast and slow growing tumour series.
This investigation started from serially in vivo transplanted tumours. The chromosome patterns of in vitro cultures and of parallelly and serially transplanted tumours were examined. Cultured cells were also used for retransplantation to mice. When comparing the chromosome counts of the cultured cells and the in vivo transplanted tumours, significant differences were revealed. When retransplanting cultured cells, it was also noticed that a similar chromosome distribution appeared, as previously found in 90Sr-induced transplanted series.
Transplanted tumours from a 90Sr-induced primary tumour were used as an experimental model to eliminate any connection between the chromosome distribution and outgrowth period of radiation-induced transplanted tumours. For this reason repeated transplantations were performed from different tumours of predetermined age from a transfer generation, to receive separate but parallel transfer generations. From each one of these generations a tumour of the same age as used to establish a particular generation was used for continued transplantation. It was then observed that the percentage of 40-chromosome cells in general decreased with the age of the tumour used for transplantation. Furthermore, early established transfer generations were often characterized by more slow growing tumours. However, it should be realized that even if a true relationship between chromosome pattern and outgrowth period cannot be eliminated, the differences found may be due to accidental variations.
A series of naphthalene diols, quinones, and related compounds were examined for their ability to inhibit mixed-function oxidase in liver microsomes obtained from rats which had been pretreated with 3-methylcholanthrene (3-Mc) or phenobarbital (PB). Using benzo(a)pyrene monooxygenase as a measure of mixed-function oxidase activity, it was found that phenanthrene-9, 10-quinone was the most active compound tested with a K1 = 0.79 microM. Phenanthrene-9, 10-quinone did not affect cytochrome c reductase but did inhibit aminopyrine N-demethylase and p-nitroanisole-O-demethylase in both 3-MC and PB-induced microsome with almost identical inhibition constants. 1,2-Naphthoquinone exerted similar effects as phenanthrene-9,10-quinone on cytochrome c reductase, aminopyrine N-demethylase and p-nitroanisole-O-demethylase. Both quinones stimulated NADPH oxidase activity but the extent of this stimulation did not explain their inhibition of microsomal oxidation. Kinetic studies using benzo(a)-pyrene monooxygenase with phenanthrene-9, 10-quinone and 1,2-naphthoquinone indicated that they were noncompetitive with benzo(a)pyrene and mixed noncompetitive with NADPH. Both of these quinones inhibited benzo(a)pyrene induced oncogenic transformation in C3H10T1/2CL8 cells in culture in a dose response manner, presumably by inhibition of the cellular microsomal enzyme which activate benzo(a)pyrene. Phenanthrene-9, 10-quinone and 1,2-naphthoquinone seem to inhibit microsomal oxidative processes by interaction at the level of cytochrome P-450 possibly with a cytochrome P-450-substrate-oxygen complex.
The adsorption of CTAB (cetyltrimethylammonium bromide) to the rat erythrocyte membrane was studied by determining the electrophoretic mobility of erythrocytes treated with CTAB and by SDS polyacrylamide electrophoresis of membrane proteins from erythrocytes treated with 14C-CTAB. At low concentrations of CTAB there was only a small reduction in the electrophoretic mobility of the erythrocytes. At lytic concentrations the electrophoretic mobility of the erythrocytes was markedly reduced. Alterations at the cell surface were found to be a more likely reason for the reduction in the electrophoretic mobility than interactions between the surfactant and charged groups at the cell surface. Very small amounts of radioactivity were found to be associated with the protein and sialoglycoprotein bands of the polyacrylamide gels. It is suggested that the adsorption of CTAB to the rat erythrocyte membrane does not involve electrostatic interactions between the surfactant and negatively charged groups of the sialoglycoproteins and that membrane proteins do not play a major role in the lytic events.
The readaptation time (RAT) is the interval during which a person exposed to a bright intense light flash cannot perceive a given target. In this study the target used was an optokinetic pattern and the elicited nystagmus (OKN) was registered with electrooculography (EOG), thus giving an objective registration of RAT. Oxazepam in therapeutic doses was given to five healthy subjects and the RAT and serum concentrations of the drug were registered simultaneously at different time intervals. An almost parallel increase of RAT and serum concentration of oxazepam was recorded. This suggests that RAT reflects the depressant effect on the CNS of this drug and it may be used as an objective method of following the clinical effect of a depressant drug as a function of time after intake.
Retest and internal consistency-reliability of the rod-and-frame task, a version which required the body be erect, and using a Rod-alone series, was satisfactory. There was a slight but statistically significant improvement over a 1-mo. interval.

20 male, alcohol-discordant twin pairs, aged 45 to 65 yr. were compared for ability to structure and articulate Rorschach inkblots. The Rorschach responses were evaluated by a new rating scale technique. Data were analyzed by means of one-sample t and multivariate Td2 tests. No relationship between alcohol consumption and tendency to articulate and structure could be established.
