Tissue-specific expression of onco-fetal antigens during embryogenesis.

It has become evident from recent literature that especially in tumor virus systems, cell transformation leads to an arrest of differentiation or to a retrodifferentiation. This may be reflected by the expression of embryonic antigens and it is therefore particularly important to characterize such antigens according to their specificity as well as to their specificity during embryogenesis. We have demonstrated the expression of embryonic antigens which are cross-reactive in avian fibroblasts transformed either by Rous sarcoma virus or by methylcholanthrene. This paper is intended to demonstrate that these embryonic antigens are detected only at a certain period of embryogenesis and particularly in muscle cells. They are detected only occasionally or not at all in cells of other tissues such as brain, liver, lung, and the digestive organs. These antigens are absent from the target cells before transformation and are consequently induced by the transforming agent, either viral or chemical. Therefore, these results suggest that by transformation mechanism, cells become specifically reverted to an earlier stage of differentiation (retrodifferentiation).

[Epidemiology of ovarian cancer].

At the Gynecological University Hospital in Marburg/Lahn, 190 women with cancer of the ovary who had been under clinical treatment from 1962--July 1976 were studied epidemiologically. Special risk factors which seem to predispose to the development of cancer of the ovary were determined. Higher social standing, having no children or having born more than four children, a shortened menarche-menopause interval, having had radiation of the pelvis, as well as having had an appendectomy seem to be factors overrepresented among cancer patients in Marburg. More patients had blood group A and fewer had blood group O than would be expected from the distribution in the population as a whole. Polyps of the corpus uteri were often seen to precede cancer of the ovary. Diabetes mellitus, adiposity, hypertension and varicosis were frequent accompanying diseases. Many patients had a fmily history of cancer.

Influence of different routes of anti-tumor immunization: alternative induction of tumor immunity and tumor enhancement.

Chickens and quails were immunized in parallel either i.v. or intramuscularly (i.m.) with lectin column-purified antigens from chick embryo cells that were transformed in vitro by avain sarcoma virus (ASV). After five to six injections, immunity of the animals was tested by challenge with ASV into the wing webs. Whereas tumor growth was inhibited after i.v. immunization with respect to incidence rate and time of tumor appearance, tumor growth was enhanced after i.m. injection. Animals that were injected with normal cell antigens served as controls. Spleen cells from only those animals that were immunized i.v. exerted a cytotoxic effect in vitro against ASV-transformed cells, whereas spleen cells from i.m. injected animals, in contrast, suppressed such cytotoxicity. The search for serum blocking or arming factors suggested that sera from i.m. injected animals block cellular cytotoxicity whereas sera from i.v. immunized animals render normal spleen cells cytotoxic (arming effect). The use of viruses from different subgroups and of antigens from gp85-lacking ASV-transformed cells indicates that immune effects were obtained against tumor cell surface antigens that differ from the antigen that is involved in virus neutralization (s-gp85).

[Inhibition of stimulated gastric secretion with simultaneous administration of cimetidine and somatostatin].

In four gastric fistula dogs we studied the effect of cimetidine (5 mg/kg/h) and somatostatin (10 microgram/kg/h), alone or in combination, on gastric secretion stimulated by histamine (160 microgram/kg/h) or pentagastrin (6 microgram/kg/h). Acid and pepsin output after histamine stimulation was more reduced by cimetidine than after pentagastrin stimulation. Somatostatin showed an inhibitory effect only after pentagastrin stimulation. The most effective reduction, both on the degree of inhibition and on duration after cessation of the inhibitors, was seen after the combination of cimetidine/somatostatin. These results give some clinical aspects for the use of these two agents.

[Emergency treatment in chest trauma (author's transl)].

The measures taken by the emergency physician can in many cases decisively affect the subsequent course of a chest trauma. In cases where the injured person has to be extricated from collapsed structures the medical officer has to decide on the mode and sequence of the rescue operation, prevent further damage and initiate shock therapy, intubation and artificial ventilation while the rescue operation is in progress. Emergency treatment also includes infusion therapy, if necessary via a central venous catheter; intubation, suction drainage and artificial ventilation; evacuation of fluid from the pleural cavity, mediastinum and pericardium combined with electrodiagnosis (ECG) and electrotherapy (defibrillation) if necessary. By closely observing the reaction and condition of the patient during transport to hospital he can provide important information regarding further treatment, especially in polytraumatized persons. Close collaboration between the trained rescue teams of the Fire Service, the emergency medical officer and the hospital staff has succeeded in reducing the mortality rate of combined chest injuries to 13%. 87% of cases who had received emergency treatment were subsequently admitted to hospital.

High-performance liquid chromatographic investigation of the amino acid, amino sugar and neutral sugar content in glycoproteins.

A methods for the simultaneous separation and determination of amino acids, amino sugars and neutral carbohydrates is described. Stepwise elution systems with sodium citrate and borate buffers have developed for the ion-exchange liquid chromatographic separation of amino acids and sugars, using 8-micrometer particle size resins and the Stein and Moore and orcinol colorimetric method for detection. With the aid of this system, the direct quantitative comparison of sugars and amino acids by liquid chromatography becomes possible for the first time.

A case of Coxsackie A9 virus infection with orchitis.

A thirteen-year-old boy with bilateral orchitis was diagnosed as being infected by Coxsackie A9 virus by isolation of the virus from a throat swab and a fecal specimen. Serologic investigations revealed the development of transitory specific neutralizing IgM antibodies against the isotate and of persistent antibodies belonging to the other immunoglobulin classes. An etiology of the orchitis by any of the infectious agents usually found with this condition could be excluded.

Bovine and ovine leukemia viruses. I. Characterization of viral antigens.

A comparative study on several virus-specific antigens of bovine and ovine leukemia viruses is presented. In addition to the major immunologically reactive, nonglycosylated antigen p21, which has a molecular weight of 21,000, both viruses share two glycoproteins of apparent molecular weights of 60,000 (gp60) and 32,000 (gp32), respectively. Immunological cross-reaction suggests that ovine and bovine leukemia viruses are genetically highly related.

Effects of dexamethasone on the morphogenesis of two mutants of Rous sarcoma virus.

Japanese quail cells transformed by the replication-defective, Bryan high-titer strain of Rous sarcoma virus, BH RSV(-)Q, clone 3, revealed intracytoplasmic A-type particles after dexamethasone treatment. In the absence of dexamethasone, or when superinfected with a helper virus in the presence or absence of dexamethasone, no such particles were observed. Chick embryo cells (CEC) infected with a temperature-sensitive sarcoma virus mutant, LA 334, coordinately defective for transformation and viral replication, showed abnormal accumulation of viral core substances and aberrant budding at the non-permissive temperature (41 degrees). CEC infected with LA 334 and treated with dexamethasone at 41 degrees resulted in more extensive accumulation of abnormal budding without increased release of viral particles. Dexamethasone, however, did not lead to abnormal morphogenesis of virus under permissive conditions. The selective effects of dexamethasone on the morphogenesis of these two mutants of avian sarcoma virus are discussed.

Rous sarcoma virus-transformed avian cells express four different cell surface antigens that are distinguishable by a cell-mediated cytotoxicity-blocking test.

Japanese quails bearing avian sarcoma virus-induced tumors develop immune spleen cells that are cytotoxic in vitro against virally and chemically transformed cells, as well as against embryonic cells. The cell-mediated cytotoxicity can be blocked by soluble antigens extracted from in vitro cultured cells. The existence of partial as well as total blocking effects in tests with extracts from various transformed and untransformed virus-producing cells makes it possible to distinguish up to four different kinds of antigens expressed on sarcoma virus transformed cells: a) a subgroup-specific determinant of the virus-envelope glycoprotein gp85 (s-gp85) is expressed at the surface of productively infected, tranformed as well as untransformed cells; b) a group-specific determinant of gp85 (g-gp85) that is only expressed on the surface of virus-transformed cells; c) embryonic antigens, also detectable on chemically transformed as well as on primary normal embryonic cells, and finally; d) a sarcoma virus transformation-specific antigen (TSSA) that is not a structural constituent of the virus.

[The effect of domperidone and metoclopramide on antral motility (author's transl)].

The effect of domperidone and metoclopramide on antral motility was studied by measuring the intraluminal pressure in the antrum of 10 normal persons and of 8 patients suffering from duodenal ulcer. Both drugs stimulated the rhythmic activity of the antrum. The antiemetic effect and the therapeutic effect of domperidone on complaints by gastrointestinal retention was the result of the influence on gastrointestinal motility, which is similar to that of metoclopramide.

The definitive technique of selective proximal vagotomy with pyloroplasty appropriate to form and function in surgery of peptic ulcer disease.

The overall efficiency of the ff-PYPL is most apparent when we compare our own large series of long-term observations of SPV, accompanied with these pyloroplasty forms with all types of DU, in contrast to the series of SPV without pyloroplasty of some other authors with a smaller number of patients and a shorter observation period (Table 1). The table shows that the combination operation of complete SPV with ff-PYPL (Fig. 17) assures the clinical permanent healing of DU. In GU the results of the nonresecting method are less satisfying. Twenty-two percent of the GU patients must have selective vagotomy with antrectomy because of antral disorder, destroyed antral innervation, or suspicion of malignancy.
