[Study on the arylaminothiourea compounds by IR spectroscopy].

In this paper, eleven new thiourea compounds were prepared and studied by IR spectroscopy. The fundamental vibrations have been assigned. The band near 1540cm(-1) was identified as thiourea I band, which mainly originated from the N-H deformation vibration coupling with a little C-N stretching vibration. Thiourea I band near 1280cm(-1) mainly originated from C-N stretching vibration. C=S stretching vibration frequency was at near 110cm(-1) and it was identified as thiourea III band.

Tolerance to mu-opioid receptor agonists but not cross-tolerance to gamma-aminobutyric acid(B) receptor agonists in arcuate A12 dopamine neurons with chronic morphine treatment.

The present study examined the potential for cross-tolerance development between mu-opioid and gamma-aminobutyric acidB receptor agonists, in hypothalamic arcuate neurons, resulting from chronic morphine treatment. Intracellular recordings were made in hypothalamic slices prepared from ovariectomized female guinea pigs. The mu-opioid receptor agonist D-Ala2,N-Me-Phe4,Gly-ol5-enkephalin and the gamma-aminobutyric acidB receptor agonist baclofen produced dose-dependent membrane hyperpolarizations of arcuate neurons. The reversal potential for both agonist-induced hyperpolarizations was near -95 mV, indicative of the activation of an underlying K+ conductance. Coadministration of maximally effective concentrations of D-Ala2,N-Me-Phe4,Gly-ol5-enkephalin and baclofen produced a response that was not additive, indicating a convergence onto a common K+ channel. In arcuate neurons, including a subset that was immunopositive for tyrosine hydroxylase, chronic morphine treatment for 4 to 7 days produced a 3.2-fold reduction in the potency, with no change in the efficacy, of D-Ala2,N-Me-Phe4,Gly-ol5-enkephalin. In contrast, it affected neither the potency nor the efficacy of baclofen. Therefore, chronic morphine exposure does not produce cross-tolerance between mu-opioid and gamma-aminobutyric acidB receptor agonists in A12 dopamine neurons, suggesting that convergence upon a common effector is not a sufficient criterion for the development of cross-tolerance between receptor systems.

Renal mRNA of PTH-PTHrP receptor, [Ca2+]i and phosphaturic response to PTH in phosphate depletion.

Available data indicate that the mRNAs of the PTH-PTH-related protein (PTH-PTHrP) receptor in the kidney, liver and heart are down-regulated in chronic renal failure (CRF). This is due, in major part, to the elevation of cytosolic calcium ([Ca2+]i) in the cells of these organs. If elevation in [Ca2+]i is indeed the culprit, one should be able to demonstrate down-regulation of the mRNA of the PTH-PTHrP receptor in situations without CRF and with low levels of PTH but with elevated [Ca2+]i. Such a combination of events occurs in phosphate depletion (PD). To test this hypothesis, we examined the [Ca2+]i and the concentration of the mRNA of the PTH-PTHrP receptor in the kidneys from 1, 3 and 6 weeks PD, pair-weighed (PW) rats and PD and PW rats treated with verapamil (PD-V, PW-V). To evaluate the effect of a potential rise in [Ca2+]i on urinary phosphate excretion, we also measured the phosphaturic response to PTH and cAMP in all groups of rats after 6 weeks of the dietary intervention. Renal function was normal in all groups of animals. Blood levels of PTH were significantly (p < 0.01) lower in PD and PD-V after 1 week of PD than in PW and PW-V rats, and they remained low throughout the study. The basal levels of [Ca2+]i in the renal proximal tubular cells were normal after 1 week of PD but rose by the third week of the study and remained elevated by the end of the sixth week. These values were significantly (p < 0.01) higher than those in PD-V, PW and PW-V rats. The concentrations of mRNA of the PTH-PTHrP receptor relative to that of the housekeeping gene G3DPH were significantly (p < 0.01) lower in PD rats after 3 and 6 weeks than in the other three groups of rats. The phosphaturic response to PTH or cAMP was significantly (p < 0.01) greater in PD-V rats than in PD animals. The data show that PD is associated with a rise in [Ca2+]i of renal proximal tubular cells and with down-regulation of PTH-PTHrP receptor in the kidney despite low levels of PTH and normal renal function; normalization of the concentration of [Ca2+]i in PD-V rats was associated with normal expression of mRNA of the receptor. These results provide strong support for the proposal that elevated [Ca2+]i down-regulates the mRNA of the PTH-PTHrP receptor even in the absence of CRF and elevated blood PTH levels. The improvement in the phosphaturic response to PTH and cAMP in PD-V rats is consistent with the notion that the elevated [Ca2+]i of the renal cell in PD rats may interfere with the coupling of PTH receptor-adenylate cyclase system and/or with the postreceptor events responsible for the inhibition of phosphate reabsorption by these agonists.

[Clinico-pathological significance of the immunostaining of myosin heavy chain isoforms in pathological human skeletal muscle].

Expression of the four myosin heavy chain isoforms (fast-twitch, slow-twitch, neonatal and embryonal isoforms) was immunohistochemically observed in 500 biopsied limb muscles of neuromuscular disorders. Fast-twitch isoform was expressed in type 2A, 2B and pathologic 2C fibers. Slow-twitch isoform was expressed in type 1 and 2C fibers. Embryonic isoform was expressed in regenerating type 2C fibers of active myopathies such as Duchenne dystrophy and polymyositis. Expression of neonatal isoform, which was longer positive than that of embryonic isoform, was noted in regenerating fibers, denervated fibers and highly atrophic fibers in chronic myopathies of limb-girdle dystrophy and myotonic dystrophy. In conclusion, the immunostaining of MHC isoforms are useful to make a clinico-pathological diagnosis to determine the stages in evolution of regeneration or degeneration of pathologic muscle fiber per se.

[Immunohistochemical localization of chymase; a mast cell marker and clinical significance in diseased human skeletal muscle].

UNLABELLED: In the advanced stage of dystrophinopathy, cardiac dysfunction is a serious complication for prognosis. Recently, an angiotensin converting enzyme (ACE), which converts angiotensin (A) 1 to A 2, has been reported to be effective for cardiac insufficiency. The A 2 is produced more dominantly in the path via the production of a neutral serine protease, chymase (MW 25,000), secreted from the mast cell. We have observed localization of chymase in diseased human skeletal muscle tissues, and evaluated its clinical significance. The frozen muscle biopsied specimens from 91 neuromuscular disorders (muscular dystrophies, inflammatory myopathies and neurogenic muscular disorders) were stained by using monoclonal antibody against the chymase, and the positive cells in a whole sectional field were counted. In the serial sections, we also performed routine histochemistry and immunostainings of immunological markers (CD4, CD8 and others) as well as the apoptotic proteins for comparison. RESULTS: The chymase-positive mast cells were scattered mainly in the endomysium, partly in the perimysium and around small vessels. Although the positivity was not disease specific, more numerous strongly positive cells were observed in dystrophinopathy and inflammatory myopathies, but less in myotonic dystrophy and neurogenic muscle disorders. In the normal control muscle, however, strongly positive cells appeared less frequently than in the above mentioned diseased muscles. The chymase-positive cells partly corresponded to the ubiquitin-positive ones, but perforin, granzyme A, Fas and Bcl-2 did not. In conclusion, the chymase-positive mast cell may play a primary or secondary role in the diseased muscle, and their more abundant appearance in dystrophinopathy and some other myopathies suggest the effectiveness of an ACE blocker, an anti-chymase drug.

Effects of antisense oligodeoxynucleotides to NR1 on suppression of seizures and protection of cortical neurons from excitotoxicity in vivo and in vitro.

OBJECTIVE: To investigate the effects of NR1 subunit on the initiation and development of seizures and protection of cortical neurons from excitotoxicity by using antisense oligodeoxynucleotides (ODN) to NR1 in vivo and in vitro. METHODS: Intracerebroventricular injection, temporal cortex slices discharge, cerebral cortical neuronal culture, induction of neurotoxicity and [3H]MK-801 binding were used in this study. RESULTS: After an antisense ODN for NR1 was administered intracerebroventricularly (i.c.v. 100 micrograms in 10 microliters) once daily, for three days in genetically epilepsy-prone rats (GEPR, P77PMC), the animals did not develop any clonic and tonic convulsions and their seizure scores were significantly lower compared to the control groups. The frequency and amplitude of early seizure-like events (SLEs) and late recurrent discharges (LRD), induced by lowering Mg2+, were reduced in entorhinal cortex (EC) of the temporal slice treated by antisense ODNs. Pretreatment with antisense ODN (2 microM) protected more than 52% of glutamate-sensitive neurons and reduced the [3H]MK-801 binding to 50% in cultured cerebral cortical neurons. CONCLUSIONS: N-methy-D-aspartate-receptors (NMDAR), specifically the NR1 subunit, may participate and play important roles in the initiation and propagation of epilepsy in the P77PMC rat.

Fibula grafting for treatment of aggressive benign bone tumor and malignant bone tumor of extremities.

OBJECTIVE: To compare the effect of vascularized and nonvascularized fibula grafting performed for the reconstruction of bone defects after the resection of bone tumors. METHODS: Vascularized fibula grafting was performed in 18 patients with malignant or aggressive benign bone tumor of the extremities. Non-vascularized fibula grafting was performed in 15 patients with giant cell tumor of the radius. All the 18 patients were followed up with radiography, single photon emission computed tomography (SPECT), colored Doppler's sonography (CDS) examinations and functional observation for 2 to 14 years. RESULTS: Vascularized fibula grafts with bone defects of 9 to 26 cm showed good blood supply in emission computerized tomography (ECT) and radiography examinations, and bone union was achieved (Table 1). In non-vascularized fibula grafting cases (Table 2), bone union was not achieved. CONCLUSIONS: The advantages of vascularized grafting are the feasibility of one-stage reconstruction of various tissues and the growth of grafted fibula. It is necessary to adopt vascularized fibula grafting to reconstruct large bone defect resulting from the resection of extensive bone tumors, such as giant cell tumor (GCT), fibrous dysplasia of bone and malignant bone tumors.

Effects of removing circulatory tumor necrosis factor by immunoadsorption on experimental endotoxin shock animals.

OBJECTIVE: To evaluate the effects of removing circulatory tumor necrosis factor (TNF) by immunoadsorption on endotoxin shock animals. METHODS: Sixty New Zealand white rabbits were injected intravenously with lethal dose of endotoxin (10 Billion cfu/kg E. Coli endotoxin) and randomly divided into 3 groups: perfusion group, hemoperfusion started at 1 hour after injecting endotoxin through immunoadsorbent columns against TNF; pseudoperfusion group, hemoperfusion through blank columns; and control group, injected with endotoxin only. The arterial pressure, microcirculation of the mesentery, plasma levels of TNF, IL-1, IL-6, IL-8, nitrite, endothelin-1 (ET-1), alanine aminotransferase (ALT), aspartate aminotransferase (AST), creatinine were measured and analyzed and finally the survival rate was observed. RESULTS: Plasma levels of TNF were sharply reduced after immunoadsorption. Moreover, release of IL-1, IL-6, IL-8, NO and ET-1 were also attenuated. Hemodynamic abnormalities could be improved and survival rate ameliorated significantly. CONCLUSION: Specific immunoadsorption of circulating TNF might be a new and effective therapy for endotoxin shock.

[Investigation on the viscoelastic properties of hepatocytes and their relavances to cytoskeleton structure].

Using micropipette aspiration technique, the authors investigated the viscoelastic properties of human fetal hepatocytes (HFH) and hepatocellular carcinoma (HCC) cells. And further, the effects of two cytoskeleton interferents, cytochalasin D and colchicine on the viscoelastic properties of HFHs were investigated. The results showed: the elastic coefficients K1 and K2 of HCC cells were significantly higher than those of HFHs; the action of cytochalasin D led to an obvious decrease in elastic coefficients K1, K2 as well as the viscous coefficient mu of HFHs; under the action of colchicine, the K1 value of HFHs increased while the K2 and mu values decreased obviously or tended to decrease. A brief discussion was presented about these results.

[Improvement of technigue in restoration of stability of thoracolumbal spine].

A new fixator for spine was designed to restore the stability of spine in improving the treatment of thoraco-lumbar dislocation. This instrument was composed of six nails, two longitudinal connecting rods and two transverse connecting rods. On the surface of the nails, there were furrows engraved, instead of screw thread. It fixed the spine through pedicle of vertebra from the posterior approach. The fixed vertebrae were limited between two vertebrae. From 1989 th 1995, twelve patients with fracture-dislocation of thoraco-lumbar spine were reduced with this instrument. After four years follow-up, it showed that the deformity was corrected and the spine firmly fixed. Compared with Dick's screws, it had the following advantages such as simple manipulation, increased strergth of nails and minimal damage to tissue during operation.

[Clinical evaluation of polyamine assay in diagnosis of malignant ascites].

Polyamine levels were determined by precolumn derivation RH-HPLC in 68 patients with ascites including 21 malignant ascites, 28 liver cirrhosis and 19 tuberculosis. The total polyamine, especially the contents of cadaverine (CA) and spermidine (SPD) in malignant ascites were significantly higher than that in cirrhosis and tuberculosis. Among them CA levels in malignant ascites and cirrhotic and tubercular ascitic fluid were 2.26 +/- 1.83 nmol.ml-1, 0.75 +/- 0.52 nmol.ml-1 and 0.76 +/- 0.47 nmol.ml-1 respectively, demonstrated the most obvious difference. Using 1.0 nmol.ml-1 as a threshold cut off point to differentiate the malignant ascites from non-malignant ascites, the from non-malignant ascites, the sensitivity, specificity and accuracy reached 76. 2%, 83.0% and 80.9% respectively. The results suggest that polymine level in ascites might be regard as one of the cancer-associated markers.

IRES bicistronic expression vectors for efficient creation of stable mammalian cell lines.

Stable transfection of mammalian cells is a widely used technique for the study of gene expression and protein purification. However, selection of cell lines expressing desired genes from a large number of candidate clones is often labor-intensive and time consuming. To improve the efficiency of stable cell line production, we have used a bicistronic mammalian expression vector, pIRES1hyg, which contains the internal ribosome entry site (IRES) of the encephalomyocarditis virus (ECMV). The IRES element permits the translation of two open reading frames from one messenger RNA: one reading frame encoding the recombinant protein of interest and the other an antibiotic resistant marker (e.g. hygromycin). We demonstrate that the use of the bicistronic vector significantly facilitates the creation of stable mammalian cell lines, because all selected antibiotic-resistant colonies express the recombinant gene of interest. Therefore, the use of the pIRES1hyg bicistronic vector for stable transfection eliminates the need to screen large numbers of colonies to find functional clones. We conclude that the IRES bicistronic vector provides a powerful tool for efficient selection of stable transformants in mammalian cells.

An enhanced green fluorescent protein allows sensitive detection of gene transfer in mammalian cells.

The green fluorescent protein (GFP) from the jellyfish Aequorea victoria has become an important marker of gene expression. However, the sensitivity of wild-type GFP has been below that of standard reporter proteins, such as beta-galactosidase, which utilize enzymatic amplification. To improve the detection of GFP in transfected mammalian cells, we have constructed a unique GFP variant which contains chromophore mutations that make the protein 35 times brighter than wild-type GFP, and is codon-optimized for higher expression in mammalian cells. These changes in the GFP coding sequence provide an enhanced GFP (EGFP) that greatly increases the sensitivity of the reporter protein. We show that the EGFP expression vector delivered into mammalian cells gives rise to bright fluorescence that is readily detectable following a 16-24 hr transfection interval. Visual detection of transfected cells with EGFP appears to be more sensitive than equivalent measurements with beta-galactosidase catalyzed conversion of the X-gal substrate. We conclude that EGFP allows sensitive and convenient detection of gene transfer in mammalian cells.

Crystallization and preliminary diffraction analysis of porcine heart mitochondrial NADP(+)-dependent isocitrate dehydrogenase.

Isocitrate dehydrogenases [isocitrate:NAD(P)(+) oxidoreductase (decarboxylating), E.C. 1.1.1.42] are ubiquitous metabolic enzymes which occur in all living organisms. The NADP(+)- dependent mitochondrial isocitrate dehydrogenase from pig heart has been crystallized from polyethylene glycol/sodium sulfate mixtures in the presence of Mg(2+) and isocitrate. The crystals belong to space group C2 with a = 137.0, b = 113.4, c = 65.0 A and beta = 98.5 degrees, and diffract to at least 2.4 A resolution. There are two protein monomers per asymmetric unit which are related by non-crystallographic twofold symmetry.

Gene transfer into mammalian cells using histone-condensed plasmid DNA.

A recombinant histone (NLS-H1) containing both the SV40 large T antigen nuclear localization signal and the carboxy-terminal domain of human histone H1(0) was produced in bacteria. NLS-H1-plasmid DNA complexes, in the presence of chloroquine, mediated reporter gene transfer into cultured cells with similar efficiencies as plasmid DNA-cationic lipid (lipofectin) complexes. NIH-3T3 or COS-7 cells transfected with NLS-H1-plasmid DNA-lipofectin complexes expressed at least 20 times more luciferase or had at least 2.5 times more beta-galactosidase-positive cells than those transfected with plasmid DNA-lipofectin complexes. Foreign gene expression was also improved by other DNA-binding proteins and cationic lipid formulations, yet the greatest enhancement was obtained with complexes containing either NLS-H1 or calf thymus histone H1. Histone H1-plasmid DNA-lipofectin complexes were internalized by a greater number of cells than plasmid DNA-lipofectin complexes.
