[Antioxidation of Pueraria lobata isoflavones (PLIs)].

Isoflavones are isolated from Pueraria lobata. By studyign Pueraria lobata isoflavones (PLIs) on lipid peroxide (LPO) and superoxide dismutase (SOD) in animal tissue, this paper suggests that PLIs is an effective antioxidant. The results in vitro show that PLIs can inhibit significantly the content of LPO. The highest inbibition rate are found to be 79.7%, 84.7%, and 86.6% in liver, kidney of mice and brain of rabbit respectively. By cold induced brain injury with chalcone, the content of LPO and activity of SOD in animal blood and brain are changed. When animals are injected with PLIs, the result indicates that the PLIs can reduce significantly the content of LPO and enhance the activity of SOD in animal blood and brain, in a dose-dependent relationship.

[Protective effect of paeonol on repeated cerebral ischemia in rats].

Protcective effect of paeonol was observed in the repeated cerebral ischemia-reperfusion model in rats. Paeonol (100, 50 mg/kg x 7 d i.p.) could inhibit the increase of Ca2+, the decrease of superoxide dimutase (SOD) activity and the content of malondialdehyde (MDA), improve Ca(2+)-ATPase activity in ischemia brain tissue. The results showed that paeonol could protect cerebral ischemia by inhibiting accumulation of Ca2+ and production of oxygen free radical.

[Transcription of antisense RNA of hepatitis B virus through retroviral mediated gene transfer].

To probe the ways by which the antisense gene of hepatitis B virus (HBV) can be transferred and transcripted in eukaryotic cells to inhibit HBV replication and expression. Two retroviral vectors that carried antisense gene of hepatitis B virus (HBV) PreC/C or PreS/S region were constructed. The HBV ayw PreC/C and PreS/S fragments were inserted into the vector pDO. R cloning site in the sense or antisense orientation and the recombinant retroviral vectors were then transfected into PA317 packaging cells by calcium phosphate coprecipitation, respectively. The stably transformed G418-resistant PA317 cells were selected and the recombinant retroviruses released from transfected G418-resistant PA317 cells were assayed by G418 selection method, using NIH 3T3 cells as target cells. Southern blot and RNA Dot blot analysis showed that the recombinant retroviral vector sequences were stably integrated into the chromosome of transfected PA317 cells and the antisense RNA of HBV PreS/S or PreC/C gene also transcripted in the transduced NIH 3T3 cells. These results suggested that antisense gene of HBV can be successfully transferred and transcripted in target cells through retroviral vector mediated gene transfer and the antisense retroviral vectors may be potentially useful for anti-HBV gene therapy.

[The inhibitory effect of Chinese herb Phyllanthus on hepatitis B virus in vitro].

Seven preparations of different species of Chinese herb Phyllarthus with different formulations and additives were studied for their ability to inhibit hepatitis B virus DNA replication, that is the HBsAg and HBeAg expression in a HBV DNA-transfected cell line (2.2.15 cell). The results showed that all the drugs were effectively against HBV replication in vitro. Capsule 1 and tuo cha zhen zhu cao were found to be the most potent anti-HBV extracts of those examined. Both extracts decreased HBaAg expression by nearly 100% at the concentration of 500 mg/L without toxicity on cell growth. Southern blot showed that Capsule 1 inhibited HBV DNA replication in a dose-dependent manner. We observed further, that under similar dosage the drug inhibition to HBsAg and HBeAg expression was lower than that to HBV DNA replication, suggesting that the production of progeny virus particles not only were inhibited at the level of HBV DNA synthesis, but also was affected probably at the level of translation of viral mRNA.

[The antithrombin action of stichopus japonicus acid mucopolysaccharide (Sjamp) is mediated by heparin cofactor II].

OBJECTIVE: To further evaluate the antithrombin mechanism of stichopus japonicus acid mucopolysaccharide (Sjamp). METHODS: The antithrombin mechanism of Sjamp was checked by assaying its effects on the thrombin activity in normal human pooled plasma, purified human heparin cofactor II (HC II) system and antithrombin III (AT III) system. RESULTS: Sjamp inhibited thrombin activity in the presence of HC II with a second order rate constant of 1.56 x 10(7) m(-1) x min(-1), which was 4.6 times as much as that of AT III. CONCLUSION: Sjamp was similar to dermatan sulfate both in the efficiency and in the mechanism of antithrombin.

[The spectrum analysis of high performance liquid chromatographic peak by ultraviolet-visible spectrophotometric detectors].

In the analysis of high performance liquid chromatography (HPLC), the qualification of chromatographic peak can be carried out by the methods of adding standard, comparing retention time and absorption spectrum etc. This paper reports a simple and practical method for measurement of absorption spectrum of HPLC peak by ultraviolet and visible variable wavelength detector. A BASIC program was edited with Shimadzu C-R4A data processor. Stopping the pump at the retention time we use the program to analyse absorption spectrum of the peak and the absorption spectrum of standard (Cr, PU, UA) by Shimadzu UV-2201 UV-VIS recording spectrophotometer and the absorption spectrum of standard (-S) with sample (-T) by this method are presented. The figure shows that the absorption spectra of the samples of Cr, PU and UA almost completely overlapped with those of the standards. The results showed that this method is reliable.

[Catalytic fluorimetric determination of trace amount of bromine ion].

A new catalytic fluorimetric method for the determination of bromine ion based on its catalytic effect on the oxidation of fluorescein by potassium bromate in the solution of dilute sulfuric acid has been established. The detection limit for Br- is 1.94 x 10(-2) microg/25 mL. The linear range of the determination is 6.0-12.0 microg/25 mL. The method has been applied to the determination of Br- in tablet pyridostigmini bromidum sample with satisfactory results.

[Investigation of the carboxymethyl hydroxypropyl guar gum cross-linked with metal ions].

In this paper the infrared-spectral changes and thermal gravimetric analysis of the gels formed with carboxymethyl hydroxypropyl guar gum (CMHPG) cross-linked with metal ions (Fe3+, Cr3+, Ti4+) have been studied. The results indicated that the coordinated active sites are mainly supplied with hydroxypropyl, neigbouring cis-OH of main chain and carboxymethyl groups of CMHPG. And the results of spectral analysis showed the strength orders and coordinated mode of the guar gum cross-linked with metal ions.

The determination of Ni, Al, Mn and Si in copper based alloy by high current and microsecond pulsed (HCMSP) glow discharge optical emission spectrometry (GD-OES) has been reported. With the laboratory built experiment set-up, the factors effecting analytical performance have been studied. The results of sample analysis are in good agreements with the recommendation values, and the stansard deviation for six determinations were 2.66% for Ni, 2.17% for Mn, 6.02% for Si, 1.00% for Al respectively.
