In planta expression of HIV-1 p24 protein using an RNA plant virus-based expression vector.

Plant viruses show significant potential as expression vectors for the production of foreign proteins (e.g., antigens) in plants. The HIV-1 p24 nucleocapsid protein is an important early marker of HIV infection and has been used as an antigen in the development of HIV vaccines. Toward developing a plant-based expression system for the production of p24, we have investigated the use of a (positive)-strand RNA plant virus, tomato bushy stunt virus (TBSV), as an expression vector. The HIV p24 open reading frame (ORF) was introduced into a cloned cDNA copy of the TBSV genome as an in-frame fusion with a 5'-terminal portion of the TBSV coat protein ORF. In vitro-generated RNA transcripts corresponding to the engineered virus vector were infectious when inoculated into plant protoplasts; Northern and Western blot analyses verified the accumulation of a predicted p24-encoding viral subgenomic mRNA and the production of p24 fusion product. Whole-plant infections with the viral vector led to the accumulation of p24 fusion protein in inoculated leaves, which cross-reacted with p24-specific antibodies, thus confirming the maintenance of key antigenic determinants. This study is the first to demonstrate that TBSV can be engineered to express a complete foreign protein of clinical importance. Strategies for optimizing protein yield from this viral vector are discussed.

Effects of photoinduced membrane rigidification on the lysosomal permeability to potassium ions.

Mechanism for the photoinduced increase in the lysosomal K+ permeability is still unknown. In this study, we investigated the effect of photodamage-induced membrane rigidification on the lysosomal K+ permeability by measuring the membrane potential with bis(3-propyl-5-oxoisoxazol-4-yl)pentamethine oxonol and by monitoring proton leakage with p-nitrophenol. Membrane fluidity was measured by the steady-state fluorescence anisotropy of 1,6-diphenyl-1,3,5-hexatriene. Methylene blue-mediated photodamage to lysosomes decreased their membrane fluidity and increased their K+ permeability. The photoinduced increase in the K+ permeability can be reversed by fluidizing the rigidified lysosomal membranes with benzyl alcohol. The results suggest that the membrane rigidification induced by photodamage may increase lysosomal K+ permeability. This conclusion is supported by the observation that rigidifying lysosomal membranes by the treatment with membrane rigidifier cholesteryl hemisuccinate also enhanced the lysosomal K+ permeability.

[The common C677T polymorphism in the methylenetetrahydrofolate reductase gene is associated with neural tube defects and preeclampsia].

OBJECTIVE: To assess whether methylenetetrahydrofolate reductase(MTHFR) gene C677T polymorphism is associated with neural tube defects and preeclampsia. METHODS: Twenty- four mothers who had given birth to normal children, 27 mothers who had given birth to NTDs children, 120 normal women, and 57 women who had suffered from preeclampsia were genotyped for C677T polymorphism by using PCR/RFLP method. RESULTS: (1) VV genotype frequency in the normal mothers group it was 0.13, while in the NTDs mothers group it was 0.33; mothers with VV genotype were at 3 times increased risk to have a NTDs child, compared with the normal mothers. (2) In the normal women group VV genotype frequency was 0.15, while in the preeclampsia women group it was 0.31; women with VV genotype were at 2.5 times increased risk to have preeclampsia, compared with the normal women. CONCLUSION: The authors believe that MTHFR VV genotype is associated with the development of NTDs and preeclampsia.

Study on DNA polymorphism at D1S8 locus in Hebei Han population.

OBJECTIVE: To study the polymorphism at D1S8 locus and to provide basic information for the construction of DNA fingerprint database and the application in forensic medicine. METHODS: Minisatellite variant repeat-polymerase chain reaction (MVR-PCR) and polyacrylamide gradient gel electrophoresis followed by silver staining were used to detect the variant repeat sequences at D1S8 locus of 240 unrelated individuals in Hebei Han population, and digital codes were obtained. RESULTS: Each individual obtained about 30 digital codes, in which none of two unrelated individuals had the same code. The probability of identity of 30 digital codes was 3.55x10(-11). The percentages of three repeat units, a-type, t-type and o-type were 54.77%, 42.54% and 2.69% respectively. The heterozygosity (H) was 0.9837. The excluding probability of paternity(EPP) and polymorphism information content(PIC) were 0.9669 and 0.9833 respectively. CONCLUSION: Because of its high polymorphic nature, D1S8 locus is a valuable marker for forensic identification and paternity testing.

[The application of fluorescence in situ hybridization performed on the decolorized G-banding chromosomes in detecting the marker chromosomes of gastric cancer].

OBJECTIVE: Using a rapid, accurate method that detects the marker chromosomes of gastric cancer and enhancing the ability of discriminating complicated chromosome rearrangements of gastric cancer. METHODS: The improved method of fluorescence in situ hybridization(FISH) performed on the decolorized G-banding chromosome was used. RESULTS: The changes of two marker chromosomes (M1, M2) of the cell line(SGC-7901) of gastric cancer and one marker chromosomes(M3) of one primary gastric cancer were respectively analyzed by this method. The M1, M2 and M3 had complicated structural chromosome aberrations: del(7)(p15)/del(7)(q22), t(1;3)(p11;q11) and del(7)(q32). CONCLUSION: This method showed strong signals, low backgrounds and well-repetitions. It may play an important part in exploring the chromosome rearrangements in the process of pathogenesis and development of gastric cancer.

[The change of serum melatonin and diagnosis value in hepatic encephalopathy].

OBJECTIVE: To explore the change of serum melatonin (MT) and its diagnosis value in hepatic encephalopathy (HE). METHODS: The change of MT in serum in cirrhotic patients with HE and without HE was determined by ELISA, and normal serum served as control. The change of serum MT during exacerbation and remission stage of HE was also determined. RESULTS: The level of MT in patients with HE was higher than that without (P<0.01). Both groups were higher than normal group (312.7 +/- 77.4) ng/L, (149.8 +/- 38.4) ng/L, and (77.3 +/- 28.4)ng/L, respectively, P<0.01. The level of serum MT was higher in exacerbation stage than remission stage, (308.5 +/- 59.1) ng/L and (147.8 +/- 23.3) ng/L, respectively, P<0.01. CONCLUSION: The elevation of MT in serum may be one of pathogenic factors for HE. Dynamic measurement of MT in serum is conducive to the diagnosis of HE.

Kupffer cells do not play a role in governing the efficacy of liposomal mitoxantrone used to treat a tumor model designed to assess drug delivery to liver.

A tumor model designed to assess liposome-mediated drug delivery to liver has been used in an attempt to better understand the mechanism of activity of liposomal mitoxantrone, a liposomal anticancer drug formulation that appears to be uniquely effective in treating this tumor model. Reductions in liposomal mitoxantrone accumulation in the liver were achieved either by use of poly(ethylene)glycol (PEG)-modified lipids or by methods designed to deplete liver phagocytes, a method referred to as hepatic mononuclear phagocytic system (MPS) blockade. A 2-fold reduction in mitoxantrone delivery to the liver was obtained using a mitoxantrone formulation with PEG-modified lipids, and a 3-fold reduction was obtained when liposomal mitoxantrone was given to animals pretreated to induce hepatic MPS blockade. Results demonstrate that the liposomal mitoxantrone formulation prepared with PEG-modified lipids was significantly less active than the formulations that did not contain PEG lipids, with median survival times of 17 days and 100% 60-day survival, respectively. In contrast, hepatic MPS blockade had no effect on the therapeutic activity of 1,2-dimyristoyl phosphatidylcholine/cholesterol (DMPC/Chol) mitoxantrone (100% 60-day survival). These data suggest that the hepatic MPS does not play a role in mediating the therapeutic activity of DMPC/Chol mitoxantrone in the treatment of liver localized disease. Results with formulations prepared with a PEG-stabilized surface, however, suggest that nonspecific methods to decrease liposome cell interactions inhibit the therapeutic activity of DMPC/Chol mitoxantrone.

[Studies on the polymorphism of MICA gene in four Chinese populations].

OBJECTIVE: To understand with greater clearness the genetic polymorphism of (GCT)n repeat of MHC class I chain-related gene A (MICA)in some Chinese populations and provide preliminary genetic evidence for the independent origin of Chinese Baima Tibetan (BMT). METHODS: ACD-blood or saliva specimens of 411 unrelated individuals from four Chinese populations were collected. A primer pair spanning exon 5 of MICA gene was used to amplify the GCT region. Alleles were detected by PCR and denaturing PAGE. Comparison of the allelic distributions among the four populations was carried out. RESULTS: Five previously reported alleles have been observed in all the four populations, but the allelic distributions are different from one another. The most frequent allele is the A5 in all the four populations (0.325 in BMT, 0.345 in Tibetans, 0.390 in Chengdu Hans and 0.319 in Qiangs). A5.1 allele is the second most frequent allele in Chengdu Hans (0.230) and in Qiangs (0.293), while the second most frequent alleles for BMT and Tibetans are A4 (0.254) and A9 (0.272) respectively. The distribution of alleles in BMT is significantly different from that in the other three populations. CONCLUSION: Alleles of MICA gene exon 5 are conservative in all populations studied so far. The results suggest that genetically BMT might be an independent ethnic population.

Comparison of the viscoelastic properties of normal hepatocytes and hepatocellular carcinoma cells under cytoskeletal perturbation.

The viscoelastic properties of both hepatocytes and hepatocellular carcinoma (HCC) cells were measured by means of a micropipette aspiration technique. Experimental results were analyzed with a three-element standard linear solid model, in which an elastic element, K1, is in parallel with a Maxwell element composed of another elastic element, K2, in series with a viscous element, mu. Further, we investigated the relevance of viscoelastic properties of these two types of cells to the cytoskeleton structures by treating cells with three cytoskeletal perturbing agents, namely cytochalasin D (CD), colchicine (Col) and vinblastine (VBL). The results showed that the elastic coefficients, but not viscous coefficient of HCC cells (K1 = 103.6 +/- 12.6 N m-2, K2 = 42.5 +/- 10.4 N m-2, mu = 4.5 +/- 1.9 Pa s, n = 30), were significantly higher than the corresponding values for hepatocytes (K1 = 87.5 +/- 12.1 N m-2, K2 = 33.3 +/- 10.3 N m-2, mu = 5.9 +/- 3.0 Pa s, n = 24). Upon treatment with CD, the viscoelastic coefficients of both hepatocytes and HCC cells decreased uniformly, with magnitudes for the decrease in elastic coefficients of HCC cells (K1: 68.7 to 81.7 N m-2, 66.3 to 78.9%; K2: 34.5 to 37.1 N m-2, 81.2 to 87.3%) larger than those for normal hepatocytes (K1: 42.6 to 49.8 N m-2, 48.7 to 56.9%; K2: 17.2 to 20.4 N m-2, 51.7 to 61.3%). There was a smaller decrease in the viscous coefficient of HCC cells (2.0 to 3.4 Pa s, 44.4 to 75.6%) than that for hepatocytes (3.0 to 3.9 Pa s, 50.8 to 66.1%). Upon treatment with Col and VBL, the elastic coefficients of hepatocytes generally increased or tended to increase while those of HCC cells decreased. The differences in either the pattern or the magnitude of the effect of cytoskeletal perturbing agent on the viscoelastic properties between HCC cells and hepatocytes might possibly reflect differences in the state of the cytoskeleton structure and function, or in the cells' sensitivity to perturbing agent treatment between these two types of cells. Changes in the viscoelastic properties of cancer cells might well affect tumor cell invasion and metastasis as well as interactions between tumor cells and their micro-mechanical environments.

Peri-implant mucosal inflammation and bone loss: clinical and radiographic evaluation of 108 dental implants after 1-year loading.

OBJECTIVE: To evaluate the clinical outcomes, and a possible correlation between peri-implant mucosal inflammation and marginal bone loss, 1 year after restoration in a group of Chinese patients with dental implants. METHODS: Observation of the clinical and radiographic parameters of 108 IMZ and Frialit-2 dental implants in 70 patients. RESULTS: One year after loading, there were no signs of mobility and peri-implant radiolucency in any of the implants. The average bone loss adjacent to implants was 0.63 mm (SD 0.78). The prevalence of peri-implant mucosal inflammation, as indicated by sulcus bleeding index (SBI) of 1.0 or more, was 32.9%. Significant positive correlation was found between mucosal inflammation and bone loss (P < 0.05). 92% of the observed sites had plaque index (PLI) scores greater than 1. CONCLUSIONS: The 1-year outcomes from this patient group indicate that IMZ and Frialit-2 implants provide satisfactory clinical results. One of the etiologic factors of alveolar bone loss around implants seems to be plaque-associated marginal inflammation. Improvement of oral hygiene, elimination of soft tissue inflammation, and proper treatment of periodontal disease of natural teeth are critical for maintaining the implants.

Changes in coagulation and tissue plasminogen activator after the treatment of cerebral infarction with lumbrokinase.

This paper aimed to investigate the effect of lumbrokinase on the anticoagulation and fibrinolysis in treating cerebral infarction. Lumbrokinase was used in patients with cerebral infarction. Patients were randomly divided into treatment group (n = 31) and control group (n = 20). Single blind method was used in this investigation. The Chinese stroke score was used to evaluate the results of treatment before and after administration of lumbrokinase. Kaolin partial thromboplastin time (KPTT), prothrombin time (PT), fibrinogen content, vWF content were analyzed, and tissue plasminogen activator (t-PA) activity, plasminogen activator inhibitor (PAI) activity, D-dimer level were assayed. In both groups, the stroke score decreased after administration, but in the treatment group, it was more obvious. In the treatment group, KPTT was prolonged, t-PA activity and D-dimer level increased, while the content of fibrinogen decreased significantly. There were no significant changes of PT and PAI activity in both groups. It is concluded that lumbrokinase is beneficial to the treatment of cerebral infarction. The effect of lumbrokinase is related to the inhibition of intrinsic coagulation pathway and the activation of fibrinolysis via an increase of t-PA activity.

[The effect of tetrahydropalmatine on ouabain-induced delayed after depolarization and triggered ventricular arrhythmia].

OBJECTIVE: To study the effect and mechanism of tetrahydropalmatine (THP) ouabain-induced on delayed after depolarization (DAD) and anti-triggered ventricular arrhythmia in in-vivo rabbit heart. METHODS: 14 rabbits were divided into two groups at random, control group and THP group. The monophasic action potential (MAP) of right ventricule and the surface lead II electrocardiogram (ECG) were simultaneously recorded. The changes of MAP and ECG in the THP group were compared with those in the control group after administration of ouabain 150-170 ug/kg and before using the drug. RESULTS: The incidence and couple interval of DAD in the two groups had no apparent difference (P > 0.05) after administration of ouabain, but the amplitude of DAD and the incidence of triggered ventricular arrhythmia in the THP group were significantly lower than those in the control group (P < 0.05). CONCLUSION: THP had the effect of decreasing the amplitude of DAD of rabbit heart induced by ouabain thus reducing the chance of triggered ventricular arrhythmia induced by DAD.

[The effects of hepatitis G virus infection on clinical features and liver pathologic lesions of chronic hepatitis C].

OBJECTIVE: To explore the clinical and pathological effect of hepatitis G virus infection on chronic hepatitis C. METHODS: Detecting HGV-RNA by reverse transcription-polymerase chain reaction from serum samples of 53 chronic hepatitis C through patients as diagnosed by liver biopsy. The clinical and pathologic features of the patients with positive HGV-RNA were compared with those of patients with negative HGV-RNA. RESULTS: The results showed that 15 patients (28.3%) were positive for serum HGV-RNA and there were no significant differences in clinical manifestations, biochemical indexes, HCV-RNA positive rates and the liver pathologic lesions between these HGY RNA positive and negative groups (P > 0.05). CONCLUSIONS: These data indicate that HGV coinfection does not affect the liver lesions and HCV replication of chronic hepatitis C.
