[FTIR study of transition metal 2-acrylamido-2-methyl-1-propanesulfonates].

Compounds of transition metal ion (Cu2+, Co2+, Ni2+) 2-acrylamidomethyl-2-methyl-1-propanesulfonates were studied by FTIR and X-ray powder diffraction techniques. The FTIR spectra of Co and Ni compound are similar but are different from that of Cu compound. The results of X-ray diffraction indicated that the crystal structure of Co and Ni compound are different.

[Effects of octreotide on implanted liver tumor after partial hepatectomy in rats].

OBJECTIVES: To investigate the effects of octreotide(OCT) on experimental live tumor after partial hepatectomy (PH) in rats and explore its intrinsic mechanism. METHODS: Sixty-four male rats were randomly divided into four groups and operated on with laparotomy. Walker-256 tumor was implanted in all the four groups. Group A served as the control, and groups B, C and D received left lobe PH. Octreotide at a dose of 50 microg/kg was intraperitoneally (i.p.) administered twice a day in groups C and D (OCT). The time of the first administration was at the 12th hour after PH in group C and the 72nd hour after PH in group D. The rats in groups A and B received a similar volume of normal saline (NS). RESULTS: Group A showed a significant reduction of implanted liver tumor volume at the 7th day versus group B (P < 0.01). Groups C and D showed a significant reduction of tumor volume at the 7th and 15th day versus group B (P < 0.01). The tumor cell apoptotic rate was significantly higher in groups C and D than in groups A and B at the 7th and 15th day (P < 0.01). CONCLUSION: The growth of experimental liver tumor is enhanced after PH bat can be inhibited by OCT.

Oxygen ion implantation at 20 to 2000 keV into polysulfone for improvement of endothelial cell adhesion.

The irradiation effects of oxygen on polysulfone have been investigated at energies of 20 keV, 150 keV and 2 MeV. The strong improvement of endothelial cell adhesion and proliferation is found on ion irradiated polysulfone at 20 keV. Such improvement is declined with increasing ion energy. The changes of surface color and free energy are strongly dependent on ion energy and dose. The formation of amorphous carbon phase is demonstrated by Raman spectroscopy and its degree is correspondent to the color changes observed. The formations of hydroxyl and carboxyl groups are confirmed by the attenuated total reflectance (ATR) FTIR spectroscopy. The depletions of heteroatoms are conjectured by detail analysis of X-ray photoelectron spectroscopy (XPS). Since no single one of these changes can be related directly to the improved adhesion and proliferation of endothelial cells on irradiated surface, we argue that the distribution of functional groups is crucial in promoting the adhesion of endothelial cells. Although the distribution cannot directly be detected at present, the irradiation effects were related to the results of TRIM simulation. The surface changes can be controlled by adjusting the size energy and dose of irradiating ion for the optimum morphology to cell adhesion.

Chemical changes of polymeric surfaces after modification with multi-source cluster deposition.

A new process for surface modification of polymers with multi-source cluster deposition apparatus has been reported in our previous work. The apparatus simultaneously supplies reactant of ammonium sulfamate and activator of energetic Ar(+) ion. In this work chemical changes are analyzed on the basis of XPS spectra and the relations of contact angle and platelet adhesion with chemical changes are discussed. Polymer film, setting on a turning holder, was irradiated by Ar(+) ions during bombardment with ammonium sulfamate clusters. The Ar(+) ion source served for activation of polymer surface and a cluster ion source supplied ammonium sulfamate molecules to react with activated surface. After thorough washing with deionized sterile water, the modified surfaces were evaluated in terms of contact angle of water, elemental composition and binding state on XPS and platelet adhesion with platelet rich plasma (PRP). The modification of polysulfone decreased the contact angle of water on surfaces from 82.6 down to 34.5 degrees. The adhesion number of platelets were decreased to one-tenth of the original surface. Ammonium, amine, sulfate and thiophene combinations were formed on the modified surfaces. The primary studies showed successful modification of polysulfone with ammonium sulfamate by assistance of Ar(+) ion irradiation. The polar groups like N-sulfate were formed on surfaces and contribute to the decrease of surface contact angle and adhesion number of platelets. Since the same process can also be applied to other polymeric materials with various substrates, combining with the features of no solvent and no topographic changes, this method might be developed in a promising way for modification of polymers.

Electrochemiluminescent detection of chlorpromazine by selective preconcentration at a lauric acid-modified carbon paste electrode using tris(2,2'-bipyridine)ruthenium(II).

A new detection scheme for the determination of adsorbable coreactants of Ru(bpy)3(2+) electrochemiluminescent reaction is presented. It is based on selective preconcentration of coreactant onto an electrode, followed by Ru(bpy)3(2+) electrochemiluminescent detection. The coreactant employed is chlorpromazine. It was sensitively detected after 5-min preconcentration onto a lauric acid-modified carbon paste electrode. The linear concentration range was found to occur from 1 x 10(-8) to 3 x 10(-6) mol L-1 with a detection limit of 3.1 x 10(-9) mol L-1. The total analysis time is less than 10 min. As a result of selective preconcentration and medium exchange, such remarkable selectivity is achieved that reproducible quantitation of chlorpromazine in urine is possible.

Cloning, characterization and chromosomal assignment of NBC4, a new member of the sodium bicarbonate cotransporter family.

We report the cloning, characterization and chromosomal assignment of a new member of the sodium bicarbonate cotransporter (NBC) family, NBC4, from human heart. NBC4 maps to chromosome 2p13 and is a new candidate gene for Alstrom syndrome. NBC4 encodes a 1074-residue polypeptide with 12 putative membrane-spanning domains. Unlike other members of the NBC family, NBC4 has a unique glycine-rich region (amino acids 438-485). In addition, NBC4 lacks the lysine-rich C-terminus of NBC1 with which it is most homologous. The first of two putative stilbene binding motifs (K(M/L)(X)K) is lacking in NBC4 (amino acids 655-658). The approximately 6 kb NBC4 transcript is moderately expressed in heart, with the highest expression in liver, testes and spleen.

Intermediates Produced in the Electron-Transfer Processes of Phenothiazine/Semiconductor Systems.

Phenothiazine radical cation PTH(+*) and phenothiazine dication PTH(2+) produced by photoinduced electron transfer in phenothiazine/semiconductor systems have been studied by using ESR, UV-visible absorption, and fluorescence spectroscopic methods. It is found that the PTH(+*) was generated by electron transfer from (1)PTH* and (3)PTH* to the conduction band of the semiconductors besides PTH(+*) resulted in the photoionization of PTH as well as PTH(2+) produced by electron transfer from PTH(+*) to the conduction band of the semiconductors successively. Very significant supplementary information provided by UV-visible absorption, ESR, and resonance Raman spectra of PTH(+*) and PTH(2+), which were obtained by oxidation of PTH with lead tetraacetate, not only confirmed the two-step mono-electron-transfer mechanism proposed for interpretation of electron-transfer processes in PTH/semiconductor systems but also demonstrated that the two successive electron-transfer steps were corresponding to the removal of a p-electron at the nitrogen atom and a p-electron at the sulfur atom in the PTH molecule, respectively. Consequently, the nonplanar configuration of the PTH molecule changed into a planar configuration of the PTH(2+). Copyright 2000 Academic Press.

Application of capillary electrophoresis in clinical chemistry: the clinical value of urinary modified nucleosides.

Urinary modified nucleosides were determined by capillary electrophoresis using a 300 mM SDS-25 mM sodium tetraborate-50 mM sodium dihydrogenphosphate buffer. The nucleosides were extracted from urine by phenylboronate affinity gel chromatography. In cancer patients the levels of the modified nucleosides are generally elevated. By an artificial neural network method breast cancer patients were differentiated from normal individuals, which indicates that the modified nucleosides could be of clinical value as tumor markers.

Holes in a Quantum Spin Liquid.

Magnetic neutron scattering provides evidence for nucleation of antiferromagnetic droplets around impurities in a doped nickel oxide-based quantum magnet. The undoped parent compound contains a spin liquid with a cooperative singlet ground state and a gap in the magnetic excitation spectrum. Calcium doping creates excitations below the gap with an incommensurate structure factor. We show that weakly interacting antiferromagnetic droplets with a central phase shift of pi and a size controlled by the correlation length of the quantum liquid can account for the data. The experiment provides a quantitative impression of the magnetic polarization cloud associated with holes in a doped transition metal oxide.

Preparative isolation and purification of notopterol and isoimperatorin from Notopterygium forbessi Boiss (Chinese traditional medicinal herb) by high-speed counter-current chromatography.

Preparative high-speed counter-current chromatography was successfully used for isolation and purification of coumarins from Chinese traditional medicinal herb Notopterygium forbessi Boiss (Qianghuo in Chinese) using stepwise elution with a pair of two-phase solvent systems composed of light petroleum-ethyl acetate-methanol-water at volume ratios of 5:5:4.8:5 and 5:5:5:4. Four major components including notopterol and isoimperatorin were isolated, each at over 98% purity.

[Dynamic study on blood and milk lead levels of pregnant women in three districts of Hubei].

In order to study the effect of lead exposure on the health during the period of pregnancy, 96 pregnant women and 105 non-pregnant women with no known occupational exposure to heavy metals from urban area(Wuhan), iron mining area(Daye) and rural area(Yidu) were selected. Blood and milk samples were collected under strict quality control and analyzed by flame atomic absorption spectrometer. The general status of subjects were collected by a medical interviewer. The results showed that the blood lead in the non-pregnant women were higher in the urban area, but the blood lead in the pregnant women and the milk (colostrum and mature milk) lead were lower in the urban area. It is concluded that the risk of exposure to low concentration of lead during pregnancy must not be ignored.

Evidence that reversed glutamate uptake contributes significantly to glutamate release following experimental injury to the rat spinal cord.

Released excitatory amino acids contribute significantly to secondary damage following spinal cord injury. Reversal of normal transport due to cell membrane depolarization may contribute to this release. We tested this by administering dihydrokainic acid (DHK), a non-transported glutamate uptake blocker, into the rat spinal cord by microdialysis in association with contusion spinal cord injury. Glutamate release in response to injury was reduced by 34% (P<0.05) when 3 mM DHK was administered within the microdialysis fiber, suggesting that reversed transport is an important contributor to glutamate release upon spinal cord injury.

Triplet waves in a quantum spin liquid

We report a neutron scattering study of the spin- 1/2 alternating bond antiferromagnet Cu(NO3)(2);2.5D(2)O for 0.06<k(B)T/J1<1.5. For k(B)T/J1<<1 the excitation spectrum is dominated by a coherent singlet-triplet mode centered at J1 = 0.442(2) meV with sinusoidal dispersion and a bandwidth of J2 = 0.106(2) meV. A complete description of the zero temperature contribution to the scattering function from this mode is provided by the single mode approximation. At finite temperatures we observe exponentially activated band narrowing and damping. The relaxation rate is thermally activated and wave vector dependent with the periodicity of the reciprocal lattice.

Effect of age on the formation and repair of UV photoproducts in human skin in situ.

Ultraviolet radiation (UVR)-induced photoproducts can be measured by a number of methods. The newly developed 32P-postlabelling method is feasible in molecular epidemiological studies due to its sensitivity, specificity and little amount DNA needed. We applied the 32P-postlabelling method to investigate the induction and repair of photoproducts (cyclobutane pyrimidine dimers (CPDs) and 6-4 photoproducts) after UVR in human skin in situ and studied the effects of age, skin type and gender. The study included 30 subjects aged 32-78 years. The photoproduct induction levels varied 7- to 15-fold between the individuals tested. All four types of photoproducts were induced at a higher frequency in the older population (>/=50 years) than in the younger population (<50 years). Individuals with skin type I and II had a higher CPD induction frequency than individuals with skin type III and IV. In both cases, the differences in thymidylyl (3'-5') thymidylyl (3'-5')-2'-deoxycytidine induction reached statistical significant levels (p<0.05). Photoproduct repair rates 24 h and 48 h after UV irradiation showed a large inter-individual variation. No clear effects of age, skin type or gender on DNA repair could be detected. Our data suggest that UV-induced DNA photoproduct levels increase with age.

Protection of nonobese diabetic mice from diabetes by gene(s) closely linked to IFN-gamma receptor loci.

Nonobese diabetic (NOD) mice carrying a segment of chromosome flanking the disrupted IFN-gamma receptor gene from original 129 ES cells are resistant to development of diabetes. However, extended backcrossing of this mouse line to the NOD mouse resulted in a segregation of the IFN-gammaR-deficient genotype from the diabetes-resistant phenotype. These results indicate that the protection of NOD mice from the development of diabetes is not directly linked to the defective IFN-gamma receptor gene but, rather, is influenced by the presence of a diabetes-resistant gene(s) closely linked to the IFN-gammaR loci derived from the 129 mouse strain.
