Investigation of a corticosteroid-binding globulin-like molecule in human placenta.

The existence of an endogenous corticosteroid-binding globulin (CBG)-like molecule in human placenta has been reported in studies which depended largely on immunological techniques. The experiments reported herein do not support that contention. Using a well characterized antibody to purified human CBG, we were unable to demonstrate CBG in placental cytosol. Although the immunological methods were too insensitive to detect CBG, the presence of small amounts of it were detected in binding experiments with [3H]cortisol. Simultaneous measurement of serum albumin indicated that the presence of CBG in the placental cytosol could be accounted for by contamination of the preparation with plasma.

Galactosyl transferase activity in human transitional cell carcinoma lines and in benign and neoplastic human bladder epithelium.

Cultured cells of human transitional cell carcinoma line MGH-U1, in suspension, were assayed for galactosyl transferase by measurement of the transfer of [3H]galactose from uridine diphosphate:[3H]galactose to desialylated ovine submaxillary mucin. The assay was optimized with respect to time and to protein, uridine disphosphate:galactose, desialyated ovine submaxillary mucin, and Triton X-100 concentrations. This assay was then applied to fresh specimens of benign, inflamed, and neoplastic bladder epithelium from 33 patients who under went cold-cup biopsies at cytoscopy. Transitional cell carcinoma specimens gave values in the range of 24.7 to 184.8 cpm [3H]galactose transferred per microgram protein per hr [72.0 +/- 44.7 (S.D.); n = 25]; normal and inflamed specimens ranged from 0.8 to 46.1 cpm/microgram protein per hr [8.3 +/- 8.4 (S.D.); n = 35]. By using a known method of cell rupture, cell ghosts, representing cell-surface membranes, were isolated both from the cultured cell line and from two biopsy specimens of transitional cell carcinoma. Although a complete enzymatic and electron microscopic analysis was not undertaken, the coincidence of an enzyme marker with the cell ghost fraction containing the elevated galactosyl transferase made it appear probable that this enzyme is located in the cell surface.

[Synthesis of the fragments A1-,, A9-15 and A16-21 of ovine insulin A chain using the S-tert-butylmercapto residue for thiol protection (author's transl)].

The following paper describes the synthesis of the fragments A1-8, A9-15 and A16-21 of the ovine insulin A chain using the S-tert-butylmercapto residue for thiol protection. The synthesized fragments, which showed a good solubility in organic solvents, were partially deprotected with trifluoracetic acid and converted to the corresponding S-sulfonates quantitatively by oxidative sulfitolyses at pH 7.6.

[Synthesis of (LysA13)bovine insulin A chain analogs as (Lys(Tfa)A13)A(SO3H)4 and Nalpha-A1-Msc-(LysA13)A(SO3H)4 derivatives using the S-tert-butylmercapto residue for thiol protection (author's transl)].

The following paper describes the synthesis of the [LysA13]bovine insulin A chain analog as [Lys(Tfa)A13]A(SO3H)4 and NalphaA1-Msc-[LysA13]A(SO3H)4 derivatives using the S-tert-butylmercapto residue for thiol protection. Although the intermediate S-tert-butylmercaptocysteinyl-peptide derivatives showed a good solubility in organic solvents the resulting fully protected A chain derivatives had a poor solubility in organic solvents and therefore were deblocked converted into the tetra(S-sulfonic acid) derivatives and purified via ionexchange chromatography.

Recent evidence for the participation of vitamin A in glycoprotein synthesis.

Recent evidence supports the concept that vitamin A plays some role in glycoprotein synthesis in a large-variety of tissues examined. Its involvement may be through participation of a retinol-linked sugar, mannosyl retinyl phosphate (MRP). Upon injection of [3H]retinol and [14C]mannose into rats, [14C, 3H]MRP could be isolated from liver and intestinal mucosa, and identified by chromatographic and hydrolytic experiments. The enzyme system that forms MRP from GDP-mannose and retinyl phosphate was located primarily in rough endoplasmic reticulum of fractionated liver cells, with some activity also in smooth membranes and Golgi apparatus. Vitamin A deficiency resulted in depressed synthesis of the rat serum glycoprotein alpha 1-macroglubin (alpha 1-MG), as shown by a decline in labeling. Analysis of the labeled alpha 1-MG from serum of normal and vitamin A-deficient rats showed this to be the result of a defect in glycosylation. The specific activity ratio (deficient:normal) of the alpha 1-MG of serum declined progressively with development of the deficiency, as a result of underglycosylation. Complete carbohydrate analysis of the alpha 1-MG of normal and deficient serum revealed a sugar loss in this glycoprotein as a result of vitamin A deficiency.

Test for oral and postingestional factors mediating differential acceptability of morphine, methamphetamine, and chlordiazepoxide drinking solutions.

The relative degrees to which aversive oral (presumably gustatory) and postingestional variables attenuate the voluntary drinking of morphine, methamphetamine, and chlordiazepoxide solutions by rats were assessed by comparing immediate acceptability with subsequent acceptability. Results indicated that morphine solutions (0.01--0.05%) are avoided primarily because of unpalatability, while methamphetamine (0.005--0.02%) and chlordiazepoxide solutions (0.02--0.08%) are avoided primarily because of aversive postingestional effects.

The influence of a new antitumor agent on hemopoietic colony forming cells.

The cytostatic and immunsuppressive agent N'-methyl-N'-beta-chloroethylbenzaldehyde hydrazone (B1) in in-vitro experiments has a stimulating effect on colony-forming culture (CFUc) of bone marrow from C57BL mice. This unusual behaviour, which is in contrast to other cytostatics, could also be observed in vitro with CFUc obtained from mice treated with therapeutic doses of B1 for 2 weeks. This stimulation is not a particular effect of B1 alone but seems to depend on a synergistic effect of the combination of B1 and the colony-stimulating activity (CSA) present in the serum from endotoxin-treated mice (MP) in the testing system. The results suggest that the described effect of B1 is due to an interference at the cell membrane of CFUc or their precursor cells.

Escape from deficits in sodium intake after thalamic lesions as a function of preoperative experience.

Saline and water intakes in response to treatments with a natriuretic agent and a mineralocorticoid were studied in intact rats and in rats with lesions centered in the taste delay of the thalamus. Intact rats responded to the treatments by increasing both saline and water intake. Rats with thalamic lesions that had never drunk saline prior to induction of the lesions generally did not increase their saline intake in response to the treatments but showed normal increases in water intake. In contrast, rats with thalamic lesions that had drunk saline prior to induction of the lesions showed normal increases in both saline and water intake. Preoperative experience of sodium need did not protect rats against the lesion-induced deficit.

Work settings and job satisfaction: a study of primary care physicians and paramedical personnel.

This paper examines job satisfaction of primary care physicians and paramedical personnel in traditional office practices and in modern medical organizations. A series of two-way analyses of variance using work settings and occupational level as independent variables showed consistent effects of setting on job satisfaction. In modern organizational settings satisfaction with the work activity, with coworkers, and with income was lower than in traditional practices. Only with respect to income, were physicians, on the average, more satisfied than paramedical workers. The relationships between job satisfaction and the variety and complexity of work as well as ownership of work settings are analyzed and discussed.

SV40-transformed hamster cells resistant to 100--250 microgram/ml of ethidium bromide.

SV40-transformed hamster cells were selected for resistance to ethidium bromide (EB). Several cell lines were established, which grew in the presence of up to 250 microgram/ml EB. The EB resistance is genetically stable. The cloned resistant cells show no difference in morphology, with the exception of the mitochondrial ultrastructure, which exhibits condensed cristae formation. The tumorigenicity of these cells in Syrian gold hamsters is considerably reduced. Incorporation of radioactive labelled thymidine into mitochondrial DNA is not influenced by the presence of the drug. Gel electrophoresis with mitochondrial proteins from wild-type and resistant cells reveals significantly different patterns. The mechanism of EB resistance is discussed.
