[The effect of antibody against vascular endothelial growth factor (VEGF) on tumor metastasis].

OBJECTIVE: To study whether manipulation of angiogenesis with antibody against vascular endothelial growth factor (VEGF) may interrupt the tumor growth and metastasis. METHODS: IV TA2MA 891 is a murine spontaneous breast cancer with high rate of metastasis in lung in (TA2x615) F1 mice, and it was chosen as an animal model in this study because of the high expression of VEGF in primary tumor as well as in lung metastatic tumor. The mice were injected with anti-VEGF polyclonal antibody at a dose of 200 micrograms/mouse/day from day 9 to 23 following s.c. tumor inoculation and sacrificed on day 26. RESULTS: The growth of primary tumor was inhibited by 44.0% and the number and size of metastatic foci in the lungs were inhibited by 73.0% and 83.7% respectively. CONCLUSION: The antibody against human VEGF may be a potentially effective antimetastasis agent for the treatment of patients with lung metastasis.

[The effect of antibody against vascular endothelial growth factor on tumor growth].

OBJECTIVE: Vascular endothelial growth factor (VEGF) is a very important factor in the process of angiogenesis, which is chosen as a target for experimental study of anticancer. METHODS: Anti-VEGF antibody was obtained from immunized rabbits and purified with affinity column. Morever, it was identified as neutralized antibody by Mile's assay. RESULTS: The anti-VEGF antibody was able to inhibit growth of S180 sarcoma in a dose-dependent manner, and the inhibition rate reached 41.0% with dose 200 micrograms per mouse per day. For human gastric cancer (MGC-803)-bearing nude mice, Anti-VEGF antibody inhibited tumor growth by 76.2%. When anti-VEGF antibody was combined with 131I-3H11, a murine monoclonal antibody conjugated with 131I for treatment, only one of five nude mice developed tumor and 84.0% more inhibition of tumor growth was obtained in comparison with treatment by 131I-3H11 only. CONCLUSION: The anti-VEGF antibody may be potentially useful for clinical treatment of cancer.

[Expression of human 17 beta-hydroxysteroid dehydrogenase gene in BmNPV expression system].

Estrogenic 17 beta-hydroxysteroid dehydrogenase (17 beta-HSD) plays a crucial role in the synthesis of estrogens, but it also produces negative action of estrogens in promoting the growth of hormone-sensitive cancers, especially in breast and prostate. The high specific activity can be taken as an important signal for the diagnosis of cancers. Recombinant rAcBm-NPV/17 beta-HSD virus which contains the human 17 beta-HSD cDNA under the control of polyhedron gene promoter is generated by cotransfection of the BmN cells with the transfer plasmid pVL/17 beta-HSD and wild BmNPV genomic DNA. 17 beta-HSD is maximally expressed 72 h and 120 h post infection in BmN cells and the 5th instar silkworm larvae respectively. At those time interval, intracellular and hemolymphic enzymatic activity reach 0.12 U/mg and 0.15 U/mg of protein which produced total activity of 0.97 U/1.5 x 10(6) cells and 4.7 U/larva. The expressed quantities in female larvae are a little higher than that in male larvae. The present data shows that Silkworm/BmNPV expression system can express 3-5 times higher than that of the richest human placenta. It also indicates that there is an apparent band with a molecular mass of 35 kDa using SDS-PAGE method, the size of which is similar to that of the crude enzyme from placenta.

[The effect of pH on anti-corrosion nature of amalgams].

Corrosion behavior of alloys are affected not only by themselves but also by environment, such as a change of pH. In this study, the corrosion behavior of four dental amalgams (L, C, DA, GK) in Fusayama saliva with different pH 6.65 and 4.00 are evaluated by electrochemical techniques. It seems that changes in pH may exert different effects on the four amalgams. In the acid environment DA exhibits best anti-corrosion nature and followed by GK, C, and L, which conforms to clinical observations. This may be resulted either from amalgams themselves or from conditions of corrosion products of amalgams' surface. Thus, consideration must be given to the changing oral environment when screening anti-corrosion alloys.

[Effects of HE particles on medicinal plant Agastache rugosus (Fisch. et Mey.) O. Ktze].

The biological effect of HE particles on the seeds of Agastache rugosus was probed in experiments on board a retrievable satellite. The result shows that the germination rate of the seeds pierced by HE particles radiation appears rather low. The seeds hit by HE particles (piercing radiation or not) start to germinate two days earlier than those in the control group, and the first leaf emerges four to five days earlier than that in the control group. The resultant seedlings grow markedly faster. Variations take place in the nuclear types of chromosome. The yield of essential oils becomes slightly higher. No marked changes have been observed in the major chemical components of these oils.

[Clinial study of "qingjunyin" detoxification for the treatment of heroin addicts].

To evaluate the efficacy of treatment of heroin addicts (n = 100) by "Qingjunyin" (QJY) detoxification (10 days program). Methadone detoxification (10 days program) group (n = 50) and colonidine treated group (n = 50) serve as controls. QJY group is given 180 ml P. O daily. The dosage of the two comparatine groups is given according to Documentation, the method of abserving the treatment protocols and detoxification standard is according to current regulation. The scores of abstinence syndrome in QJY detoxification group are lower than those in colonidine treated group in the first three days of potocol, but this difference disappear in the late stage of treatment. While QJY detoxification is effective as methadone detoxification in the control of abstinece syndrome during the first five days of treatment but the difference in the scores of abstinence syndrome between QJY and methadone group is observed during the late five days of protocol. QJY does not result in potential dependence and has definite curative effect in the treatment of heroin addiction.

[Identification of G6PD gene mutations in several nationalities from Yunnan province].

OBJECTIVE: To analyze the G6PD gene mutations in several nationalities from Yunnan province. METHODS: G6PD gene mutation of 29 samples from G6PD deficient patients were analyzed by using common and mis-matched primering PCR followed by restriction enzyme digestion. The samples were collected from Yunnan province among several nationalities. RESULTS: In these 29 cases, 20 were of two known mutations, including 19 of 1388 (G-->A) and 1 of 1376 (G-->T) mutation. The 7 cases of national minorities, including 3 of Yi ([symbol: see text]), 2 of Bai ([symbol: see text]), 1 of Dai ([symbol: see text]), and 1 of Naxi ([symbol: see text]), were all 1388 mutations. No report of the G6PD gene 1388 mutation in these ethnic groups was published excepting for Dai. CONCLUSION: The mutation probably had been prevalent before the emergence of these national minorities. The approximate rate of G6PD gene mutations provided in Yunnan province may, in some respects, help to interpret the molecular evolution.

Cyclin-dependent kinase inhibitor expression in pulmonary Clara cells transformed with SV40 large T antigen in transgenic mice.

Expression of cell cycle regulatory genes in mouse lung was investigated in transgenic models for Clara cell transformation. Clara cells were transformed by generating transgenic mice in which the SV40 large T antigen was expressed under the control of the mouse Clara cell M(r) 10,000 protein promoter. The resulting lung tumors express the large T antigen in normal Clara cells and in tumors, and these tumors express reduced levels of CC10 mRNA. The expression of cell cycle regulatory protein, p53, and the cyclin-dependent kinase inhibitors was analyzed by Northern blot analysis and in situ hybridization throughout the progression of Clara cell transformation in the lung. Increases in specific cyclin-dependent kinase inhibitor steady-state mRNA levels were detected in p15, p18, p27, and p57 during tumor progression. The expression of p15, p57, and p21 mRNAs were verified by in situ hybridization. Using this approach, regulatory genes have been identified that may be involved in the regulation of Clara cell differentiation.

[Sweat gland carcinoma of the vulva, report of 7 cases].

OBJECTIVES: To report the clinical and pathologic features and to evaluate the treatment results of 7 cases of sweat gland carcinoma of the vulva. METHODS: The clinical records of the 7 cases of sweat gland carcinoma of the vulva, admitted to our hospital for treatment between December, 1974 and December, 1994, were reviewed. The data including clinical and pathologic features, treatments and posttreatment follow-ups. Five of the 7 cases were referred from other hospitals, 4 followed by local resection and 1 with recurrence 7 years after surgery. RESULTS: 5/7 of the cases were alive for more than 5 years. One of the 5 cases recurred twice at 7 and 11 years after the initial treatment and each time remedied by radiotherapy, but eventually died of tumor with pulmonary metastasis at the 17th year. Two died within 5 years after treatment; one from cerebral hemorrhage and the other from extensive cancer metastasis 1 year and 9 months after surgery. CONCLUSIONS: The prognosis of sweat gland carcinoma of the vulva is fairly good. Wide local resection of the vulva or radical vulvectomy is the treatment of choice.

[Effects of oxidatively modified low density lipoprotein and very low density lipoprotein on the expression of MCP-1 in macrophages].

OBJECTIVE: To examine whether oxidatively modified low density lipoprotein (OX-VLDL) and very low density lipoprotein (OX-VLDL) induce the expression of MCP-1 mRNA and protein by rabbit peritoneal exudate macrophages, and to clarify what a role of both lipoproteins play in atherogenesis. METHODS: After exposure of the macrophages to 25 micrograms/ml of LDL, VLDL, OX-LDL and OX-VLDL respectively, and a 24 hour incubation at 37 degrees C, the total RNA was extracted from the cells by guanidinium isothiocyanate method, and the media conditioned by the cultured macrophages were collected. Meanwhile, MCP-1 protein in the conditioned media was determined by using sandwich ELISA. Monocyte migration induced by the media was assayed by micropore filter method using modified Boyden chamber. RESULTS: After a 24 hour exposure to OX-LDL and OX-VLDL, the MCP-1 mRNA expression in macrophages was markedly increased (3.2-fold and 3.4-fold, respectively), and the level of MCP-1 protein was also increased (2.2-fold and 2.5-fold, separately), and furthermore, the monocyte migration distance was significantly increased. However, the expression of MCP-1 mRNA and protein was only slightly increased when exposed to LDL or VLDL. CONCLUSIONS: Rabbit peritoneal exudate macrophages can express MCP-1 mRNA and protein, and OX-LDL and OX-VLDL induce stronger MCP-1 mRNA and protein expression in the cells.

Effects of removing circulatory tumor necrosis factor by immunoadsorption on experimental endotoxin shock animals.

OBJECTIVE: To evaluate the effects of removing circulatory tumor necrosis factor (TNF) by immunoadsorption on endotoxin shock animals. METHODS: Sixty New Zealand white rabbits were injected intravenously with lethal dose of endotoxin (10 Billion cfu/kg E. Coli endotoxin) and randomly divided into 3 groups: perfusion group, hemoperfusion started at 1 hour after injecting endotoxin through immunoadsorbent columns against TNF; pseudoperfusion group, hemoperfusion through blank columns; and control group, injected with endotoxin only. The arterial pressure, microcirculation of the mesentery, plasma levels of TNF, IL-1, IL-6, IL-8, nitrite, endothelin-1 (ET-1), alanine aminotransferase (ALT), aspartate aminotransferase (AST), creatinine were measured and analyzed and finally the survival rate was observed. RESULTS: Plasma levels of TNF were sharply reduced after immunoadsorption. Moreover, release of IL-1, IL-6, IL-8, NO and ET-1 were also attenuated. Hemodynamic abnormalities could be improved and survival rate ameliorated significantly. CONCLUSION: Specific immunoadsorption of circulating TNF might be a new and effective therapy for endotoxin shock.
