Serum leptin levels and adiposity in adult Chinese: a preliminary observation.

OBJECTIVE: To establish normal serum leptin levels in Chinese and investigate the relationship between serum leptin levels and body fat, gender, age and androgen. METHODS: Serum leptin levels were measured in 77 lean (BMI < 25) and 28 overweight or obese (BMI > or = 25) subjects by a radioimmunoassay (RIA) method. RESULTS: The serum leptin levels in lean Chinese were 2.15 +/- 1.46 ng/ml in male and 7.85 +/- 3.60 ng/ml in female, which are similar to those of Caucasians, while in overweight or obese ones, the levels were 4.87 +/- 3.47 ng/ml and 16.59 +/- 6.92 ng/ml respectively, lower than those in Caucasians. A 2-3 times higher leptin concentrations were found in women than in men in both conditions. Even when the number of lean males was expanded to 79 subjects aged from 17-80 years, no significant leptin-age relationship was found. Despite 25% of obese subjects manifested a relative deficiency of leptin, as a whole, leptin levels in both men and women were significantly correlated with BMI (r = 0.69, P < 0.001 in male and r = 0.63, P < 0.001 in female). CONCLUSIONS: Serum leptin levels in Chinese lean people are similar to those in Caucasians and in both lean and obese groups, the leptin levels are correlated with BMI, suggesting that the great majority of the obese patients are resistant to endogenous leptin. Those who are relatively deficient of leptin may become a group of good candidates for leptin treatment in the future.

[Effect of zinc-intensified milk powder on hematopoiesis in mice].

OBJECTIVE: To observe the effect of Zn-intensified milk powder and zinc on hematopoiesis in mice. METHODS: Kunming mice were randomly divided into four groups (n = 9 each) (control group; milk powder group; equal zinc group; high zinc group) and fed on experimental diet for 24 weeks. RESULTS: 1. In high zinc group: the mice hemoglobin (Hb) declined (P < 0.05) after 12 wk of supplementation and were significantly lower (P < 0.01) than those in control group at 16 wk. At 24 wk, the mice showed lower Hb, red blood cell(RBC), hematocrit (HCT), and mean corpuscular volume (MCV) compared with control group (P < 0.05). Bone marrow aspirates revealed erythroid hyperplasia. Zinc concentrations in serum, liver and kidney were higher (P < 0.01) and copper was lower than that of control group(P < 0.01). 2. At 16 wk, no change occurred in milk powder group and equal zinc group. At 24 wk, parameters changed in milk powder group similar to those in high zinc group, and in equal zinc group liver and kidney copper was lower than that in control group (P < 0.05). CONCLUSION: Ingestion of over-dosed zinc could induce anemia. Generally, intake of Zn-intensified milk powder is safe, but long-term intake of the milk powder may interfere with hematopoiesis.

[Study on serotyping of Haemophilus influenzae and the positive rates of beta-lactamase].

OBJECTIVE: To investigate the serotypes of both infectious and carriage strains of Haemophilus influenzae (Hi) and to evaluate the prevalence of beta-lactamase of major serotypes in Shanghai. METHODS: The 534 specimens of sputum and swabs from patients with acute upper or lower respiratory tract infection and 255 cases of control group were collected and incubated on improved Collumbia chocolate medium. All isolated Haemophilus influenzae strains were serotyped by slide agglutinating assay or reverse indirect blood agglutinating method. beta-lactamase were measured by iodine test and paper disc method. RESULTS: In the patient group, of the 164 Hi strains there were 69 Hib strains (42%, 69/164) and 91 NTHi(56%, 91/164). In the control group, there were 14 strains of Hib and 49 strains of NTHi among 66 strains of Hi, which were isolated from 255 specimens(26%, 66/255). 24%(38/160) of Hi in the patient group produced beta-lactamase. In this group, the positive rate of beta-lactamase was 29%(20/69) for Hib strains and 20%(18/91) for NTHi strains. Sixteen strains resisting to ampicillin were beta-lactamase negative. CONCLUSIONS: These results indicate that Hib and NTHi are the major serotypes of Hi in both the patient and control group. The prevalence of Hib is higher in patients than in the control. Hib strains are more likely to produce beta-lactamase than NTHi.

[Studies on the spectrum features of nasalized vowels].

OBJECTIVE: To study the spectrum features of nasalized vowels and set up effective indexes to reflect the features. METHODS: The acoustic features of 100 children's speech, aged 5 to 12 years old, consisted of 40 unrepaired cleft palate children, 28 VPI children and 32 normal children, were studied by the application of digital spectrograph. RESULTS: No significant changes in the frequency of inherent formants of each vowel, appearance of one relatively fixed extra formant in the low frequency area, and the energy of high frequency area being damped obviously were the main spectrum features of nasalized vowels. CONCLUSION: There are three main features on the spectrums of nasalized vowels compared to that of oral vowels. Nasal resonance index (NRI) can effectively distinguish the extra formant in low frequency area of oral vowel /a/ from nasalized vowel /a/. Energy damping index (EDI) can effectively reflect that the energy of high frequency area of each nasalized vowel is damped obviously.

[An experimental study on the effect of azithromycin treatment in bleomycin-induced pulmonary fibrosis of rats].

OBJECTIVE: To evaluate the therapeutic effects and mechanism of azithromycin treatment in bleomycin-induced pulmonary fibrosis of rats. METHODS: Animal model of bleomycin-A5-induced pulmonary fibrosis was established in rats.36 animal models were divided into two groups: a bleomycin-induced pulmonary fibrosis group and a azithromycin group in which the animal models were treated with azithromycin (80 mg/kg once a day for three continuous days in a week). The animals of the two groups were killed at the first, second and fourth week respectively. Another six rats constituted a normal control group, instillated withsaline intratracheally and killed at the first week. Pathological changes activity of nuclear factor kappaB (NF-kappaB) of alveolar macrophage, cytokine tumor necrosis factor (TNF) alpha, transforming growth factor-beta (TGF-beta) mRNA expression and protein levels of alveolar macrophage and lung tissue were studied or measured. RESULTS: Amelioration of alveolitis and lung fibrosis after treatment with azithromycin was shown in pathological section (P < 0.05). The activity of NF-kappaB was significantly higher in one-week pulmonary fibrosis model than that in normal control and its level in alveolar macrophage reduced (67.2%) after treatment with azithromycin. The level of protein and mRNA of TNFalpha, TGF-beta in lung tissue and alveolar macrophage was increased in the early stage of pulmonary fibrosis and reduced after treatment with azithromycin (P < 0.05). CONCLUSION: It is suggested that azithromycin might be a therapeutic drug for pulmonary fibrosis in the future. Azithromycin reduced the degree of alveolitis and fibrosis through inhibition of the activity of NF-kappaB and the expression of TNFalpha, TGF-beta mRNA and lowering the level of protein in alveolar macrophage and lung tissue in the early stage of pulmonary fibrosis. This might be one of the mechanisms of azithromycin treatment in pulmonary fibrosis.

[The effect of inhaled ipratropium bromide on airway and lung tissue muscarinic receptors in a rat model of COPD].

OBJECTIVE: To observe the effect of inhaled ipratropium bromide on airway and lung tissue muscarinic receptors (MR) in a rat model of chronic obstructive pulmonary disease (COPD). METHODS: The rat model of COPD was established by chronic exposure with high concentrations of SO(2) gas, COPD rats inhaled aerosolized 0.025% ipratropium bromide solution 10 ml (twice/d, 20min/time). The MR was determined by radioligand binding studies with (3)H-QNB as the ligand. RESULTS: Compared with that of control, there was a marked increase in MR density on day 30 of treatment with ipratropium but not on day 5. The MR density decreased and close to normal level on the 6th day after cessation of regular therapy with inhaled ipratropium for 30 days. No significant difference was observed in antagonist affinity (Kd) among different time groups of ipratropium therapy. CONCLUSIONS: Chronic exposure to the anticholinergic agent ipratropium bromide produces an up-regulation of airway MR. The airway MR density can recover to normal when treatment stopped. Up-regulation of airway MR is associated with transient bronchoconstriction following cessation of regular ipratropium therapy and lung function deteriorated following long-term inhaled ipratropium bromide.

[Experiment and clinical application of targeting treatment with adriamycin magnetic albumin microspheres in human gastric carcinoma].

OBJECTIVE: To study the pharmacokinetics in experimental animals and observe the concurrent anticancer effects in human advanced gastric cancer as an adjuvant chemotherapy, when adriamycin magnetic albumin microspheres (ADM-MAM) were combined with external static magnetic fields. METHODS: The drug concentration of targeting tissues in the animals and the stability of ADM-MAM in human gastric juices were determined. Clinical and histopathological changes were observed in 55 cases of advanced gastric carcinomas after targeting treatment. RESULTS: The peak concentrations of the targeting tissue were highest after ADM-MAMs were administered for 2 hours and remained at the high level for a long time. ADM-MAM maintained its stability in human gastric juices. Targeting treatment improved the patients' symptoms, raised the rated of tumor resection, prolonged patients' lifetime, and enhanced histopathological effects (P < 0.01). CONCLUSIONS: The administration of ADM-MAM combined with the external static magnetic fields is effective for targeting location and is of clinical value as a preoperative adjuvant chemotherapy for human advanced gastric carcinoma.

[A study on the relationship between lymphocyte micronucleus rates and blood plasma benzo(a)pyrene levels in coking workers].

OBJECTIVE: To study the relationship between lymphocyte micronucleus rates and blood plasma levels of benzo(a)pyrene [B(a)P] in coking workers. METHODS: One hundred coking workers and 100 unexposed workers were selected, and their lymphocyte micronucleus rates and plasma levels, as well as workplace air concentrations, of B(a)P were determined. RESULTS: It showed that there was significant difference in plasma B(a)P level between coking workers and unexposed workers, and it correlated with air concentration of B(a)P in the workplace. Positive rates of micronucleus and rates of micronucleus lymphocyte were 60% and 1.79per thousand in coking workers and 29% and 1.10per thousand in controls, respectively, with very significantly statistical difference. It also showed that positive micronucleus rate correlated with plasma level of B(a)P in dose-effect pattern. Multivariate analysis showed that factors contributing to positive micronucleus rate ranked in such an order: plasma level of B(a)P, economic status, regularity of their lifestyle and smoking status of the coking workers. CONCLUSION: Accumulation of B(a)P in blood plasma played a major role in the formation of lymphocyte micronucleus. Results of micronucleus test in coking workers could reflect preliminary damage to their health caused by it.

[Production and identification of anti-paraoxon monoclonal antibodies].

Paraoxon (E600) was conjugated to bovine serum albumin(BSA) or tachypleus tridentatus hemocyanin (TTH) by diazotization. Two hybridoma cell lines secreting monoclonal antibodies(McAb) against paraoxon have been established by fusing mouse myeloma cells and splenocytes from Balb/c mice immunized with E600-BSA. The chromosomes of the hybridoma cell 2B10 were analyzed. The immunoglobulin of the McAb was classified. The affinity and specificity of this antibody were determined. The hybridoma cells have fairly reserved the antibody-producing capacity after continuously growing or stored in liquid nitrogen for 10 weeks.

[Effect of fluoride-arsenic exposure on the neurobehavioral development of rats offspring].

In order to explain the effects of fluoride-arsenic exposure on the neurobehavioral development of rats, a two generations-one nest reproductive test was used. Wistar rats were exposed to fluoride and arsenic for two generations. The results demonstrated that: with the increasing concentrations of fluoride and arsenic, the positive rates of early physiological development and nerve reflex were decreased obviously, and the abilities of learning and memory of the offspring were decreased, the swimming and enduring anoxia time gradually shortened. The pathological changes of brain under electron microscope were as follows: decreased number of nerve cells, irregular cell nucleus, organellae degeneration, and obvious decreased number of synapses. It was demonstrated that fluoride-arsenic displayed distinct damage on the nerve system of the offspring.
