Rsp5 ubiquitin-protein ligase mediates DNA damage-induced degradation of the large subunit of RNA polymerase II in Saccharomyces cerevisiae.

Rsp5 is an E3 ubiquitin-protein ligase of Saccharomyces cerevisiae that belongs to the hect domain family of E3 proteins. We have previously shown that Rsp5 binds and ubiquitinates the largest subunit of RNA polymerase II, Rpb1, in vitro. We show here that Rpb1 ubiquitination and degradation are induced in vivo by UV irradiation and by the UV-mimetic compound 4-nitroquinoline-1-oxide (4-NQO) and that a functional RSP5 gene product is required for this effect. The 26S proteasome is also required; a mutation of SEN3/RPN2 (sen3-1), which encodes an essential regulatory subunit of the 26S proteasome, partially blocks 4-NQO-induced degradation of Rpb1. These results suggest that Rsp5-mediated ubiquitination and degradation of Rpb1 are components of the response to DNA damage. A human WW domain-containing hect (WW-hect) E3 protein closely related to Rsp5, Rpf1/hNedd4, also binds and ubiquitinates both yeast and human Rpb1 in vitro, suggesting that Rpf1 and/or another WW-hect E3 protein mediates UV-induced degradation of the large subunit of polymerase II in human cells.

Novel, non-radioactive, simple and multiplex PCR-cRFLP methods for genotyping human SP-A and SP-D marker alleles.

We have previously identified an allele of the human SP-A2 gene that occurs with greater frequency in an RDS population [12]. Because of the importance of SP-A in normal lung function and its newly emerging role in innate host defense and regulation of inflammatory processes, we wish to better characterize genotypes of both SP-A1 and SP-A2 genes. It has been determined that SP-D shares similar roles in immune response. Therefore, in this report we 1) describe a novel, non radioactive PCR based-cRFLP method for genotyping both SP-A and SP-D; 2) describe two previously unpublished biallelic polymorphisms within the SP-D gene; 3) present the partial sequence of one new SP-A1 allele (6A14) and describe other new SP-A1 and SP-A2 alleles; and 4) describe additional methodologies for SP-A genotype assessment. The ability to more accurately and efficiently genotype samples from individuals with various pulmonary diseases will facilitate population and family based association studies. Genetic polymorphisms may be identified that partially explain individual disease susceptibility and/or treatment effectiveness.

Feline immunodeficiency virus vectors persistently transduce nondividing airway epithelia and correct the cystic fibrosis defect.

Several problems limit the application of gene transfer to correct the cystic fibrosis (CF) Cl(-) transport defect in airway epithelia. These include inefficient transduction with vectors applied to the apical surface, a low rate of division by airway epithelial cells, failure of transgene expression to persist, and immune responses to vectors or vector-encoded proteins. To address these issues, we used a feline immunodeficiency virus-based (FIV-based) vector. FIV vector formulated with a calcium chelator transduced fully differentiated, nondividing human airway epithelia when applied to the apical surface. FIV-based vector encoding the cystic fibrosis transmembrane conductance regulator cDNA corrected the Cl(-) transport defect in differentiated CF airway epithelia for the life of the culture (>3 months). When this approach was applied in vivo, FIV vector expressing beta-galactosidase transduced 1-14% of adult rabbit airway epithelia. Transduced cells were present in the conducting airways, bronchioles, and alveoli. Importantly, gene expression persisted, and cells with progenitor capacity were targeted. FIV-based lentiviral vectors may be useful for the treatment of genetic lung diseases such as CF. This article may have been published online in advance of the print edition.

KGF and FGF-10 stimulate liquid secretion in human fetal lung.

During fetal life, the pulmonary epithelium secretes liquid that distends the airways and is important for normal lung growth and development. The factors regulating human fetal lung liquid secretion are poorly understood; however, recent studies in murine models show that keratinocyte growth factor (KGF, FGF-7) and fibroblast growth factor 10 (FGF-10) stimulate liquid secretion. We asked whether KGF and FGF-10 stimulate liquid secretion in human fetal lung. First trimester fetal lung explants developed dose-dependent increases in intraluminal volume in response to KGF and FGF-10. Although there were no acute changes in explant transepithelial potential difference in response to KGF (0.1-1000 ng/mL), exposure to 5-50 ng/mL KGF over 60 h depolarized transepithelial potential difference compared with controls. We used ribonuclease protection assays to quantitate the ontogeny and regulation of mRNA expression for KGF and its receptor. Both mRNA were expressed in fetal and postnatal lung. Because the promoter region of the human KGF gene contains cAMP and IL-6 response elements, we asked whether cAMP or IL-6 stimulated expression of KGF or its receptor. We have previously shown that cAMP stimulates liquid secretion in this model. Both cAMP and IL-6 significantly increased expression of KGF but not KGF receptor during a 48-h experiment. Thus, stimulation of liquid secretion in explant models by cAMP may be mediated in part by induction of KGF expression. KGF and FGF-10 may be important paracrine factors regulating liquid secretion in human fetal lung.

Techniques of CT colonography (virtual colonoscopy).

Colorectal cancer is a leading cause of death in older adults, which usually involves a long-term progressive change of normal mucosa into adenomatous polyps and then cancer. The detection and treatment of this disease in an early stage can lead to a cure in most cases by simply removing the polyp. Computed tomographic colonography (CTC), also referred to as virtual colonoscopy (VC), is a recent advance that gives an intraluminal visualization of the colon that is similar to endoscopy. VC requires fast 3D display (at least 10 frames/sec) of the colon's mucosal surface on a computer screen. Spiral/helical computer tomography is used to gather 3D volume data prior to display. CTC has been demonstrated to be promising for colorectal cancer screening. Studies on unraveling of the colon are underway to map the convoluted tubular structure into a straightened and flattened image volume for global visualization. In this article, we review the current status of CTC with an emphasis on image processing and visualization algorithms. Clinical assessment results of existing techniques are summarized. Practical issues and future perspectives are also discussed.

The health effects of swimming in ocean water contaminated by storm drain runoff.

Waters adjacent to the County of Los Angeles (CA) receive untreated runoff from a series of storm drains year round. Many other coastal areas face a similar situation. To our knowledge, there has not been a large-scale epidemiologic study of persons who swim in marine waters subject to such runoff. We report here results of a cohort study conducted to investigate this issue. Measures of exposure included distance from the storm drain, selected bacterial indicators (total and fecal coliforms, enterococci, and Escherichia coli), and a direct measure of enteric viruses. We found higher risks of a broad range of symptoms, including both upper respiratory and gastrointestinal, for subjects swimming (a) closer to storm drains, (b) in water with high levels of single bacterial indicators and a low ratio of total to fecal coliforms, and (c) in water where enteric viruses were detected. The strength and consistency of the associations we observed across various measures of exposure imply that there may be an increased risk of adverse health outcomes associated with swimming in ocean water that is contaminated with untreated urban runoff.

[Detection of TTV DNA in liver tissue of patients with non A-G hepatitis].

OBJECTIVE: To study the existence of TTV (transfusion-transmitted virus) in liver tissue of patients with non A-G hepatitis. METHODS: 19 cases of paraffin-embedded liver tissues of the patients with non A-G hepatitis were studied by in sity hybridization with Dig-TTV DNA probe. RESULTS: Out of 19 cases, TTV DNA were detected in 6 cases, including 4 cases of acute mild hepatitis (30.80%) and 2 cases of chronic hepatitis (33.33%), respectively. Hybridization signals were detected within the nuclei or cytoplasm of the hepatocytes. The positive cells were scattered in the intralobular areas in acute mild hepatitis and more aggregated in periportal areas in chronic hepatitis. Among the 6 positive cases, 5 had a raised ALT. CONCLUSION: The result showed that the TTV detected in liver tissue of patients with unknown etiology is a novel virus, which belongs to an hepadnavirus.

Cytosolic fatty acid binding protein enhances rat hepatocyte [3H]palmitate uptake.

Liver cytosolic fatty acid binding protein (FABP) represents the intracellular equivalent to extracellular serum albumin, participating in the intracellular transport of long-chain fatty acids. In this study we observed the effect of increasing and decreasing FABP levels on hepatocyte [3H]palmitate uptake in male Sprague-Dawley rats. We also were interested to determine whether uptake, from either the unbound or unbound and protein-bound fractions, was fundamentally different at the different FABP levels. FABP levels were modified by hypophysectomy and clofibrate treatment (50 mg/100 g body weight for 10 days). Results showed that the [3H]palmitate clearance rates paralleled the 54% decrease and 73% increase in FABP levels in hypophysectomy and clofibrate-treated animals, respectively. In the presence of 2 and 20 microM albumin, hepatocyte clearance rates of unbound [3H]palmitate from hypophysectomized animals (0.16+/-0.01 and 0.64+/-0.01 mL x s(-1) x 10(-6) cells, respectively) were significantly lower (p<0.01) than those of the sham group (0.30+/-0.02 and 1.00+/-0.06 mL x s(-1) x 10(-6) cells, respectively). However, the unbound [3H]palmitate clearance rates from the clofibrate-treated group (0.39+/-0.04 and 1.18+/-0.12 mL x s(-1) x 10(-6) cells) were significantly higher (p<0.01) than the control group (0.29+/-0.02 and 0.81+/-0.05 mL x s(-1) x 10(-6) cells) for 2 and 20 microM albumin, respectively. To investigate whether uptake was fundamentally different between the hypophysectomized and clofibrate-treated groups, we expressed the clearance rates as enhancement factors, i.e., EF = CL20 microM/CL2microM. No statistical difference was observed between EF of the hypophsectomized (3.8+/-0.4) and EF of the clofibrate-treated (3.1+/-0.3) groups, suggesting that the extracted ligand originated from similar fractions.

[Long-term evolution of C-terminal of HCV NS3 protein containing the predicted CTL epitopes in two HCV patients].

OBJECTIVE: To investigate the Long-term evolution of NS3 C-terminal protein containing the predicted CTL epitopes. METHODS: The HLA typing was determined serologically in two chronic HCV patients. Based on this, the HLA restricted CTL epitopes were predicted by HLA binding motif. Sera samples were amplified by PCR, HCV was sequenced and translated into proteins. The changes of predicted CTL epitopes were analysed. RESULTS: No obvious change was observed in predicted CTL epitopes in two chronic HCV patients during 5 and 3 years. CONCLUSION: It demonstrated that the C-terminal of HCV NS3 protein containing the predicted CTL epitopes may have no relationship with HCV persistence.

[Molecular epidemiology of the human immunodeficiency virus type 1 in Santiago, Chile].

BACKGROUND: Most of the studies of HIV-1 infection in South America have been limited to Brazil and little is known about the viral variants that are causing disease elsewhere in the continent. AIM: To determine the characteristics of the viral variants present in Chile as well as patterns of viral transmission. MATERIAL AND METHODS: Viral sequences were obtained from 21 HIV-1 infected people from Santiago, Chile who were infected either via sexual contact or intravenous drug use. Cloned sequences obtained from both the third variable and conserved regions of the envelope as well as the viral protease were evaluated. RESULTS: We found only clade B subtype viruses in Santiago. An evaluation of the envelope gene revealed no evidence that the sequences were monophyletic by risk group. A number of the protease sequences were predicted to encode amino acid substitutions commonly found during selection for protease inhibitor resistance. CONCLUSIONS: The HIV-1 strains studied in Chile, belong to the subtype B. There is no molecular evidence of separate introductions of the virus into the different risk groups. A number of substitutions in the protease gene that may confer resistance to protease inhibitors were found in patients with no previous exposure to this class of drugs.

The orthodontic treatment need and demand of Hong Kong Chinese children.

OBJECTIVE: To investigate the need and demand for orthodontic treatment of Chinese school children in Hong Kong. METHODS: The material consisted of study casts and questionnaires collected from 765 randomly selected 12-year-old school children in Hong Kong. The need for orthodontic treatment was assessed by the Index of Orthodontic Treatment Need, and questionnaires were used in assessing demand for orthodontic treatment. RESULTS: The results indicate that 12% of the 12-year-old school children had no need of orthodontic treatment, 18% had little need (Grade 2), 33% moderate need (Grade 3), 33% great need (Grade 4), and 4% of children had very great need for orthodontic treatment (Grade 5). About two thirds of the children were not satisfied with their dental appearance, but only 40% of these would like to have orthodontic treatment. A definite relationship was found between negative self-appraisal of dental appearance and demand for orthodontic treatment. CONCLUSION: The study demonstrates that the need of orthodontic treatment among Chinese children is similar to that of Caucasian children; the attitude and demand toward treatment are also similar.

[Biological activity of recombinant human hepatopoietin].

We examined the effects of rhHPO on the cell growth and DNA synthesis of both rat primarily cultured hepatocytes and hepatic carcinoma cell line in vitro by MTS and 3H-TdR in corporation methods. It was indicated that rhHPO is an important stimulating factor of regeneration, which may be developed as a potential drug for the treatment of severe hepatic diseases. We also found an inhibitory effect of rhHPO on the DNA synthesis of lung cancer cell lines GLC-82 in vitro, which might provide a valuable indicator for the study of its specificity and mechanisms.

[Surveillance and diagnosis of laryngeal burn].

OBJECTIVE: To explore early diagnosis and treatment of laryngeal burn, in order to prevent laryngeal obstruction, define the prevention of laryngemphraxis, and elect optimal time of treatment and appropriate treatment. METHODS: A total of 18 patients suffering from laryngeal burn were repeatedly examined with a fiberlaryngoscope. Each part of the larynx was assessed for the degree of congestion, swelling, blister formation and impairment of laryngeal function, and the effects of these pathologies on breathing. RESULTS: Our observations showed that classification of inhalation injury according to anatomical division could not reflect the severity of the injury. For this reason, every changes in each part of the larynx was analyzed. A classification consisting of 3 types of laryngeal injury was proposed in respect to 3 predisposing factors of laryngeal obstruction. This classification could serve as a guide for treatment. CONCLUSION: A clearcut diagnosis of laryngeal inhalation injury is a pre-requisite for the choice of the correct treatment and the decision on the time for surgical intervention, thus laryngeal obstruction could be avoided, and death rate of early stage of inhalation injury could also be lowered.

Factors involved in the neuronal death during postischemic reperfusion: experimental study in rabbits.

OBJECTIVE: To explore the main pathogenic factors in the development of neuronal death during normothermic reperfusion in rabbits. METHODS: Ninety-six New Zealand rabbits were randomly allocated into two groups: group I served as non-ischemic controls; group II served as postischemic normothermic reperfusion models. Complete cerebral ischemia was induced by the four-vessel model for 30 minutes. After ischemia, rabbits in group II were further divided into three subgroups according to the duration of reperfusion: subgroup A, 30 minutes; subgroup B, 180 minutes and subgroup C, 360 minutes. Twenty-eight biochemical parameters in the brain were measured, and neuronal changes were observed by histomorphological assessment. Neurons of 12 regions were differentiated into four types: type A (normal), type B (mildly damaged), type C (severely damaged) and type D (necrotic). Bivariate correlate analysis between the levels of biochemical parameters and the percentages of each type of neurons was carried out. RESULTS: The main parameters involved in the progressive decrement of type A neurons were VIP, beta-EP, PGI2, T3, T4 and Na+, K(+)-ATPase; in the increment of type B were beta-EP and TXB2; in the increment of type C were GLU and TXB2/PGI2 respectively; in the stepwise increment of percentages of type D neurons were T4, Na+, K(+)-ATPase, GLU, T3 and VIP (P < 0.05). CONCLUSION: The main factors involved in the development of neuronal death during postischemic normothermic reperfusion in rabbits include hypermetabolism, deactivation of Na+, K(+)-ATPase, release of excitatory amino acids and disorder of neuropeptides.

[An experimental study of metacresol burn].

OBJECTIVE: To study the characteristics of metacresol burn. METHODS: Metacresol of 99.6% was used. The animals were randomly separated into control group, excision group, topical SD-Ag group, saturated sodium sulfate wet compress group, and warm running water flushing group. RESULTS: It was proved that metacresol could not only cause deep burns to skin but also penetrate skin and to produce impairment of functions of the main internal organs of the body, as shown by abnormalities in blood BUN, GPT, GOT, GGT, and AKP. Pathological changes were also found in the liver, heart, brain, kidney, and skin with light and electron microscopy. CONCLUSION: Through comparison of the four different treatment methods for metacresol burns, it is concluded that excision of the wound followed by skin grafting should be done as early as possible to minimize absorption of the chemical. Topical application of SD-Ag or sodium sulfate wet compress, and flushing with running water can alleviate direct injury to skin. Drugs to protect the heart, liver, and kidney should be used for a longer period than usual.

[Association of serum leptin concentrations with blood pressure].

OBJECTIVE: To explore the relationship between serum leptin, hypertension, body mass index (BMI) and energy metabolism. METHODS: Serum leptin concentrations were measured by radioimmunoassay. Blood pressure (BMI) blood sugar (Glu), triglycerides (TG), total cholesterol (TC), LDL, HDL, BUN, Cr and Ur were determined in 64 men with essential hypertension (EHT) and 54 male normotensives (NT). The correlation between leptin and other parameters were analyzed. RESULTS: (1) Serum leptin concentrations n = 118 (3.3 +/- 1.9) micrograms/L in 118 subjects were significantly correlated with SBP, DBP, BMI, Glu, TG, TC and LDL(P < 0.05-P < 0.001). (2) Serum leptin concentrations were strongly correlated with BMI in both EHT and NT groups (P < 0.001). The concentration level was significantly higher in EHT group n = 64 (3.8 +/- 1.9) micrograms/L than in NT group n = 54, (2.8 +/- 1.3) micrograms/L, P < 0.01). The difference in serum leptin concentrations between the two groups disappeared when BMI and age were corrected by analysis of covariance. In NT group, serum leptin concentrations were correlated with SBP (P < 0.05). (3) If the cases of overweight, underweight, hyperglycemia, hyperglyceridemia were excluded, serum leptin concentrations were still highly correlated with BMI, but no longer related to blood pressure. CONCLUSION: Serum leptin concentrations are not directly related to blood pressure, but the concentration levels are actually correlated with the degree of obesity and energy metabolism.
