Separation of human lymphoid cells by preparative cell electrophoresis. III. Studies on cells of lymphoid organs in hematological normal persons.

Blood and bone marrow lymphocytes and cells of lymphoid organs in hematologically normal persons have been studied using free-flow electrophoresis by which a preparative separation of cells according to their electrophoretic mobility is achieved. The results are demonstrated as electrophoretic distributions. Blood lymphocytes have a peak corresponding to relative high electrophoretic mobility, whereas the majority of marrow lymphocytes is found in fractions corresponding to low electrophoretic mobility. The distributions of small lymphocytes from tonsil, spleen and lymphnode either resemble the distribution of blood lymphocytes or have a bimodal fashion with peaks corresponding to blood lymphocytes or marrow lymphocytes. Large lymphoid cells capable of DNA synthesis are concentrated in fractions of intermediate mobility. The results confirm the concept that peripheral T cells have a high electrophoretic mobility whereas most B cells are of low mobility. In contrast to peripheral T cells, cells of the thymus have an intermediate or low electrophoretic mobility. Thus, it appears that in the lymphoid cell system differences in the electrophoretic mobility not only reflect different functional properties but also differences in maturation stages.

[Comparative studies on influences of physical and psychological strain on the ECG in coronary disease patients].

In 7 patients suffering from heart infarction the electrocardiogram was telemetrically registered during telecast of the world championship in football 1974. The same patients were tested on a bicycle ergometer with standardized loads of 50, 75, 100 and 125 Watt for 3 minutes. Heart rate under psychial strain did not exceed the rate of 50 Watt on bicycle ergometer. Only 1 patient showed some extrasystoles during ergometry. Looking at television all patients developed extrasystoles, in one case 122. If the FRG took part in the game, ventricular extrasystoles were two times, auricular extrasystoles were four times more frequent than during games between foreign countries. It is concluded that psychical strain is able to produce dangerous arrhythmias in patients suffering from coronary heart disease. This danger cannot be proved by bicycle ergometry.

Regulation of cell cycle stage-specific transcription of histone genes from chromatin by non-histone chromosomal proteins.

RNA transcripts from chromatin of S phase but not G1 cells contain histone-specific sequences. Chromatin reconstituted with S phase non-histone chromosomal proteins transcribes histone messenger RNA sequences whereas chromatin reconstituted with G1 non-histone proteins does not. These results suggest that transcription of histone genes is regulated during the cell cycle and that non-histone proteins have a key role in this regulation.

[Treatment of psoriasis. New possibility of psoriasis treatment using long-wave UV light and a photosensitizer].

Report about the effect of the therapy of psoriasis with topical application of Meladinine and blacklight as inaugurated by Mortazawi and Oberste-Lehn as well as Tronier. In a high percentage of patients treated a regression of the lesions could be observed. As this therapy is possible for ambulatory patients the number of cases in need of stationary treatment may be reduced.

[The keloid. Thoughts on therapy].

The possibilities of treatment of prophylactic treatment of keloids are described based on results in 96 patients. First therapeutic trials should be conservative. If the external treatment is not successful, the treatment with X-ray in combination with surgical measures in the best therapy.

[Therapy of malignant melanoma].

The treatment of malignant melanoma is described (preoperative, one-time radiation with 10 000 roentgen, excision, endolymphatic radionuclid therapy in localisation of the tumor on the legs, following operative and radiological treatment in metastatic tumors). The statistical reports are analyzed. It seems that the survival rats is not dependent alone on the extent of the surgical intervention.

[Efficacy of a new slow-release isosorbide dinitrate preparation in myocardial infarction (author's transl)].

A new slow-release isosorbide dinitrate preparation (ISDN retard, Boehringer Mannheim) was investigated in a controlled study (double blind/cross over) in 20 patients with clear symptoms of coronary insufficiency after myocardial infarction, comparing the ECG on effort with that under placebo treatment. A favourable effect on the symptoms of coronary insufficiency could be demonstrated under standardised conditions on a bicylce ergometer 4 hours after oral ingestion of ISDN. The ST interval of the ECG was significantly lowered (p less than or equal to 0.025) under comparable cardiac work. The prolonged action of ISDN retard was also shown by a significant reduction in systolic and diastolic blood pressure at rest, during and after effort. Simultaneously, there was a slight but not-significant rise in heart rate.

Separation of human lymphoid cells by preparative cell electrophoresis. II. Free-flow electrophoretic separation of human blood cells.

By means of free-flow electrophoresis the different types of human blood leukocytes can be characterized by their electrophoretic distributions. In addition a separation of cells according to their electrophoretic mobility and at least an enrichment of certain cell types is achieved. The data given in this paper confirm that reproducible results can be obtained using this separation method and indicate that it can best be used to study populations of lymphocytes.

Comparison between Wilson's thoracic leads and the telemetrically transmitted lead CM6 in patients with heart diseases during exertion.

In 76 patients suffering from heart diseases and 19 healthy men simultaneously the chest leads V(2), V(4) and V(5) were recorded and the bipolar lead CM(6) (Manubrium sterni-Apex cordis, 6th intercostal space) transmitted telemetrically during increasing exercise load on the bicycle ergometer. The lead CM(6) was most similar to chest lead V(5) with regard to R/S-ratio, ST-depression, form and amplitude of the T-wave, QX-QT-ratio and ST-elevation. ST-depressions of V(5) were found nearly without exception in CM(6), mostly more pronounced. Ascending ST-depressions are more frequent in CM(6) than in V(5), in single patients also ST-depressions with horizontal or downward slope. ST-elevations registrated in V(2) and V(4) are not reliably reproduced in CM(6). It is concluded that the wireless transmission of the bipolar chest lead CM(6) is sufficient for the diagnosis of coronary insufficiency during exercise,
