Comparative study of biochemical technique and radioimmunoassay for the measurement of serum prostatic acid phosphatase. Interest in the diagnosis of prostatic cancer?

The radioimmunoassay of prostatic acid phosphatase and the measurement of L-tartrate labil acid phosphatase by biochemical technique are compared in the diagnosis of prostatic cancer. This study concerning in 122 patients bearing prostatic cancers (40), prostatic adenomas (30) and other solid tumors (52) shows that the sensibility of RIA technique is better than the biochemical one. The positive predictive value of PAP-RIA is 93 p. cent However, seeing that the percentage of positivity of RIA in intracapsular stages rarely exceeds 40 p. cent, this test does not allow to increase detection power of early stages. The RIA technique, if it is better than biochemical method will not be effective as a sole screening tool for prostatic cancer and its principal application consists in the follow-up of the therapy of prostatic cancer.

[Radioimmunoassay for prostatic acid phosphatase in human serum. Methodologic aspects (author's transl)].

We propose a double antibody radioimmunoassay for human prostatic acid phosphatase (PAP) in serum for diagnosis and management of prostatic adenocarcinoma under treatment. The antigen is purified from human prostatic fluid by a gel-filtration on Sephadex G 100 followed by affinity chromatography on Con A Sepharose. A specific antibody is raised in rabbits and purified by immunoadsorption with a female serum. The described technique offers both radioisotopic sensibility and immunologic specificity. Physiological values determined in the serum of 125 healthy males are below 2 ng/ml. No significative differences are observed with age. The proposed technique also shows significant differences between values evaluated for benign prostatic hyperplasia and prostatic adenocarcinoma.

Intrahepatic polyamine levels during rat liver carcinogenesis induced by N-2-fluorenylacetamide.

During a period of 200 days, the chronological changes of polyamine levels (putrescine, spermidine and spermine) were observed in the liver of adult female Sprague Dawley rats submitted to hepatocarcinogenesis by N-2-fluorenylacetamide (FAA). Three groups of 70 rats each were used: (1) Control 1: normal diet; (2) Control 2: low protein and low riboflavin diet; and (3) EXPERIMENTAL: 0.06% FAA added to the diet. No significant differences were noted for tissue levels of the three polyamines when the two control groups were compared. In contrast, considerable variations of these molecules were observed as a function of time in the FAA treated group: (a) an early and constant rise was seen in putrescine, with 3 maxima at days 10, 60 and 150. This last peak was the highest: 25 +/- 6 nmol/g (8 times the value for the controls at this time), and coincided with the appearance of cancerous lesions. (b) While spermidine levels varied during the experiment, no significant differences were noted in comparison with the control groups. Mean levels (nmol/g) were: 535 +/- 108 Control 1; 552 +/- 95 Control 2; 633 +/- 160 FAA-treated group. (c) Spermine levels were significantly lowered, with 3 minima corresponding to the putrescine maxima. The lowest minima was observed on day 60: 114 +/- 67 nmol/g, i.e., 4 times lower than the controls. This work shows that polyamine metabolism is profoundly modified during chemical carcinogenesis, but the possible effect of polyamines on tumorigenesis itself cannot be assessed at this point since modifications of polyamine levels are probably also associated with phenomena of liver necrosis and compensatory tissue proliferation observed during the experiment.

[Gamma glutamyl transferase and cancer (author's transl)].

Attention has been drawn in recent years to the diagnostic interest offered by increases in serum gamma GT in the detection of liver metastases and various cancers in man. Although this analysis is not specific and presents a few false positive and negative results, it seems however profitable to include it either in detection or in supervision tests. Furthermore, in the rat and mouse, this enzyme practically absent from the adult liver, reappears during liver cancer Whatever the cause: this onco-foetal character may be useful to exploit in studies of experimental carcinogenesis, but does not seem to be a valid model for the increases observed in man.

[Determination of circulating ectopic beta HCG in solid tumors (author's transl)].

The beta subunit of human chorionic gonadotropin (betaHCG) was performed on the serum of 1 850 patients with microscopically documented non trophoblastic carcinomas. 96 patients (5,2%) had elevated betaHCG plasma levels. The incidence of elevated plasma values varied little from one studied malignancy to another; the highest circulating levels were observed in patient's with bladder carcinomas (22,2%), squamous cell carcinomas of the bronchus (14,2%), pancreatic (20%) and colo-rectum (15,7%) adenocarcinomas. BetaHCG determinations did not improved the diagnostic sensibility of CEA values except for the bladder carcinomas. The short mean survival of patients with positive betaHCG values suggests that elevated ectopic betaHCG, sometime during the clinical course of metastatic maligancies worsens the prognosis.

[Changing relative proportions of apolipoproteins of VLDL in chronic male alcoholics (author's transl)].

Apoproteins of very low density lipoproteins of plasma from male chronic alcoholics and healthy men serving as controls were investigated. VLDL were delipidated using diethyl ether-ethanol and were separated by polyacrylamide gel electrophoresis. The average relative concentration of apo Cll, Clll1 and Clll2 are investigated. The proportion of apo Cll is reduced, and that of apo Clll increased (particularly Clll1) in male chronic alcoholics compared to the normal and the ratio of apo Clll1 to apo cll was significantly increased. The Cll/Clll1 + Clll2 ratio is highly significantly reduced in patients. Changes in apo Clll1/Cll ratio in relation to hypertriglyceridaemia are envisaged. Furthermore apo Cll is believed to activate and apo Clll to inhibit lipoprotein lipase activity "in vitro".

The measurement of total lactic dehydrogenase and its isoenzymes in solid tumours.

The routine practice of making enzymatic assessments for the purposes of cancer diagnosis, including the determination of total amount of LDH provides only limited information. However, the total amount of LDH may be used to monitor the effect of chemotherapy or radiotherapy in certain tumours, such as epithelial tumours and germ-cell tumours of the ovaries, non Hodgkin's lymphomas or malignant tumours of the testicles. The differentiation of LDH isoenzymes, with a view to linking their pattern to precise localizations has been studied. In germ-cell testicular tumours, there is a marked increase in LDH 1. In liver metastases, LDH 5 and/or LDH 4 are disturbed in 90% of the cases, while the total amount of LDH is raised in only 72% of the cases. In many cases, the association of a disturbance in the serum ratio of the total amount of LDH and a disturbance in the isoenzymes of LDH 4 and LDH 5 associated with a high level of GGT indicate the presence of liver metastases.

Biochemical modifications of pulmonary surfactant after bromhexine derivate injection.

The authors study the influence of bromhexine metabolite VIII on phospholipid and fatty acid composition comparatively to controls. The subcutaneous injection of NA 872 produces on pulmonary surfactant an increase of 43% of total phospholipids. This change in the distribution of phospholipids is shown by the increase of 53% of phosphatidylcholines. A rise of 61.9--74.4% of palmitic acid appears in the phosphatidylcholines of the pulmonary surfactant. It can be noticed that the specific activity of the phosphatidylcholines rises by 31%.

Fatty acid composition of aortic lipids in male and female rabbits.

Aortic tissue of adult New-Zealand male and female white rabbits was studied. After thin-layer chromatography of cholesterol esters, triglycerides, free fatty acids, phosphatidylethanolamines and phosphatidylcholines, their fatty acid compositions are determined. Phosphatidylcholines and phosphatidylethanolamines contain important quantities of arachidonic acid. The level of linoleic acid in these phospholipids is higher in male animals than in female. Conversely the level of arachidonic acid is higher in females than in males.

Inhibition of lecithin:cholesterol acyltransferase in human plasma by dicarboxylic lecithins.

The effect of dicarboxylic lecithins on the lecithin:cholesterol acyltransferase [LCAT] activity was studied with a partially purified preparation [approximately 24-fold]. The presence of dicarboxylic lecithins is followed by the inhibition of LCAT. The inhibition is proportional to the concentration of glutaryl-lecithin and is about 50% with 0.16 mM glutaryl-lecithin. For a higher concentration, the inhibition is total. The inhibition of LCAT activity was of a mixed type. It can be explained by a combination of competitive inhibition and interaction of glutaryl-lecithin with phospholipid, which changes the physical properties of lipid, making it less available for conversion into cholesteryl ester.

[Chicken erythrocyte nuclei: composition and analysis of phospholipids].

While many works analyse lipid composition of cellular nuclei, essentially of liver, few describe bird erythrocyte nuclei. For them very discordant results are reported due to very different methods of preparation. In the method we adopted a good reproducibility is controlled by electron microscopy, evaluation of specific enzymatic activities and by protein, RNA and DNA determinations. Total lipid content and phospholipid composition were determined showing, in agreement with other works, the predominance of phosphatidylcholine and phosphatidylethanolamine. On the contrary, the nuclear fraction is poor in sphingomyelin, which is essentially a plasma membrane phospholipid.

[Incorporation of 32P-orthophosphate in aortic phospholipids in chronic lathyrims in the rat].

Study of 32P orthophosphate incorporation into rat aortic phospholipids after B.A.P.N. treatment, which was followed or not by a lipid-supplemented diet (4.5 p. 100 cholesterol and 37 p. 100 butter). The 32P orthophosphate turnover in the total phospholipid was twice as slow among B.A.P.N.-treated rat than among control-rat. Such a decrease of the 32P orthophosphate turnover could be found as well in phosphatidylethanolamines, phosphatidylcholines and lysophosphatidylcholines. Sphingomyelins showed a very weak metabolic activity among two groups of rats. 32P orthophosphate incorporation into inositolphosphatides was slowed down by B.A.P.N. treatment. Maximal specific activity of inositolphosphatides after injection of 32P orthophosphate was reached within 4 hours in control rats, and 8 hours in B.A.P.N.-treated rats. Three months after the end of B.A.P.N. treatment, no difference in specific activity could be found between control-rats and B.A.P.N.-treated rats. Lipid supplemented diet modified the metabolic activity of phospholipids equally in control rats and B.A.P.N.-treated rats. Specific activity decrease in total phospholipids, phosphatidylcholines, phosphatidylethanolamines, and increase in lysophosphatidylcholines were noted in both groups of animals under a high rat diet. These modifications of the metabolic activity of different phospholipids were most marked fourteen hours after 32P orthophosphate injection. B.A.P.N. treatment only slowed the incorporation of 32P phosphate into phospholipids. Administration of a single lipid supplemented diet or a diet associated with B.A.P.N. treatment induced a slower turnover of phosphatidylcholines and phosphatidylethanolamines, and a faster turnover of lysophosphatidylcholines.
