Cholecystokinin octapeptide purified from brains of Australian marsupials.

Cholecystokinin octapeptides (CCK8s) have been purified from methanol extracts of two brains from each of two Australian marsupials, Tammar Wallaby and Eastern Quoll, containing 3 nmol and 2 nmol of the peptides, respectively. Immunoreactive CCK was concentrated on QMA SepPak cartridges and purified by two successive HPLC steps on Nova C18 radial-pak cartridges. The sequence of each of the peptides is identical with that previously reported for Old World mammals (DYMGWMDF). This is in contrast to the previously reported sequence for CCK8 from the South American hystricomorphs, guinea pig and chinchilla, which differs in a substitution of valine for methionine in position 3 from the NH2-terminus. Although evolutionary history suggests that marsupials migrated from South America into Australia before the two continents separated, this peptide resembles that found in Old World mammals rather than that of South American hystricomorphs. Such molecular data are useful in assessing phylogenetic relationships among taxa.

Neural tube defects and maternal weight reduction in early pregnancy.

We present data from a case series in France of pregnant women who lost 2 to 14 kg during the first month after conception and whose fetuses developed neural tube defects (NTDs). Preliminary epidemiologic evidence from other data sets suggest that relative lack of weight gain during pregnancy may be associated with NTDs. The role of starvation, diabetes, and valproic acid in the etiology of NTDs is established. This etiologic information coupled with our data suggest an hypothesis that ketoacidosis induced by weight loss in early pregnancy is a risk factor for NTDs.

Management of renal-vein perforation during a challenging percutaneous nephrolithotomy.

A 54-year-old man with complex urinary anatomy as the result of previous surgery sustained a renal-vein injury during percutaneous nephrolithotomy for a staghorn calculus with a blood loss of 1.5 L. He was managed with antibiotics, bed rest, heparin, and a 28F nephrostomy catheter, which was withdrawn gradually as the tract sealed. This case highlights the importance of early diagnosis of this complication and the possibility of conservative management.

A prospective study of septic complications of endoscopic retrograde cholangiopancreatography.

Prophylactic antibiotics are used in an attempt to avoid the septic complications of endoscopic retrograde cholangiopancreatography (ERCP). We prospectively performed blood cultures and surveyed patients for complications. The aims were first, to determine the incidence of bacteraemia associated with ERCP, second, to assess the incidence of clinical sepsis following the procedure and third, to evaluate the effectiveness of our antibiotic prophylaxis. One hundred and fifty successive patients underwent 179 ERCP. Bacteraemia related to the procedure or the underlying pathology was found in nine procedures (5.2%). Bacteraemias were more likely to complicate therapeutic procedures (P = 0.015), biliary obstruction (P = 0.045) or underlying pathology (P = 0.022). Although 61% of ERCP received antibiotics, 22 septic events occurred. Five bacteraemic patients were septic despite antibiotics. Septic complications were associated with the same factors as bacteraemia. It was concluded that patients with biliary obstruction and undergoing therapeutic endoscopic procedures are at greatest risk of bacteraemia. Single dose prophylactic antibiotics may not prevent sepsis in these patients and longer-acting drugs or repeated dosing may be necessary.

A statistical view of FMRFamide neuropeptide diversity.

FMRFamide-like peptide (FLP) amino acid sequences have been collected and statistically analyzed. FLP amino acid composition as a function of position in the peptide is graphically presented for several major phyla. Results of total amino acid composition and frequencies of pairs of FLP amino acids have been computed and compared with corresponding values from the entire GenBank protein sequence database. The data for pairwise distributions of amino acids should help in future structure-function studies of FLPs. To aid in future peptide discovery, a computer program and search protocol was developed to identify FLPs from the GenBank protein database without the use of keywords.

Radionuclide transport above a near-surface water table: III. Soil migration and crop uptake of three gamma-emitting radionuclides, 1990 to 1993.

This paper summarizes the vertical distributions of 22Na, 137Cs, and 60Co above controlled water tables in deep and shallow lysimeters during a four-year experiment. The activity concentration profiles were all determined at the time of harvest of a winter wheat (Triticum aestivum L. cv. Pastiche) crop. Activity concentrations in different crop tissues were determined and crop uptake expressed as both an inventory ratio (IR) and a transfer factor (TFw), weighted to account for root and radionuclide distributions within the soil profile. Experimental variates were subjected to analysis of variance to determine the single and combined effects of the soil depth and the year of the experiment on the results obtained. Each radionuclide showed significant variations in activity concentration with soil depth, but the significance of these variations from year to year was dependent on radionuclide. A distinction in the behavior of weakly sorbed (22Na) and more highly sorbed (137Cs and 60Co) radionuclides was observed. The former exhibited significant variations in its distribution in the soil profile from year-to-year whereas the latter did not. Relatively high TF, values for 22Na were maintained throughout the experiment, whereas for 137Cs and 60Co, the highest TFw values were recorded in 1990 followed by a significant decline in 1991, with TFw remaining low in 1992 and 1993. The TFw values were, in general, significantly higher for deep lysimeters than for shallow lysimeters. This is thought to provide evidence of enhanced radionuclide absorption by the relatively small fraction of roots in the vicinity of the deeper water table.
