Sickling and Vinblastine.

After incubation of sickle anaemia (SS) erythrocytes for 1 hour at room temperature with Vinblastine (VBL) doses (0.2 mM) far lower than those capable of causing spherostomatocytosis of normal red blood cells (0.6 mM), the percent of erythrocytes which underwent swelling resulted to be overlapping the percentage of cells which showed irreversible sickling (IS) after adequate deoxygenation. Thus VBL can reveal in the SS erythrocytes the most liable cohort of cells that show IS; this may be accounted for by the fact that in both irreversibly sickled and VBL-incubated erythrocytes there are overlapping disorders of the membrane cationic traffic. The increase of the drug concentrations induced an exponentially related cohort of SS erythrocytes to swell. The observed failure of SS erythrocytes previously incubated with VBL to sickle after adequate deoxygenation is accounted for by the close interaction of the drug with the red cell membrane subunits, which renders the red cell surface unliable to undergo further morphological alterations.

[A new case of hemoglobin Lepore-beta-thalassemia disease].

The A. relate on a case of disease from Hb Lepore-heterozygous beta thalassemia observed in an italian family originated from Campania. The patient is a boy 16-year old who shows a clinical picture similar to that of Cooley's disease. The electrophoretic study of the hemoglobin has evidenced a double heterozygosis Hb-Lepore-beta thalassemia. Enlarging the examination to the other components of the strain, also the sister of the propositus shows the same disease, while the brother and the mother get a little amount of Hb Lepore. The father gets heterozygous beta-thalassemia. The negatively of the sickling test "in vitro" excludes tha presence of HbS in all components of the family. The radiological examination of the skeleton of the propositus evidences interesting analogies with the skeleton alterations observed in the subjects affected with Cooley's disease and scarcely blood-transfused.

Why inclusion bodies do assume different locations in thalassaemic erythrocytes.

The reported scanning (SEM), transmission (TS), and freeze-etching (FE) electron microscopic studies have agreed in confirming that in thalassaemic erythrocytes, previously incubated with brilliant cresyl blue (BCB), the unpaired alpha chains precipitate in the central portions of the cell whereas excess beta chains locate in the submembranous regions. This is due to the fact that beta chains, possessing two thiols instead of only one (as in alpha chains), are more liable to bind to similar groups contained in the inner red cell leaflet. Less soluble alpha chains tend to form inter-chain bridges and thus precipitate centrally. SEM observations have given evidence that on the surface of the affected red cells denaturated alpha chains give rise to large and shallow invaginations whereas denatured beta chains lead to diffuse wrinkled appearance. The causes of the different SEM aspects have been suggested.

A simple method for implanting electrodes in freely moving neonatal rats.

A simple method for implanting cortical and subcortical electrodes in very young rats is described, which does not interfere with the behavior of the animal during the experiment. The cortical electrodes are anchored to the skull by a spiral whose first loop is introduced between the inner layer of the skull and the dura mater. The subcortical electrodes have a loop which rests above the skull. Both the spirals of the cortical electrodes and the loops of subcortical electrodes are then embedded in dental acrylic.

Further considerations on alcohol intake and coronary risk factors in a Rome working population group: HDL-cholesterol.

A study on the relationships between habitual alcohol consumption and the main coronary risk factors was carried out in 520 consecutive high-risk men examined during an annual rescreening of a working population group within the Rome Project of Coronary Heart Disease Prevention. Alcohol intake was determined by means of a semiquantitative questionnaire. The mean daily wine intake was about 0.5 liter, the consumption of other alcoholic beverages being negligible. No statistically significant correlations were found between average daily wine consumption and the main coronary risk factors, while a strongly positive correlation (p less than 0.001) was observed with plasma HDL-cholesterol levels. Also differences in HDL-cholesterol between occasional drinkers and moderate drinkers were statistically significant. No differences were observed in total cholesterol and triglycerides. The findings suggest that habitual moderate amounts of alcohol may increase HDL-cholesterol between occasional drinkers and moderate drinkers were statistically significant. No differences were observed in total cholesterol and triglycerides. The findings suggest that habitual moderate amounts of alcohol may increase HDL-cholesterol levels.

Left ventricular size following endurance, sprint, and strength training.

Left ventricular size following endurance, sprint, and strength training. Med. Sci. Sports Exercise, Vol. 14, No. 5, pp. 344-347, 1982. Left ventricular dimensions in adolescent boys were determined before and after three types of training regimens: endurance (END), N = 8, means = 16.8 yr; sprint (SPR), N = 8, means = 16.3 yr; strength (STR), N = 12, means = 18.7 yr. With training the END group significantly increased VO2max in 1 X min-1 (3.71 +/- 0.27 to 4.16 +/- 0.57, P less than 0.05) and in ml X min-1 X kg-1 (58.4 +/- 5.6 to 64.2 +/- 5.5, P less than 0.05). The SPR group increased VO2max in 1 X min-1 (3.63 +/- 0.63 to 3.98 +/- 0.78, P less than 0.05) but not in ml X min-1 X kg-1 (59.5 +/- 4.1 to 63.2 +/- 5.4) because body weight increased from 61.2 +/- 10.5 to 63.1 +/- 10.7 kg (P less than 0.05) with no change in percent body fat. The STR training group significantly improved upper body strength. Despite these specific training adaptations no significant modifications were found for interventricular and left ventricular posterior wall thickness or for left ventricular internal diameter in either training group. However, calculated left ventricular mass was slightly but significantly higher by 10% and 4% in the END and STR training groups, respectively. These small increases in calculated left ventricular mass with short-term training are probably caused by small but insignificant increases in left ventricular internal diameter secondary to a training bradycardia (END group: 76 +/- 8 to 64 +/- 1 beats X min-1) and to increased diastolic filling time rather than to true cardiac hypertrophy. Significant increases in aerobic capacity and in strength can occur without modification of left ventricular dimensions.

L-lanthionine oxidation by snake venom L-amino acid oxidase.

L-Lanthionine is oxidized by snake venom L-amino acid oxidase with the release of one mole of ammonia per mole of lanthionine. Spectrophotometric, chromatographic, and analytical properties are all consistent with the identification of the mono-keto derivative of lanthionine as the first enzymatic product of the reaction. This then cyclizes into a dihydrothiazine dicarboxylic acid which is further subject to spontaneous changes. Authentic samples of the thiazine derivative have been prepared by interacting L-cysteine with bromopyruvic acid. The properties of the synthetic product are described and compared with those exhibited by the enzymatic product.

Multiple forms of bovine liver rhodanese.

The heterogeneity of crystalline rhodanese from bovine liver has been studied. The enzyme was separated by preparative disc electrophoresis into four stable forms designated I(3.5%), II(17.0%), III(62.0%) and IV(17.5%). All these components have the same molecular weight of about 35,000 both in native and denaturing conditions. No differences in the primary structure have been detected in amino acid composition and by peptide mapping. Although the isolated forms show similar values of specific activity, except form I, their content of transferable sulfur is different. The enzyme-sulfur complexes of the four components have slightly different circular dichroism spectra in the near ultraviolet, though their spectra in the sulfur-free state are identical. We conclude that these forms are conformational isomers originated from form IV which is predominant in the mitochondrial extract of liver cells.
