Molecular and cellular effects of antisickling concentrations of alkylureas.

Alkylureas are capable of inhibiting sickling in vitro and the gelation of solutions of hemoglobin S at concentrations between 0.05 and 0.1 M with increasing effectiveness that is directly proportional to the length of the alkyl chain (butyl greater than propyl greater than ethyl greater than methyl). 6The inhibitory effect is independent of pH between 6.5 and 7.5 and is a process driven by entropy. The alkylureas at concentrations of 0.1 M have minimal effects on several erythrocyte functions. Oxygen equilibria, osmotic fragility, reduced glutathione content, and glutathione reductase activity are totally unaffected, while pyruvic kinase activity is decreased only by butylurea by about 20%, and glucose-6-phosphate dehydrogenase activity is decreased progressively to a maximum of 30% in direct proportion to the length of the alkyl chain. Alkylureas not only inhibit sickling but are also capable of desickling erythrocytes that have been maintained in the deoxygenated state. They have little effect on several erythrocyte functions at antisickling concentrations, but their toxicity must be evaluated before they can be examined as potential therapeutic agents for the treatment or prevention of acute episodes in sickle cell anemia.

Erythrocytes, Abnormal

[Metabolism of essential and endogenous serum fatty acids in healthy persons, coronary disease and patients with lipid metabolism disorder. Partial aspect of high risk problem patients with age-related lipid and fatty acid concentrations in serum and regulation of long-chain products from essential and endogenous precursors in healthy subjects].

Total lipid, free and esterified cholesterol and triglycerides significantly increase in concentration with increasing age. Palmitic, palmitoleic, oleic and linoleic acid behave in the same way. Free fatty acids, phospholipids and stearic acid show the same concentrations in all groups of age. C20 fatty acids demonstrate a competitive regulatory mechanism of desaturase activity in their age dependant distribution. While the 20:3(n-9), a product of the endogenous (n-9) family, increases, the 20:3(n-6) and 20:4(n-6), both desaturation and chain elongation products of the essential n-6 family, decrease. Such behavior has been demonstrated in many feeding studies (12, 13, 15, 16, 17, 29) and is interpreted as a result of competitive substrate interaction on desaturase activity.

8,11,14-Eicosatrienoic Acid

[Non-invasive myocardial scanning with thallium-201: basic principles and methods (author's transl)].

201Tl scanning demonstrated viable myocardium in the area supplied by the anterior interventricular branch (on frontal scanning) and viable myocardium in the interventricular septum and left ventricular lateral wall (scanning in the second oblique position) in 9 of 11 patients. Akinetic myocardial segments in the left ventricular anterior wall and left ventricular apex could be recognized by absent activity when scanning from in front. But hypokinetic segments, which contrary to the appearance with akinesia cannot be equated with myocardial scar, could not according to present experience be recognized by 201TL scanning. The special advantage of 201Tl scanning is is that, except for its intravenous injection, no further invasive procedure has to be undertaken, and it can be repeated. No side effects of complications have been observed.

Angiocardiography

[Thoracic gunshot-injury with penetration of the pericardium in a 15-year-old student].

On account of the exceptionality is reported on a 15-year-old adolescent who suffered a gunshot into the thorax by a device which was built by himself. More than 12 hours after the accident for the first time a physician was consulted. Then the patient was transported to Leipzig for operation. The missile had penetrated the anterior wall of the pericardium, had passed through the cleft between exterior layer of the myocardium and the pericardium and lodged at the trangression into the adjacent lung. The postoperative course was without complications. The patient could be dismissed without any complaints.

Lipid metabolism of developing central nervous tissues in organotypic cultures. I. Lipid distribution and fatty acid profiles of the medium for rat brain cortex in vitro.

The distribution and concentration of lipid components of different media used for organotypic cultures of rat brain cortex have been measured quantitatively. Five cholesterol ester species have been fractionated from chicken embryonic extract. Their fatty acid profiles have been determined by combined gas liquid chromatography-mass spectrometry. The depletion characteristics of medium cholesterol esters and some fatty acids during development of brain cortex were not symmetrical. Polyunsaturated fatty acids of the medium were lowered prior to the saturated and monoenoic fatty acids. The phenomenon of depletion may be correlated with enhanced esterase activity mediated by the tissue.

[The epidemiology of lung cancer in the German Federal Republic, with special reference to regional differences (author's transl)].

11 per cent more men and 23 per cent more women died of lung cancer in the German Federal Republic during 1960-1971 than would have been the case if the age structure had remained unchanged. This increase involves exclusively (men) or predominantly (women) the over-65 years age group. It is still showing an upward trend in men and is most pronounced in the 45-74 age group. Mortality from lung cancer differs significantly between the Länder of the Federal Republic. It is highest for both men and women in the City States and is lowest in the less industrialized areas, particularly South Germany. This difference is presumably not entirely population: the particularly marked increase (especially in men) in the Saarland and North-Rhine-Westphalia-Länder with a predominance of heavy industry-suggests that occupational noxae may play a part.

Adult

[The costs of cancer screening (author's transl)].

By the cancer screening programme of the German legal health insurance 4,830,717 females were examined in 1973. The yield of cytologic "positive" cases has been 0.23 percent. The sum for finding one of these cases has been in the range from 14,388.-- DM to 16,132.--DM. As can be calculated by the available data on the incidence of cancer of the cervix uteri only about one third of these cases with "positive" findings can be a proven case of cancer (carcinoma in situ included). Thus one proven carcinoma of this site can be detected by the costs of 40,656.--DM to 80,586.--DM and this sum shows an increasing tendency. In spite of these high costs for the cancer detection programme the total expenses for early detection measures amounted only to 0.6 percent of the total expenditues of the legal health insurance in the first half-year 1974. The problem of a high rate of false-positive smears is discussed.
