Glycoproteins of the AKR leukaemia cell surface and their relevance to leukaemia-specific surface antigens.

An attempt has been made to prepare antibodies against leukaemia-specific surface antigens by immunizing (C57 B1/6 X C3H/He)F1 mice with formaldehyde-stabilized AKR leukaemic cells. The presence of antibodies was examined by indirect immunofluorescence microscopy (IFM) and the indirect antiglobulin rosetting reaction (IARR). Galactose oxidase treatment destroyed the ability of leukaemic cells to react with antibodies prepared in the hybrid mice, an effect that was reversed by treating the enzyme-modified cells with borohydride. Analysis by immunoprecipitation and polyacrylamide gel electrophoresis of leukaemic cells, labelled by the galactose oxidase/[3H]-NaBH4 technique, indicated that a group of glycoproteins of apparent molecular weight greater than 70 000 was involved. Antibodies could be raised in AKR mice to the same group of glycoproteins by immunization with irradiated leukaemic cells or irradiated neuraminidase-treated leukaemic cells. The level of antibody raised in AKR mice had no effect on the growth of leukaemic lymphoblasts introduced subcutaneously into the host. Antibodies prepared in hybrid mice against leukaemic cells also were absorbed by lymphoid cells of pre-leukaemic 6-month-old AKR mice, indicating that contrary to previous claims in the literature antigens detected by such antisera are not related to malignancy. Hybrid mouse serum cross-reacted with antigens from purified RNA virus isolated from Abelson lymphoma, as demonstrated by the immunoelectrophoretic blotting technique. The pattern of reactivity was not appreciably altered following the absorption of antibodies directed against leukaemic cells. It is concluded that the glycoproteins detected by us may not be viral antigens but normal high molecular weight lymphoid glycoproteins with altered glycosylation patterns that are induced when the viral genomes are expressed.

Kinetic parameters of a beta-D-galactoside alpha 2 leads to 6 sialyltransferase from embryonic chicken liver.

A beta-D-galactoside alpha 2 leads to 6 sialyltransferase was purified 500-fold in 14% yield from 14-day embryonic chicken liver. Characterization of the product of the sialyltransferase catalysis was accomplished by separation and permethylation of double-labelled ([14C]NeuAc, [3H]Gal) oligosaccharides following their release from the glycoprotein fetuin by hydrazinolysis. The enzyme transfers NeuAc to Gal(beta 1 leads to 4)GlcNAc(beta 1 leads to)R-terminated oligosaccharides; no activity was found towards Gal(beta 1 leads to 3)GalNAc(alpha 1 leads to)R structures. The trisaccharide. NeuAc(alpha 2 leads to 6)Gal(beta 1 leads to 4)Glc, was shown to be a good inhibitor of the sialyltransferase. Kinetic investigations of the enzyme indicate it to have a sequential, random bi-bi mechanism.

Glycoprotein biosynthesis in quiescent and stimulated thymocytes and a T-cell lymphoma.

Quiescent thymocytes, mitogen-stimulated thymocytes and acute-leukaemic lymphoblasts provide a model for the study of protein glycosylation in quiescent cells, mitotically active non-malignant and malignant cells respectively. The biosynthesis of both complex and high-mannose-type oligosaccharides was monitored by metabolic labelling with [6-3]fucose and [2-3H]mannose. Bio-Gel P6 elution profiles of [6-3H]fucose-labelled glycopeptides showed that quiescent thymocytes and stimulated thymocytes synthesized qualitatively and quantitatively similar glycopeptides; however, higher-molecular-weight glycopeptides were synthesized by the acute-leukaemic lymphoblasts. The amount of [2(-3)H]mannose incorporated into glycopeptide by quiescent thymocytes was less than 10% of that incorporated by stimulated thymocytes. The Bio-Gel P6 elution profile of [2(-3)H]mannose-labelled glycopeptides from acute leukaemic lymphoblasts was qualitatively similar to that of stimulated thymocytes, with about 40% of the radioactivity incorporated into one glycopeptide peak. This glycopeptide was characterized by Bio-Gel P6 and concanavalin A affinity chromatography, radioactive-sugar analysis, sensitivity to alpha-mannosidase and endoglycosidase H and resistance to beta-glucosaminidase as containing a high-mannose oligosaccharide, possible of Man7-8GlcNAc2 structure. Pulse/chase experiments indicated that this high-mannose oligosaccharide was an end product and not a biosynthetic intermediate. It is concluded that higher-molecular-weight fucose-labelled glycopeptides are characteristic of the malignant cell type, and the synthesis of high-mannose oligosaccharide, Man7-8GlcNAc2, in stimulated thymocytes and acute-leukaemic lymphoblasts is associated with mitotically active cells.

A case-control study of diet and colo-rectal cancer.

A case-control study of cancer of the colon and rectum has been conducted in Calgary, Alberta and Toronto, Ontario, Canada. A total of 348 cases of cancer of the colon and 194 cases of cancer of the rectum were individually matched by age, sex and neighbourhood of residence to 542 population controls and frequency match to 535 hospital controls who had undergone an abdominal operation. Each subject received a personal medical history questionnaire and a quantitative diet history questionnaire. Data on a number of potential non-nutrient risk factors for bowel cancer and on the consumption of 9 nutrients in the 2-month period up to 6 months before interview were analysed. The dietary data thus refer to recent diet consumed in a period antedating the diagnosis of, and in most cases symptoms from, large-bowel cancer in the cases, and a corresponding time period in the controls. The major findings were an elevated risk for those with a history of bowel polyps, and for those with an elevated intake of calories, total fat, total protein, saturated fat, oleic acid and cholesterol. No association was seen with an elevated intake of crude fibre, Vitamin C and linoleic acid. The nutrients for which an increased risk was demonstrated were highly correlated, though multivariate analysis using logistic regression indicated highest risk for saturated fat, with evidence of a dose-response relationship. The findings in both cancer sites, both sexes and with both sets of controls were quantitatively very similar. The population-attributable risk for colon and rectal cancer combined was estimated from the neighbourhood controls to be 41% for males and 44% for females for saturated fat intake and 9.8% and 6.4% respectively for any history of polyps.

Tobacco use, occupation, coffee, various nutrients, and bladder cancer.

In a Canadian population-based case-control study of 480 males and 152 female case-control pairs, the relative risk for development of bladder cancer for ever used versus never used cigarettes was 3.9 for males and 2.4 for females, with a dose-response relationship in both sexes. A reduced risk was associated with the use of filter cigarettes compared to nonfilter cigarettes. After control for cigarette usage, a significant risk was noted for male pipe smokers. For male ex-smokers the risk after 15 years of no smoking was less than one-half that of current male smokers. Bladder cancer risk was found for workers in the chemical, rubber, photographic, petroleum, medical, and food processing industries among males and for workers occupationally exposed to dust or fumes among both sexes. Bladder cancer risk was elevated for males consuming all types of coffee, regular coffee, and instant coffee and for females consuming instant coffee, but no dose-response relationship was found. Risk was found for males consuming water from nonpublic supples but not for females. No risk was observed in males or females consuming nitrate-containing foods, beverages other than coffee, or fiddlehead greens. Hair dye usage in females and phenacetin usage in males and females carried no risk. Divergent findings by area for aspirin suggested that an overall association was not causal. Reevaluation of the data on artificial sweeteners confirmed a significant bladder cancer risk in males and a dose-response relationship. The cumulated population attributable risk for bladder cancer was 90% for males from cigarette smoking, industrial exposure, and exposure to nonpublic water supplies and 29% for females from cigarette smoking, industrial exposure, and instant coffee consumption.

Solubilization of human erythrocyte membrane glycoproteins by triton X-100.

1. The enzymic removal of sialic acid residues from the glycoproteins of the human erythrocyte decreases the solubilization of membrane glycoprotein by Triton X-100. 2. The solubilization of asialoglycoprotein by Triton X-100 may be restored by the addition of borate. 3. Use of this non-ionic detergent in the presence of borate, as a general procedure for the mild solubilization of membrane glycoproteins deficient in sialic acid residues, is discussed.

The relationship of detergent-solubilization plasma-membrane components of rabbit alveolar macrophages to an isolated inhibitor of phagocytosis.

1. A plasma-membrane fraction prepared from rabbit alveolar macrophages by hyposmotic borate lysis is described. 2. Rabbit lung lavages, containing a glycoprotein inhibitor of phagocytosis, may be fractionated by preparative isoelectric focusing in the presence of Triton X-100. 3. Chemical analysis indicates that the glycoproteins of the lung lavage contain sialic acid, fucose, mannose, galactose, hexosamine and appreciable quantities of glucose. 4. The relationship of macrophage membrane glycoproteins, solubilized with Triton X-100 in the presence of borate, to the lung lavage glycoproteins is demonstrated immunoelectrophoretically.

A lectin which binds specifically to beta-D-galactoside groups is present at the earliest stages of chick embryo development.

Extracts obtained from chick embryos at the pre-gastrula and gastrula stages are able to agglutinate trypsinized rabbit erythrocytes fixed with glutaraldehyde. Agglutination is inhibited by saccharides sharing a beta-D-galactopyranoside configuration. Agglutinin activity is also inhibited by desialysed fetuin which bears terminal galactose residues but not by native fetuin, desialysed agalacto-fetuin, bovine submaxillary mucin and desialysed bovine submaxillary mucin. Lectin activity is present in extracts obtained from the embryonic area of the blastoderms as well as in extracts from extra-embryonic endoderm and ectoderm. In extracts subjected to gel filtration on ECD Sepharose, lectin activity eluted between ovalbumim (mol. wt 45 000) and chymotrypsinogen (mol. wt 25 000). Under some experimental conditions, thiodigalactoside, the most potent inhibitor of lectin activity, inhibited the aggregation of cells of the extra-embryonic endoderm of the primitive chick embryo.

Discharge patterns of residents from halfway houses for male alcoholics.

Length of stay (LOS) data for male alcoholics discharged from eitht halfway houses were examined in the light of the hypothesis that the rate of discharge would be a linear function of time. In general, the hypothesis was confirmed, although the rate of discharge for four houses was significantly accelerated during the first two weeks of residence. Further analysis showed that both resident characteristics and house structure (defined in terms of the number of hours of formal activities each week) contributed significantly to LOS. Resident characteristics accounted for 5.3% of the variance in LOS when house structure was controlled, and structure accounted for 1.6% of variance when resident characteristics were controlled. No significant interaction effects were detected. Implications of these results are discussed.

Isolation of a Golgi-apparatus-enriched fraction from leukaemic cells.

1. A Golgi-apparatus-enriched fraction was isolated from acute leukaemic lymphoblasts of AKR mice by using an homogenate stabilized with 1 mM-glutaraldehyde. 2. The isolated fraction, which was shown morphologically to be enriched in dictyosomes, possessed between 44- and 76-fold increase in specific activity, compared with the tumour homogenate, of UDP-galactose-glycoprotein galactosyltransferase and between 3- and10.5-fold increase in relative specific activity of UDP-N-acetygalactosamine-polypeptide N-acetylgalactosaminyltransferase. 3. Plasma membranes isolated from the leukaemic lymphoblasts also possessed glycoprotein galactosyltransferase activity, though in contrast with Golgi-apparatus-enriched material had no detectable polypeptide N-acetygalactosaminyltransferase. 4. The difficulties associated with maintaining the morphological integrity of the Golgi apparatus in subcellular fractionation are discussed.

Comparison of early embryonic and differentiating cell surfaces. Interaction of lectins with plasma membrane components.

1. Cells of the unincubated as well as those of primitive streak chick blastoderm, which are preparing for or are involved in morphogenetic movements, are agglutinated by wheat germ agglutinin, Ricinus communis agglutinin and concanavalin A, but not by fucose-binding protein. 2. Agglutination of these cells with soybean agglutinin occurs only after neuraminidase treatment, while that induced by concanavalin A, wheat germ and Ricinus communis agglutinins is not affected. 3. Trypsin treatment of blastoderm cells had no effect on lectin-mediated agglutination. 4. In contrast, cells derived from 10-and 12-day differentiating chick liver were agglutinated by wheat germ agglutinin only after trypsinization. 5. Mechanically dissociated embryonic liver cells, which are not agglutinated, bind more 3H-labelled wheat germ agglutinin per cell than trypsinized cells, suggesting that during differentiation there may be a spatial reorganization of wheat germ agglutinin receptors within the plasma membrane. 6. Membranes isolated from the above cell types were examined by analytical polyacrylamide gel isoelectric focusing and, in combination with affinity chromatography using wheat germ agglutinin conjugated to agarose, membrane material in the differentiating liver membrane, which binds to this lectin, was identified.
