[Utilization of municipal sludge in agriculture and sustainable development].

Based on the ideas of sustainable development, this paper analyzed the features and disposal situations of municipal sludge and its influence on agricultural and urban development. It is suggested that the land application of municipal sludge is significant and urgent for urban and agricultural sustainable development. Land application of sludge will also be basic outlet for sludge disposal in the future. The key of land application of sludge lies in controlling the content of toxic pollutants in sludge and utilizing sludge scientifically.

[Impact of starvation on predation by male adult Chrysopa septempunctata].

The predation on Schizaphis graminum by male adult Chrysopa septempunctatata at various levels of starvation was examined in this paper. Th results showed that the functional reactive types of predator were not altered by its starvation duration, and the amount(Na) of catched S. graminum by C. septempunctata within 24 hrs varied with duration time(t), which could be expressed by the model: Na = 100/(1 + e-0.3088 - 0.0996t). The relationship between predation rate(v) and time(x) could be described as v = 6.7117 x-0.7928. The male adult predators preferred feeding on the young preys.

A study of Hodgkin/Reed-Sternberg cells using single cell polymerase chain reaction.

OBJECTIVE: To investigate the characteristics of Hodgkin/Reed-Stemberg (H/R-S) cells found in patients with various types of Hodgkin's disease (HD). METHODS: H/R-S cells were micropicked from frozen sections of tissues affected by HD. The DNA from these cells was amplified by polymerase chain reaction (PCR) using immunoglobulin heavy chain gene FR III a/JH primers and light chain gene family-specific primers. RESULTS: A total of 52/135 (35.8%) isolated cells showed the specific products in the reactions. IgH and V kappa 4 rearrangements were repeatedly found in many cells from a lymphocyte predominance type sample; repeated V kappa 4 and individual IgH/V kappa 2,4 rearrangements and individual IgH, V lambda 3/ V kappa 4 rearrangements were found in two different cases of the nodular sclerosis type; repeated IgH/ V lambda 3 and individual V lambda 2,4 rearrangements, repeated V kappa 2,4 rearrangements, repeated V kappa 4 and individual IgH/ V kappa 3 rearrangements, repeated IgH and individual V kappa 3/ V lambda 4 rearrangements were detected in 3 cases of the mixed cellularity type. Repeated and individual IgH rearrangements were found in other 2 cases. CONCLUSION: The H/R-S cells isolated from the lymphocyte predominance subtypes of HD have IgH and V lambda 4 gene rearrangements. This suggests that the lymphocyte predominance type is a proliferation of neoplastic B cells. The cells isolated from the mixed cellularity and nodular sclerosis types derive from B lineage cells at various stages of differentiation because of the presence of their IgH, kappa and/or lambda gene rearrangements. To our knowledge, this is the first time that the lambda gene rearrangement was detected in H/R-S cells.

A cDNA located on chromosome 7q32 shows loss of expression in epithelial cell line of nasopharyngeal carcinoma.

OBJECTIVE: To isolate and clone the tumor suppressor gene on chromosomal region 7q32 correlated with the carcinogenesis of human nasopharyngeal carcinoma (NPC). METHODS: The genotypes of polymorphic microsatellite markers on 7q32 in DNA from 24 biopsies of nasopharyngeal carcinoma and matched normal blood cells were identified. The expression levels of 20 expressed sequence tags (ESTs) on 7q32 between human nasopharyngeal carcinoma epithelial 1 (HNE1) and primary cultures of normal nasopharyngeal epithelial (PNNE) cells were compared using differential RT-PCR and Northern hybridization. The quantity of AA070437 DNA and mRNA was detected by differential PCR and differential RT-PCR, respectively. RESULTS: Loss of heterozygosity (LOH) was found in 25%-46% of NPC biopsies. AA070437 EST expression was down-regulated in HNE1 cell compared to PNNE cells. The down-regulation of AA070437 was found in 30.7% of NPC biopsies and allelic loss of AA070437 was observed in 29.1% of NPC biopsies. CONCLUSION: Our results show that AA070437 EST is negatively related with the occurrence of human NPC and may represent a candidate tumor suppressor gene of NPC on 7q32.

c-myc gene inactivation during induction of nasopharyngeal carcinoma cells with retinoic acid.

OBJECTIVE: To investigate the effects of retinoic acid (RA) on the growth, morphology, oncogene expression and regulation of nasopharyngeal carcinoma cells. METHODS: Nasopharyngeal carcinoma cell line (HNE1) was induced by RA. The RA-treated and control cells were established and cellular morphology and growth patterns were defined. Oncogene expression and regulation were detected by Northern hybridization and DNase-I hypersensitive site analysis. RESULTS: RA markedly inhibited cell growth. The growth of HNE1 cells was reduced to 50% of the control level on the 4th day of RA (10(-4) mol/L) treatment. After 4 days of treatment, the rapidly growing polygonal cells were reversed into a slow growing phenotype, with flattened morphology similar to fibroblast-like cells. Northern hybridization showed that c-myc and c-Ha-ras expression was high in HNE1 cells and undetectable in normal blood cells. c-myc was down-regulated at 48 h of RA treatment. In contrast, the c-Ha-ras was not affected. DNase I hypersensitive site analysis detected changes in the regulatory elements of c-myc and c-Ha-ras genes. 5 hypersensitive sites were found in the c-myc of HNE1 cells, while 3 hypersensitive sites disappeared upon HNE1 induction. However, only 1 hypersensitive site was found in c-Ha-ras of RA treated cells and controls. In normal peripheral white blood cells, no DNase I hypersensitive sites were found in the inactive c-myc and c-Ha-ras gene. CONCLUSION: RA can induce differentiation in a nasopharyngeal carcinoma cell line at high concentration of RA; HNE1 shows some similar patterns of DNase I hypersensitive sites with the common one in other types of cells expressing c-myc. The repression of c-myc expression with induction is accompanied by the loss of 3 DNase-I hypersensitive sites; c-myc has more than one inactive conformation.

Design and clinical application of cervical hollow threaded fixator.

OBJECTIVE: Cervical hollow threaded fixator (CHTF) was designed and applied to stabilize the cervical spine following anterior decompression due to cervical spondylosis or acute cervical disc herniation. METHODS: Medical use titanium alloy with no magnetism was made into a hollow threaded structure with holes on the wall of the fixator. Forty-two patients with cervical spondylosis and acute cervical disc herniation were stabilized using the CHTF technique after anterior decompression. X-ray films and CT scans were taken after the operation to observe the stability and fusion rate of the segments. RESULTS: Patients were allowed to walk one day after the operation and were followed up for 6 to 36 months. The operated segments were stable and solid fusion was observed 3-6 months after surgery. CONCLUSIONS: Immediate stability of the operated segments can be obtained using the CHTF technique. Patients were easy to manage after the operation and returned to their jobs in a short time. The CHTF technique is safe and simple to perform and eliminates the complications of an autograft. It may be an alternative to traditional cervical interbody fusion with bone graft.

[Total wrist arthroplasty].

OBJECTIVE: To review the history and current status of total wrist arthroplasty. METHODS: The original articles about wrist arthroplasty in recent years were reviewed. RESULTS: The properties of wrist prosthesis of different generations were reviewed, with the emphasis on the prosthesis design and biomechanical behaviours. The surgical techniques, complications and salvage procedures were also discussed. CONCLUSION: Although wrist arthrodesis is highly successful in treating the end-stage wrist arthritis, total wrist arthroplasty using well-designed prosthesis remains an alternative for the patients with special demands of hand functions.

[The effects of local stress on bone mass of mandibles in ovariectomized rats].

OBJECTIVE: To observe the relationship between the change of stress environment of mandibles and their bone mass in normal and ovariectomized(OVX) rats. METHODS: The high and low stress environment models of mandibles were established by unilateral maxillary molars extraction. The masticatory sides of the mandibles were regarded as the high stress sides and the non-masticatory sides were regarded as the low stress sides. Ovariectomized rats were utilized as postmenopausal osteoporosis models and Sham-ovariectomized(Sham-OVX) rats as control group. According to these ideas, 48 female rats were divided into 4 groups: 1) OVX + extraction group; 2) OVX + non-extraction group; 3) Sham-OVX + extraction group; 4) Sham-OVX + non-extraction group. The bone mineral densities (BMD) of all mandibles were measured by single-photon absorptiometry bilaterally 1.5 months and 3 months after extraction. RESULTS: The mandibles of ovariectomized rats had lower BMD than those of normal rats(P < 0.05). To normal rats, the BMD of mandibles in low stress sides were lower than the high stress sides 3 months after extraction(P < 0.05), but the BMD of the two sides were not significantly different to those of the non-extraction control ones(P > 0.05). To ovariectomized rats, the BMD of mandibles low stress sides were much lower than the high stress sides(P < 0.01). The BMD of mandibles in low stress sides were much lower than non-extraction ones of OVX group after 3 months(P < 0.05). The BMD of the high stress sides were much higher than those of the non-extraction control ones of OVX group(P < 0.05), but they were still lower than those of the non-extraction control ones of Sham-OVX group(P < 0.05). CONCLUSION: The results indicate that the masticatory functions(the high stress environment) tend to conserve the mandibular bone mass in patients with systemic osteopenia such as postmenopausal osteoporosis and the masticatory deficiency (the low stress environment) tends to aggravate mandibular bone loss induced by systemic osteopenia. The masticatory functions should be rehabilitated for these patients as early as possible.

[Clinical and experimental study on treatment of infantile autumn diarrhea by retention enema with Qilian liquid].

OBJECTIVE: To find out the effective treatment for infantile autumn diarrhea (IAD). METHODS: Retention enema with Qilian Liquid (QLL) was applied to the patients of IAD with positive antigen of rotavirus in stool, and the clinical effect in the treated group was compared with that in the control group in aspects of negative conversion rate of rotavirus and change of immunoglobulin. Animal experimental study was also conducted. RESULTS: The disappearance time of principal symptoms, negative conversion rate of rotavirus and serum levels of IgA and IgG in the treated group were significantly higher than those in the control group (P < 0.01, P < 0.05). Experimental study showed that QLL not only has the the anti-viral effects to stop diarrhea, but also has effects in enhancing immune function and protecting intestinal mucous membrane. CONCLUSION: Retention enema with QLL is an effective therapy for the treatment of IAD.

[Effect of kidney replenishing Chinese herbs on insulin-like growth factor-1 and its receptor in androgen induced sterile rats].

OBJECTIVE: To observe the effect of Kidney replenishing Chinese herbs (KRCH) on insulin-like growth factor-1 and its receptor (IGF-1/IGF-1R) in androgen induced sterile rats (ASR). METHODS: ASR model was set up and screened by vaginal smear. The level of serum IGF-1 was detected by radioimmunoassay, and IGF-1R by immunohistochemical method. RESULTS: (1) Serum IGF-1 level in model rats group > model rats treated with KRCH group > normal control group, but no statistical difference was found among them. KRCH could lower the level of serum IGF-1. (2) Level of IGF-1R in pancreas, liver, ovary and adrenal gland showed an opposite sequence to that of serum IGF-1. CONCLUSION: ASR shows a high IGF-1 level in serum and low IGF-1R level in main target organs, and KRCH could adjust the levels in certain degree. From this, it is inferred that the ovulation effect of KRCH on ASR may come true partly by adjusting the level of IGF-1/IGF-1R.

[Therapeutic effect of weikangfu on gastric precancerous disorder with spleen deficiency syndrome and its effect of gastric mucosal zinc, copper, cyclic adenosine monophosphate, superoxide dismutase, lipid peroxide and 3H-TdR lymphocyte conversion test].

OBJECTIVE: To observe the clinical therapeutic effect of Weikangfu granule (WKF) on intestinal metaplasia (IM), atypical proliferation (ATP) and its therapeutic mechanism. METHODS: Sixty-one patients (treated group) were diagnosed by integrated TCM and western medicine (WM), and treated according to Syndrome Differentiation, the amount of WKF was adjusted according to various Syndromes. The control group consisted of 54 patients, took Weisu granule without considering their Syndromes. Besides, 15 healthy subjects comprised of the control group. Gastroscopy was conducted both before and after the treatment, mucosal sample was taken from antral region of stomach for pathological changes, for IM classification through histochemical staining and for determination of gastric mucosal Zn, Cu, cAMP, SOD, serum LPO and 3H-TdR lymphocyte conversion test (LCT). RESULTS: (1) The symptomatic and pathologic therapeutic effect of the treated group were markedly better than those of the control group (P < 0.05, P < 0.01). (2) The levels of gastric mucosal Zn, Cu, cAMP, SOD and 3H-TdR LCT of the treated group before treatment were all lower than that of the healthy control group, but they were markedly increased after treatment, while serum LPO, however, increased before treatment, significant difference existed between the above groups (P < 0.05, P < 0.01). CONCLUSION: WKF can reverse IM and ATP of Spleen Deficiency Syndrome through the mechanism of improving the level of gastric mucosal Zn, Cu, cAMP and SOD, promoting cytodifferentiation, improving cytoimmunity, reducing superoxide and LPO.

[Observation on short-term effects of Angelica injection on chronic obstructive pulmonary disease patients with pulmonary hypertension].

OBJECTIVE: To study the effects of 25% Angelica sinensis injection on hemodynamics, endothelin-1 (ET-1), angiotensin-II (AT-II), endogenous digitalis-like factor (EDF), pulmonary function and arterial blood gas in the patients with chronic obstructive pulmonary disease (COPD) complicated pulmonary hypertension. METHODS: Sixty COPD patients complicated with pulmonary hypertension in remission stage were randomly divided into two groups, 30 cases in each. The Angelica group and the control group were treated with Angelica sinensis injection and 5% glucose injection (250 ml, intravenous dripping per day for 10 days) respectively. It was designed to investigate the changes of hemodynamics, ET-1, AT-II, EDF, pulmonary function and arterial blood gas. RESULTS: The levels of mean pulmonary arterial pressure (mPAP), pulmonary vascular resistance (PVR), blood ET-1, AT-II and EDF were reduced by (18 +/- 5)%, (27 +/- 8)%, (20 +/- 6)%, (36 +/- 9)%, (38 +/- 11)% respectively, and PaO2 was increased in Angelica group (P < 0.05 or P < 0.01). There were insignificant differences of the above parameters in the control group, and no changes of pulmonary function in both groups. CONCLUSION: Twenty-five Percent of Angelica injection can improve pulmonary hemodynamics through influencing the metabolism of ET-1, AT-II and EDF as well as increase PaO2 of the body.

[The genetic instability of mismatch repair gene linked microsatellite and the evolution of CML].

OBJECTIVE: To explore the relationship between the genetic instability of microsatellite linked with mismatch repair gene and the evolution of CML to blast crisis. METHODS: The loss of heterozygosity (LOH) and microsatellite instability (MSI) of two polymorphic microsatellite markers, D2s123 and D3s1298, linked with mismatch repair gene hMSH2 and hMLH1 respectively, were detected by PCR-silver staining method on the bone marrow cells of 18 CML patients, who clinically progressed from the chronic phase to accelerated phase or blast crisis. RESULTS: Differences in microsatellite D2s123 and D3s1298 at the CML accelerated phase or blast crisis in 5 (27.8%) of the 18 patients were demonstrated compared with chronic phase. For D2s123, MSI and LOH were observed in 1 of 8 patients with accelerated phase (12.5%) and 2 of 10 patients in blast crisis (20%). For D3s1298, MSI and LOH were observed in 2 of 10 patients in blast crisis (20%). CONCLUSION: The genetic alteration of microsatellite D2s123 and D3s1298 may play a role in the progress of some CML cases.

[CO2 laser-microscopy technique in treating tumors near the sella turcica].

OBJECTIVE: To assess the laser-microscopy technique in dealing with tumors near the sella turcica. METHODS: Sixteen patients with tumors near the sella turcica who had been treated surgically by laser-microscopy technique from October 1996 to June 1998 in Haikou municipal hospital were analyzed retrospectively. RESULTS: In these patients, 11 patients had tumors bodies excised totally, and 5 had tumors bodies excised partially. All of then were followed up for 4 to 30 months. The curative effects were satisfactory. In 9 patients with defect in vision before operation, 6 showed obvious improvement, 2 moderate improvement, and 1 unmarked improvement. In 6 patients with endocrinopathy before operation, 4 returned to normal, and 2 showed improvement. In 4 patients with intracranial hypertension before operation, symptoms were retrieved. Only one patient who had diabetes insipidus after operation, and recovered after medication. CONCLUSIONS: CO(2) laser-microscopy technique the treatment of tumors near the sella turcica is advantageous: few accidental injuries because of its high-accuracy; less harassment to the hypophysis and subthalamus; less complication; small exposure of operative field; less bleeding and clear operative field; no stimulation of metastasis of carcinoma;and no interference of bioelectric current.

[Clinical use of acellular allogenic dermis or acellular porcine dermis with split-thickness autologous skin graft in 119 cases].

OBJECTIVE: To observe the effect of acellular allogenic dermis or acellular porcine dermis together with split-thickness autologous skin graft on coverage of deep burn wound and the wound of scar excision. METHODS: Acellular allogenic dermis or acellular porcine dermis produced by our unit, with split-thickness autologous skin graft, were used in repairing various wounds in 119 cases. The take rate of various wounds was compared, and the take rate of wounds in which autologous skin from different sites was used to cover either acellular allogenic dermis or acellular porcine dermis was also compared. Histological examination and follow-up were made in some cases. RESULTS: After tangential excision, eschar excision and scar excision, the wounds were covered with either allogenic acellular dermis or porcine acellular dermis and autologous split-thickness skin. The take rate was found to be (93.4 +/- 3.4)%, (92.1 +/- 4.6)%, (94.5 +/- 3.5)%, respectively. There was no a significant difference in take rate (P > 0.05). No significant difference in take rate between the transplantation of allogenic acellular dermis with autologous split-thickness skin and transplantation of porcine acellular dermis with autologous split-thickness skin was found (P > 0.05). When autologous split-thickness skin harvested from the trunk or extremities was used, the take rate was (93.1 +/- 4.8)%, (89.0 +/- 6.2)%, respectively, whereas the take rate was only about 70% - 80% when skin harvested from the scalp was used. The take rate was only about 40% when acellular allogenic dermis and autologous micro-skin graft were used to cover the wound. Histological examination 19 months after the transplantation revealed normal structure. Collagenous fibers were orderly arranged. There was no apparent hyperplasia of collagenous fibers. Hair follicles, sweat glands and other skin appendages were not found in the healed area. Grossly, no obvious pigmentation was seen, the surface was smooth with slight wound contraction, and the consistency was flexible. CONCLUSION: Acellular allogenic split-thickness dermis or acellular porcine split-thickness dermis with autologous split-thickness dermis for coverage of deep burn wound and the wound of scar excision is an ideal material.

[Reversal of severe left ventricular dilatation after operation and its correlation with cardiac function].

OBJECTIVE: To investigate the relationship between the reversal of left ventricular dilatation and postoperative cardiac function, and prognosis. METHODS: 16 patients with severely dilated left ventricle underwent echocardiographic examinations preoperatively and at discharge (7 - 10 days after operation). Left ventricular (LV) diameters were measured (LVEDD, LVESD) and matched to body surface area (LVEDDI, LVESDI) and fractional shortening (FS); left ventricular ejection fraction (EF) was calculated. RESULTS: Left ventricular dimensions significantly decreased at discharge after operation. Preoperative LVEDD was (78.54 +/- 6.30) mm and postoperative LVEDD (63.77 +/- 13.69) mm (P < 0.01). Preoperative LVESD (60.03 +/- 8.95) mm and postoperative LVESD (53.23 +/- 12.68) mm (P < 0.05). Preoperative LVEDDI was (44.10 +/- 5.24) mm/m(2) and postoperative LVEDDI (36.01 +/- 9.53) mm/m(2) (P < 0.01). Preoperative LVESDI was (34.78 +/- 6.22) mm/m(2) and postoperative LVESDI (30.90 +/- 9.19) mm/m(2) (P < 0.05). Left atrium (LA) dimension was shorter after operation than before operation. Left ventricular construction didn't improve postoperatively, even temporarily decreased at discharge. Preoperative EF was (48.00 +/- 11.71)% and postoperative EF (40.15 +/- 8.69)% (P < 0.01). Preoperative FS was (22.82 +/- 6.27)% and postoperative FS (19.63 +/- 3.42) (P > 0.05). The incidence of postoperative cardiac arrhythmia was 75%. No dimension changes were observed in 2 patients with severely dilated left ventricles after operation: one (LVEDDI 56 mm/m(2)) died, and the other had severe low cardiac output. Six of 14 who had reduced dilated left ventricle didn't reach normal limits, still in the range of severe dilated left ventricule (LVEDDI > 37 mm/m(2)). CONCLUSIONS: Most patients decreased the dimension of left ventricular size early after operation, but some had partial recovery from severe ventricular dilatation. Although left ventricular dimensions (LVEDD, LVESD, LVEDDI, LVESDI) significantly reduced, cardiac function didn't improve at once with the reversal of left ventricular dilatation, but temporarily decreased after operation and the incidence of cardiac arrhythmia was high.

[The protective effect of ligustrazine on optic nerve axons in rabbit eyes with continuous elevated IOP].

OBJECTIVE: To investigate the effects of ligustrazine on optic nerve axons in rabbit eyes with continuous elevated intraocular pressure (IOP). METHOD: Thirty-six NZ rabbits with continuous elevated IOP induced by injection of alpha-chymotrypsin into anterior chamber were divided into two groups by randomization: treatment and control group. Ten mg/kg ligustrazine injected into the treatment group every day for 3 weeks began on the 7(th) day after IOP elevated continuously. The changes of conjunctival microcirculation and optic nerve axonal ultrastructure were observed at different time points. RESULTS: The destruction of conjunctival microcirculation and axonal ultrastructure was milder in treatment group than that in the control group. In addition, the total number of axons and the percentage of axons in optic nerve area were higher, whereas the axonal diameter was smaller in the treatment group. CONCLUSION: It is suggested that ligustrazine protect optic nerve axons from more damage induced by elevated IOP through improving ocular microcirculation.

[Analysis of TCR-gamma gene rearrangement in single H/R-S cells of Hodgkin disease].

OBJECTIVE: To detect the rearrangement of T cell receptor gamma chain gene in single H/R-S cell as an indication of early T lymphocyte differentiation. METHODS: CD30 positive H/R-S cells were isolated with micromanipulator from the frozen sections and analyzed by single-cell PCR. TCR-gamma chain gene rearrangement was detected through semi-nested PCR. In the first round of PCR, a pair of TCR-gamma primers which amplify the segments between Vgamma and Jgamma region (including the N region) of TCR-gamma gene mixing with a pair of beta-globin primers were used. In the second round, only the products positive beta-globin were amplified with the primers for TCR-gamma gene. RESULTS: A 268 bp beta-globin gene specific product was obtained in 24 out of 56 H/R-S cells from 6 cases of Hodgkin's disease. After the first round of PCR, no rearrangement of the TCR-gamma gene was noticed at a single cell level. CONCLUSION: H/R-S cells from the selected cases were not derived from the T-cell lineage but possibly from the B-cell lineage.
