Semipermissive replication of Tipula iridescent virus in Aedes albopictus C6/36 cells.

Comparative studies were carried out using two different insect cell lines, Aedes albopictus and Estigmene acrea, for Tipula iridescent virus (TIV) propagation. Light microscope autoradiography showed viral DNA present in viroplasmic centers (VCs) and an inhibition of nuclear DNA synthesis. These VCs appeared to be morphologically similar in both cell lines when examined by light and electron microscopy. Radiolabeled cDNA was synthesized from RNA samples obtained from infected cells at different times after infection and hybridized to TIV DNA digested with various restriction endonucleases. The results indicated that the pattern of transcription and the kinetics of TIV infection were qualitatively similar in both cell lines. The major TIV DNA components, L (greater than 174 kbp) and S1 (10.8 kbp) that are found in virions in approximately equivalent amounts, were made in both infected cell lines. However, the infected cell lines produced S1 DNA at higher levels relative to L than in virions. The cDNA hybridization studies also revealed that the S1 DNA has sequences that are transcribed and are TIV specific. While VC morphology, levels of L and S1 DNA synthesis, transcription, and capsid protein synthesis were similar in both cell lines, time course electron microscope studies revealed that progeny virions were detected only in the VCs of E. acrea cells and not in the VCs of A. albopictus cells, even by 96 hr p.i. These data suggest that the A. albopictus C6/36 cell line is semipermissive for TIV replication.

[Diagnosis and therapy of injuries of the lamina fibrocartilaginea of the hand (palmar plate)].

Authors studied a frequent injury of the finger joints. The anatomy, the mechanism of the injuries, the diagnostic possibilities of the lamina fibrocartilaginea ("palmar plate") and of the palmar ligaments of the joints are described. In the diagnosis a special significance is attached to the stressed and tangential X-ray pictures. They take the part of the conservative treatment of the injury. In cases of dislocations, that cannot be reduced, of open "palmar plate" injuries and of major hyperextension and dislocations of the broken bone scale, operative treatment is thought necessary. In the treatment of the injuries of the "palmar plate" of the PIP joint they use the so called rein method, that keeps the palmar cartilaginous joint capsule, torn at the injury, on its original place, it enables however the active joint movements, the gymnastics and hinders this way the development of the joint contracture.

[Demonstration of antibodies against gliadin using an immunofluorescence method in childhood celiac disease].

IgA and IgG type antibodies against gliadin were demonstrated in 396 serum samples of 350 patients with immunofluorescence method. The procedure was used in cases of clinically suspected celiac disease and for the control of the diet of patients with diagnosed celiac disease. The sensitivity of the patient material was 83% in the case of antibodies against IgA and 85% with antibodies against IgG. The specificity value with IgA antibodies was 96% and 91% with IgG antibodies. The authors consider the immunofluorescence technique suitable for the demonstration of immune complexes developed against gliadin and circulating in blood. On this basis the method is recommended both for the screening test of patients with suspected celiac disease and for the control of patients on gluten-free diet.

[Investigations of skin damage caused by detergents (author's transl)].

With the aid of an electric impedance test developed by himself, the author studied the effects of the washing ingredients on the skin. From his investigations he came to the conclusion that the different ingredients only cause slight irritation when the same concentration as that found in water used for washing. Their effects is however cumulative and depending on the patient's resistance capability, still may cause dermatitis.

Relationship between chromatin structure and replication in mouse L-cells.

Mouse L-cells treated with cytosine arabinoside, hydroxyurea, fluorodeoxyuridine, methotrexate, or mitomycin C rapidly cease DNA synthesis and stop dividing. Such inhibition of DNA replication is followed by interruption of formation of lysine- and arginine-containing proteins, including chromatin-bound histones, and by a major reorganization of the heterochromatin of the central nucleoplasm, manifest as disaggregation of large clumps of this condensed chromatin. Morphometric analysis revealed both cell and nuclear enlargement in cells treated with such antimetabolites of DNA replication. These observations are in contrast to those made with WT-4 cells starved of isoleucine or treated with cycloheximide. Isoleucine depletion was associated with inhibition of DNA synthesis and continued increase of cell and nuclear volume, but not with massive disaggregation of heterochromatin. Cycloheximide produced inhibition of DNA synthesis and protoplasmic growth, and also prevented structural reorganization of chromatin. A model is presented which suggests that initiation of chromatin replication is associated with a process, dependent upon de novo protein synthesis, which results in chromatin disaggregation. This can be revealed by inhibition of the correct replication of chromatin DNA and chromatin protein.
