Topical cyclosporin A in nickel contact hypersensitivity: results of a preliminary clinical and immunohistochemical investigation.

Four out of eighteen (22%) patients with nickel contact sensitivity showed inhibition of skin patch test responses to the allergen in the presence of topical cyclosporin A (CsA; 5% v/v). No systemic drug absorption or side effects were detected. The clinical response to CsA was accompanied by marked diminution of the T cell infiltrate, although no alteration in the helper/suppressor cell ratio was observed. Expression of the Leu 6 marker on epidermal Langerhans cells and of major histocompatibility complex (MHC) class II antigens (HLA-DR, DQ and DP) on lymphocytes and Langerhans cells was unaffected by topical CsA. The incidence of IL-2 receptor positive lymphocytes in all biopsies was too small to ascertain the influence, if any, of CsA. The prospective use and method of application of CsA in immune contact dermatitis and other immunologically-based skin disorders warrants further evaluation.

Macrophage chemotaxis in experimental tooth movement in the rat.

Extracts from tissue surrounding teeth being moved contained chemotactic activity for peritoneal exudate cells (PEC). The tissue was homogenized, extracted and diafiltered using a 100,000 mol. wt cut-off filter. The filtrate contained statistically-significant chemotactic activity, whereas the retentate did not. Further fractionation of the filtrate using an isoelectric focusing column produced a filtrate fraction in the pH range between 5.0 and 6.8 which was chemotactically active. This sample was analysed for purity and mol. wt using sodium dodecylsulphate polyacrylamide-gradient gel electrophoresis (SDS-PAGE). Gels of the active fraction stained with Coomassie blue showed two distinct bands with molecular weights of 69,000 and 60,000.

Relationship of age to neuropsychological deficit in Tourette's syndrome.

A previous study suggested that neuropsychological deficit in patients with Gilles de la Tourette's syndrome (TS) may increase with age. This question was studied by examining the relationship between age and neuropsychological deficit in 21 TS patients. It was found that increased age was related to greater neuropsychological deficit on a pattern of measures consistent with previous reports. The use of age corrected t-scores ruled out the general relationship of age with test performance as an explanation of these results. In addition, when the subjects were grouped on the basis of age, older patients were significantly worse than younger patients on several measures.

Neuropsychological abnormalities in Gilles de la Tourette's syndrome.

Seven patients with Gilles de la Tourette's syndrome underwent an extensive psychological and neuropsychological examination. On psychological examination, it was found that consistent with previous reports these patients had a high incidence of Verbal IQ-Performance IQ discrepancies on the Revised Wechsler Intelligence Scale for Children, and had average mental arithmetic but poor written arithmetic. On neuropsychological examination, no single measure was performed poorly by all subjects, but there was a high incidence of poor performance on several measures from the Halstead-Reitan Battery. High frequency of abnormal performance for age was observed on the Category, Trail Making and Tactual Performance Tests. In addition, the patients had an interesting pattern of average auditory attention span but poor visual attention span. The pattern of visuomotor and visuospatial deficits was discussed with respect to possible underlying mechanisms.

Potentiation of cigarette smoking and radiation: evidence from a sputum cytology survey among uranium miners and controls.

To assess the effect of cigarette smoking and of exposure to radon daughters, a prospective survey consisting of periodic sputum cytology evaluation was initiated among 249 underground uranium miners and 123 male controls. Sputum cytology specimens showing moderate atypia, marked atypia, or cancer cells were classified as abnormal. As compared to control smokers, miners who smoke had a significantly higher incidence of abnormal cytology (P = .025). For miner smokers, the observed frequencies of abnormal cytology were linearly related to cumulative exposure to radon daughters and to the number of years of uranium mining. A statistical model relating the probability of abnormal cytology to the risk factors was investigated using a binary logistic regression. The estimated frequency of abnormal cytology was significantly dependent, for controls, on the duration of cigarette smoking, and for miners, on the duration of cigarette smoking and of uranium mining.

Studies on gonococcus infection. XI. Comparison of in vivo and vitro association of Neisseria gonorrhoeae with human neutrophils.

The levels of association between Neisseria gonorrhoeae and neutrophils as determined by microscopic study of smears of urethral exudate from men with acute gonorrhea and by in vitro assay of N. gonorrhoeae cultivated from those same individuals showed statistically significant correlation. The percentage of N. gonorrhoeae attached to or ingested by neutrophils varied from 30% to 90% in the clinical smears. Neither the apparent in vivo nor the in vitro association with neutrophils could be correlated with piliation of the N. gonorrhoeae studied.
