Elevated titer of antibodies to Simian sarcoma virus envelope antigen (gp70) and normal response to influenza virus in untreated Danish Hodgkin's patients.

Untreated Danish Hodgkin's disease (HD) patients and paired age- and sex-matched controls were tested for serum antibodies to Epstein-Barr virus (EBV), six strains of influenza virus and Simian sarcoma virus/Simian sarcoma-associated virus [SSV(SSAV)] antigens. HD sera showed significantly elevated titers against EBV and both increased incidence and mean titer of antibodies to the envelope glycoprotein of SSV(SSAV), whereas testing against the influenza viruses revealed no differences between HD and controls. A focus reduction assay demonstrated a low incidence in HD and controls of sera with neutralizing effects against SSV(SSAV) and the "baboon" C-type virus component of the human HL-23 virus complex, which seemed coupled to HL-A type W19.

Pilot study on the growth inhibition by estramustine phosphate (Estracyt) of rat mammary tumours sensitive and insensitive of oestrogen.

Rat mammary tumours induced by 7,12-dimethylbenz(a)anthracene had a higher concentration of 3H and 14C than muscle after injection of estramustine phosphate labelled with 3H in the oestrogen moiety and 14C in the alkylating moiety. Thin-layer chromatography showed that dephosphorylated estramustine phosphate was present in the tumours but no free oestradiol-17beta. The uptake of the drug in the tumours was parallelled by a dose dependant retardation of tumour growth and a prevention of tumour number increase. Estramustine phosphate also retarded growth of mammary tumours resistant to treatment with oestradiol-17 beta. It is concluded that estramustine phosphate has a greater effect on tumour growth than oestrogen.

Presenile corneal arcus in healthy persons. A possible cardiovascular risk indicator in younger adults.

The results of blood-lipid screening of 200 39--49-year-old participants with incidentally diagnosed corneal arcus from the Copenhagen City Heart Study are reported. A large sample of study-participants of the same age-group without arcus served as controls. In general, the arcus-group had higher se.-cholesterol than the control-group (P less than 0.01 in males and less than 0.02 in females). The se.-triglycerides in the two groups did not differ. A trend towards positive correlation between arcus-intensity and cholesterol-level was demonstrated by non-parametrical statistics. By use of the same statistics, an association between lipids and arcus-localisation was searched out. Dense arcus in the nasal/lower limbal regions occurred together with hypercholesterolemia more often than did a marked upper/outer arcus. Consequently, based on the present investigation, se.-cholesterol-screening of middle-aged persons with marked lower/nasal arcus seems justified, since hypercholesterolemia might will be an underlying cause. Finally, the importance of diagnosing and treating hypercholesterolemia is briefly discussed.

Mammary tumour inhibition and subacute toxicity in rats of prednimustine and of its molecular components chlorambucil and prednisolone.

Prednimustine, a chlorambucil ester of pregnisolone, retarded growth of DMBA-induced mammary tumours in rats and reduced the number of tumours. A combination of chlorambucil and prednisolone (C + P) in the same proportion as in prednimustine, had similar effects, 8 and 26 mg per kg of the C + P combination being equipotnet to 16 and 64 mg per kg of prednimustine, respectively. The mortality figures suggested that prednimustine was considerably less toxic than equipotent doses of C + P. This toxicity difference was confirmed in a parallel investigation of the subacute toxicity in rats of prednimustine and C + P. This study showed that the mortality, reduction of lymphocytes and platelets, and bone marrow depression was much lower after pregnimustine than after equimolar amounts of the C + P combination. The results suggest that the low toxicity of prednimustine makes this drug a better cytostatic agent than the C + P combination treatment.

Generality of learning differences in brain-weight-selected mice.

Eight lines of mice from two independent selection programs for high, medium, and low brain weight were raised in enriched environments and tested on active avoidance, water maze, operant discrimination, and passive avoidance tasks. Although statistically reliable performance differences were found on each task within at least one selection, there was no consistent relation within selections and across tests or within tests and across selections between brain weight and performance. The results emphasize the necessity of eliminating confounded performance variables in behavior genetic research on learning phenotypes and suggest that brain weight-learing corrleations in mice are either small or nonexistent.

Haemodynamic effects of intravenous disopyramide in heart failure.

The haemodynamic effects of disopyramide have been studied in 11 patients with manifest or imminent heart failure. Disopyramide, 2 mg per kg body weight, was given intravenously during right and left heart catheterisation. The cardiac index decreased by an average of 28% (p less than 0.01); mean maximal left and right ventricular end-diastolic pressures were increased by 5.0+/-0.9 mm Hg (p less than 0.01) and 5.0+/-0.6 mm Hg (p less than 0.05), respectively; and left ventricular systolic pressure fell slightly but significantly (p less than 0.05). No significant change in right ventricular systolic pressure was seen. Pulmonary wedge pressure rose on average by 2.7 mm Hg (p less than 0.05). No significant change in heart rate was observed in 5 patients with sinus rhythm. In 6 patients with atrial fibrillation, there was a significant (p less than 0.01) increase in heart rate; the average increase in heart rate for the entire group was 19.6 beats per minute. The maximum effect on all the parameters occurred 7-11 minutes after the injection, and it gradually subsided during the following 10 minutes. It was concluded that disopyramide had a potentially serious myocardial depressant effect.

Growth patterns and chromosome constitutions of human malignant tumours after long-term serial transplantation in nude mice.

Five human malignant tumours, heterotransplanted to athymic nude mice, have been re-investigated after long-term serial transplantation, and their growth patterns and chromosomal constitutions compared with their early appearance. After 27-56 passages over 3 1/2--5 1/2 years, all of the tumours--two adenocarcinomas of the colon, two malignant melanomas, and one Burkitt's lymphoma--retained the cytological and histological appearance of the inoculated human material. All the mitoses observed in the chromosome studies were of human karyotype. The fact that no total chromosomal species shift, no interspecies hybridization, and no changes in biological properties appeared--as observed in some other heterotransplantation systems--would indicate that human tumours grafted to nude mice are more suitable models in, e.g., studies concerning cancer chemotherapeutics.

Adams-Stokes seizures due to ventricular tachydysrhythmias in patients with heart block: prevalence and problems of management.

One hundred and twelve patients with heart block and chronic tendency to syncope were ECG-monitored during syncope. Ventricular tachycardia and/or fibrillation (VT-VF) was observed as the cause of syncope in 11 patients: in 6 of 20 patients with chronic third degree A-V block, in 3 of 65 with paroxysmal A-V block and in 2 of 27 with S-A block. The R-R interval preceding the escape beat which initiated VT-VF varied between 1.2 and 2.2 seconds. The cerebral attacks were amenable to long-term pacemaker treatment. However, relapses of VT-VF were observed during pacing with a low rate of 55 per minute and during short interruptions in pacing, as produced by intermittent pacemaker failure or threshold determination. In one patient, supplementary treatment with a beta-blocking agent had to be given to suppress exercise-induced attacks of VT-VF after pacemaker implantation.

On thorotrast leucaemia. Evolution of clone of bone marrow cells with radiation-induced chromosome aberrations.

Results are presented of chromosome studies of bone marrow cells from a 62-year-old woman. The patient had been given 40 ml of Thorotrast in connection with a neuro-radiological examination 34 years earlier. Clinically, the patient was now considered to be in an incipient myeloid leucaemic phase. Ninety-seven per cent of the bone marrow cells belonged to a clone with characteristic marker chromosomes induced by radiation. Few data are available on chromosome analysis of bone marrow cells from Thorotrast patients. Therefore, the results of the present study are compared with data from chromosome analyses of 32P-treated patients with polycythaemia vera, in whom large clones were found in the bone marrow. The results of chromosome analysis of bone marrow cells from the Thorotrast patient support previous hypotheses concerning the carcinogenesis in radiation-induced leucaemia in patients with polycythaemia vera, treated with 32P. In the latter patients, the clone cells are pressumed to represent cell populations with selective qualities, originating from radiation-damaged cells, which possibly possess an increased tendency to malignant transformation because of a more pronounced sensitivity to carcinogenic agents. The cells might also be potentially malignant and manifest themselves as leucaemic cells, if the patient's immunological defence mechanism is broken.
