Krestin (PSK).

A polysaccharide preparation isolated from Coriolus versicolor (Fr.) Quél. of Basidiomycetes (PSK) predominantly consists of glucan and approximately 25% tightly bound protein. PSK was effective against various allogeneic and syngeneic animal tumors and has been given orally to cancer patients. Various suppressed or enhanced immune responses of tumor-bearing animals were restored to normal levels by the administration of PSK in the tumor models tested. The killer T cell activity was augmented in tumor-bearing mice by intraperitoneal or oral administration of PSK, and there was correlation between the PSK associated antitumor effect and the killer T cell activity. It was found that PSK competed with immunosuppressive substances isolated from tumor-bearing mice and that the intestinal immune system appeared to be modulated by oral administration of PSK. After oral administration of 14C- or 35S-labeled PSK to normal rats, it was found that small or large molecular substances appeared in the serum depending on the time elapsed after administration, an indication that large molecular size products were from the digestive tract.

[Combined therapy with microwave hyperthermia and adriamycin in MM2-bearing mice].

C3H/He mice were inoculated i.p. with synergic MM2 tumor cells(2 X 10(6) cells/mouse), and subsequently treated either by systemic hyperthermia induced by microwave irradiation (2450 MHz)alone, administration of ADM alone, or a combination of both. The mice were exposed to hyperthermia for 5 minutes everyday at an output of 10, 20 or 30 watts. ADM was administered i.p. for 3 days at dose levels of 0.025 mg/kg, 0.05 mg/kg or 0.075 mg/kg body weight. The mean survival time and the mean relative body weight were recorded. From these data, tumor-suppressing effects of the treatments were analysed by one-way or two-way variance analysis. Mice treated with 30-watt microwave irradiation survived significantly longer than control mice without treatment (P less than 0.05), as did mice treated with ADM alone (P less than 0.01). The mean relative body weight was similar among the microwave, ADM and control groups. When the mice were given microwave treatment combined with ADM, significant difference was observed in the mean survival time by one-way variance analysis (P less than 0.01), but no significant difference was observed by two-way variance analysis. The best condition of mice and the optimal dose of ADM still remain to be determined to facilitate more efficient tumor-suppression using this combination therapy.

[Tumor suppressing effect of systemic hyperthermia with 2350 MHz microwaves combined with 5-fluorouracil in mice].

C3H/He mice were inoculated i.p. with syngeneic MM2 tumor cells (2 X 10(6) cells/mouse), and they were subsequently treated by systemic hyperthermia with microwave irradiation (2450 MHz) alone, administration of 5-FU alone, or a combination of the both. The mice were exposed to hyperthermia for 5 minutes everyday with an output of 10, 20 or 30 watts. 5-FU was administered i.p. for 3 days at dose levels of 2.5 mg/kg, 5 mg/kg or 10 mg/kg body weight. The mean survival time and the mean relative body weight were recorded. From these data, tumor-suppressing effects of the treatments were analyzed by one-way variance analysis or two-way variance analysis. The mice treated with microwave irradiation alone or 5-FU alone survived significantly longer than the control mice receiving no treatment (P less than 0.01). However, no significant difference in mean survival time was observed between the microwave group and the 5-FU group. The mean relative body weight was similar among the microwave, the 5-FU and the control group. When the mice were treated with microwave irradiation and 5-FU, significant difference was observed in the mean survival time (P less than 0.01), and the combination of microwave irradiation with an output of 20 W and 10 mg/kg of 5-FU seemed to be optimal. Determination of an optimal combination was difficult with the mean relative body weight, because body weight decreased due to side effect of the combination therapy.

[Experimental study on the anti-tumor effect of microwaves].

C3H/He mice bearing a mammary carcinoma, MM2, were treated by systemic hyperthermia with microwave irradiation (2450 MHz). An output of 30 watts of microwaves for 5 min at a distance of 20 cm from the animal raised and maintained the rectal temperature of mice to higher than 42 degrees C for 2.2 min and higher than 40 degrees C for 10.2 min. The mice were inoculated i. p. with MM2 tumor cells (2 X 10(6) cells/mouse), and then they were exposed to hyperthermia every day; 10 watts for 2, 4, or 6 min, or 20 watts for 2, 4, 6 min. The control mice receiving no irradiation and the mice treated with irradiation of 10 watts for 2 min or 20 watts for 2 min died within 21-23 days. Whereas, 40 percent of the mice treated with microwave irradiation of 10 watts for 4 or 6 min, or 20 watts for 4 or 6 min survived longer than 30 days. The 6 mice, which survived longer than 120 days, were challenged by reinoculation with MM2 tumor cells (2 X 10(6)cells/mouse). All the mice but one survived longer than 30 days with no accumulation of ascites. These experimental data suggest that the systemic hyperthermia with microwave irradiation (24 50 MHz) might suppress the tumor growth in vivo.

Forssman antibody levels in sera of cancer patients.

By means of SRBC lysis in agarose gel and Forssman (F) liposome lysis assay, serum levels of F antibodies were determined in various cancer patients. F antibody titers of the patient's sera were shown to be lower than that of normal controls. Following radical surgery, F antibody titers were elevated significantly in most of the patients with advanced gastric, colorectal and lung cancer. Similar tends of anti-A antibody titers were observed in group O and B patients with advanced cancer.

Immune response to alien histocompatibility antigens on Epstein-Barr virus transformed cells.

Expression of alien histocompatibility antigens on Epstein-Barr virus transformed, cultured lymphoblastoid cell lines (LCL), which were established from normal peripheral blood lymphocytes (PBL), was studied by means of mixed lymphocyte reaction (MLR), cell mediated lysis (CML) and primed lymphocyte typing (PLT). Stimulation of PBL by autologous LCL resulted in some MLR responses and generation of cytotoxic effector cells against autologous LCL. Restimulation of PBL by 16 individual allogeneic PBL failed to prime an individual lymphocytes against autologous LCL in PLT tests. However, stimulation of PBL by a pooled normal PBL resulted in generation of cytotoxic cells against autologous LCL. Culturing of stimulated PBL in the presence of T cell growth factor (TCGF) for 30 days was shown to maintain cytotoxic effector cells in the cell population.

[Effect of levamisole hydrochloride on advanced and recurrent breast cancer--LMS randomized controlled study of a group responding to CAF (cyclophosphamide, adriamycin, and 5-FU) therapy].

The effect of levamisole HCL on advanced and recurrent breast cancer was investigated cooperatively by a randomized controlled study using envelope method. Patients with advanced and recurrent breast cancer who had showed either complete response or partial response to the previous CAF therapy (cyclophosphamide 100 mg/body/day p.o. from day 1 to 14, adriamycin 30 mg/body/day i.v. on day 1 and 8, 5-FU 500 mg/body/day i.v. on day 1 and 8) were entered in this study and divided randomly into a control group and a levamisole group. No further treatment was given to patients of the control group until they fell into progressive disease. Patients of the levamisole group were given the drug at a daily dose of 150 mg for 3 consecutive days every fortnight. Fifty-nine cases were entered in this protocol, but 9 cases of them were ruled out as exclusions or dropouts. Therefore, the number of eligible cases was 31 in the levamisole group and 19 in the control group. Both the duration of remission and the survival time were significantly (p less than 0.05) prolonged in the levamisole group compared with the control group. There was no difference in side effects between these two groups.

Inhibition of leukocyte migration in agar by soluble extract from a human ovarian carcinoma cell line.

A soluble membrane extract was prepared by a hypotonic extraction method from a cultured cell line, CKS, derived from serous cystadenocarcinoma of the ovary. The leukocyte migration inhibition assay in agar was used to determine the cell-mediated immune response with this extract in patients with ovarian carcinoma in comparison with patients with other benign or malignant tumors and normal healthy subjects. With 100 micrograms protein/ml of the extract, leukocytes from 15/25 patients with ovarian carcinoma showed positive response, whereas 5/30 patients with benign tumors, 2/23 patients with non-ovarian carcinomas, and 0/13 normal healthy controls did so. These findings suggested that the soluble membrane extract from the CKS cells specifically inhibited migration of leukocytes from patients with ovarian carcinoma. There was a tendency that the stage III or IV patients responded to the extract more frequently than the stage I patients irrespective of histological type of ovarian carcinoma. The migration inhibition assay with a basic encephalitogenic protein (100 micrograms protein/ml) from bovine brain was performed simultaneously with the ovarian cancer extract. One of 20 patients with ovarian carcinoma, 1/29 patients with benign tumors, 1/22 patients with non-ovarian carcinomas, and 0/13 normal healthy controls responded positively. Therefore, cross-reactivity between the basic protein and tumor-associated antigens could not be demonstrated in the present study.

Effect of anticancer agents on the susceptibility of tumor cells to immune cytolysis.

Effect of anticancer agents on the immune cytolysis of tumor cells was investigated. Cell of ascites hepatoma line of C3H/He strain of mouse, MH134, were treated with anticancer agents and used for target cells in immune cytolysis experiments. In cytotoxicity tests, MH134 cells pretreated with mitomycin C or Adriamycin were lysed by xenogeneic or allogeneic antisera and complement at significantly higher rates than non-treated MH134 cells. To test cytostatic effect of antisera, target MH134 cells were pretreated with much lower concentration of anticancer agents than that in the cytotoxicity tests, the anticancer-agent-treated tumor cells were damaged with complement and the xenogeneic and allogeneic antisera at higher rates than the non-treated tumor cells. Allogeneic spleen cells from DDD mice sensitized with MH134 tumor damaged anticancer-agent-treated MH134 cells at higher rates than non-treated MH134 cells. These results suggested an enhancing effect of anticancer agents on immune cytolysis of tumor cells in hosts who could actively and/or passively immunologically respond against tumor.

A newly established cel line derived from human ovarian adenocarcinoma.

A continuous cultured cell line, CKS, was established from the ascites of a serous cystadenocarcinoma of the ovary and has been maintained in vitro for more than 3 years. The cells grew in a medium containing human umbilical cord serum and form a monolayer in a mosaic fashion with polyhedral or angular cells of varying size together with occasional giant cells. Hemicyst formation was evident in confluent culture. The ultrastructural features noted, were well developed microvilli and poorly developed junctional complex on the cell surface associated with abundant secretory vesicles in the cytoplasm. Population doubling time obtained from the growth curve was about 79 hr. The chromosomal modal number was near-diploid with minimal aberrations. Heterotransplantation of the cells was successful in ATS-treated hamsters and athymic nude mice. Histological feature of the heterograft much resembled those of the original tumor.

Cross-reactive antibodies against human cultured B cells and bovine erythrocytes in human renal transplantation sera.

Sera of 58 recipients of renal allografts were studied for the presence of antibodies against cell cultures of human B lymphoid cell lines (B-LCL) and bovine erythrocytes (BRBC). Cytotoxic anti-B-LCL antibodies were found in 13% of the sera from recipients with the grafts and in 67% of the sera obtained after removal of the rejected grafts. Most of these sera also contained BRBC lysins of high titers. Absorption studies showed that the anti-B-LCL antibodies are directed against antigens shared by BRBC and that they can be absorbed with corresponding graft tissues. The specificity of BRBC lysins found in some of the transplantation sera was shown to be similar to that of Hanutziu-Deicher antibodies.

Establishment and characterization of a human neuroblastoma cell line in tissue culture.

A new continuous cell line, GOTO, has been established in tissue culture from a human neuroblastoma arising from the adrenal and maintained for 5 years. The cells are small and fibroblast-like, and grow in dense layer. Population doubling time was approximately 48 hr. The modal chromosome number was near-diploid. Neither marker chromosomes nor double-minute chromosomes were abserved. Inoculum of 10(7) cells produced tumors in ATS-treated hamsters and athymic nude mice, and the histological appearance of tumors was consistent with undifferentiated neuroblastoma. Neural features of the cells were also confirmed by ultrastructural examination. Biochemical analysis showed this cell line to be cholinergic. Morphological differentiation leading to neurite formation was induced by cultivation in serum-free medium.

An ascites form of human mammary carcinoma transplantable in nude mice.

Mammary carcinoma cells from pleural effusion of a patient were inoculated into the peritoneal cavity of nude mice, and a large amount of ascites was produced about 120 days later. From the ascites, serial passages in the same form were successful in nude mice by intraperitoneal injection of 10(7) or more cells. The ascites cells retained the morphology almost similar to that of the patient tumor cells, whereas specific estrogen-binding proteins in the cytoplasm disappeared after growing in male nude mice. The results were compared with those of other established human cancer cell lines in nude mice.

Establishment of cultured cell lines derived from a human gastric carcinoma.

A human gastric carcinoma cell line, KATO-II, and its subline, KATO-III, have been established in vitro from a pleural effusion of a 55-year-old male patient. Kato-II cells have been maintained using a culture medium containing human cord serum and KATO-III cells have been cultured with a medium containing fetal bovine serum. Both cell lines are morphologically quite similar; they grow in vitro floating free and have cytological features of signet ring cells. Population doubling times for KATO-II and KATO-III were 74 hours and 36 hours, repsectively. The modal chromosomal numbers of these cell lines fell in a tetraploid range. Heterotransplantation of KATO-II and KATO-III cells was successfully done in the cheek pouches of antithymocyte serum-treated hamsters. Microscopical appearance of the resultant tumors was consistent with poorly-differentiated adenocarcinoma comparable to the original tumor.

Effects of the combination of tumor-specific and nonspecific immunization on tumor suppression.

The crushed whole Aoyama B cells attached to oil droplets were found to be most suitable for the adjuvant effect of Mycobacterium tuberculosis against tumor growth when judged by the criterion of effectiveness and a clinical application with minimal side effects. The immunogenicity of tumor cells was well stabilized and maintained with a suitable concentration of formalin; this was less than 1% and the optimum concentration was 0.05% or 0.1%. When the tumor cells treated with the optimum concentration of formalin were combined with suitable adjuvant, immunoprotection against the tumor progress was accelerated.
