PCR amplification products are of limited use for the study of DNA/protein interaction.

Conventional methods for labeling double-stranded DNA lead to high specific activity. Yet they often alter the target DNA sequence to such an extent as to prevent a meaningful protein/DNA interaction analysis. Therefore we tried to establish a polymerase chain reaction (PCR)-based method which allows radiolabeling to high specific activity and should maintain the protein binding capability of small double stranded DNA fragments. By using PCR it is possible to label double stranded DNA to high specificity, but the protein binding capability of such DNA is drastically reduced.

The granuloma pouch: an in vivo model for pharmacokinetic and chemotherapeutic investigations. I. Biochemical and histological characterization.

The "granuloma pouch" model in rats was evaluated on the basis of biochemical, haematological and histological factors. Seven days after formation of the pouch following the intramuscular injection of air and a mixture of 1% croton oil in olive oil as an irritant, the pouch was filled by approximately 5 ml of haemorrhagic exudate. Biochemical assessment of the exudate revealed that its main characteristics were increased lactate dehydrogenase levels as well as increased alpha 2- and gamma-globulin fractions. Histological sections of the surrounding tissue showed a typical granulomatous inflammatory response. The granuloma pouch can therefore be characterized as a local, chronic inflammation caused by the creation of a body cavity. The biochemical data of the pouch exudate were compared with those of human wound fluid obtained after surgery; since the values were in very good agreement, it is assumed that the granuloma pouch model reflects the clinical situation in inflamed wound cavities following the extirpation of organs.

The granuloma pouch: an in vivo model for pharmacokinetic and chemotherapeutic investigations. II. Microbiological characterization.

In this paper we have described the microbiology of the granuloma pouch model in rats. We studied the biochemical parameters of pouch exudates infected with Escherichia coli. Data revealed that the inflammatory response increased during the course of infection since lactate dehydrogenase levels as well as alpha 2 and gamma-globulin fractions were increased in comparison to uninfected controls. Infection of the pouches did not spread. We also monitored the growth characteristics of four different E. coli strains. In vitro incubation of these strains in human and rat serum as well as in pouch exudate, and the in vivo growth rate in infected pouches revealed that the degree of serum sensitivity was clearly related to viability in vivo. Serum-resistant strains grew well in pouch exudate, whereas serum-sensitive strains were eliminated from the infected pouches. Since elimination of these strains was dependent on the challenge dose, we concluded that cellular and/or humoral host defense mechanisms became locally exhausted or inactivated. Thus, the granuloma pouch represents a local bacterial infection of a poorly defended, inflamed body cavity.

The measurement of radioactive noble gases by DWD in the frame of the Global Atmospheric Watch programme of WMO.

The Deutscher Wetterdienst (DWD, German Meteorological Service) is integrated into the Global Atmospheric Watch programme (GAW) of the World Meteorological Organization (WMO). According to this programme a variety of chemical compounds and radionuclides are measured at global stations. At the research platform "Schneefernerhaus" 7Be, 222Rn, and its decay products, 14C in CO2, tritium as HTO, 85Kr and 133Xe are continuously monitored by the DWD or are sampled and then measured at the central laboratory in Offenbach. The results are used as additional information for studying atmospheric mixing processes or on the other hand as information about the background level of radioactivity. As a main subject of this paper the integration and partly the optimization of sampling and measuring procedures for the detection of noble gases are described. In particular, the methods of quality assurance are discussed for Kr and Xe.

[Clinical experiences with Wilhelm's method of wrist joint denervation].

Wrist joint denervation for treatment of painful arthrosis was carried out in 36 patients in the last five years. 33 Patients could be followed up. Besides subjective reports on pain and maximum stress, strength, mobility and the X-ray of the wrist-joint were also considered in the evaluation of the results. As a whole, a success rate of 84% could be attained. We have not observed disorders of sensitivity or trophic alterations. An increase of symptoms after the second postoperative year was noteworthy.

Ultrasonically tissue-mimicking liver including the frequency dependence of backscatter.

Tissue-mimicking (TM) liver, previously reported by our laboratory, mimics the speed of sound, density, and attenuation coefficient (including frequency dependence) of liver; however, scatter properties are only qualitatively simulated. Two new versions of TM liver are reported here which not only mimic liver with respect to sound, density, and attenuation coefficient, but also with respect to backscatter coefficients, including the frequency dependence of the latter. Compositions, methods of production, and comparisons of ultrasonic properties with those found in the literature for human liver are presented.

The primary structures of the core antenna polypeptides from Rhodopseudomonas marina.

The antenna complex B880 of Rp. marina has been isolated by applying ion-exchange chromatography on Whatman DE-52 resin and sucrose density centrifugation of LDAO-solubilized photosynthetic membranes. The antenna polypeptides B880-alpha and B880-beta were prepared by organic solvent extraction of extensively dialyzed and freeze-dried B880 antenna complex material or photosynthetic membranes. Gel filtration on Sephadex LH-60 and ion-exchange chromatography on Whatman DE-32 resin in the presence of organic solvents and an additional step on a C-8 reversed phase column yielded pure alpha- and beta-apoproteins. Their complete primary structures have been elucidated using automated Edman degradation and carboxypeptidase digestion. According to quantitative Edman degradation the ratio of B880-alpha and B880-beta has been determined as 1:1 in the isolated antenna complex as well as in the photosynthetic membrane. B880-alpha of Rp. marina, presumably N-formylated, consists of 52 amino acid residues and is 75, 56, 52 and 44% homologous to the corresponding core antenna polypeptides of Rs. rubrum, Rp. viridis, Rb. capsulatus and Rb. sphaeroides. In contrast, B880-beta (56 amino acid residues) is less homologous to the corresponding core beta-antenna polypeptides of the same strains (57, 51, 41 and 42%). It shows an extended N-terminal domain as compared to the B880-alpha polypeptide. Apart from the typical structural features of bacterial membrane-bound antenna polypeptides (three domain structure. His-residue in the hydrophobic stretch) the antenna polypeptides of Rp. marina are structurally related to polypeptides of core antenna complexes with strong near infrared circular dichroism signals.

The primary structure of the presumable BChl d-binding polypeptide of Chlorobium vibrioforme f. thiosulfatophilum.

In addition to the previous isolated and sequenced polypeptides from green photosynthetic sulfur bacteria, which are presumably involved in binding BChl c and e, an analogous polypeptide has been purified from the BChl d-containing bacterium Chlorobium vibrioforme f. thiosulfatophilum. The primary structure of this 6.15 kDa polypeptide was determined. It shows an extremely high homology (98.3%) to the corresponding polypeptide from Pelodictyon luteolum, indicative of an important functional role.

The New Stories/New Cultures after-school enrichment program: a direct cultural intervention.

This article describes the organization, curriculum, and outcomes for New Stories/New Cultures, an activity-based program for after-school enrichment in five schools in the low-income neighborhood near a major American university. The program encourages students (70% Hispanic-American, 30% African-American) to experience themselves as producers of culture, not just as consumers. Its methods include (a) creative team use of video equipment and other expressive media and (b) lessons about media literacy (i.e., making critical choices about images and activities depicted in popular culture and commercials). Outcome measures with the cohort of fifth and sixth graders support the programs occupation-based philosophy. They show that students are more likely to experience themselves as building skills when engaged in activities that are both challenging and enjoyable. The students reported greatest engagement and enjoyment in activities that were creative, team-based, and involving media production. These same activities were correlated with increased self-esteem. The term direct cultural intervention is used to describe the application of occupational principles and critical perspectives to provide a population with conceptual tools and skills for interpreting and successfully navigating the social world.

[Acquired aphasia with convulsion anomalies in the developmental age: clinical neuropsychological and electroencephalographic study of a case].

A four year old boy presented three epileptic seizures of psychomotor type; immediately after he began to show a progressive and rapid dissolution of speech, until he became completely aphasic after few weeks. Since then, repeated EEG examinations have always shown anomalies of epileptic type, located on the left hemisphere, at times on the right, at times bilaterally asynchronous. The neurological, psychic, audiological, chemical-biological and neuro-radiological (bi-lateral carotidogram and penumoencephalogram) exams did not show any other anomalies. After an observation period three years, the AA. underline the following evolutive aspects of the case: 1) The aphasic syndrome is on the way to slow improvement both in its expressive component and in its perceptive component. The recovery of speech seems to follow, with a slower rhythm, the stages of acquisition of the speech in the normal subject. 2) Diversely from other cases of the literature, no positive correlation exists between the gravity of the aphasic syndrome and that of the EEG anomaly: they have worsened while the disturbance of the speech have partially regressed.

Stereotactic radioisotope implantations in the treatment of inoperable low malignancy neoplasms.

The authors report their experience of 53 patients with low grade malignancy cerebral neoplasms treated by means of stereotactic 125-I permanent interstitial radiotherapy. The method has been employed from 1981 to date. The authors confirm the validity of the technique in this brief report, and stress the greater efficacy of this treatment in young patients (less than 40 y/o) than in adults.

Clinical course of post-traumatic coma.

The Authors describe the clinical course of 331 cases of post-traumatic coma, studied during the two-year period 1970-1971 at the 2nd Division of Neurosurgery (Ospedale Maggiore, Bologna). Among them, 42% died from trauma-related causes; 40% survived and had a complete reintegration into society; 18% survived at a lower social level or remained totally excluded from society. The quality of survival does not depend on the presence and degree of organic sequelae only, but also on psycho-social factors.
