Purification of a bovine counterpart to human prealbumin and its interaction with a bovine retinol-binding protein.

A bovine counterpart to human prealbumin was purified from bovine serum by thiol-disulfide exchange chromatography on thiol-Sepharose 4B and affinity chromatography on human retinol-binding protein linked to Sepharose 4B. The bovine prealbumin had alpha1-mobility on agarose gel electrophoresis at pH 8.6. It has the same molecular weight as human prealbumin on gel filtration and consisted of subunits with a molecular weight of 12 500. This is compatible with a tetrameric structure for the bovine protein. Antiserum against human prealbumin cross-reacted with bovine prealbumin and vice versa. The bovine prealbumin formed at high ionic strength complexes with another bovine serum protein which were dissociated at low ionic strength. This property was used to isolate a protein from bovine serum, by chromatography on bovine prealbumin linked to Sepharose which cross-reacted with antiserum against human retinol-binding protein; had a molecular weight of 21 000 and alpha 2-mobility on agarose gel electrophoresis. It was concluded that the latter protein was a bovine retinol-binding protein.

Purification of prealbumin from human and canine serum using a two-step affinity chromatographic procedure.

1. A two-step-method is described by which human and canine prealbumin have been isolated from serum in good yield and high purity. The first step involves binding and release of prealbumin by a thiol-disulfide interchange reaction with thiol-Sepharose 4B. The second step consists of reversible binding of prealbumin to retinol-binding protein covalently linked to Sepharose 4B. 2. The canine prealbumin was immunochemically identified as an alpha2-globulin. Like human prealbumin it is a tetramer with some 10% smaller subunits than human prealbumin. Both proteins are devoid of carbohydrate and both bind thyroxin.

Low molecular weight proteinuria and slight hyperlipoproteinemia in three mentally retarded brothers.

Mental retardation in combination with proteinuria and a slight hyperlipoproteinemia was found in three brothers. The increased urinary protein excretion was dominated by albumin and the low molecular weight proteins retinol-binding protein (RBP) and beta2-microglobulin, indicating the presence of proximal tubular dysfunction. However, there was no glucosuria, phosphaturia or amino aciduria and the renal concentrating and acidification capacities were normal. A kidney biopsy in one of the patients revealed morphologic evidence of glomerular damage but a normal tubular structure. A mild hyper-beta-lipoproteinemia was found in the patients but not in their healthy siblings. The cause of this syndrome, hitherto not described, is unknown.

Phospholipids and creatinine in amniotic fluid in relation to gestational age. II. Complicated pregnancy.

176 samples of amniotic fluid were obtained by abdominal amniocentesis from 69 women with complicated pregnancies (toxemia, diabetes mellitus or rhesus isoimmunization) in the 27th-43rd week of pregnancy. The concentration of creatinine (172 cases), the ratio of lecithin/sphingomyelin (L/S-ratio, 155 cases) and phospholipid phosphorus (155 cases) were determined and related to gestational age. The results were compared with normal pregnancy. The mean level of creatinine in toxemia was significantly higher from the 32nd week till term (p less than 0.05) and the L/S-ratio was significantly higher (p less than 0.05) in the 34th-35th week compared with normal pregnancy. Creatinine concentration was significantly elevated in diabetic pregnancy in the 36th-37th week (p less than 0.05). Creatinine concentration tended to be low in Rh-isoimmunization. A creatinine concentration greater than or equal to 1.8 mg% and a L/S-ration greater than or equal to 2.25 always corresponded to a gestational age greater than or equal to 35th week. To get increased precision in estimating gestational age in complicated as well as in normal pregnancy the determination of both creatinine concentration and L/S-ratio in amniotic fluid is recommended.

The value of amniotic fluid lecithin/sphingomyelin determination in prediction of hyaline membrane disease.

246 samples of amniotic fluid were obtained from 245 women with normal or complicated pregnancy near or at parturition. The lecithin/sphingomyelin (L/S) ratio was determined and retrospectively correlated with the clinical course of the neonate. Ten neonates developed hyaline membrane disease (HMD). The highest L/S-ratio in this group was 1.82. Cesarean section was not associated with HMD if L/S-ratio indicated pulmonary maturity (L/S greater than 2). With the exception of children born by women with toxemia or chronic hypertensive disease, where no case of HMD appeared among nine infants children with birth weights less than 2000 g developed HMD in about 50% (6/11 cases). The results show that the L/S-ratio correlates well to the pulmonary maturity of the neonate. Determination of L/S-ratio is therefore of great importance in cases of high risk pregnancy or doubtful gestational age prior to induction or Cesarean section.

Effects of insulin secretagogues on phospholipid metabolism in pancreatic beta-cells.

The effect of insulin secretagogues on the incorporation of [32-P] orthophosphate into phospholipids was studied in microdissected islets from obese-hyperglycemic mice. Increased 32-P-labelling was observed after incubation for 60 min with 10 mM L-leucine, 10 mM L-arginine or 20 mM D-glucose. Most of the label occurred in the phosphatidyl inositol fraction. The effect of L-leucine was additive to that induced by D-glucose while the effect of L-arginine was not. Glibenclamide (0.05 mM) was ineffective whether or not D-glucose was present. The results suggest that there is no direct correlation between the releasing actions of insulin secretagogues and changes in the metabolism of certain phospholipids and that phospholipid metabolism may be stimulated through more than one mechanism.

Rate of restoration of cardiolipin and other major phospholipids during liver regeneration in rat.

The rate of restoration of liver phospholipids, especially cardiolipin, during liver regeneration after two-thirds partial hepatectomy, was studied. Preoperative content of cardiolipin was not restored during the first week of regeneration, while this was the case for the other major liver phospholipids. The observations are discussed in relation to the morphogenesis of mitochondria during liver regeneration.

Phospholipids and creatinine in amniotic fluid in relation to gestational age.

185 samples of amniotic fluid were obtained from 174 healthy women with uncomplicated pregnancies in the 15th-43rd week of pregnancy by abdominal amniocentesis or by puncture of the amniotic sac via an amnioscope. The concentrations of creatinine (183 cases), phospholipid phosphorus and the ratio of lecithin/sphingomyelin (L/S-ratio, 130 cases) were determined. The treatment of the amniotic fluid samples (centrifugation, filtration and lipid extraction) and the methods for the determination of L/S-ratio were examined and the accuracy and limitations of the latter method were defined and discussed. The relative amounts of lecithin and sphingomyelin as well as the L/S-ratio and creatinine concentration were closely correlated to gestational age. A L/S-ratio of greater than or equal to 2.25 indicated all women to be greater than or equal to 34th week of pregnancy; 64% of them had reached the 38th week. A creatinine concentration of greater than or equal to 1.8 mg% corresponded to X greater than or equal to 35th week or pregnancy, while 75% of the women had reached at least the 38th week. With a L/S-ratio greater than or equal to 2.25 and a creatinine concentration greater or equal to 1.8 mg% of all women were greater than or equal the 36th week. 71% of the women greater or equal the 38th week fulfilled these critiera. A combination of the two parameters seems to be of a great value in discrimination between gestational ages less than the 36th week and greater than or equal to the 38th week.
