Calcium dobesilate (Doxium) as a prostaglandin synthetase inhibitor in pregnant human myometrium in vitro.

This comparative study on the effect of calcium dobesilate and indomethacin on prostaglandin biosynthesis was performed on microsomal fractions of pregnant human myometrium. Both drugs inhibited prostaglandin synthesis, indomethacin being more potent. Calcium dobesilate inhibited, in a dose-dependent manner, the synthesis of 6-oxo-PGF1 alpha, PGF2 alpha, PGE2 and TXB2. Its inhibitory action is comparable to that of etamsylate.

[Hyperprolactinemia in children with celiac disease].

The serum prolactin level was determined serially in patients with coeliac disease during gluten uptake and on gluten free diet, and in one to 14 years old healthy children serving as controls. The prolactin levels in healthy controls and in coeliac patients on diet were within normal limits whereas in children with coeliac disease taking gluten in their meals significant hyperprolactinaema was found.

Beta-adrenergic receptors in early human placenta characterization of [3H]-dihydroalprenolol binding.

The binding characteristics of beta-adrenergic ligand [3H]-dihydroalprenolol (DHA) were determined in particulate membranes of early human placenta (8-12 weeks of gestation). [3H]-DHA binding to crude membrane fractions was rapid, reversible, saturable and linearly correlated with the membrane protein concentration. Scatchard analysis of saturation experiments showed a KD of 2.80 +/- 0.9 nM and a density of binding sites of 330.30 +/- 93.5 fmol/mg protein. Agonist potency isoproterenol greater than epinephrine greater than norepinephrine indicated that early human placenta contains an adrenergic receptor of beta-2 subtype.

Characterization of luteal function by measuring the cumulative concentration of serum progesterone and urine pregnanediol-3 alpha-monoglucuronide.

The corpus luteum function of healthy, fertile women was established by measuring the serum progesterone and the early-morning urine pregnanediol-3 alpha-monoglucuronide levels during the ovulation cycles. Integration of the results led to a parameter, the luteal index (Ll), which represents the overall hormone production that characterizes the luteal function quantitatively much better than the criteria used so far. The average of the integrated hormone concentrations (Ll) for progesterone and pregnanediol were 451.4 nmol/l X time and 151.3 mumol/l X time respectively. Statistical analyses show that values of luteal index below 348.9 nmol/l X time progesterone and 134.5 mumol/l X time pregnanediol can be regarded, with very high probability, as symptomatic of luteal dysfunction. The course of the production of both progesterone and pregnanediol is characterized. The importance of these parameters in functional infertility is discussed.

Prolactin synthetizing capacity of the human decidua during the first trimester of pregnancy.

In vitro prolactin (PRL) synthesis by human decidua of early pregnancy (5-12 weeks gestation) was investigated. Tissues obtained during therapeutic abortions were incubated in oxygenated Gey's buffer supplemented with 20% fetal calf serum at 37 degrees C for 24 h. PRL production increased rapidly to 42.3 mU PRL/100 mg tissue wet weight until 8 weeks gestation and then slightly declined but these differences were not significant. There was not PRL production when chorion was incubated under similar conditions. A significant correlation was found when the initial PRL content in the decidua prior to incubation and the capacity of decidua to synthesize PRL were compared (r = 0.76, P less than 0.001). These data provide further evidence that human decidua can produce PRL in vitro. The finding that the rate of the PRL synthesis increases progressively in early pregnancy is in correlation with the intensive decidualization of the endometrium.

Etamsylate as inhibitor of prostaglandin biosynthesis in pregnant human myometrium in vitro.

The effects of etamsylate on prostaglandin (PG) biosynthesis in microsomes of pregnant human myometrium in vitro have been determined, and compared with those of indomethacin. Both drugs inhibited PG biosynthesis, indomethacin being the more potent inhibitor of the two. Etamsylate inhibited synthesis of 6-oxo-PGF1 alpha, PGF2 alpha, PGE2, and thromboxane B2; increasing the concentration of etamsylate increased the inhibition of synthesis. It is suggested that etamsylate has no anti-cyclo-oxygenase activity, but acts by inhibiting the activity of prostacyclin synthetase, endoperoxide reductase, endoperoxide isomerase, and thromboxane synthetase.

Peripheral concentrations of gonadotrophins and sexual steroids in infertile males.

Relationships between the spermiogram and sex hormones in 104 andrologic patients were studied. Endocrine connections in normal and in pathologic spermiogenesis have been studied. A rather close correlation was found between FSH and ICSH levels. ICSH increases parallel with the increase of FSH level. There is also a close relationship between sperm concentration and FSH level. Between ICSH and sperm count the correlation is not significant. Between steroid hormone level and sperm concentration no correlation ws found. It is only the plasma FSH level which show a close relationship with the disorder of spermiogenesis and it has diagnostic significance.

Prostaglandin synthesis and metabolism in the human uterus and midtrimester fetal tissues.

Prostaglandin (PG) synthesis and metabolism was studied in human fetal kidney, lung, small intestine, heart, brain and liver (gestational ages: 10, 12, 14, 18 and 23 weeks) and pregnant uterus (4-40 weeks of pregnancy). PG synthesis was increased in the myometrium during pregnancy while the capacity of metabolism did not change. PG synthesis increased in lung and kidney (4-fold), brain (20-fold) and small intestine (2-fold) but not in heart or liver. Metabolic activity increased only in fetal kidney and lung.

Bromocryptine therapy in luteal insufficiency.

Fifteen cases of female infertility, probably connected with luteal insufficiency, are reported. This abnormality was found to be fairly common in hyperprolactinaemic and clomiphene-treated normoprolactinaemic women with galactorrhoea. Bromocryptine reduces the prolactin-level, prolongs the luteal phase and increases the secretion of progesterone. According to the observations thus far reported, some optimum prolactin concentration seems to be desirable for an adequate luteal function. In the case of luteal insufficiency, estimation of the prolactin and progesterone level in the luteal phase is advocated. The basal temperature curve the hyperthermic phase to be followed up before and after treatment allows.

Effect of steroid prophylaxis and of elective caesarean section on maternal and fetal cortisol values.

The cortisol level was studied during and after delivery in the mothers and their newborns after normal deliveries, elective Caesarean sections, and steroid pre-treated vaginal deliveries. The suppressive effect of Caesarean section, and steroid pre-treated vaginal deliveries. The suppressive effect of exogenous steroid was observed in both the mother and the newborn but its extent was not significant and its duration transitory; in 48 hours it disappeared. In the newborns delivered by elective Caesarean section, an extreme increase in the cortisol level was observed. The cortisol level in the amniotic fluid was not suitable for evaluating the function of the hypophysis-adrenal axis, especially not after steroid prophylaxis.
