Inhibitors of prostaglandin synthesis, tracheal fluid, and surfactant in fetal lambs.

To study their effects on tracheal fluid (TF) production and surfactant flux, we gave 12-h infusions of prostaglandin synthetase inhibitors (PGSI) on 16 occasions to 10 fetal lambs at gestational ages (GA) of 125--141 days. Results were similar with both sodium meclofenamate (13.9 +/- 3.4 mg.kg-1, 12 studies) and indomethacin (33.6 +/- 8.0 mg.kg-1, 4 studies). All studies were done at least 6 days after surgery and 4 days before spontaneous birth. During infusions of PGSI, there were no changes in fetal arterial blood pressure, pH, PaO2, PaCO2, TF production or its concentration of sodium, potassium, and chloride; calcium concentration in TF increased slightly. We expressed tracheal surfactant flux as micrograms.kg-1.h-1 of saturated phosphatidylcholine (SPC). If control SPC flux was less than 5 micrograms.kg-1.h-1 (10 studies at GA of 125--141 days), it did not change during infusion of PGSI; however, if control was greater than 5 micrograms.kg-1.h-1 (6 studies at GA 132--140 days), SPC flux decreased during the infusions in all studies. The results suggest that prostaglandins do not strongly influence TF production up to 4 days before birth and that prostaglandins are involved in the increased flux of surfactant which occurs in late gestation.

Excessive ingestion of iodide by the rat during pregnancy and lactation. Effects on ribonucleic acid transcription in the pups brain.

The effects of an excessive ingestion of iodide by the rat during pregnancy and lactation on the transcription of RNA were studied in the brain of the pups at 10, 15 and 20 days of age. Cerebral weight, DNA content and synthesis of heterogeneous RNA were not affected during the period studied. The activity of endogenous RNA polymerase I, alpha-amanitin resistant, measured in isolated cell nuclei was significantly reduced in twenty day old rats. Similar conditions meaningfully diminished body weight at the same age.

Oral prostaglandin E2 for induction of labour at term.

A low dose regimen of prostaglandin E2 tablets, 0.5 mg hourly for up to 10 doses, was used in an attempt to induce labour without prior amniotomy in 69 patients between 35 and 44 weeks gestation. The induction was considered successful if the patient delivered within 36 hours after therapy began. The success rate was 62 percent. The incidence of undesirable side effects was low and no serious fetal complications were related to treatment.

Development of hepatic acetyl coenzyme A carboxylase in hormone-treated chicks.

It has been suggested that the carbohydrate-rich diet of chicks after hatching is responsible for the emergence of hepatic enzymes involved in lipogenesis; the injection of glucose to newly hatched chicks gives rise to an appreciable elvation on the activities of acetyl coenzyme A carboxylase and fatty acid synthetase. The present study shows that during the first hours after hatching, there is a natural elevation of glycemia which parallels the increase in acetyl coenzyme A carboxylase activity. However, the administration of hormones which alter the blood glucose levels considerably (insulin, tolbutamide, glucagon and hydrocortisone) did not influence the enzyme activity. The administration of thyroxine, estradiol and cyclic AMP, was also without effect. These results do not support the theory that the increased amount of blood glucose is the natural effector of the induction acetyl coenzyme A carboxylase. They also show that different lipogenic enzymes are not regulated via the same 'operon' since thyroxine or glucagon which alter the level of some enzymes on this pathway did not modify that of the acetyl coenzyme A carboxylase.

[Griseocarnin, an antifungal antibiotic isolated from Streptoverticillium criseocarneum (IA-7527)].

We have been studying the antagonistic activity of the Streptoverticillium griseocarneum, strain IA-7527. It produces a strong antifungal substance which we named griseocarnin. Its antimicrobial power to Neurospora crassa, Rhodotorula, rubra, Hansenula suaveolens and Candida albicans deserves to be exceled. The isolated substance was compared with other antifungal antibiotics presenting close similarity with the heptaens Ascosin and PA-150. The production, extraction and purification methods as well as physical, chemical and biological properties are included in this work.

Role of leukotriene C4 and edema in the acute allergic bronchoconstriction in the guinea pig.

In vitro studies have suggested that leukotrienes are involved in acute allergic bronchoconstriction, though this has not been definitively corroborated yet in in vivo studies. On the other hand, edema production during antigenic challenge could be an additional factor favouring such bronchoconstriction. In the present work we quantified immunoreactive leukotriene C4 (iLTC4) concentrations in bronchoalveolar lavages during allergic bronchoconstriction induced by 1 mg/kg i.v. ovalbumin (OA) in immunized guinea pigs, as well as water content in guinea pig lung fragments obtained before and during this bronchoconstriction. We found that basal concentrations of iLTC4 (median 1.06 ng/ml) were not significantly modified at 2, 5 and 10 min (median 1.10, 0.29 and 1.37 ng/ml, respectively) of the bronchoconstrictor response. Water content in lung fragments did not change among non-immunized guinea pigs, immunized ones and at 15 min of bronchoconstriction (mean +/- SEM 79.32% +/- 0.18, 79.10% +/- 0.31 and 79.13% +/- 0.40%, respectively). In addition, isoproterenol (20 micrograms/kg, i.v.) rapidly reverted about 70% of the bronchoconstriction induced by a higher antigenic dose (OA, 3.1 mg/kg i.v.); residual obstruction was not associated with increased water content in lung fragments (78.13% +/- 0.43). These results suggest that in this model, acute allergic bronchoconstriction is not due to an increased iLTC4 release or to edema production, and that airway smooth muscle contraction is the main component of this response.
