A procedure for the assay of DNA damage in mammalian cells by alkaline elution and microfluorometric DNA determination.

The microfluorimetric assay of Kissane and Robins (J. Biol. Chem. 233, 184-188, 1958) has been modified to monitor DNA content in alkaline elution fractions. The elution profiles of DNA from treated and control cultured cells demonstrated a good correlation whether DNA in the elution fractions was determined by the fluorometric method or by measuring its radioactivity. The DNA recovery was of about 80% and the measured material was DNase digestible.

Reduction of DNA repair efficiency during the initial phase of rat liver carcinogenesis.

Chemically induced DNA fragmentation and unscheduled DNA synthesis (UDS) were determined in hepatocyte primary cultures (HPC) obtained from the entire liver during the development of hyperplastic lesions induced in rats by the following treatment: 200 mg/kg i.p. of N-nitrosodiethylamine (DEN) on day 0; 2 ml/kg i.g. of CCl4 on day 21; dietary administration of 0.02% 2-acetylaminofluorene (2-AAF) during the 3rd and the 4th week, and of 0.05% phenobarbital (PB) from the 6th week. At 4, 5, 6, and 7 weeks after DEN injection, the level of DNA fragmentation elicited by either the activation-independent carcinogen methyl methanesulfonate (MMS) or the activation-dependent carcinogen N-nitrosodimethylamine (DMN) was significantly lower than that in HPC from age-matched normal rats. In the same HPC, a statistically significant decrease was observed in the capability of repairing DNA damage induced by MMS, while the reduction of the DNA repair efficiency did not reach the level of significance after exposure to DMN.

[Dermatopathies in the domestic worker: the Lombard experience].

In order to identify the etiopathogenesis of cutaneous diseases in housewives, a questionnaire was made, and the gathered data are here described. 1,824 people have answered this questionnaire: 1,007 have reported lesions with a suspicious immunological pathogenesis. The commercial products which were reported as a cause of the dermatitis have been 834. In a group of 142 housewives (86 with allergic contact dermatitis and 56 with irritative contact dermatitis), the allergic sensitizations to the chemicals in the detergents were tested. 21% of the cases had positive cutaneous reactions for nickel salts and 8.4% for chromium salts. The analysis in atomic absorption method of these metals in 34 commercial products has shown their presence in almost all the samples. Hypothesis are formulated to find the source of these polluting substances. The aims and the methods of a investigation carried out on 1.800 housewives, who are going to be controlled in order to estimate the prevalence, the clinical kinds and the etiopathogenesis of dermatitis related to house work, are described. The study will be carried out at the Occupational Health Institute "Clinica del Lavoro L. Devoto" of Milan and at the Dermatological Departments of the Universities of Bari and Florence.

[Informatics in cardiology: proposal for standardized terminology in clinical records and in permanent cardiac electrostimulation].

To make operative two medical computerized programs, one dedicated to the cardiological clinical record (PAC) and the other to permanent cardiac stimulation (PGP), we codified about 4000 terms concerning the general medical field and particularly the cardiological area. The importance of standardization of the medico-cardiological language is emphasized, specially in relation to the systematization of the information, necessary to work with computerized systems.

[Aneurysms of the hepatic artery. Clinico-radiologic study].

A series of 7 cases of hepatic artery aneurysm is reported, pointing out the different features for etiology, localization, shape and size. The diagnostic imaging methods are then analyzed. Conventional radiology is actually useless. Sonography is hardly ever conclusive, even if the association of real time-US with echo-Doppler is likely to be promising. CT often concludes the diagnostic procedure, though the small size of aneurysms may limit its efficacy. Arteriography is the only method fit to occasionally find out small aneurysms and clear eventual diagnostic doubts; it's also mandatory before surgical treatment and allows percutaneous embolization. MR is likely to offer interesting perspectives.

[Changes in CSF dynamics and performance tests after head injuries (author's transl)].

In a large series of head-injured patients (132 cases), we have selected 30 patients (mean age 43 years) with only primary school education: they have been investigated with isotope cisternography and psycodiagnostic tests. 19 underwent neurosurgical procedures (6 extradural haematomas, 8 subdural haematomas, 5 cerebral lacerations). These investigations have been carried at least six months after the head injury. We have used for isotopic cisternography RISA and In111-DTPA. Raven's Progressive Coloured Matrices and Immediate Memory Tests were performed. Cisternographic pictures were classified as 1) normal, 2) with asymmetrical diffusion, 3) with ventricular reflux and were correlated with the neuropsychological data. We have found ventricular reflux in 10 patients (5 underwent surgical procedures), asimmetrical diffusion in 15 patients (11 operated); in 5 cases cisternographic pictures were normal. Neuropsychological tests give quantitative data about progressive mental deterioration, in full agreement with cisternographic results.

[Vascularized renal neoplasms: does the angiographic study enable the differential diagnosis? Cluster analysis of 65 cases].

65 selective renal arteriographies showing vascularized renal tumors are reviewed and a series of 18 parameters is collected. By means of an unusual statistical analysis, the significance of the presence of such parameters in the various histological kinds of tumors is verified in order to find out whether patterns of arteriographic signs permitting a sure differential diagnosis exist. Finally, the present role of arteriography in the diagnostic iter of renal masses is discussed.
