Effects of pildralazine alone and in combination on severe hypertension and cerebrovascular lesions in saline-drinking spontaneously hypertensive rats.
The efficacy of long-term treatments with the vasodilator antihypertensive pildralazine alone and in combination with propranolol or dihydrochlorothiazide in preventing blood pressure increase and cerebrovascular lesions in saline-drinking spontaneously hypertensive rats (SHR) was evaluated. Groups of 20-40 animals were treated daily for 13 weeks. At the end of the experiments the brains were examined histologically and vascular lesions, ranging from arterial wall damage to cerebral infarction, were checked. In the control groups (n = 60), systolic blood pressure gradually rose to over 200 mmHg, and about 74% of the animals had lesions. Pildralazine at a dose of 1 mg/kg p.o. significantly inhibited the onset of severe hypertension; the combination of pildralazine with moderately effective doses of propranolol (10 mg/kg p.o.) or dihydrochlorothiazide (5 mg/kg p.o.) completely prevented the blood pressure increase. When blood pressure control was achieved, the incidence of cerebrovascular lesions was also significantly reduced.
[Radiographic and xeroradiographic case finding in asbestosis].
Statistical researches were carried out on subject samples exposed to asbestosis risk with the aim of investigating the advantages of xeroradiographic methods when applied to the early diagnosis of this occupational disease. In the initial phase of the research, the aim was to quantify the statistical incidence of minimum asbestosis. Thus the frequency of initial forms was compared with developed forms in 750 subjects working with amianthus cement. These subjects had been homogeneously exposed to asbestosis risk (as research on the environment in the factory confirmed), and over a period of 10 years had been checked radiographically at the Clinica del Lavoro of the University of Pavia. This first phase of the research confirmed the remarkable incidence of the disease. In the subsequent phase, the radiographs of 200 subjects with asbestosis were studied. The aim was to quantify the frequency of the different radiological symptoms of the disease. In the final phase of the research, both traditional radiographs and xeroradiographs were obtained on 51 subjects exposed to asbestosis risk with pulmonary alterations, although with no (or not significant) radiographic techniques in the early diagnosis of asbestosis would thus become clear. For protective reasons, the use of xeroradiography is proposed not for mass screening but only for controlled investigations. Xeroradiography can evidence plaques or pleural fibrous alterations, which would not be shown on traditional radiographs. It also allows more satisfactory examination of the small fibrous parenchymal opacities at the base of the lungs. On the other hand, additional controlled examinations of the same subjects, even when use was made of projections for pleural backgrounds and lordotic projections for the parietal pleurae, reduced earlier favorable statistics by half. However, the wide range of xeroradiography can evidence nearly all radiographic symptoms in a single xeroradiograph. This greatly facilitates routine examination while providing more satisfactory iconographic data for subsequent medico-legal discussion.
Prophylactic antimetastatic treatment with aryldimethyltriazenes as adjuvants to surgical tumor removal in mice bearing Lewis lung carcinoma.
The effects of two benzenoid dimethyltriazenes (1-p-(3,3-dimethyl-1-triazeno)benzoic acid potassium salt [DM-COOK] and 1-p-tolyl-3,3-dimethyltriazene), which have been previously shown to reduce the formation of spontaneous lung metastases in mice bearing subcutaneous Lewis lung carcinoma, have been investigated in mice implanted im with the same tumor in the calf of the hind leg. Primary tumor was removed surgically by amputation in mice treated with the tested compounds preoperatively, and the survival time of the animals was then determined. A high rate of cures, ranging from 23% to 43%, has been observed when the treatment consisted of eight or three daily doses prior to tumor removal; a single immediately preoperative dose was less effective and caused a 10% cure rate. Survival time of uncured mice was also significantly increased, particularly by DM-COOK. The division of the dose into two daily injections did not modify the activity of the triazenes, thus indicating that the antimetastatic coverage between two subsequent doses is not limited by fast pharmacokinetics or decomposition of the drugs. These results show that, at least in one animal system, aryldimethyltriazenes can be used also on palpable tumors as prophylactic adjuvants to surgery in order to reduce preoperative and intraoperative tumor spread.

[Role of platelet serotonin in hyperperistalsis after digestive hemorrhages].
Intestinal transit accelerates during gastroduodenal haemorrhage. To test a personal theory that the reason is to be found in serotonin released by platelets and absorbed by the intestine, the behaviour of serotoninaemia has been studied. This proved to be 1.35-9.7 microgram/l (mean: 4.175 microgram/l; E.S. +/- 1.39) in patients with haematemesis and melena, compared to 0.49-0.9 microgram/l (mean: 0.728 microgram/l; E.S. +/- 0.07) in the controls (p less than 0.001). The increase in circulating serotonin to significant values is probably due to hormone release. This dose not occur in cases of melena alone, probably owing to the organism's high capacity for catabolism.

[Primary lung neoplasm of rare incidence: giant cell carcinoma].
Primary giant-cell carcinoma of the lung is an infrequent observation. It is distinguished clinically solely its fairly rapid development but otherwise it does not seem to have any predilection for localization, age or sex. Histologically, differential diagnosis with othe tumours such as angiosarcoma, rhabdomyosarcoma, chorioepithelioma and others is of interest. Three cases of primary giant-cell carcinoma of the lung are reported and the various clinical and anatomopathological aspects discussed.
