[Diagnosis of chylous ascites with oral administration of 13C-palmitic acid].

12 cases of chylous ascites in PUMC hospital in the recent 30 years were analysed. The etiology of this disease includes non-traumatic (83.3%) and traumatic (16.7%) causes. All the patients received isotope examination or lymphangiography, but only in 8 patients the site of the lesion was found. As these two kinds of examination can only show the lesions in the right and left lumbar lymph trunks, cisterna chyli and throacic duct but not the intestinal lymph truck, the authors set up a method by administering orally 13C-palmitic acid to detect the lesion of intestinal lymph trunk. Palmitic acid is a long-chain fatty acid, which enters directly into the intestinal lymph trunk after absorption. Palmitic acid labelled with 13C could be detected in the ascitic fluid if there is a leak from intestinal lymph trunk. This new method was used to examine a patients who had negative results with isotope examination and lymphangiography; 13C could be detected in the ascitic fluid 30 minutes after oral administration but not in exhaled air. It can be concluded that cyhle leaked into the peritoneal cavity from the intestinal lymph trunk. This method is also of help to determine the possible site of leakage and the degree of obstruction, so it is quite useful for the diagnosis of chylous ascities.

A hybrid-hybrid matrix method for 3D NOE-NOE data analysis.

A hybrid-hybrid matrix method is described that quantitatively analyzes 3D NOE-NOE NMR data. Experimental 3D data are merged with simulated 3D data to create a hybrid 3D NOE-NOE spectrum. This is then deconvoluted into a 2D hybrid NOESY spectrum. The deconvoluted, 2D hybrid NOESY spectrum can then be merged with other 2D NOESY experimental data along with additional simulated 2D data as necessary to create a hybrid-hybrid 2D NOE volume matrix. This hybrid-hybrid volume matrix is then used with the complete relaxation program, MORASS, to calculate a rate matrix, and the resulting distances taken from the off-diagonal cross-relaxation rates can then be utilized in a distance geometry or restrained molecular-dynamics refinement of the structure. This process is repeated until a satisfactory agreement between the calculated and observed 3D volumes is obtained. This hybrid-hybrid matrix method retains computational efficiency and utilizes the resolution of the 3D data set while retaining any information content of the available 2D data. The initial tests of the deconvolution algorithm give high correlation results even with the introduction of random error into the 3D data set. Our results suggest that the hybrid-hybrid matrix method for analysis of 3D NOE-NOE spectra may provide a viable tool in the refinement of large molecules.

[Efficacy of praziquantel combined with albendazole in the treatment of clonorchiasis].

This paper reported on the clinical efficacy of praziquantel-albendazole combination in the treatment of clonorchiasis in Guangxi. Two hundred and two cases were divided into three groups. The first group comprised 66 cases who were treated with praziquantel at 180 mg/kg bwt for three days. The second group consisted of 62 cases who were treated with albendazole at 90 mg/kg bwt for three days. In the third group, 74 cases were treated with 90 mg/kg bwt praziquantel plus 45 mg/kg albendazole for three days. The results showed that the negative conversion rates were 98.5%, 61.3% and 87.8% in groups one, two and three, respectively, one month after treatment. In some cases who continued to be egg positive, the reduction rates in faecal egg counts were 99.7%, 65.0% and 97.3%, respectively. Although the cure rate in group three was not as high as in group one, the side effects were more mild and transient and the cost of treatment could be reduced by 60% as compared with group one. In cases with light and moderate infections, the praziquantel-albendazole combination proved more effective. In addition, the praziquantel-albendazole combination was also highly effective for treating those cases who were infected with Ascaris, Trichuris or hookworm.

Experimental study on electrochemical treatment of cancer in mice.

Electrochemical treatment of cancers in mice resulted in at least partial regression or disappearance of transplanted SRS solid tumours and increased the survival time. Six days after treatment, lymphocyte infiltration in the tumour tissue was found. Seventy-one days after treatment, tumour cells were rarely found in the tissue. The application of direct current was found to facilitate the destruction of tumour cells in a shear stress field.

Analysis of a herpes simplex virus 2 fragment from the open reading frame of the large subunit of ribonucleotide reductase with transcriptional regulatory activity.

Expression of herpes simplex virus 2 (HSV-2)-encoded ribonucleotide reductase (RR) is required for growth in nondividing cells. The functional enzyme is composed of a large and a small subunit. In virus-infected cells, RR is expressed temporally as a delayed early protein. However, the promoter regulatory region of the large subunit can function as an immediate early promoter in transient transfection assays, suggesting that expression may be quite complex. In this study, a 95-bp fragment derived from the open reading frame of the large subunit of RR (RR-A) functioned as a silencer when placed adjacent to a heterologous promoter. If the fragment was placed distal to the promoter, repression was relieved and in human keratinocytes promoter activity was consistently higher than control constructs. Exonuclease III protection assays revealed that nuclear factors from human keratinocytes as well as other primate cells specifically bind to this fragment. A 30-bp motif containing a consensus SP-1 binding site and an alternating Pu/Py element was protected in all cell lines. These results suggest that a 95-bp fragment in the open reading frame of HSV-2 RR-A plays a role in regulating viral gene expression.

[Influences of acupuncture on gastroduodenal mucosal lesion and electrical changing induced by stress in rats].

Experiments were performed in 57 SD rats. The stress was induced by binding plus revolving (5.5 xg, 170 c/min, 10 min) or cold (0-4 degrees C, 30-60 min). Selected acupuncture points were "Zusanli" and "Weishu". Bipolar Ag-AgCl recording electrodes were respectively implanted on the serous membrane surface of the antrum, duodenum and on the abdominal skin corresponding to above areas. The results were as follows: (1) There are 63.2% individuals in the stress group which showed a large quantity of bleeding points spreading over the gastroduodenal mucosal membrane. The area density of lesion was 15.8-27.7%. Under microscope the signs emerged including mucosal blood vessel dilatation and hyperaemia, gland cell swelling, virlus broken and peeling off and lymph cell accumulating, etc. 84% individuals showed obvious inhibition of gastroduodenal electroactivities including that the amplitude of slow wave was lower, the frequency decreased even the rhythm was disordered, the number of fast wave reduced and the IDMEC III cycle was prolonged. (2) In the group of acupuncture plus stress only 16.7% individuals showed bleeding or hyperaemia, and the lesion area density was only 1.7%. Also the pathohistological changes were obviously decreased. Gastroduodenal electrical changes were hardly seen (27.2%) or the inhibitory state above were significantly reduced (54.5%) when compared with the stress group. There is strong reason to consider that acupuncture could effectively reduce the mucosal lesion and the electrical inhibition of stomach and duodenum induced by stress in rats.
