Effects of amiloride on oral glucose loading, serum potassium, renin, and aldosterone in diet-controlled diabetes.

The effects of amiloride on oral glucose loading, serum potassium, renin, and aldosterone were evaluated in 10 patients with diet-controlled diabetes. Eight had mild hypertension, and 2 had normal blood pressure. Prior to receiving amiloride all were studied for renin and aldosterone responses while supine and after 2 hr ambulation. All had a normal response to change in position in the renin and aldosterone systems. Before administration of amiloride glucose tolerance tests were carried out, with simultaneous determinations for potassium and insulin. Amiloride 5 to 10 mg was given orally for 6 wk. Blood glucose and serum potassium levels were monitored weekly. After 6 wk renin and aldosterone responses were again determined, as were oral glucose tolerance and serum potassium and serum insulin levels. Amiloride did not induce hyperkalemia in these diabetic patients and did not alter the postamiloride relationship. It is concluded that amiloride is safe for patients with an intact renin aldosterone system, more especially those with normal renal function and diet-controlled diabetes mellitus.

Flexor tendon healing and adhesion formation after Sterispon wrapping: a study in the rabbit.

The influence of Sterispon (Allen & Hanbury) wrapping on the healing of the rabbit flexor digitorum profundus tendon sutured within the digital sheath was studied. Control tendons healed in 2 weeks in association with thick adhesions. The wrapped tendons were surrounded with fewer adhesions, but 50% separated. Healing was delayed until adhesions reached the suture site. A sheath of a single layer of mesothelial cells formed on the surface of the Sterispon opposite the tendon and this markedly reduced adhesions at the suture site. The cellular reaction was bland. Sterispon wrapping had a similar effect on tendon healing and adhesion formation as wrapping with other non-absorbable substances and the results support the theory that healing in the digital sheath is necessarily mediated through the formation of adhesions. Sterispon wrapping has been helpful after tenolysis operations.

The observer interaction variable in evaluation of socialising properties of pimozide (Orap).

Eighteen chronic, institutionalised patients were randomly assigned to two groups for the purpose of a double blind trial of pimozide (Orap). The results of the study indicated an increase in socialisation, as measured by the Venables scale, for both groups, and no significant difference in this variable between placebo and active preparation groups. It is suggested that greater observer interaction may be responsible for the observed increase in socialisation previously attributed to pimozide.

Periodic acid-Schiff (PAS) staining for glycogen in clinically normal psoriatic and non-psoriatic skin.

PAS staining was used to assess the glycogen content of clinically normal forearm epidermis in 32 patients with active psoriasis, 30 patients with inactive psoriasis and 40 non-psoriatic subjects. Glycogen accumulation was greater in patients with active than in those with inactive psoriasis. The inactive psoriatics showed significantly less glycogen accumulation than the controls.
