[Liver involvement in lymphogranulomatosis].

Of 146 patients with lymphogranulomatosis biochemical parameters were tested for their diagnostic valency concerning the recognition of a liver infiltration. In patients with histologically proved affection of the liver the AP, GGTP, AAP, LAP and LDH show a significant increase in comparison to the enzyme values of the patients without any hepatic manifestation. In an increased result of 4 enzyme values with a probability of 85% muste be reckoned with a participation of the liver. The enzyme SGOT, SGPT, GDH, LDH-isoenzymes, choline esterase, beta-GC, the De Ritis quotient and the quotient (Formula: see text), on the other hand, do not give any additional differential-diagnostic information.

[Zinc activated tartrate resistent phosphatases in the brains of different animal species and their characterization].

A zinc activated tartrate resistent phosphatase (ZnTP) of the brain of different animal species was separated by electrofocusing in polyacrylamide gels. It is demonstrable selectively in the presence of 20 mM zinc acetate and 10 mM D,L-sodium tartrate or of 100 mM zinc acetate only. The ZnTP hydrolyzes 1-naphthyl phosphate and 4-nitrophenyl phosphate, respectively. High activity of ZnTP is evident in the brains of rats and rabbits. The activity is moderate or absent in the brains of mice, syrian-hamsters, sheeps, cats, rhesus monkeys, and of human beings. The isoelectric points of the enzyme from the various species are different, but the molecular weight is identical (65 000 estimated by gelfiltration on Sephadex G 100 in the brain of rat, rabbit, syrian-hamster, and man). A method of quantitative evaluation of ZnTP activity is described.

[The significance of the enzyme histochemistry in the diagnosis of malignant non-Hodgkin's lymphomas (author's transl)].

The significance of histo- and cytochemical enzyme investigations in the diagnosis of malignant Non-Hodgkin's lymphomas (NHL) is evaluated. Histochemical enzyme methods complete the morphological diagnosis if some general principles of diagnostic histochemistry are observed. This is particularly true for the diagnosis of the hairy cell leukemia, the T-lymphoblastoma and the histiocytic reticulosarcoma.

[Histochemical demonstration of the zinc activated tartrate resistant phosphatase (ZnTP) in the glial reaction of rat brains following stabbing injuries (author's transl)].

ZnTP shows a characteristic pattern of activity in experimentally induced cerebral tumours of rats. Our previous investigations demonstrated a high activity of this enzyme in astrocytomas and in blood vessels within the tumours. The question arose, whether the increased activity is specific for tumours of astrocytic origin or occurs in reactive astrocytic proliferations as well. The present investigation is aimed at the elucidation of this problem.

[Early stages of experimental brain tumours in rats. Investigation of serial sections (author's transl)].

The results of transplacental induction of brain tumours by a single intravenous injection of ethylnitrosourea in a dosage of 20 or 30 mg/kg body weight to pregnant BD IX and Albino rats on the 17th day of gestation are presented. From 46 animals of the offspring 43 rats developed 185 intracranial neoplams. 157 of them were early stages. Most of the animals had multiple tumours. One rat showed 13 separate tumour nodules. The average number of brain tumours was 4.3 per rat. 101 tumours had a diameter of less than 1 mm. The neoplasms were classified as 179 glial tumours (mainly polymorphous gliomas, oligodendroastrocytomas and oligodendrogliomas), 4 sarcomas and 2 gliosarcomas. All the oligodendrogliomas and about two thirds of the polymorphous gliomas were early stages. The following preferential localizations in the rat brain were established: White matter of cerebrum, hippocampus, basal ganglia, cortex of cerebrum and brain stem. 25.4% of the tumours were localized in the subependymal area. Bulbus and tractus olfactorius, leptomeninx, and cerebellum were very rare sites of tumour growth.

[Diagnostic significance of morphological changes in biopsies of the pelvic crest (author's transl)].

Pelvic crests of 265 autopsies were histologically investigated. In nearly all cases with chronic diseases a remodelling of bones and especially processes of bone resorption were found. A fibroosteoclastic bone resorption was evident in 28 of 40 cases with chronic renal failures, in 5 of 26 cases with chronic lung diseases and/or chronic insufficiency of the right heart ventricle, in 4 of 12 cases with chronic diseases of the gastro-intestinal tract, and in a case of lung cancer. Processes of bone formation were less prominent. The findings indicate that several chronic diseases caused by different overlapping pathogenetic mechanisms can principally lead to the same spectrum of alterations of bone structure. This and the aetiological ambiguity of typical patho-histological reaction patterns of bones require a critical reserve in the interpretation of adequate findings in bone biopsies. It has been pointed out that the use of morphometric methods and the use of slices of undecalcified bones are necessary to obtain a reliable diagnostic tool in bone biopsies.

[Treatment of chronic infantile hepatitis with D-penicillamine (author's transl)].

Detailed discussion of action, indication and side effects of D-Penicillamine which was used for the treatment of chronic hepatitis of infancy. Of 18 patients, 7 had chronic-active hepatitis, 6 chronic persisting hepatitis, 2 subacute hepatitis and 3 fibrosis of the liver. Control of results was based on numerous clinical chemical investigations and repeated liver biopsies. The transaminases and histology of the biopsies were the essential parameters. Doses between 15 and 35 mg/kg of body weight gave very favorable results in these 18 patients, treated over 5 to 24 months. Australia antigen-negative, chronic active hepatitis appeared to be particularly suited for this type of treatment.

[The histochemical demonstration of a zinc activated, tartrate resistant acid phosphatase (author's transl)].

An acid phosphatase which is highly activated by zinc acetate is demonstrated histochemically in the medulla of adrenal gland of the rat. It is shown that the acid phosphatases demonstrable histochemically in the medulla of adrenal gland are strongly inhibited by sodium tartrate while acid phosphatase demonstrable histochemically in presence if zinc acetate is tratrate resistant.

[The presence and isolation of a highly zinc activated acid phosphatase in the rat brain (author's transl)].

The paper describes an acid phosphatase from rat brain extracts, which is highly activated by zinc acetate. The separation of the different acid phosphatases was performed by gel electrofocusing. After staining with 1-naphthyl phosphate and hexazonium pararosanilin or stabilized diazonium salts in the presence of zinc acetate and sodium tartrate is this zinc activable enzyme fraction nearly selectively demonstrable. With the method of gelfiltration on Sephadex G 100 two fractions of acid phosphatase splitting 4-nitrophenyl phosphate with a very different molecular weight were obtained. In the presence of zinc acetate a third peak of acid phosphatase appears, which is highly activated by zinc and is different from the two other phosphatases. The gel electrofocusing of the last peak showed that it is identical with the above described, though not yet pure. By the fact that it can hydrolyze both 1-naphthyl phosphate and 4-nitrophenyl phosphate at pH = 4.8 and that it is activated by zinc acetate but resistant to tartrate, we would call it zinc activable tartrat resistant acid phosphatase. It is possible to demonstrate this enzyme histochemically. The enzyme activity is especially high in the stromal cells of the plexus chorioideus. After further purification and characterization of this enzyme it could be possible to distinguish it from other zinc activable acid phosphatases in different tissues and body fluids.

[Histochemical demonstration of a zinc activated tartrate resistant acid phosphatase in experimentally induced glial microtumors of the rat brain (author's transl)].

The tartrat resistant zinc activated acid phosphatase (ZT-AP) was investigated in 17 glial microtumours of the rat brain. The tumours were induced by N-ethyl-N-nitrosourea injected to newborn rats subcutaneously (85 mg/kg body weight). The morphology of these microtumours corresponds to the statements in the literature. In regard to their behaviour of the investigated enzyme the microtumours may be divided in 2 groups: 1) Microtumours with an activity of acid phosphatases but without any histochemically demonstrable activity of ZT-AP. Histologically they correspond to oligodendric microgliomas. 2) Microtumours with a high activity of acid phosphatases and a remarkable activity of ZT-AP especially in the perivascular cell proliferations. This type corresponds histologically to the astrocytic and mixed glial proliferations including glioblastomas. According to our present findings the ZT-AP was found in astrocytes and especially in mesenchymal cells. In some cases a focal increased activity of ZT-AP could also be observed before histological alterations appeared. We regard it as an early stage of the development of tumours.
