Influence of the trapidil derivative AR 12463 on serum and tissue lipids and development of aortic lesions during experimental atherosclerosis in rabbits.

Daily administration of the trapidil derivative AR 12463 (20 mg/kg body weight i.p.) to cholesterol-fed rabbits diminished statistically significantly the increase in serum total cholesterol. After 8 weeks of treatment all measured lipoprotein fractions were significantly lower in animals treated with AR 12463 (group 3) compared with the values of the untreated control (group 1) or vehicle-treated group (group 2). The reduction of serum levels was associated with statistically significantly reduced levels of cholesterol esters in kidney, liver and aorta. The levels of free cholesterol in the liver of group 3 animals were statistically significantly lower compared with the levels in the two other groups, whereas in kidney and aortas the levels of free cholesterol remained unchanged under the influence of AR 12463. The area of aorta covered with fatty streaks was significantly smaller in group 3 versus group 1. The results of this study indicate that treatment of rabbits with AR 12463 while feeding a cholesterol-rich diet prevents hypercholesterolemia as well as the cholesterol incorporation into tissues. The mode of action of AR 12463 on serum and tissue lipids as well as on the development of atherosclerosis is discussed.

Influence of high density lipoprotein (HDL), prepared from human blood, on prostanoid formation, serum and tissue lipids and development of arteriosclerosis in cholesterol rich fed rabbits.

The i.v. administration of high density lipoprotein (HDL) into cholesterol fed rabbits decreased statistically significantly the serum level of total cholesterol and of low density lipoprotein cholesterol after a feeding period of 8 weeks. These diminished levels of cholesterol were associated with a statistically significant reduction in the levels of cholesterol esters in kidneys and platelets but not in hepatic tissue or in aorta. Macroscopically detectable arteriosclerosis was not statistically significantly diminished. The formation of prostanoids by the aorta remained unchanged. The atherogenic role of immunologic factors acting against the heterologous HDL may have compensated for the antiatherogenic HDL action on plasma and tissue lipids.

[Effect of the oral administration of zinc on metastasis after intravenous application of benzpyrene-induced rat sarcoma cells].

In experimental investigations it was shown that rats having received drinking water enriched by zinc acetate (22.8 mmol/l) developed significantly more pulmonary metastases after an intravenous infection of cells of a benzpyrene-induced rat sarcoma than rats receiving normal drinking water. This increase is caused by an increase of the number of rats developed metastases as well as the number of metastases of the individual rat.

[Subcutaneous metastasis of an atrial myxoma? Case report and literature review].

A 25 year old woman developed 2 subcutaneous myxomas one month after removal of an atrial myxoma. The pathogenesis of this event is discussed. Special attention was given to distinction between true myxomas and myxoid sarcomas as well as to myxomatous diathesis, caused by a widespread, genetically determined specific abnormality of certain mesenchymal cells (Swiss-syndrome). With all aspects considered, this appears to be the best interpretation of the case under review.

[Prostacyclin and thromboxane synthesis in liver tissue in chronic liver diseases].

This paper reports on investigations of the formation of PGI2 and TXA2 using their stabile products 6-keto-PGF1 alpha and TXB2 (RIA) in liver biopsy specimens of 46 patients suffering from fatty liver (n = 19), chronic hepatitis B (n = 11), liver cirrhosis (n = 13), and miscellaneous diseases (n = 3). The measured formation rates in chronic liver disease were evaluated in comparison to a reference group (n = 19) consisting of minimal liver lesions. The 6-keto-PGF1 alpha formation correlating to the degree of the portal inflammation in the liver (morphometric evaluation). The same trend existed in relation to the intralobular inflammation. The results presented suggest in respect of analogous data in animal experiments that PGI2 is predominantly generated in mesenchymal cells of the liver and, presumably influences the course of liver diseases.

Enzyme histochemical investigation of ITO's cells in semithin sections of human liver biopsy specimens.

Human liver specimens obtained by needle biopsy were embedded into glycolmethacrylate and enzyme histochemical investigations were carried out on semithin sections. By this method, we could demonstrate an activity of acid phosphatase (EC 3.1.3.2) by azo-coupling as well as by metal precipitation methods, of alkaline phosphatase (EC 3.1.3.1) by metal precipitation method, and of ATPases (EC 3.6.1.3 and EC 3.6.1.4) in ITO's cells. Special aspects of the application of enzyme histochemical methods to resin-embedded liver tissue are explained.

[Vitamin A and the liver--current knowledge].

A short survey of metabolism and effects of vitamin A as well as of the role of Ito cells is presented. The Ito cells of the liver represent a special form of fibroblasts, about 80% of the vitamin A (VA) content of the normal liver is stored in these cells. Following to the absorption of VA in the intestine, VA (retinol) is transported in chylomicrons and their remnants, and taken up by hepatocytes through receptor-mediated endocytosis. Along with retinolbinding protein VA is transported from the hepatocytes to the Ito cells where is is being stored as lipid droplets. The VA content of the liver tissue in chronic, especially alcoholic liver disease is distinctly lowered. The VA deficiency of the chronic alcoholic liver disease is caused by an accelerated microsomal metabolism of VA. Furthermore, investigations elucidating the therapy of VA-deficiency in liver diseases are explained.

[Fatal course of basalioma terebrans caused by brain abscess with brain compression and hemorrhagic anemia].

Report on a 79 years old female patient with a giant basalioma terebrans which has been growing for 15 years at the forehead. Its rapid exophytic and invasive growth at the final stage resulted in an extend destruction of the squamofrontal bone with infiltration into the dura mater. In consequence of this brain abscess with brain compression and a high grade hemorrhagic anemia inducing at last the fatal outcome were developed.

[Cytogenetic relationship between portal fibroblasts and Ito cells of the liver].

Portal fibroblasts of normal human and rat liver were morphologically investigated to estimate their capability of storing vitamin A. Vitamin A levels were semiquantitatively estimated by evaluation of the number and size of intracellular lipid droplets. Vitamin A storage was found to be low in more than 10% of normal portal fibroblasts in man and rat. Lipidstoring fibroblasts of portal tracts exhibited remarkable morphological similarities to "activated" Ito cells and were accordingly designated as portal transitional cells.

The ability of cultivated human skin fibroblasts to store vitamin A. A contribution to the cytogenesis of the Ito cells of the liver.

Cultivated skin fibroblasts were fed with retinol palmitate in a dosage of 100,000, 200,000, or 300,000 I.U. In dependence on the dosage a remarkable uptake and storage of vitamin A can be observed in vitro (27.2, 53.0 or 60.7% of the fibroblasts). Distinctly retinol palmitate storing cells have lost their typical fibroblast-like appearance and are transformed into cells which are comparable to the Ito cells of the liver from the cytomorphological point of view.

[Adenosine deaminase activity--an indicator of inflammation in liver disease].

Inflammatory liver diseases display higher levels in serum ADA activity compared to non-inflammatory ones. The most pronounced increases in activity are found in acute virus-induced hepatitis, in active liver cirrhoses, extremely high levels in some liver tumours. Due to correlative relations, the ADA is mainly attributed to the mesenchymal parameters by factor analysis. This can be validated by assessment of histological criteria. The highest correlation coefficient could be demonstrated for ADA in relation to all parameters under investigation by means of the semi-quantitatively evaluated mesenchymal reaction within the histological section. The determination of the serum ADA is valuable in assessing the inflammatory reaction of the liver and for diagnosing active cirrhosis in particular. Increases in activity based on an elevated nucleic acid metabolism in tumours and regenerative processes have to be taken into consideration. This interpretation is evidenced by correlative relations between the activity of ADA in liver biopsy homogenate specimen and the hepatic inflammatory reaction.

[Effect of orally administered zinc on the inducibility of experimental brain tumors in the rat].

The effect of orally administered zinc on induction and growth of brain tumours in rats was investigated. The brain tumours were induced experimentally by N-ethyl-N-nitrosourea injected subcutaneously in a single dose (17 mg/kg body weight) into newborn BD IX rats. The rats were killed after 180 days. Their brains were cut in serial sections and investigated histologically. The incidence of brain tumours of a control group of rats (group 1) was compared with that of rats which received drinking-water containing 22.8 mmol/l (5.0 g/l) zinc acetate from the 4th week (group 2) or from the 150th day (group 3) after birth. The rats of group 2 developed three times more brain tumours than the rats of the control group. This difference is statistically significant. The incidence of brain tumours in rats of group 3 corresponds to that of the control group. The average size of the tumours was the same in all groups. In the brains of the rats that received zinc with the drinking-water up to the 180th day but without administration of a cancerogen immediately after birth (group 4) we could not find any tumour, neither after 180 nor after 380 days. Our results indicate a cocancerogenic (promotion) effect of zinc in experimental neurooncogenesis of the rat.

The histochemical behaviour of zinc-activated tartrate-resistant phosphatase (ZnTP) in early stages of experimental tumors in the rat trigeminal nerve.

The activity of the ZnTP (Felicetti und Rath 1975; Rath and Felicetti 1975) was investigated histo- and biochemically in neurinomas of trigeminal nerves of rats. The tumors were induced by a single transplacental pulse of 30 mg ENU/kg on the 17th day of gestation. The rats were killed at the age of 40 to 160 days. The activity of the ZnTP is demonstrated histochemically by a coupling azo dye technique with 1-naphthylphosphate after Rath and Felicetti (1975) as well as a lead nitrate method with 4-nitrophenyl phosphate at the age of 40 to 140 days. Biochemical investigations of the trigeminal nerves are performed at the age of 140 and 160 days only. The separation of ZnTP is carried out by isoelectrofocusing in polyacrylamide gels after Felicetti and Rath (1975). Together 14 neurinomas are found in the trigeminal nerves of 53 rats. All neurinomas show a high activity of the ZnTP. The ZnTP activity is absent in normal Schwann cells. Three trigeminal nerves of rats subjects to the transplacental pulse of ENU harbour small foci of ZnTP activity without detectable neoplastic cell proliferation. Similar spots of enhanced ZnTP activity have never been found in control rats. This rises suspicion that the neoplastic proliferation of cells is preceded by the activation of ZnTP.
