The use of gastric ultrasonography in the evaluation of a new antiflatulent preparation in human volunteers.

A method is described for inducing experimental aerogastria (flatulence) in human volunteers by the ingestion of "whipped" egg-white. Abdominal ultrasonography was used to measure the distension of the stomach and its interference with the visualization of organs such as gall bladder, pancreas and left kidney lying behind the stomach. A double-blind crossover comparison of Flatoril, a mixture of dimethicone (200 mg) and clebopride (0.5 mg) against placebo clearly demonstrated the superior antifoaming activity of the former both in terms of reduced gastric distension and of improved visualization of organs otherwise hidden by the foam-filled stomach.

Comparison of the antacid properties of almagate and aluminium hydroxide against pentagastrin-induced gastric secretion in healthy volunteers.

The potency of a new antacid, almagate (hydrated aluminium-magnesium hydroxycarbonate, Al2Mg6 (OH)14(CO3)2 X 4 H2O, Almax) has been compared with that of aluminium hydroxide in neutralizing pentagastrin-induced hyperacidity in healthy human volunteers. Almagate (1 g) produced a significantly (p less than 0.01) greater reduction (87.5%) in the titratable acid content of gastric aspirates than did an identical dose of aluminium hydroxide (45.1%). Similarly the pepsin proteolytic activity in the samples of gastric juice was reduced 58.9% by almagate and only 27.5% by aluminium hydroxide. Furthermore the neutralizing effect of almagate was more prolonged (90 min) than that of the standard antacid (30 min) and these first studies in man have therefore confirmed the findings of in vitro and in vivo preclinical experiments.

Gastric emptying of solid and liquid meals in healthy young subjects.

Gastric emptying was measured in 50 young healthy subjects by means of sequential scintigraphy with two radioactive markers. Gastric emptying of the solid component of the meal was slower than that of the liquid component, corresponding to a linear model (T 1/2, 112 min). Gastric emptying of the liquid component was different, following an exponential model, although only after 45 min (100% of the liquid marker retained in the stomach after 15 min, 56.7% after 45 min, 43.3% after 75 min, and 35.5% after 105 min). Significant differences were found in gastric emptying of both solids and liquids among sexes; women emptied the stomach more slowly than men regardless of age, weight, height, or body surface. A relationship was found between gastric emptying and the phase of the menstrual cycle. Furthermore, a quicker emptying of the liquid component was observed in smokers. These factors should be considered in the evaluation of gastric emptying studies.
